Genome-wide association study identifies protective genetic factors in active blood donors against multiple diseases.

The healthy donor effect (HDE) refers to the lower mortality observed among blood donors compared to the general population. While HDE arises due to healthier individuals being more likely to donate, the extent to which it is influenced by genetic differences remains unclear. To elucidate the genetic basis of HDE, we conducted a genome-wide association study (GWAS) involving 53,688 active blood donors with extensive donation histories and 228,060 controls from biobank cohorts within the FinnGen project. We identified 46 fine-mapped genome-wide significant loci associated with several health-re

Identification of ZmPH1 as a plant height novel gene in maize.

Plant height is a key agricultural trait for lodging resistance in maize. To explore key genes regulating plant height, a genome-wide association study (GWAS) was conducted using a large population of 1149 inbred lines. Plant height (PH), ear height (EH), and ear height coefficient (EH/PH) in this population followed a standard normal distribution, with heritability of 86%, 85%, and 75%, respectively. The GWAS identified two significant SNPs (Chr3:163963128 and Chr3:163963155) associated with all three traits. Within a 100-kb genomic region spanning upstream and downstream of the two SNPs, fou

Genomic Predictors of Diapause Polymorphism in the Bark Beetle Ips typographus With Implications for Voltinism Potential and Outbreak Dynamics.

Diapause polymorphisms shape insect life cycles, affecting their responses to seasonal environmental fluctuations. In the spruce bark beetle Ips typographus, diapause phenotypes, classified as facultative or obligate, significantly impact voltinism and thus outbreak potential, yet the genetic mechanisms underlying this trait remain incompletely understood. Here, we combined reduced-representation genome-wide association study (rGWAS) and machine learning (ML) to identify genetic polymorphisms associated with diapause phenotypes. Using ddRADseq, we analysed genomic data from diapause-phenotyped

Dissecting genetic architecture of growth and yield traits in horsegram using GWAS.

Horsegram (Macrotyloma uniflorum), a member of the Fabaceae family, is a nutritious and low-cost legume used for both grain and fodder. This study employed a genome-wide association approach to identify loci linked to key agronomic traits in horsegram. Plant height, seed size, and shoot fresh weight were evaluated in a panel of 96 diverse genotypes. GBS was performed using the Illumina HiSeq platform, yielding 20,241 high-quality SNPs after filtering at a 5% minor allele frequency. Population structure analysis classified genotypes into three admixed subgroups. Phenotyping was conducted over t

Multi-Taxa Genome-Environment Associations With Contemporary and Historical Climate in the Brazilian Atlantic Forest.

Multi-taxa genomic data offer valuable opportunities to assess the extent to which co-distributed taxa exhibit parallel responses to shared environmental regimes and climatic history. In this study, we use genome-environment associations (GEA) to examine how present and past climate may have shaped spatial patterns of adaptive genomic variation across 27 taxa in the Atlantic Forest biodiversity hotspot. Our sampling repurposes existing data from a wide array of taxonomic groups, including frogs, lizards, birds, butterflies, flowering plants, and a conifer, spanning multiple bioclimatic domains

Suggestive associations between genetically predicted gut microbiota and endometriosis: a two-sample Mendelian randomization study.

Introduction. Endometriosis affects 10-20% of reproductive-age women. Emerging evidence links the gut microbiota to endometriosis pathogenesis, but observational studies are limited by confounding, reverse causation and uncertainty about whether reported microbial signals represent reproducible aetiological associations.Hypothesis/Gap Statement. Whether genetically predicted gut microbial genera are associated with endometriosis risk remains unclear, and available observational evidence does not establish robust causal effects after accounting for multiple testing.Aim. This study aimed to expl

Locus- and Gene-Level Insights into the Inverse Association Between Alzheimer's Disease and Cancer.

Alzheimer's disease (AD) and cancer are both age-related conditions, yet numerous large-scale epidemiological studies have consistently documented an inverse association, with individuals diagnosed with cancer exhibiting a reduced risk of AD and vice versa. Although this relationship has been replicated across diverse populations, its biological basis remains poorly understood. To address this gap, the present study applies a framework that integrates locus-level genetic correlation (rg) with genetically regulated gene expression to clarify the molecular factors contributing to the inverse epi

Exploring the potential of naked barley to manage deoxynivalenol accumulation from Fusarium head blight.

Developing and deploying multi-use naked barley varieties that accumulate lower levels of deoxynivalenol (DON) resulting from Fusarium head blight (FHB) disease could help farmers mitigate economic risks and expand market possibilities. Previous work established that a substantial amount of DON in covered barley accumulates in the hull and can be removed by pearling. We studied a diverse panel of 244 naked barley lines to determine if there is a genetic variation for the distribution of DON in Fusarium-infected spikes. We evaluated the panel genotyped with a 50K Barley SNP array for disease se

Metabolic-renal disease genetic architecture revealed by genomic structural equation modeling.

Physiologic mechanisms underlying metabolic and renal diseases interact and are highly comorbid, yet their genetic associations and underlying mechanisms have not been systematically elucidated. Using genome-wide association study (GWAS) summary statistics from UK Biobank, FinnGen, and CKDGen, we integrated and analyzed gout, serum urate, chronic kidney disease, kidney stones, and metabolic syndrome. Using genomic structural equation modeling (GSEM), we created the first genetic linkage map for 'gout-urate-kidney-metabolism' (GUKM) to explore shared genetic architecture. Post-GWAS analyses for

PON1 haplotypes show genotype-dependent associations with dysglycemia and metabolic liver risk beyond paraoxonase activity.

INTRODUCTION: Paraoxonase 1 (PON1) is a liver-derived HDL-associated enzyme with key antioxidant functions implicated in cardiometabolic disease. Genetic variation in PON1 strongly influences enzyme activity; however, whether specific genetic configurations contribute to dysglycemia and metabolic liver risk beyond enzymatic activity remains unclear. METHODS: We analyzed 922 individuals from the PREVADIAB2 cohort to investigate the relationship between PON1 genetic variation, serum paraoxonase (PONase) activity, and dysmetabolic phenotypes. Genetic determinants of PONase activity were identifi

PLCH2 gene modifies the U-shaped association between environmental selenium exposure and blood total cholesterol: A gene-environment interaction study.

Exposure to excessive selenium (Se) is hazardous to human health, while Se is also an essential trace element. Therefore, exploring the appropriate exposure dose of Se has received widespread attention. Currently, the impact of Se exposure on blood lipids remains uncertain, and the potential mechanism is still unclear. Evidence from our earlier study pointed to PLCH2 as a potentially pivotal element in the association between Se exposure and total cholesterol (TC). This study aims to explore the appropriate exposure dose of Se on blood lipids, and effects of PLCH2 gene on the relationship betw

The stability of fatty acid composition in sunflower oil is dependent on environment and affected by structural variation.

In sunflower (Helianthus annuus L.), the composition of fatty acids in the seeds, primarily oleic, linoleic, stearic and palmitic acid, is of utmost importance for oil quality. Despite this, the genetic basis of this trait and its interaction with the environment is poorly understood. Understanding this interaction is critical to improvement of sunflower within the context of climate change. In this work, we incorporated fatty acid composition measurements from the sunflower SAM population and eight environments across an extensive geographic cline into GWAS. The SAM panel consists of 287 vari

Meta-GWAS of Pig Semen Quality Traits Reveals Conserved Genes Regulating Mammalian Fertility.

Semen quality serves as a vital indicator of male fertility, yet its underlying genetic and regulatory mechanisms remain poorly understood. Here, 1.15 million records of six semen quality traits from 14 210 boars in four distinct breeds were collected. These traits have low to moderate heritability (0.12-0.26), and are genetically correlated with growth traits like average daily gain. Genome-wide association study (GWAS) and multi-breed meta-analysis detected 234 loci associated with semen quality. Systematic integration of the Pig Genotype-Tissue Expression resource with these GWAS loci allow
