Detection and characterization of a nucleoside transport system in human fibroblast lysosomes.

Lysosomes contain enzymatic activities capable of degrading nucleic acids to their constituent nucleosides, but the manner by which these degradation products are released from the lysosome is unknown. To investigate this process, human fibroblast lysosomes, purified on Percoll density gradients, were incubated with [3H]adenosine at pH 7.0, and the amount of adenosine taken up by the lysosomes was measured. Adenosine uptake by fibroblast lysosomes attained a steady state by 12 min at 37 degrees C and was unaffected by the presence of 2 mM MgATP or changes in pH from 5.0 to 8.0. An Arrhenius pl

Interplay of ELF5, GATA-3 and FLI-1 regulate malignant transformation of keratinocytes.

Cutaneous squamous cell carcinoma (cSCC) arises from epidermal keratinocytes and is the second most common epithelial malignancy. E74-like factor 5 (ELF5) is a member of the epithelium specific ETS subfamily of transcription factors and is essential for epithelial development, homeostasis and prevention of epithelial tumourigenesis. Transformation of keratinocytes involves the disruption of the progenitor differentiation program causing keratinocytes to fail to complete the process. This can lead to dysplastic epithelium and precancerous cells in skin. However, the precise mechanisms that caus

An AAV-based bioluminescent reporter reveals dynamic YAP/TAZ-TEAD transcriptional activity during liver injury.

YAP/TAZ are central mechanosensitive regulators of liver injury, fibrosis, and regeneration; however, their dynamic activity in vivo remains challenging to monitor. Here, we establish an AAV8-delivered bioluminescent reporter system (rAAV-8 × GTIIC-Luc) that enables real-time, non-invasive, and longitudinal visualization of hepatic YAP/TAZ-TEAD transcriptional activity. Small-molecule inhibitors targeting the upstream kinases LATS1/2 were employed to pharmacologically perturb YAP/TAZ activity, resulting in robust induction of reporter activity both in cultured cells and in mouse liver. Using a

Cancer stem cells are hyper-responsive sensors of the tumour microenvironment and regulate metastasis dynamics through YAP/TAZ.

Cancer stem cells (CSCs) drive metastasis and therapy resistance, yet their behaviour within the complex tumour microenvironment remains poorly understood. Here we use a fluorescent reporter that marks CSCs to show that CSCs and their more differentiated progeny display strikingly different population dynamics during metastatic lung colonization in breast cancer models. CSC expansion is rapidly curtailed early in colonization, suggesting a strong negative feedback mechanism acting selectively on this subpopulation. We showed that CSCs are exceptionally sensitive to local microenvironmental cue

Gain of function p53 mutant R273H confers distinct methylation profiles and consequent YAP/TAZ signaling mediated activation of partial or full EMT states to colon tumours.

BACKGROUND: Mutations in tumour suppressor p53 are frequently implied in aggressive progression and metastasis in colorectal cancer. But the distinct phenotypes exhibited by site-specific mutations of p53 are not well elucidated. Here, we investigate the epigenetic and transcriptional impact of three major p53 hotspot mutations (R175H, R273H and R282W), through DNA methylation changes and single cell transcriptomics. RESULTS: We observed that the p53 R273H mutation is associated with a partial epithelial-mesenchymal transition (pEMT) and increased metastatic progression. Analysis of DNA methy

Blocking PSMD14-mediated E2F1/ERK/AKT signaling pathways suppresses the progression of anaplastic thyroid cancer.

Anaplastic thyroid cancer (ATC) is the most aggressive subtype of thyroid cancer with few effective therapeutic strategies. Recent studies have identified the deubiquitinating enzyme (DUB) 26S proteasome non-ATPase regulatory subunit 14 (PSMD14) as a promising therapeutic target for multiple cancers; however, the role of PSMD14 in ATC remains largely unknown. Here, we found that PSMD14 was upregulated in ATC tissues and that its aberrant expression was negatively associated with the overall survival of patients with ATC. Functionally, PSMD14 promotes the proliferation and invasiveness of ATC c

UCMSCs-Derived Exosomal circHIPK3 Restrains Oxidative Stress and Inflammation by Downregulating the Stability of HMGB1 mRNA via Recruiting UPF1 in Diabetes Foot Ulcer.

Excessive oxidative stress and inflammation in human umbilical vein endothelial cells (HUVECs) are significant barriers to wound healing in diabetic foot ulcers (DFUs). Umbilical cord mesenchymal stem cells (UCMSCs)-derived exosomal circular RNA homeodomain-interacting protein kinase three (circHIPK3) exerts protective effects on HUVECs under high-glucose (HG) conditions. Our objective was to elucidate the downstream mechanisms through which UCMSCs-derived exosomal circHIPK3 modulates oxidative stress and inflammation in cellular and mice models of DFU. Single-cell RNA sequencing reveals that MYBL2 in malignant epithelial cells is involved in the development and progression of ovarian cancer.

BACKGROUND: Ovarian carcinoma (OC) is a prevalent gynecological malignancy associated with high recurrence rates and mortality, often diagnosed at advanced stages. Despite advances in immunotherapy, immune exhaustion remains a significant challenge in achieving optimal tumor control. However, the exploration of intratumoral heterogeneity of malignant epithelial cells and the ovarian cancer tumor microenvironment is still limited, hindering our comprehensive understanding of the disease. MATERIALS AND METHODS: Utilizing single-cell RNA sequencing (scRNA-seq), we comprehensively investigated th

The ketogenic diet modulates tumor-associated neutrophil polarization via the AMOT-YAP/TAZ axis to inhibit colorectal cancer progression.

Despite significant advances in the diagnosis and treatment of colorectal cancer (CRC), the prognosis for late-stage patients remains poor, highlighting the urgent need for new preventive and therapeutic strategies. Recent studies have focused on the ketogenic diet (KD) and its metabolite, β-hydroxybutyrate (BHB), for their tumor-suppressive effects and modulation of inflammatory responses. Using the azoxymethane (AOM) / dextran sulfate sodium (DSS)-induced mouse CRC model, we found that the ketogenic diet and BHB inhibit pro-tumor N2-type tumor-associated neutrophils (TANs) while promoting th

YAP1 inhibits ovarian endometriosis stromal cell invasion through ESR2.

Endometriosis is an estrogen-dependent disease, and estrogen receptor 2 (ESR2) plays a critical role in the pathogenesis of ovarian endometriosis by promoting cell invasion. Yes-associated protein 1 (YAP1) plays suppressive roles in several types of tumors. However, the relationship between YAP1 and ESR2 is not fully understood. The aim of this study was to investigate the regulatory mechanism of YAP1 in terms of ESR2 and YAP1 regulation of endometriotic stromal cell (ECSC) invasion in ovarian endometriosis. Our results demonstrated that YAP1 mRNA and protein levels in eutopic endometrium (EU)

Bacillus anthracis Virulence Regulator AtxA Binds Specifically to the pagA Promoter Region.

Anthrax toxin activator (AtxA) is the master virulence gene regulator of Bacillus anthracis It regulates genes on the chromosome as well as the pXO1 and pXO2 plasmids. It is not clear how AtxA regulates these genes, and direct binding of AtxA to its targets has not been shown. It has been previously suggested that AtxA and other proteins in the Mga/AtxA global transcriptional regulators family bind to the curvature of their DNA targets, although this has never been experimentally proven. Using electrophoretic mobility shift assays, we demonstrate that AtxA binds directly to the promoter region

RAC2 promotes abnormal proliferation of quiescent cells by enhanced JUNB expression via the MAL-SRF pathway.

Radiation-induced lung injury (RILI) occurs most often in radiotherapy of lung cancer, esophageal cancer, and other thoracic cancers. The occurrence of RILI is a complex process that includes a variety of cellular and molecular interactions, which ultimately result in carcinogenesis. However, the underlying mechanism is unknown. Here we show that Ras-related C3 botulinum toxin substrate 2 (RAC2) and transcription factor jun-B (JUNB) were upregulated in non-small cell carcinoma (NSCLC) tissues and were associated with poor prognoses for NSCLC patients. Parallel Regulation of von Hippel-Lindau Disease by pVHL-Mediated Degradation of B-Myb and Hypoxia-Inducible Factor α.

pVHL, the protein product of the von Hippel-Lindau (VHL) tumor suppressor gene, is a ubiquitin ligase that targets hypoxia-inducible factor α (HIF-α) for proteasomal degradation. Although HIF-α activation is necessary for VHL disease pathogenesis, constitutive activation of HIF-α alone did not induce renal clear cell carcinomas and pheochromocytomas in mice, suggesting the involvement of an HIF-α-independent pathway in VHL pathogenesis. Here, we show that the transcription factor B-Myb is a pVHL substrate that is degraded via the ubiquitin-proteasome pathway and that vascular endothelial growt

Hepatitis B Virus X Protein Induces Hepatic Steatosis by Enhancing the Expression of Liver Fatty Acid Binding Protein.

UNLABELLED: Hepatitis B virus (HBV) has been implicated as a potential trigger of hepatic steatosis although molecular mechanisms involved in the pathogenesis of HBV-associated hepatic steatosis still remain elusive. Our prior work has revealed that the expression level of liver fatty acid binding protein 1 (FABP1), a key regulator of hepatic lipid metabolism, was elevated in HBV-producing hepatoma cells. In this study, the effects of HBV X protein (HBx) mediated FABP1 regulation on hepatic steatosis and the underlying mechanism were determined. mRNA and protein levels of FABP1 were measured b
