Neurocognitive contributions to motor skill learning: the role of working memory.

Researchers have begun to delineate the precise nature and neural correlates of the cognitive processes that contribute to motor skill learning. The authors review recent work from their laboratory designed to further understand the neurocognitive mechanisms of skill acquisition. The authors have demonstrated an important role for spatial working memory in 2 different types of motor skill learning, sensorimotor adaptation and motor sequence learning. They have shown that individual differences in spatial working memory capacity predict the rate of motor learning for sensorimotor adaptation and

Connectional heterogeneity of the ventral part of the macaque area 46.

We found that the ventral part of the prefrontal area 46 (46v) is connectionally heterogeneous. Specifically, the rostral part (46vr) displayed an almost exclusive and extensive intraprefrontal connectivity and extraprefrontal connections limited to area 24 and inferotemporal areas. In contrast, the caudal part (46vc) mostly displayed intraprefrontal connectivity with ventrolateral areas and robust connectivity with frontal and parietal sensorimotor areas. Based on a topographic organization of these connections, 3 fields were identified in area 46vc. A caudal field (caudal 46vc) was preferent

Neurofunctional correlates of expressed vocal affect in social phobia.

We investigated the neural correlates of expressed vocal affect in patients with social phobia. A group of 36 patients performed an anxiogenic public-speaking task while regional cerebral blood flow (rCBF) was assessed using oxygen-15 positron emission tomography. The patients' speech was recorded and content masked using low-pass filtering (which obscures linguistic content but preserves nonverbal affective cues). The content-masked speech samples were then evaluated with regard to their level of vocally expressed nervousness. We hypothesized that activity in prefrontal and subcortical brain

User-friendly software for the analysis of brain lesions (ABLe).

We previously developed a software package called ABLe (analysis of brain lesions) which characterizes brain lesions in terms of lesion volume and intersection with cytoarchitecture (e.g. Brodmann areas). Since our previous publication, there have been significant improvements to this software package which utilize methods standard to the neuroimaging community. These features include spatial normalization to the MNI template brain (standard of the international consortium for brain mapping), and use of the volume occupancy Talairach labels (VOTL) and automated anatomical labeling (AAL) atlase

Sleepiness enhances distraction during a monotonous task.

STUDY OBJECTIVES: Although sleepiness appears to heighten distraction from the task at hand, especially if the latter is dull and monotonous, this aspect of sleep loss has not been assessed in any systematic way. Distractions are a potential cause of performance lapses (as are micro-sleeps). Here, we investigate the effects of sleepiness on a monotonous task, with and without distraction. DESIGN: Repeated Measures 2 x 2 counterbalanced design, comprising Sleepiness (night sleep restricted to 5 hours x normal sleep) and Distraction (distraction x no distraction). SETTING: Participants underwe

Anterior asymetrical alpha activity predicts Iowa gambling performance: distinctly but reversed.

Animal research indicates that the prefrontal cortex (PFC) plays a crucial role in decision making. In concordance, deficits in decision making have been observed in human patients with damage to the PFC. Contemporary accounts of decision making suggest that emotion guides the process of decision making by ways of providing for reward-punishment contingencies. A task capable of assessing the influence of reward and punishment on decision making is the Iowa gambling task. In this task decisions become motivated by inherent punishment and reward schedules. Insensitivity for punishment together w

Object vision and visual awareness.

Conscious experience involves perceiving, attending, remembering, and recognizing. Recent neuroscientific research has made significant contributions to our understanding of the mechanisms that mediate such capacities. Physiological and neuropsychological investigations have provided increasingly detailed descriptions of the location and functional properties of the brain structures involved in conscious perception, in attentive behavior and working memory, and in the recognition of objects. Such studies suggest that awareness of a visual stimulus probably reflects the interconnectivity and th

Restricted lesions to ventral prefrontal subareas block reversal learning but not visual discrimination learning in rats.

Previous studies have shown that extensive damage to the medial prefrontal cortex (mPFC) of rats causes reversal learning deficits. The mPFC of rats, however, consists of several subareas that are different from each other in both cytoarchitecture and neural connectivity, suggesting a functional dissociation among the mPFC subareas. In the present study, selective lesions of the mPFC of rats were made with a specially designed microknife whose intracranial placement could be controlled stereotaxically. Restricted lesions were made to each of the 3 parts of the mPFC: the anterior cingulate area

Efficacy of dual-target iTBS on gait function and brain activation in stroke patients: a randomized, single-blinded, sham-controlled study.

OBJECTIVE: This study aimed to assess the efficacy of intermittent theta burst stimulation (iTBS) applied to the lower limb motor cortex and the contralateral cerebellar hemisphere, both individually and in combination, on walking function in stroke patients. Secondarily, the study analyzed the effects of iTBS on brain functional connectivity across the three groups. METHODS: A total of 63 participants were randomly assigned to one of three groups, with 21 participants in each group: a sham stimulation group, a single-target iTBS group that stimulated the affected lower limb motor cortex, and

Effects of Psilocybin on Mouse Brain Microstructure.

BACKGROUND AND PURPOSE: There is surging interest in the therapeutic potential of psychedelic compounds like psilocybin in the treatment of psychiatric illnesses like major depressive disorder (MDD). Recent studies point to the rapid antidepressant effect of psilocybin; however, the biologic mechanisms underlying these differences remain unknown. This study determines the feasibility of using diffusion MRI to characterize and define the potential spatiotemporal microstructural differences in the brain following psilocybin treatment in C57BL/6J male mice. Network Communications Flexibly Predict Visual Contents That Enhance Representations for Faster Visual Categorization.

Models of visual cognition generally assume that brain networks predict the contents of a stimulus to facilitate its subsequent categorization. However, understanding prediction and categorization at a network level has remained challenging, partly because we need to reverse engineer their information processing mechanisms from the dynamic neural signals. Here, we used connectivity measures that can isolate the communications of a specific content to reconstruct these network mechanisms in each individual participant (N = 11, both sexes). Each was cued to the spatial location (left vs right) a

Abnormal Regional Homogeneity in Patients With Obsessive-Compulsive Disorder and Their Unaffected Siblings: A Resting-State fMRI Study.

Objective: Previous studies suggest that abnormal brain structure and function may be neuroimaging endophenotypes of obsessive-compulsive disorder (OCD). Comparing the intrinsic brain activity of OCD patients and their unaffected siblings will help to further understand the susceptibility to, and pathological mechanisms of, OCD. We used a case-control study design aiming to establish whether the abnormal regional homogeneity (ReHo) found in OCD patients also exists in their unaffected siblings. Method: Fifteen unmedicated OCD patients, 15 of their unaffected siblings, and 30 healthy controls (

Behaviorally Relevant Abstract Object Identity Representation in the Human Parietal Cortex.

UNLABELLED: The representation of object identity is fundamental to human vision. Using fMRI and multivoxel pattern analysis, here we report the representation of highly abstract object identity information in human parietal cortex. Specifically, in superior intraparietal sulcus (IPS), a region previously shown to track visual short-term memory capacity, we found object identity representations for famous faces varying freely in viewpoint, hairstyle, facial expression, and age; and for well known cars embedded in different scenes, and shown from different viewpoints and sizes. Critically, thes

Effective Connectivity Reveals an Interconnected Inferotemporal Network for Three-Dimensional Structure Processing.

Disparity-defined 3D shape is processed in both the ventral and the dorsal visual stream. The network of cortical areas that is activated during the processing of disparity-defined 3D shape includes, in addition to parietal and premotor areas, three clearly distinct regions in inferotemporal cortex (ITC). To investigate the connectivity of the latter regions, we combined electrical stimulation with fMRI in male macaque monkeys. Electrical stimulation of each of the 3D-structure nodes in ITC mainly elicited increased fMRI activations in the other 3D-structure nodes and more variably in other pa

Multiple movement representations in the human brain: an event-related fMRI study.

Neurovascular correlates of response preparation have been investigated in human neuroimaging studies. However, conventional neuroimaging cannot distinguish, within the same trial, between areas involved in response selection and/or response execution and areas specifically involved in response preparation. The specific contribution of parietal and frontal areas to motor preparation has been explored in electrophysiological studies in monkey. However, the associative nature of sensorimotor tasks calls for the additional contributions of other cortical regions. In this article, we have investig

Sumoylation profiles and mitochondrial dynamics in in vivo and in vitro models of Parkinson's disease.

Parkinson's disease (PD) is characterized by progressive dopaminergic neuron loss and motor and non-motor impairments. Mitochondrial dysfunction and altered post-translational modifications, including SUMOylation, have emerged as key mechanisms in PD pathogenesis. Here, we investigated global SUMOylation changes in in vivo and in vitro PD models induced by MPTP and MPP+, respectively. Intranasal MPTP administration in rats induced region- and time-dependent changes in Ubc9 and SENP3 across the olfactory bulb, prefrontal cortex, striatum, and hippocampus. In primary cortical neurons, MPP+ reduc

SpaDiff denoises sequence-based spatial transcriptomics via diffusion process.

Spot-swapping in sequence-based spatial transcriptomics redistributes RNA molecules across neighboring spots, distorting spatial gene expression and biasing downstream analysis. We introduce SpaDiff, a mass-conserving denoising framework that corrects this artifact through score-guided reverse diffusion. Across simulated and real spatial transcriptomics datasets, SpaDiff consistently improves the recovery of spatial gene-expression distributions. In particle-level simulations with known ground truth, SpaDiff robustly restores gene-level spatial patterns across diverse contamination regimes. In
