Laminarin suppresses PD-L1 translation by targeting YTHDF1 to exert antitumor immunity in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is characterized by low response rates and rapid acquired resistance to current immunotherapies, highlighting an urgent unmet clinical need for novel therapeutic strategies. Herein, we demonstrated that laminarin, a biocompatible β-1,3-glucan extracted from brown algae, exerts significant antitumor immune effects against TNBC with no evident toxicity under the tested conditions. In addition, laminarin suppressed pulmonary metastatic progression in a TNBC metastasis model. Via an innovative in silico target screening framework integrating AI-driven knowledge

Natural Polymer-Based Hydrogels for Localized Therapy in Inflammatory Bowel Disease: From Smart Materials to Clinical Translation.

BACKGROUND: Inflammatory bowel disease (IBD) remains difficult to manage in a substantial proportion of patients because conventional systemic therapies may fail to achieve sufficient drug accumulation at inflamed intestinal sites while causing systemic adverse effects. Therefore, localized delivery systems capable of protecting therapeutic agents during gastrointestinal transit and releasing them at disease sites are urgently needed. Natural polymer-based hydrogels have attracted increasing attention because of their biocompatibility, biodegradability, mucoadhesiveness, and tunable responsive

Organelle-Targeted Nanocarriers for Cancer Therapy: Design Principles, Intracellular Trafficking, and Translational Barriers in Precision Drug Delivery.

The clinical translation of cancer nanomedicine remains constrained by inefficient intracellular delivery, poor subcellular specificity, and limited tumor accumulation, highlighting the need for more precise drug delivery strategies. Organelle-targeted nanocarriers have emerged as a promising platform for enhancing therapeutic efficacy by directing drugs to specific intracellular compartments, including the nucleus, mitochondria, lysosomes, and endoplasmic reticulum. However, the design principles governing efficient organelle-specific delivery and the barriers limiting clinical translation re

Hyaluronic acid-based tissue-engineered biomaterials for infected diabetic foot ulcers: mechanisms, evidence, and translational challenges.

Diabetic foot ulcers (DFUs) have difficulty healing clinically when infection and mature biofilms coexist with hyperglycemic injury, immune dysregulation, oxidative stress, ischemia-hypoxia, repetitive mechanical loading, and extracellular matrix (ECM) degradation. Debridement, infection management, offloading, vascular assessment and revascularization, when indicated, constitute the foundation of care; biomaterials can be evaluated only as adjuncts to this pathway. Hyaluronic acid (HA) is an attractive hydrated ECM-associated and chemically adaptable matrix that can support local retention, c

Mechanism-based precision pharmacology of natural products for triple-negative breast cancer: translational opportunities for targeted therapeutics.

Triple-negative breast cancer (TNBC) remains one of the most therapeutically challenging malignancies because of its pronounced molecular heterogeneity, aggressive progression, and frequent development of resistance to chemotherapy, targeted agents, and immunotherapy. Natural products offer a distinctive medicinal chemistry resource because their structurally diverse scaffolds can engage multiple oncogenic and immune-regulatory targets simultaneously; however, their potential in TNBC has largely been discussed in conventional descriptive reviews that catalog individual compounds and mechanisms

Plant-Derived Peptide-Polymer Therapeutics for Cutaneous Infections and Inflammation: Mechanistic Basis, Delivery Design and Translational Considerations.

Cutaneous infections and chronic inflammatory wounds remain difficult to treat because antimicrobial resistance, polymicrobial biofilms, excessive protease activity, oxidative stress, and impaired barrier repair collectively reduce the effectiveness of conventional topical therapies. Plant-derived antimicrobial peptides (AMPs) and peptide-associated bioactives offer antimicrobial, antibiofilm, immunomodulatory, and tissue reparative potential; however, their clinical translation is limited by proteolytic instability, poor stratum corneum penetration, short cutaneous residence time, formulation

Protective role of Bergenia ciliata in artesunate-induced nephrotoxicity: a translational pharmacology study integrating in-silico and experimental evidence.

INTRODUCTION: Artesunate (ART) is a widely used antimalarial agent; however, dose-dependent nephrotoxicity limits its clinical utility. Bergenia Ciliata, a medicinal plant with established renoprotective traditional use, was investigated as a potential nephroprotective agent against ART-induced kidney injury. METHODS: Bioactive metabolites from B. ciliata root extract were identified via Gas Chromatography-Mass Spectrometry (GC-MS) and filtered through in silico ADME profiling. Network pharmacology was employed to identify key hub genes implicated in ART-induced renal injury, with PPARγ emerg

Postpartum breast cancer as a distinct oncologic entity: linking physiological tissue remodelling to tumor biology and clinical translation.

Postpartum breast cancer (PPBC), defined as breast cancer diagnosed within 10 years after childbirth, is increasingly recognized as a biologically distinct and clinically aggressive subtype affecting young women worldwide. The postpartum mammary gland undergoes extensive remodelling during involution, characterized by inflammation, extracellular matrix reorganization, and immune suppression, collectively creating a tumor-promoting microenvironment. Recent studies have revealed unique molecular features of PPBC, including alterations in immune composition, stromal activation, and selective path

The dual role of ion channels in diabetic kidney disease: a translational paradigm for biomarkers and target discovery - reviews and prospects.

The clinical heterogeneity of diabetic kidney disease (DKD) poses a major challenge to current treatment strategies. This review proposes a novel translational paradigm: viewing ion channels as dual-function entities that serve both as pathogenic mediators and as rich sources of clinically actionable biomarkers. We systematically elucidate how hyperglycemia, oxidative stress, and inflammation disrupt sodium, calcium, potassium, and chloride channel networks through interconnected pathways such as AMP-activated protein kinase (AMPK)/mammalian target of rapamycin (mTOR), NLR family pyrin domain

Inside from outside: head-to-skull-and-muscles translation with statistical shape relation and multiple region-of-interest learning.

From skin to musculoskeletal prediction is a current challenge in biomechanics. Previous studies attempted head-to-skull prediction using statistical shape relationships, but they did not leverage local shape geometries and reduced muscles to simple 1-D action lines. We introduced Inside from Outside, a novel approach that reconstructs the bones of the skull and the facial muscles from external head surfaces. The method coupled Region-of-Interest (ROI) learning with Statistical Shape Relationship (SSR) modeling to capture detailed anatomical correspondences. We reconstructed 329 head and skull

Therapeutic Translation of Epicardial Adipose Tissue Targeting in Heart Failure With Preserved Ejection Fraction: How Far Are We?

Heart failure with preserved ejection fraction (HFpEF) exhibits an inflammatory-metabolic phenotype in association with multiple factors. Among them, epicardial adipose tissue (EAT), a type of visceral adipose tissue surrounding the heart, has been gaining attention. EAT expansion is associated with the development of HFpEF, the increased risk for heart failure-related mortality in patients with HFpEF, independent of age, body mass index, diabetes, and sex. Thus, reducing EAT expansion has been proposed as a promising therapeutic strategy for treating HFpEF. Although specific treatment for EAT

The m6A reader YTHDF1 modulates adipocyte mitochondrial remodeling and diet-induced adiposity through translational control of NOD1.

The excessive expansion of white adipose tissue (WAT) that characterizes obesity is a major global health concern. N6-methyladenosine (m6A) modification and its readers regulate metabolic homeostasis, but how m6A-dependent translational control shapes mitochondrial remodeling in white adipocytes during obesity remains unclear. In this study, we examined the function of the m6A reader YTHDF1 in diet-induced adiposity using whole-body and adipocyte-specific Ythdf1 knockout mice, together with primary adipocyte models. Under high-fat diet (HFD) feeding, both Ythdf1-/- and Ythdf1AKO mice developed
