Buried disc drusen have hypo-reflective appearance on SD-OCT.

PURPOSE: Buried disc drusen are an important differential diagnosis for papilledema. Spectral domain optical coherence tomography (SD-OCT) affords clinicians with new non-invasive opportunities to probe below the surface of the optic nerve. Clinicians may use the knowledge of this appearance to rule out buried disc drusen in patients with irregular optic nerve borders or a bulging, hyperemic appearance. METHODS: SD-OCTs were obtained in a patient with one surfacing disc druse, identifying the nature of the appearance of this disc druse and others in this and the contralateral eye when imaged

An aggregation-induced emission-active theranostic agent for selectively detecting and intervening pathological Tau protein.

The accumulation of Tau aggregates is commonly linked with various neurodegenerative diseases, such as Alzheimer's disease, Pick's disease, and corticobasal degeneration. Notwithstanding substantial investments in the development of clinical strategies for effective intervention, traditional design paradigms are predominantly confined to molecules featuring either a solitary function or single-dimensional mode of intervention, ignoring the necessity of personalized and precise medicine. Herein, we design and synthesize a dual-functional aggregation-induced emission-active agent to serve as bot

Chlorophyll fluorescence imaging for early visual detection and measurement of postharvest discoloration: Potato greening and broccoli yellowing.

Chlorophyll serves as both physiological basis and fundamental pigment underlying healthy coloration of numerous fruits and vegetables. Yet conventional visual inspection, point-level color measurement, and destructive chlorophyll assays lack sensitivity for early, non-destructive detection of discoloration driven by abnormal chlorophyll metabolism in postharvest produce. This study developed a simple chlorophyll fluorescence imaging system to enable early detection and measurement of two representative postharvest discoloration phenomena: light-induced greening in potatoes and senescence-driv

Time-Resolved Native Fluorescence Imaging of the Migration and Band Broadening of Monoclonal Antibody During Capillary Gel Electrophoresis With Sodium Dodecyl Sulfate.

We developed an imaging-based approach to directly visualize and monitor molecular migration during capillary electrophoresis and applied it to the separation of monoclonal antibodies (mAbs) by capillary gel electrophoresis with sodium dodecyl sulfate (CGE-SDS). Proteins were detected through their intrinsic fluorescence at 350 nm following excitation at 280 nm, enabling real-time observation of migration behavior without additional labeling. Time-resolved imaging allowed direct visualization of both migration and band broadening of mAbs under reducing and non-reducing conditions throughout th

A green lifetime biosensor for calcium that remains bright over its full dynamic range.

Fluorescent biosensors toggle between two states, and for the vast majority of biosensors, one state is bright and the other state is dim. As a consequence, there is a substantial difference in the signal-to-noise ratio (SNR) for the two states. The dim state has a low SNR, which is problematic when precise, quantitative measurements are needed. During the engineering of a red-shifted variant of an mTurquoise-based calcium sensor, we serendipitously generated a green-emitting sensor that shows high brightness in both the calcium-bound and -unbound state, while still showing a calcium-dependent

Alkyl Chain Engineering for High-Brightness Near-Infrared-II Aggregation-Induced Emission Nanoprobes toward High-Resolution In Vivo Bioimaging.

Fluorescence imaging in the second near-infrared window (NIR-II, 1000-1700 nm) has emerged as a powerful tool for in vivo bioimaging, offering deep-tissue penetration alongside high spatiotemporal resolution and contrast. Owing to structural advantages, aggregation-induced emission (AIE) luminogens are recognized as an ideal platform for constructing advanced NIR-II organic probes. However, their brightness remains suboptimal for practical applications due to intrinsic molecular constraints and environmental quenching effects. Addressing these issues, a high-brightness NIR-II AIE nanoprobe, TT

Imaging of porphyrin-specific fluorescence in pathogenic bacteria in vitro using a wearable, hands-free system.

Fluorescence imaging is an effective method for detecting porphyrin production in bacteria, leveraging the natural fluorescence properties of porphyrins. Here we use a simple, lightweight, hands-free device for rapid, non-invasive assessments in clinical settings, microbial research, and diagnostic applications. Specifically in this study, we examined 15 bacterial and 2 fungal strains commonly associated with skin, oral, and/or multi-site infections at wound sites for their ability to autofluoresce based on their porphyrin production. We utilized Remel Porphyrin Test Agar and blood agar plates

Image-guided in vivo evaluation and comparison of bone-targeting peptides for therapeutic intervention.

Hydroxyapatite (HA)-binding peptides hold strong potential for bone-targeted therapies due to their high affinity for mineralized tissues. However, most existing studies have primarily focused on in vitro binding characterization, offering limited insight into their in vivo biodistribution and bone-binding performance. In this study, we evaluated the in vivo behavior of four HA-binding peptides, including D8, E8, YD8, and YE8, using fluorescence imaging to assess biodistribution in both healthy and pathological bone environments. In healthy animals, D8 showed the strongest bone-binding capacit

Pathologic myopia.

Pathologic myopia (PM) represents a predominant cause of irreversible visual loss worldwide, characterised by progressive axial elongation and profound structural alterations in the posterior segment, notably posterior staphyloma. Posterior staphyloma is a cardinal feature of PM and must be differentiated from physiological myopia, as it indicates localised scleral thinning and ectasia directly impacting the macula and optic nerve. Vision-threatening complications such as myopic traction maculopathy (MTM), frequently driven by vitreoretinal traction on retinal vasculature, and macular atrophy

Choroidal Layer Analysis in OCT images via Ambiguous Boundary-aware Attention.

Optical Coherence Tomography (OCT) is a commonly used retina imaging technique, and it is capable of revealing the morphology of the choroid. However, the segmentation and quantitative analysis of the sublayers and vessels in choroid are rarely explored, primarily due to the indistinct boundaries of choroidal sublayers, and imbalanced distribution of vessels observed in OCT imagery. In this paper, we propose a novel two-stage architecture called Choroidal Layer Analysis network (CLA), that may be considered the first attempt in this research community for joint segmentation of choroidal sublay

Design of donor-acceptor conjugated polymers based on diketopyrrolopyrrole for NIR-II multifunctional imaging.

Diketopyrrolopyrrole (DPP) is an excellent photosensitizer and photothermal agent with the advantages of good planarity, strong electron affinity, high electron mobility, easy purification, easy structural modification and high molar absorption coefficient. It is regarded as one of the ideal choices for the design and synthesis of efficient organic photovoltaic materials. Therefore, two kinds of donor-acceptor (D-A) conjugated polymers were designed and synthesized with DPP as the acceptor, and their optical properties and applications in the near-infrared region were studied. The quantum yiel

Reversible Fluorescent Probes for Dynamic Imaging of Liver Ischemia-Reperfusion Injury.

Hepatic ischemia-reperfusion injury (HIRI) is an inevitable complication of clinical surgeries such as liver resection or transplantation, often resulting in postoperative liver dysfunction, hepatic failure in up to 13% of postresection patients, and early graft failure in 11-18% of liver transplantation patients. HIRI involves a series of biochemical events triggered by abnormal alterations in multiple biomarkers, characterized by short lifespans, dynamic changes, subcellular regional distribution, and multicollaborative regulation. However, traditional diagnosis, including serology, imaging,

[Intraoperative Evaluation of Ureteral Blood Flow Using Indocyanine Green Fluorescence imaging on Living-Donor Kidney Transplantation].

Ureteral stenosis occurs in 2-10% after kidney transplantation. Most are caused by ischemia of the distal ureter and are difficult to manage. There is no standard method for evaluating ureteral blood flow during surgery, and it is left to the judgement of the operator. Indocyanine green (ICG) is used not only for a liver or cardiac function test but also for an assessment of tissue perfusion. We evaluated the intraoperative ureteral blood flow under a surgical light and by ICG fluorescence imaging in 10 living-donor kidney transplant patients between April 2021 and March 2022. No ureteral isch

Near-infrared fluorescent imaging for parathyroid identification and/or preservation in surgery for primary hyperparathyroidism.

INTRODUCTION: Near infrared autofluorescence (NIRAF) is a novel intraoperative technology that has shown promising results in the localisation of parathyroid glands (PGs) over the last decade. This study aimed to assess the potential utility of NIRAF in first time surgery for primary hyperparathyroidism (PHPT). METHODS: An observational study over a period of 3 years in patients who underwent surgery for PHPT was designed. Data on the use of NIRAF and fluorescent patterns in different organs (thyroid and parathyroid) and parathyroid pathology (single versus multi-gland disease) were explored.

A near infrared two-channel fluorescent probe for the detection of hydrogen sulfide and viscosity with a negligible crosstalk influence.

Both imbalance of H2S production and the change of viscosity in cells are associated with many diseases such as inflammation, Alzheimer's disease, and Parkinson's disease. Thus, the development of two-channel fluorescent probes for the detection of H2S and viscosity is of great significance for the study of pathogenic mechanisms. Herein, we design a two-channel NIR fluorescent probe RHO-DCO-DNP, which was able to selectively respond to H2S in one channel (λex = 580 nm, λem = 760 nm) and to viscosity in another channel (λex = 400 nm, λem = 585 nm). It should be emphasized that there is a neglig

Parathyroid gland detection using an intraoperative autofluorescence handheld imager - early feasibility study.

INTRODUCTION: Parathyroid glands may be compromised during thyroid surgery which can lead to hypoparathyroidism and hypocalcemia. Identifying the parathyroid glands relies on the surgeon's experience and the only way to confirm their presence was through tissue biopsy. Near infrared autofluorescence technology offers an opportunity for real-time, non-invasive identification of the parathyroid glands. METHODS: We used a new research prototype (hANDY-I) developed by Optosurgical, LLC. It offers coaxial excitation light and a dual-Red Green Blue/Near Infrared sensor that guides anatomical landma

Heterogeneous parathyroid near-infrared autofluorescence patterns are associated with single adenomas in primary hyperparathyroidism.

BACKGROUND: Primary adenoma (PA) and multi-gland hyperplasia (MGH) account for 85% and 15% of primary hyperparathyroidism (PHPT) cases, respectively. Near-infrared autofluorescence (NIRAF) enhances intraoperative parathyroid identification. We hypothesized that PA would display a more heterogeneous NIRAF pattern compared to MGH. METHODS: Patients undergoing surgery for sporadic PHPT were categorized based on the presence of PA or MGH. To quantify heterogeneity, we utilized ratios of (1) mean parathyroid gland (PG) NIRAF over background NIRAF (mean ratio), (2) minimum and (3) maximum PG NIRAF
