Running an electron microscopy core facility.

As electron microscopes became more costly, technically complex, and integral to a wide range of scientific fields, centralised electron microscopy (EM) core facilities have become essential for maintaining accessibility, performance, and quality. Drawing on the experiences of running two EM core facilities in Finland, we outline key considerations for establishing and operating a biological and biomedical EM facility. We cover facility infrastructure and technical requirements, user training, sample preparation, safety and occupational considerations, personnel and cost management, and finish

Natural Killer Cell Membrane Isolation with Minimal Nuclear, Mitochondrial, and Cytosol Contamination.

Cell membrane isolation is essential for diverse biological investigations, ranging from fundamental research to advanced therapeutic applications. This study compared two methods-differential centrifugation and discontinuous sucrose density gradient ultracentrifugation-for isolating cell membranes from the human natural killer (NK) cell line (KHYG-1). The aim was to identify the method that minimizes contamination from nuclear, mitochondrial, and cytosolic components. Differential centrifugation yielded approximately 8 mg of cell membrane, whereas sucrose density gradient ultracentrifugation

Hormonal mechanisms of women's risk in the face of traumatic stress.

Women are underrepresented in biomedical research, limiting understanding of their disproportionate rates of stress-related disorders. Although men experience more trauma, women are twice as likely to develop posttraumatic stress disorder (PTSD). We tested whether ovarian hormones contribute to this disparity via their influence on threat-related brain activity. In a randomized, double-blind crossover study of 110 young women, we examined how exogenous estradiol (E2) modulates threat neurocircuitry, timed precisely to ovarian phase. E2 increased engagement of the ventromedial prefrontal cortex

Network pharmacology-based investigation and experimental validation of the mechanism of metformin in the treatment of acute myeloid leukemia.

Metformin, a widely used anti-diabetic agent, has shown significant anti-cancer properties as reported in in various cancers, including acute myeloid leukemia. However, the detailed mechanisms by which metformin influences acute myeloid leukemia remain unrevealed. Employing a synergistic approach of network pharmacology and experimental validation, this study systematically identifies and analyzes potential metformin targets and AML-related genes. These findings are then cross-referenced with biomedical databases to construct a target-gene network, providing insights into metformin's pharmacod

In vitro analysis of iron oxide (Fe3O4) nanoparticle mediated degradation of polycyclic aromatic hydrocarbons (PAHs) and their antimicrobial activity.

To degrade anthracene, magnetite nanoparticles were produced using a simple co-precipitation process. The fabricated nanoparticles have been analyzed for structural and optical properties. XRD examination revealed that the produced Fe3O4 nanoparticles were cubic phase, having a mean crystallite dimension of 18.84 nm. DLS determined the hydrodynamic diameter of Fe3O4 nanoparticles to be 182 nm. UV-Vis research revealed that Fe3O4 nanoparticles absorb at 390 nm. A peak at 895 cm-1 in the FT-IR study indicated the metal-oxygen connection. The synthesized Fe3O4 nanoparticles demonstrated an effect

Chemical transformation and cytotoxicity of iron oxide nanoparticles (IONPs) accumulated in mitochondria.

Iron oxide nanoparticles (IONPs) have been widely used as a nanoscale tool in biomedical research. However, it remains largely unknown how IONPs are transformed at a subcellular level to elicit distinct biological effects. In the present study, we prepared three different IONPs, including two IONPs targeting mitochondria (IONP-TPP) and lysosomes (IONP-APM), respectively, and a control with no specified target (IONP). By MTT assay and JC-1 staining, mitochondria-targeted IONP-TPP was found to produce significant cytotoxicity and severe mitochondrial membrane depolarization in MCF-7 cells. Furth

Spondyloarthritis in Taihangshan Macaques (Macaca mulatta tcheliensis).

The rhesus macaque (Macaca mulatta) is the most widely distributed nonhuman primate species, and captive populations play an important role in biomedical research due to close phylogenetic and physiological similarity to human beings. However, to our best knowledge, the spondyloarthritis (SpA) in rhesus macaques has been exclusively reported in captive or semicaptive populations rather than wild counterparts. In the present study, we report 2 cases of SpA observed in Taihangshan macaques (Macaca mulatta tcheliensis) inhabiting the Taihangshan Macaque National Nature Reserve, Henan Province, Ch

Ancient hybridization and strong adaptation to viruses across African vervet monkey populations.

Vervet monkeys are among the most widely distributed nonhuman primates, show considerable phenotypic diversity, and have long been an important biomedical model for a variety of human diseases and in vaccine research. Using whole-genome sequencing data from 163 vervets sampled from across Africa and the Caribbean, we find high diversity within and between taxa and clear evidence that taxonomic divergence was reticulate rather than following a simple branching pattern. A scan for diversifying selection across taxa identifies strong and highly polygenic selection signals affecting viral processe

Vitamin E-TPGS stabilized self-assembled tripeptide nanostructures for drug delivery.

Self-assembled peptides and specifically small peptide based nanostructures have been the focus of research in past decade due to their potential biological applications. In this study, we prepared a protected peptide, Boc-Pro-Phe-Gly-OMe, which self-assembled in aqueous solutions leading to the formation of nanostructures and ability to act as a drug carrier. Dynamic light scattering (DLS) measurements showed nanostructures with average size of 119.6 nm containing hydrophobic core, wherein hydrophobic drugs, viz, eosin, aspirin and curcumin, were successfully encapsulated. These encapsulated

Engineering functional human vasculature: iPSC-derived vascular cells and organoids for disease modeling and translation.

Human vascular function depends on tightly coordinated structural, mechanical, and cellular interactions, yet these features remain difficult to recapitulate in vitro. Induced pluripotent stem cells (iPSCs) enable efficient generation of vascular cell types, including endothelial cells, smooth muscle cells, and pericytes, but current systems often lack functional maturity and physiological relevance. Recent advances in vascular organoid engineering provide new opportunities to address this limitation. By integrating self-organization, co-culture, and bioengineering approaches, iPSC-derived sys

Targeting DYRKs in Cardiovascular Diseases: From Biological Mechanisms to Therapeutic Translation.

Cardiovascular diseases (CVDs) remain the leading cause of mortality globally, characterized by myocardial injury, pathological structural remodeling, and progressive deterioration of cardiac function. Clinical manifestations include post-infarct functional impairment, pathological cardiac hypertrophy, interstitial fibrosis, malignant arrhythmias, and end-stage heart failure. Although the dual-specificity tyrosine-regulated kinase (DYRK) family has been extensively investigated in cancer and neurodegenerative disorders, emerging evidence highlights DYRKs as critical upstream regulators in a wi

Translational assessment of butyrylcholinesterase activity as a diagnostic biomarker for depression using a chemiluminescent probe.

Depressive episode is a globally prevalent psychiatric condition with a complex, multifactorial etiology involving genetic and environmental factors and despite advances in biomarker research, clinical applications remain limited due to methodological inconsistencies and population variability. Butyrylcholinesterase (BChE), an enzyme involved in cholinergic transmission, has been less studied in the context of depression compared to acetylcholinesterase. Utilizing a BChE-responsive chemiluminescent probe (BCC), BChE activity was measured in a translational approach: human serum, rat plasma, an

Gastric retention for cardiovascular therapy: matching drug properties, dosage-form design, and translational evidence.

Gastroretentive delivery offers a selective strategy for cardiovascular drugs the performance of which depends on upper-gastrointestinal (GI) residence, sustained or modified exposure, or timed release. In this review, I synthesize the rationale, candidate drugs, platform-selection logic, and translational evidence for gastroretentive cardiovascular systems. Particularly strong cases involve narrow absorption windows, poor lower-GI absorption, pH-dependent solubility or instability, low or variable bioavailability, distal-GI efflux, and circadian cardiovascular risk. I proposes a drug-first fr

On Burden of Diseases, Prevention, Medical Research and Health Service Delivery: Grampian Case Study.

Burden of diseases measured as disability-adjusted life years (DALYs) per 100,000 people can be mined from public domain data, when they are made available by population health surveillance systems. This can be analysed to allow insightful comparisons with the national average, and to understand differences in trends between the sexes, age groups, time periods, geographic regions, and sub-regions. In this illustrative case study, we have analysed the Scottish burden of disease database to understand what ailed the population of the Grampian region before the COVID-19 pandemic. We have identifi

Determination of Human Effective Dose via Translational Ocular Modeling: Clinical Translation of a Novel Topical Agent Using PBPK.

Wet age-related macular degeneration (wAMD) is a leading cause of vision loss, and human dose selection for ocular therapeutics remains a key translational challenge. We developed a physiologically based pharmacokinetic (PBPK) model for EB-203 to guide dose selection by translating preclinical ocular tissue exposure to human. PK data were obtained from mouse and rabbit studies and human plasma PK from a phase I trial. The model incorporated species-specific ocular physiology, drug permeability, and pharmacokinetics, and was implemented using an open-source PBPK modeling software (MoBi, version

FGF/FGFR signaling at the immune-stromal interface in autoimmune rheumatic diseases: from tissue remodeling to translational opportunities.

The fibroblast growth factor (FGF)/FGF receptor (FGFR) signaling system regulates tissue repair, angiogenesis, extracellular matrix remodeling, fibrosis, and bone metabolism. Although FGF/FGFR biology has been extensively studied in oncology, development, and fibrotic lung disease, its relevance to systemic autoimmune rheumatic diseases remains scattered across disease-specific and unevenly supported literatures. This narrative, evidence-informed review synthesizes current evidence on FGF/FGFR signaling in rheumatoid arthritis (RA), systemic lupus erythematosus/lupus nephritis (SLE/LN), system
