Postnatal Pdzrn3 deficiency causes acute muscle atrophy without alterations in endplate morphology.

PDZ domain-containing RING finger family protein 3 (PDZRN3) is expressed in various tissues, including the skeletal muscle. Although PDZRN3 plays a crucial role in the terminal differentiation of myoblasts and synaptic growth/maturation in myogenesis, the role of this molecule in postnatal muscles is completely unknown despite its lifelong expression in myofibers. In this study, we aimed to elucidate the function of PDZRN3 in mature myofibers using myofiber-specific conditional knockout mice. After tamoxifen injection, PDZRN3 deficiency was confirmed in both fast and slow myofibers of Myf6-Cre

Four-Dimensional Microvascular Analysis Reveals That Regenerative Angiogenesis in Ischemic Muscle Produces a Flawed Microcirculation.

RATIONALE: Angiogenesis occurs after ischemic injury to skeletal muscle, and enhancing this response has been a therapeutic goal. However, to appropriately deliver oxygen, a precisely organized and exquisitely responsive microcirculation must form. Whether these network attributes exist in a regenerated microcirculation is unknown, and methodologies for answering this have been lacking. OBJECTIVE: To develop 4-dimensional methodologies for elucidating microarchitecture and function of the reconstructed microcirculation in skeletal muscle. METHODS AND RESULTS: We established a model of comple

Pompe disease: from pathophysiology to therapy and back again.

Pompe disease is a lysosomal storage disorder in which acid alpha-glucosidase (GAA) is deficient or absent. Deficiency of this lysosomal enzyme results in progressive expansion of glycogen-filled lysosomes in multiple tissues, with cardiac and skeletal muscle being the most severely affected. The clinical spectrum ranges from fatal hypertrophic cardiomyopathy and skeletal muscle myopathy in infants to relatively attenuated forms, which manifest as a progressive myopathy without cardiac involvement. The currently available enzyme replacement therapy (ERT) proved to be successful in reversing ca

[Myoglobin content and oxygen tension in the muscle tissues in allergic reactions].

The influence of sensitization to horse serum, brucelia and BCG, and the allergic reactions induced by these antigens, on the oxygen tension and myoglobin content in the muscle tissues was studied. The lowering of myoglobin concentration associated with higher sensitivity formation was noted only in sensitization to living microbial strains. The allergic responses of immediate and delayed types provoked the decrease in myoglobin content both in the heart and skeletal muscles. The tension of oxygen in the skeletal muscle was reduced.

The effect of arsenic on mitochondrial fatty acid metabolism via inhibition of carnitine palmitoyltransferase 1B and choline kinase beta in C2C12 cells.

Arsenic can enter the human body through environmental exposure via food, drinking water, and chemotherapy for cancer. Prolonged and excessive exposure to arsenic causes various toxic reactions, leading to diseases that significantly impact health and lifespan. Increasing evidence suggests that arsenic damages skeletal muscle tissue by reducing muscle mass and causing atrophy, thereby contributing to conditions such as respiratory and cardiovascular diseases, as well as diabetes. Fatty acid β-oxidation is the most efficient mechanism for ATP production and serves as a primary energy source for

Screening of hub genes for sepsis-induced myopathy by weighted gene co-expression network analysis and protein-protein interaction network construction.

Sepsis-induced myopathy is one of the serious complications of sepsis, which severely affects the respiratory and peripheral motor systems of patients, reduces their quality of life, and jeopardizes their lives, as evidenced by muscle atrophy, loss of strength, and impaired regeneration after injury. The pathogenesis of sepsis-induced myopathy is complex, mainly including cytokine action, enhances free radical production in muscle, increases muscle protein hydrolysis, and decreases skeletal muscle protein synthesis, etc. The above mechanisms have been demonstrated in existing studies. However,

A link between mitochondrial damage and the immune microenvironment of delayed onset muscle soreness.

BACKGROUND: Delayed onset muscle soreness (DOMS) is a self-healing muscle pain disorder. Inflammatory pain is the main feature of DOMS. More and more researchers have realized that changes in mitochondrial morphology are related to pain. However, the role of mitochondria in the pathogenesis of DOMS and the abnormal immune microenvironment is still unknown. METHODS: Mitochondria-related genes and gene expression data were obtained from MitoCarta3.0 and NCBI GEO databases. The network of mitochondrial function and the immune microenvironment of DOMS was constructed by computer algorithm. Subseq

Water-soluble dietary fiber alleviates cancer-induced muscle wasting through changes in gut microenvironment in mice.

Cancer cachexia and the associated skeletal muscle wasting are considered poor prognostic factors, although effective treatment has not yet been established. Recent studies have indicated that the pathogenesis of skeletal muscle loss may involve dysbiosis of the gut microbiota and the accompanying chronic inflammation or altered metabolism. In this study, we evaluated the possible effects of modifying the gut microenvironment with partially hydrolyzed guar gum (PHGG), a soluble dietary fiber, on cancer-related muscle wasting and its mechanism using a colon-26 murine cachexia model. Compared wi

Folic acid is necessary for proliferation and differentiation of C2C12 myoblasts.

Folic acid, a water soluble B vitamin, plays an important role in cellular metabolic activities, such as functioning as a cofactor in one-carbon metabolism for DNA and RNA synthesis as well as nucleotide and amino acid biosynthesis in the body. A lack of dietary folic acid can lead to folic acid deficiency and result in several health problems, including macrocytic anemia, elevated plasma homocysteine, cardiovascular disease, birth defects, carcinogenesis, muscle weakness, and walking difficulty. However, the effect of folic acid deficiency on skeletal muscle development and its molecular mech

Mitochondrial myopathy with autophagic vacuoles in patients with the m.8344A>G mutation.

BACKGROUND AND AIMS: In mitochondrial myopathy, autophagy is presumed to play an important role in mitochondrial dysfunction. Rimmed vacuoles (RVs), a sign of autophagy, can be seen as a secondary phenomenon in muscle ragged-red fibres (RRFs), whereas the uncommon presentation is that some fibres contain RVs, but without any mitochondrial abnormalities. To investigate the pathogenesis beneath this pathological phenomenon. METHODS: We reviewed 783 skeletal muscle specimens and selected five obtained from patients with suspected mitochondrial myopathy, characterised by clearly visible autophagi

Impaired exercise capacity and exercise training in maintenance hemodialysis patients.

Patients with advanced chronic renal failure, including those undergoing maintenance hemodialysis (MHD), commonly suffer from sarcopenia and reduced physical exercise capacity. The causes of these disorders include anemia, cardiovascular and pulmonary diseases, other comorbid conditions, altered skeletal muscle metabolism, myopathic changes that are associated with chronic renal failure per se, malnutrition, and physical deconditioning. Exercise training has been shown to increase exercise capacity and, in some studies, to increase muscle mass. In our experience, cardiopulmonary fitness traini

Channelopathies.

In patients with mutations in the genes that encode the chloride, sodium, and calcium channels in skeletal muscle, there is abnormal function of the muscle membrane, which can cause myotonia or attacks of weakness. Mutations in the chloride and sodium channels can lead to myotonia, which typically begins in early childhood. Mexiletine is usually effective in controlling myotonia in these patients. Mexiletine is also effective in preventing attacks of cold-provoked muscle paralysis in patients with paramyotonia congenita, a sodium channel disorder. Certain mutations in the sodium channel cause

Ryanodine receptor gene expression thymomas.

Ryanodine receptor (RyR) antibodies are present in sera of myasthenics with thymoma, and their titer correlates with morbidity and mortality. We investigated whether skeletal muscle RyR expression in thymic tissues could be the source of immune sensitization to the RyR. Skeletal muscle RyR gene expression was investigated using reverse transcription followed by semiquantitative polymerase chain reaction. Hyperplastic and normal thymuses expressed significant levels of RyR, but RyR gene transcripts was statistically less likely in thymomas than in hyperplastic and normal thymus (P < 0.05). The

Myotoxic components of snake venoms: their biochemical and biological activities.

Necrosis of skeletal muscle is produced by two types of snake venom components: single chain peptides consisting of 42-44 amino acid residues and phospholipases A2 representing either single chain proteins or existing as complexes of several enzyme subunits or combined with other nonenzymatic proteins. Vacuolation, lysis and necrosis of skeletal muscle cells are the major pathological effects of these myotoxins. Although the exact mode of action of these toxins is not clear, interactions with the plasma membrane leading to permeability changes for ions and to their complete destruction is evid

Specific force of the rat extraocular muscles, levator and superior rectus, measured in situ.

Extraocular muscles are characterized by their faster rates of contraction and their higher resistance to fatigue relative to limb skeletal muscles. Another often reported characteristic of extraocular muscles is that they generate lower specific forces (sP(o), force per muscle cross-sectional area, kN/m(2)) than limb skeletal muscles. To investigate this perplexing issue, the isometric contractile properties of the levator palpebrae superioris (levator) and superior rectus muscles of the rat were examined in situ with nerve and blood supply intact. The extraocular muscles were attached to a f

NODULAR POLYMYOSITIS IN RHEUMATOID ARTHRITIS.

The regular findings of a nodular perineuritis in peripheral nerve trunks and of a nodular polymyositis in skeletal muscles indicates the systemic nature of this disease. Rheumatoid arthritis is not solely a disease of the joints but a systemic disease, characterized by a widespread distribution of a nodular lymphocytic inflammation in peripheral nerves and skeletal muscles. The new findings in the nerves and muscles offer an explanation of the generally encountered clinical manifestations of pain, tenderness, trophic changes and muscular atrophy.

Bi-allelic mutations in MYL1 cause a severe congenital myopathy.

Congenital myopathies are typically characterised by early onset hypotonia, weakness and hallmark features on biopsy. Despite the rapid pace of gene discovery, ∼50% of patients with a congenital myopathy remain without a genetic diagnosis following screening of known disease genes. We performed exome sequencing on two consanguineous probands diagnosed with a congenital myopathy and muscle biopsy showing selective atrophy/hypotrophy or absence of type II myofibres. We identified variants in the gene (MYL1) encoding the skeletal muscle fast-twitch specific myosin essential light chain (ELC) in b

Association between morphological and mechanical properties of lower limb muscles and metabolic syndrome in type 2 diabetes.

Metabolic syndrome (MS), a group of metabolic disorders including obesity, elevated blood pressure, abnormal blood lipid and blood glucose, is closely related to insulin resistance. Skeletal muscle is one of the important target organs of insulin, and is closely related to glucose uptake. In diabetic patients, weakened insulin signaling and elevated insulin resistance will accelerate muscle catabolism, while a high glucose state can also increase insulin resistance, thus leading to skeletal muscle atrophy and resulting in reduced skeletal muscle content or hardness. The study aims to explore t
