Comparative single-cell atlases reveal injury-driven tubal epithelial regeneration as a window for ovarian carcinoma initiation.

High-grade serous carcinoma (HGSC), the most lethal form of ovarian cancer, preferentially originates in the tubal epithelium (TE) of the distal uterine tube (also known as Fallopian tube or oviduct). Mouse models are widely used to study how HGSC initiates in humans; however, the extent to which mouse and human uterine tubes are comparable remains unclear. Here, we conduct cross-species single-cell transcriptomic comparative analyses and organoid assay validations to reveal conserved differentiation trajectories from bipotent progenitors to secretory or ciliated cell fates. Regional analyses

Small Extracellular Vesicles in Cardioprotection, Cardiac Repair, and Regeneration: Cargo Mechanisms, Producer Cell Sources, and Translational Development.

Cardiovascular diseases remain a major cause of death and disability, and the limited regenerative capacity of the adult myocardium continues to constrain recovery after myocardial infarction, ischemia/reperfusion injury, cardiomyopathy, and heart failure. Small extracellular vesicles (sEVs), often described as exosome-enriched vesicle preparations, have emerged as promising cell-free mediators of cardioprotection and cardiac repair. This review evaluates the current experimental and translational evidence on sEVs in cardiac injury, including regulatory RNA and non-RNA cargo mechanisms, produc

Cell pellets promote periodontal bone regeneration in furcation involvement by regulating macrophage polarization through TGF-β1 secretion.

BACKGROUND: Furcation involvement, a severe complication of chronic periodontitis, poses significant challenges for clinical regeneration because of failure to remodel the inflammatory microenvironment. Current strategies such as guided tissue regeneration are limited by two-dimensional architecture and insufficient immunomodulation, resulting in failure to restore functional periodontal tissues. Cell pellet, a novel scaffold-free technology, features self-assembled three-dimensional structures and endogenous extracellular matrix storing growth factors; however, its immunomodulatory mechanism,

Bioactive Hydrogels and Scaffolds for Oral Mucosal Regeneration After Oral Squamous Cell Carcinoma Therapy: A Comprehensive Review.

Oral squamous cell carcinoma (OSCC) therapy frequently produces acute and chronic injury to the oral mucosa, including surgical lining defects and radiochemotherapy-associated oral mucositis (OM). Beyond pain and ulceration, these injuries compromise nutrition, speech, oral hygiene, and feasibility of dental/implant rehabilitation, and may disrupt oncologic treatment delivery. The oral cavity imposes stringent constraints on regenerative biomaterials-continuous salivary flow, high microbial load, and repeated mechanical shear-such that clinical success depends on reliable mucoadhesion/wet adhe

Stem cells from human exfoliated deciduous teeth as an alternative cell source for cartilage regeneration and repair.

PURPOSE: Stem cells from human exfoliated deciduous teeth (SHED) hold promise for cartilage regeneration due to their chondrogenic potential and minimally invasive procurement. However, the in vivo cartilage regenerative capacity of SHED remains insufficiently explored. This study aims to compare the proliferative, migratory, and chondrogenic capacity of SHED and dental pulp stem cells (DPSCs) in vitro, and to investigate the therapeutic efficacy of SHED delivered via alginate hydrogel in a rat model of knee cartilage defect. METHODS: SHED was isolated from human exfoliated deciduous teeth, a

Insulin gene-modified 3D stem cells for enhanced islet regeneration and systemic reversal of type 1 diabetes.

Type 1 diabetes (T1D) is a chronic autoimmune disease characterized by the destruction of pancreatic β cells, leading to lifelong insulin dependence and an increased risk of severe complications. Three-dimensional stem cells (3D SCs) culture systems have emerged as a superior alternative by more accurately mimicking the in vivo microenvironment and enhancing stemness maintenance, regenerative efficiency, and paracrine secretion. However, studies exploring the application of 3D SCs in T1D remain limited. Here, we developed a novel serum- and cytokine-free orbital-shaking system. It enables effi

Pathology-Responsive Self-Assembling Hydrogel Enabling Spatiotemporally Controlled Exosome Release and Redox Regulation for Intervertebral Disc Regeneration.

Intervertebral disc degeneration (IVDD) arises from disrupted metabolism and redox imbalance, severely impairing nucleus pulposus (NP) cells' ability to repair the extracellular matrix (ECM). The early stage of IVDD is marked by excessive oxidative stress, lipid peroxidation, iron dysregulation and sustained catabolic enzyme activity, while late-stage cellular aging ultimately renders NP cells highly susceptible to iron-dependent cell death. To address these stage-specific challenges, we developed a pathology-adaptive self-assembling hydrogel that exploits dynamically varying catabolic enzyme

Orchestrating Diabetic Wound Healing by a Sunlight-Activated AIEgen-Selenide Nanospray: From Infection Control to Tissue Regeneration.

Diabetic wounds, particularly diabetic ulcers (DUs), are life-threatening complications driven by bacterial infection, persistent inflammation, oxidative stress, and a dysfunctional immune microenvironment. Addressing these interconnected barriers requires a therapeutic strategy capable of coordinating infection control with tissue repair. Here, we report a sunlight-activated nanospray formulation (C@PSe) based on a covalently engineered aggregation-induced emission luminogen (AIEgen)-selenide triblock copolymer micelle that encapsulates curcumin (Cur). This system integrates three complementa

Hybrid endometrial-derived hydrogel and human endometrial organoids synergize for uterine regeneration in an immunocompetent murine model.

BACKGROUND: The human endometrium is a regenerative tissue essential for fertility, but pathological conditions like Asherman syndrome, endometrial atrophy, and thin endometrium can impair its function. Current therapies lack efficacy, driving demand for innovative regenerative therapies. In this context, endometrial-derived hydrogels and organoids have shown promise individually for tissue regeneration, but their combined therapeutic potential has not been previously evaluated in vivo. This study explores a dual regenerative strategy combining a hybrid hydrogel - composed of synthetic PuraMat

Naringin promotes hair regeneration via wnt/β-catenin pathway: A dose-dependent study in C57BL/6J mice.

BACKGROUND: Alopecia, especially androgenetic alopecia (AGA), is a common condition affecting many individuals globally, impacting both men and women. Current treatments, such as minoxidil and finasteride, have limitations regarding efficacy and side effects. Traditional Chinese Medicine (TCM) offers alternative treatments, with naringin-a flavonoid found in Citrus species-emerging as a potential candidate due to its influence on the Wnt/β-catenin signaling pathway, a key regulator of hair follicle growth. MATERIALS AND METHODS: This study assessed the effects of naringin on hair regeneration

Ultrasound-activated piezoelectric Silk-PVDF hydrogel reprograms the osteoimmune microenvironment via NRF2 signaling for accelerated bone regeneration.

Bone defects pose significant clinical challenges due to the limited regenerative capacity of adult bone and the shortcomings of existing biomaterials, which lack dynamic electromechanical signaling crucial for repair. Here, we present an injectable, ultrasound-responsive piezoelectric hydrogel engineered to synergize silk fibroin's (SF) structural adaptability with polyvinylidene fluoride's (PVDF) bioelectrical activity. Methacrylated silk fibroin (SM) enables rapid UV-triggered crosslinking via a cost-effective photoinitiator system, while electrospun PVDF nanofibers, cryosectioned into micr

Effects of acute and chronic gamma irradiations on regeneration of tissue types from selected Ugandan cassava genotypes.

Acute and chronic gamma irradiations (ACGIs) has facilitated generation of new varieties that can withstand abiotic and biotic stresses. This study examined the impact of ACGI doses on the regenerative ability of in vitro embryogenic tissues into complete plants. Tissues from nine genotypes (Alado, NASE 13, NASE 19, NAROCASS 1, CV_193, CV_98/0505, CV_60444, BAO, and reference Ubi Putih) were subjected to acute gamma irradiation (AGI) across six dose levels (0, 5, 10, 15, 20, 25, 35 Gy). Five genotypes (Ubi Putih, CV_60444, Alado, NASE 13, NAROCASS 1) were exposed to chronic gamma irradiation (

3D-printed BaTiO3-modified PCL/β-TCP scaffolds with multimodal optimization for enhanced critical-size bone regeneration.

Critical-size bone defects (CSDs) remain a major clinical challenge. Although three-dimensional (3D) printing enables precise structural control and defect-specific scaffold fabrication, conventional polymer-based scaffolds often exhibit insufficient mechanical strength and limited osteogenic activity. In this study, poly(ε-caprolactone)/β-tricalcium phosphate (PCL/β-TCP) scaffolds containing 0, 1, 2, 4, or 8 wt% barium titanate (BaTiO3, BT) were fabricated by direct ink writing. Their rheology, microstructure, mechanical properties, local electromechanical response, cytocompatibility, angioge

SIRT1 promotes endothelial activity and osteogenesis in vitro and ameliorates age-related skeletal regeneration defects in vivo.

Understanding age-driven bone weakening mechanisms is essential for developing precision therapeutic strategies. This study systematically investigated skeletal aging, focusing on the critical decline of SIRT1 and its impact on type H vessels and osteogenic cells. We found that SIRT1 markedly enhanced both endothelial tube formation and osteogenic differentiation in senescent bone marrow-derived mesenchymal stem cell (BMSC)/human umbilical vein endothelial cell (HUVEC) co-cultures. In vivo validation studies revealed that pharmacological activation of SIRT1 significantly improved bone regenera

Development of Microfluidic Culture System for Spatially Regulated Differentiation of Mesenchymal Stem Cells Toward Osteochondral Interface Regeneration.

Regeneration of the osteochondral interface is essential for effective articular cartilage repair and the prevention of graft delamination. In this study, we developed a microfluidic culture device for the spatial regulation of mesenchymal stem cell (MSC) differentiation within a single-phase hydrogel by controlling concentration gradients of differentiation-inducing factors. The system comprised a PDMS culture chamber with dual-flow of culture media to generate concentration gradients of chondrogenic and osteogenic inductive factors. Numerical simulations were performed to characterize the fl

Self-powered electro-activated MgxFe1-xS2 scaffold for gas-ion therapy in osteosarcoma suppression and bone regeneration.

Postoperative treatment of osteosarcoma faces the dual challenges of tumor recurrence and extensive bone defect repair, necessitating the development of a synergistic strategy capable of simultaneously eliminating residual tumor cells and promoting bone regeneration. Existing prosthetic implants are often limited by their singular functionality, leading to issues such as high recurrence rates and non-union. This study innovatively constructs a functional titanium alloy scaffold system synergistically driven by a biodegradable "shell-like" triboelectric nanogenerator (BS-TENG). The BS-TENG effi

HCC-Derived Alpha-Fetoprotein Impairs Bone Regeneration by Impairing Autophagic Flux and Promoting Runx2 Degradation in Osteoblasts.

Hepatocellular carcinoma (HCC) is frequently complicated by bone metastasis, leading to severe skeletal-related events. While alpha-fetoprotein (AFP) is the gold-standard biomarker for HCC diagnosis, its functional role as a systemic or paracrine mediator in the bone metastatic microenvironment remains largely unexplored. In the present study, we investigated whether HCC-secreted AFP impairs bone formation by disrupting autophagic flux, the glycogen synthase kinase 3β (GSK3β)/β-catenin signaling pathway, and Runt-related transcription factor 2 (Runx2) stability in osteoblasts. Our results show

Commensal skin bacteria interact with the innate immune system to promote tail regeneration in Xenopus laevis tadpoles.

BACKGROUND: Tadpoles of the clawed frog Xenopus laevis can regenerate their tails following partial amputation, replacing the missing spinal cord, muscles, and fin. However, for a brief period of development this response becomes unstable, leading to a proportion of tadpoles that undergo wound healing rather than regenerative programme. Inspired by a growing number of links between the microbiome and human inflammatory disease, we asked how the tadpole skin microbiome and innate immunity influence the regeneration of a complex appendage. We previously showed that tadpoles raised in antibiotics

Collagen-polyurethane-maltodextrin hydrogels as bioactive scaffolds for soft and hard tissue regeneration, immunomodulation, and drug delivery.

Developing multifunctional biomaterials capable of supporting diverse therapeutic applications remains a major challenge in regenerative medicine. In this study, semi-interpenetrating polymer network (semi-IPN) hydrogels composed of collagen (C), polyurethane (PU), and maltodextrin (MD) were synthesized to investigate the influence of MD content (0-50 wt%) on their physicochemical and biological performance. The hydrogels exhibited an amorphous morphology with embedded MD granules, enhanced swelling capacity (up to 1100 %), and reduced crosslinking density at higher MD contents. Formulations c

Gelatin methacryloyl-phenylboronic acid/Hydroxyadamantane self-healing microgels for the periodontitis treatment by promoting alveolar bone regeneration.

Periodontitis is a chronic inflammatory condition mainly caused by the interaction between the host immune system and periodontal tissue pathogens, and may lead to consequences, such as alveolar bone defects and tooth loss. Incomplete bacterial removal, persistent inflammation and high reactive oxygen species (ROS) environment are the main challenges for periodontal tissue repair and alveolar bone regeneration. In this study, an injectable composite microgel (Gelatin methacryloyl-Phenylboronic acid/Hydroxyadamantane, GPH) loaded with antimicrobial peptide (AMP) and cerium dioxide (CeO2) micros
