SARS-CoV-2 growth, furin-cleavage-site adaptation and neutralization using serum from acutely infected hospitalized COVID-19 patients.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of coronavirus disease 2019 (COVID-19), emerged at the end of 2019 and by mid-June 2020 the virus had spread to at least 215 countries, caused more than 8 000 000 confirmed infections and over 450 000 deaths, and overwhelmed healthcare systems worldwide. Like severe acute respiratory syndrome coronavirus (SARS-CoV), which emerged in 2002 and caused a similar disease, SARS-CoV-2 is a betacoronavirus. Both viruses use human angiotensin-converting enzyme 2 (hACE2) as a receptor to enter cells. However, the SARS-CoV-

Current status and future development of infectious cell-culture models for the major genotypes of hepatitis C virus: Essential tools in testing of antivirals and emerging vaccine strategies.

In this review, we summarize the relevant scientific advances that led to the development of infectious cell culture systems for hepatitis C virus (HCV) with the corresponding challenges and successes. We also provide an overview of how these systems have contributed to the study of antiviral compounds and their relevance for the development of a much-needed vaccine against this major human pathogen. An efficient infectious system to study HCV in vitro, using human hepatoma derived cells, has only been available since 2005, and was limited to a single isolate, named JFH1, until 2012. Successiv

Zinc: A Potential Antiviral Against Hepatitis E Virus Infection?

Hepatitis E virus (HEV) is a major cause of viral hepatitis worldwide. Owing to its feco oral transmission route, sporadic as well as epidemic outbreaks recurrently occur. No specific antiviral therapy is available against the disease caused by HEV. Broad spectrum antivirals such as ribavirin and interferon alfa are prescribed in severe and chronic HEV cases. However, the side effects, cost, and limitations of usage render the available treatment unsuitable for several categories of patients. We recently reported the ability of zinc to inhibit viral replication in mammalian cell culture models

Antiviral Immunity and Virus-Mediated Antagonism in Disease Vector Mosquitoes.

More than 100 pathogens, spanning multiple virus families, broadly termed 'arthropod-borne viruses (arboviruses)' have been associated with human and/or animal diseases. These viruses persist in nature through transmission cycles that involve alternating replication in susceptible vertebrate and invertebrate hosts. Collectively, these viruses are among the greatest burdens to global health, due to their widespread prevalence, and the severe morbidity and mortality they cause in human and animal hosts. Specific examples of mosquito-borne pathogens include Zika virus (ZIKV), West Nile virus (WNV

A Functional Role of Fibroblast Growth Factor Receptor 1 (FGFR1) in the Suppression of Influenza A Virus Replication.

Influenza A virus causes annual epidemics and occasional pandemics in humans. Here, we investigated four members of the fibroblast growth factor receptor (FGFR) family; FGFR1 to 4, and examined their expression patterns in human lung epithelial cells A549 with influenza A virus infection. We identified a functional role of FGFR1 in influenza A/Puerto Rico/8/1934 (PR8) and A/Anhui/01/2005 (H5N1) virus replication. Our results showed that FGFR1 silencing by siRNA interference promoted influenza A/PR8 and H5N1 virus replication in A549 cells, while lentivirus-mediated exogenous FGFR1 expression s

Cytoplasmic translocation, aggregation, and cleavage of TDP-43 by enteroviral proteases modulate viral pathogenesis.

We have previously demonstrated that infection by coxsackievirus B3 (CVB3), a positive-stranded RNA enterovirus, results in the accumulation of insoluble ubiquitin-protein aggregates, which resembles the common feature of neurodegenerative diseases. The importance of protein aggregation in viral pathogenesis has been recognized; however, the underlying regulatory mechanisms remain ill-defined. Transactive response DNA-binding protein-43 (TDP-43) is an RNA-binding protein that has an essential role in regulating RNA metabolism at multiple levels. Cleavage and cytoplasmic aggregation of TDP-43 s

Adenovirus small E1A employs the lysine acetylases p300/CBP and tumor suppressor Rb to repress select host genes and promote productive virus infection.

Oncogenic transformation by adenovirus small e1a depends on simultaneous interactions with the host lysine acetylases p300/CBP and the tumor suppressor RB. How these interactions influence cellular gene expression remains unclear. We find that e1a displaces RBs from E2F transcription factors and promotes p300 acetylation of RB1 K873/K874 to lock it into a repressing conformation that interacts with repressive chromatin-modifying enzymes. These repressing p300-e1a-RB1 complexes specifically interact with host genes that have unusually high p300 association within the gene body. The TGF-β,

Safe, long-term hepatic expression of anti-HCV shRNA in a nonhuman primate model.

The hepatitis C virus (HCV) chronically infects 2% of the world population and effective treatment is limited by long duration and significant side-effects. Here, we describe a novel drug, intended as a "single-shot " therapy, which expresses three short hairpin RNAs (shRNAs) that simultaneously target multiple conserved regions of the HCV genome as confirmed in vitro by knockdown of an HCV replicon system. Using a recombinant adeno-associated virus (AAV) serotype 8 vector for delivery, comprehensive transduction of hepatocytes was achieved in vivo in a nonhuman primate (NHP) model following a

Poliovirus pathogenesis in a new poliovirus receptor transgenic mouse model: age-dependent paralysis and a mucosal route of infection.

We constructed a poliovirus receptor (PVR) transgenic mouse line carrying a PVR delta cDNA driven by a beta-actin promoter. We refer to this model as the cPVR mouse. The cPVR mice express Pvr in a variety of tissues (including small intestines, brain, spinal cord, muscle, blood and liver) and are susceptible to infection after intraperitoneal, intracerebral or intramuscular inoculation of poliovirus. After intraperitoneal inoculation, poliovirus replication is observed in cPVR muscle, brain, spinal cord and, notably, small intestine. The cPVR mice exhibit a striking age-dependent paralysis aft

Zika NS2B Protein: In Vitro Formation of Large Multimeric Networks.

Flaviviruses are responsible for significant morbidity and mortality worldwide. Despite intensive research, the structure and oligomerization properties of non-structural (NS) proteins, like NS2 or NS4, are still uncertain because of their high hydrophobicity. Solution NMR has shown that NS2B protein has two hydrophobic domains, organized as two short α-helical hairpins that contribute to both viral RNA replication and particle formation. These are separated by a hydrophilic loop that is a cofactor of protease NS3. However, the oligomerization behavior of NS2B has not been explored in detail.

Multi-omics Mendelian randomization and experimental validation identify PRKAB1 as a regulator of phosphatidylcholine metabolism in IBD.

Phosphatidylcholines (PCs) are implicated in inflammatory bowel disease (IBD), yet the genetic and pathophysiological mechanisms underlying phosphatidylcholine metabolism in IBD remain poorly characterized. Here, we integrated multi-omics data with Mendelian randomization (MR) and genetic colocalization to dissect causal genetic associations between phosphatidylcholine metabolism-related genes and IBD. We conducted two-sample MR using discovery data from the FinnGen study and replication data from the IBD Genetics Consortium to evaluate causal links across 78 lipid species: 5 lysophosphatidylc

Biophysical analysis reveals low‑affinity, multivalent interactions and conformational modulation of the SARS‑CoV‑2 nucleocapsid protein by berberine and palmatine.

The Nucleocapsid (N) protein of the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) is an intrinsically dynamic, multifunctional protein critical for viral replication, RNA binding and the immune response. A consensus sequence of the N protein (P13L; Δ31-33; R203K; G204R; and S413R mutations in relation to the Wuhan sequence) was recombinantly expressed using E. coli BL21(DE3)-T7 express cells, purified and characterised using Far Ultraviolet (UV) Circular Dichroism (CD), Intrinsic Tryptophan Fluorescence (ITF) Spectroscopies as well as Blue Native Polyacrylamide Gel Electrophores

Epigenetic regulation of HIV-1 transcription: insights into latency mechanisms and therapeutic strategies.

HIV-1 is one of the most serious pathogens currently threatening human health worldwide. Although antiretroviral therapy (ART) effectively suppresses HIV-1 replication, it fails to eliminate the latent reservoir of transcriptionally silent proviruses that persist in long-lived cells, making HIV-1 latency the major obstacle to achieving a functional cure for AIDS. The establishment and reversal of HIV-1 latency are largely regulated by epigenetic regulatory mechanisms involving the viral long terminal repeat (LTR) promoter. Following proviral integration into the host genome, the viral DNA is r

Antiviral Mechanisms of Justicidin B and Tuberculatin from Phyllanthus brasiliensis against Oropouche Virus Infection.

Oropouche virus (OROV) has emerged as a growing public health concern in Latin America, with an unprecedented surge in Oropouche fever cases since 2023. Previously considered a sporadic and neglected arboviral infection, Oropouche fever now represents one of the most rapidly expanding arboviral threats in the region. Despite its increasing incidence, no approved vaccines or antiviral therapies are available, highlighting the urgent need for effective interventions. Natural compounds, particularly those derived from Phyllanthus brasiliensis, have shown promising antiviral properties against sev

Targeting HsDHODH: Shape and machine-learning guided discovery and structural validation of SARS-CoV-2 antivirals.

The frequent emergence of novel RNA viruses and the rapid development of viral resistance highlight the urgent need for new antiviral therapies. In this study, we employed an integrated computational pipeline combining shape-based models, machine-learning (ML)-based models, and hotspot analysis to identify novel human dihydroorotate dehydrogenase (HsDHODH) inhibitors. A virtual screening (VS) campaign of the H3D chemical library identified H3D-002856 as the primary hit (IC50 = 2.92 ± 0.07 μM), which subsequently guided a similarity-based hit expansion (Tanimoto coefficient≥0.7) of 17 structura

Lipopolysaccharide induces a delayed iron-handling transcript pattern in primary bovine macrophages.

OBJECTIVE: To characterize the timing and between-cow consistency of iron-handling transcript changes after lipopolysaccharide (LPS) exposure in primary bovine monocyte-derived macrophages. METHODS: Macrophages from 5 adult Holstein donor cows were exposed to Escherichia coli LPS or PBS and profiled by RNA sequencing at 30 minutes and 6 hours in a paired design. The LPS-versus-PBS effects were estimated with cow-blocked negative-binomial models. Per-cow direction was examined, and 4 focal transcripts were compared directionally with a separate published cohort of 8 cattle sampled 48 hours aft

Heterologous Expression of Cryphonectria Hypovirus 1 in Fusarium verticillioides Reveals Stable Replication and Reduced Fumonisin Production.

Mycoviruses represent promising tools for the biological control of phytopathogenic fungi. However, their application is often constrained by host specificity and low natural prevalence in certain pathogens. Fusarium verticillioides, a major maize pathogen and fumonisin B1 (FB1) producer, harbors very few known mycoviruses, a circumstance that limits the development of virus-based control strategies. In this study, we explored a virocontrol approach for F. verticillioides based on the use of a heterologous mycovirus, Cryphonectria hypovirus 1 (CHV1). For the artificial transfection of F. verti

Type I interferon signature strength correlates with alloimmunization-associated transcriptomic programs in systemic lupus erythematosus: A multi-cohort analysis.

Red blood cell (RBC) alloimmunization is a clinically significant complication in transfused patients whose immunological determinants remain incompletely understood. Type I interferon (IFN-I) signaling drives RBC alloimmunization in murine models, and systemic lupus erythematosus (SLE) is characterized by constitutive IFN-I hyperactivation alongside elevated alloimmunization rates. We analyzed three publicly available SLE RNA-seq cohorts (GSE72509, GSE112087, GSE122459; whole blood and PBMC; total n = 150 SLE) in a pre-specified discovery-replication-validation design.
