IL-12 secretion by Langerhans cells stimulated with Candida skin test reagent is mediated by dectin-1 in some healthy individuals.

OBJECTIVE: Our group and others have shown that serial intra-lesional injections of common warts with skin testing reagents such as Candida, mumps and Trichophyton are effective in regressing injected and non-injected warts. Anti-HPV T-cell responses appear to be induced. The goal of this study was to understand the mechanisms of how Candida skin testing reagent enhances immune responses. METHODS: The following immunological features were studied to understand how Candida induces immune responses in healthy subjects: (1) proliferative capacity of T-cells upon exposure to Candida through monoc

[Inhibitory activity of hydrosols prepared from 18 Japanese herbs of weak aromatic flavor against filamentous formation and growth of Candida albicans].

Leaf hydrosols prepared from 18 weakly aromatic Japanese herbs used traditionally were tested on the filamentation-inhibitory activity of Candida albicans. These hydrosols were divided into two classes, A and B. The inhibitory activity of 13 hydrosols belonging to class A was markedly altered depending on the drying process of the parent herbs. On the other hand, the remaining 5 hydrosols belonging to class B showed no significant change on the composition and inhibitory activity upon drying. The change of the bioactivity was correlated with the change and concentration of the respective major

Lipidomics of Candida albicans biofilms reveals phase-dependent production of phospholipid molecular classes and role for lipid rafts in biofilm formation.

Candida albicans-associated bloodstream infections are linked to the ability of this yeast to form biofilms. In this study, we used lipidomics to compare the lipid profiles of C. albicans biofilms and planktonic cells, in early and mature developmental phases. Our results showed that significant differences exist in lipid composition in both developmental phases. Biofilms contained higher levels of phospholipid and sphingolipids than planktonic cells (nmol per g biomass, P<0.05 for all comparisons). In the early phase, levels of lipid in most classes were significantly higher in biofilms compa

Comparison of gene expression profiles of Candida albicans azole-resistant clinical isolates and laboratory strains exposed to drugs inducing multidrug transporters.

Azole resistance in Candida albicans can be due to upregulation of multidrug transporters belonging to ABC (ATP-binding cassette) transporters (CDR1 and CDR2) or major facilitators (CaMDR1). Upregulation of these genes can also be achieved by exposure to fluphenazine, resulting in specific upregulation of CDR1 and CDR2 and by exposure to benomyl, resulting in specific CaMDR1 upregulation. In this study, these two different states of gene upregulation were used to determine coregulated genes that often share similar functions or similar regulatory regions. The transcript profiles of a laborator

Facial Candida albicans cellulitis occurring in a patient with oral submucous fibrosis and unknown diabetes mellitus after local corticosteroid injection treatment.

Facial cellulitis caused by odontogenic bacterial infection is frequently encountered; however, facial cellulitis caused by Candida albicans infection is rarely found. A patient with oral submucous fibrosis (OSF) and unknown diabetes mellitus (DM) was treated in our out-patient dental clinic by biweekly submucosal injection of 40 mg triamcinolone acetonide into bilateral buccal mucosae plus forced mouth opening performed by the two hands of the clinician. The interincisal distance of the patient improved from 28 to 48 mm after four times of steroid injection. The symptoms and signs of OSF also

Longitudinal study of mucosal Candida species colonization and candidiasis among human immunodeficiency virus (HIV)-seropositive and at-risk HIV-seronegative women.

Acquisition and loss rates and estimates of duration of oral and vaginal Candida species colonization and candidiasis were examined among 868 human immunodeficiency virus (HIV)-seropositive and 437 at-risk HIV-seronegative women monitored prospectively during 1993-1999. Colonization and candidiasis acquisition rates, both oral and vaginal, were significantly higher among HIV-seropositive women; the magnitude of increase in candidiasis outcomes for HIV-seropositive women was greater for oral candidiasis than for vaginal candidiasis. Loss rates and estimates of duration of incident outcomes indi

Significance of the isolation of Candida species from respiratory samples in critically ill, non-neutropenic patients. An immediate postmortem histologic study.

The diagnosis of pulmonary candidiasis is still controversial. We undertook a prospective study on 25 non-neutropenic, mechanically ventilated (> 72 h) patients who died in our ICU with the aim of assessing the incidence and significance of the isolation of Candida species from quantitative cultures of immediate postmortem lung biopsies and different respiratory sampling techniques. Immediate postmortem respiratory samples (endotracheal aspirate, protected specimen brush [PSB], bronchoalveolar lavage [BAL], blind biopsies [average 14/patient], and bilateral bronchoscopically guided biopsies [t

Association between gastrointestinal tract carriage of Candida, blood group O, and nonsecretion of blood group antigens in patients with peptic ulcer.

Of 112 patients with peptic ulcer disease examined for oral carriage of Candida, 66 (59%) were carriers. Candida carriage was associated with blood group O (P < 0.05) and, independently, with nonsecretion of blood group antigens (P < 0.01). For each subject, the presence or absence of yeasts was found to be a constant characteristic, and only among patients positive for Candida was blood group O or nonsecretion more frequent than expected in the general population. The quantity of yeasts isolated was significantly greater in patients than in normal subjects (P < 0.002), as was the frequency of

Amphotericin B and amphotericin B methyl ester ascorbate. I. Chemotherapeutic activity against Candida albicans, Cryptococcus neoformans, and Blastomyces dermatitidis in mice.

Amphotericin B methyl ester (AME) has been reported to possess in vitro antifungal activity similar to that of amphotericin B and to have less intrinsic toxicity in mice and dogs. For these reasons AME has been porposed as an alternative to amphotericin B in the therapy of deep mycoses. For comparison of the therapeutic efficacy of the two polyenes in laboratory animals before initiation of studies in humans, groups of mice were infected with Candida albicans, Cryptococcus neoformans, and Blastomyces dermatitidis. Treatment consisted of two or more doses of each drug given by the intravenous r

Evaluating a Flexible Workflow for Candida auris Surveillance on the AltoStar Automation System AM16 with Commercial and In-House Real-Time PCR Assays.

BACKGROUND: Multidrug-resistant yeast Candida (Candidozyma) auris, responsible for healthcare-associated outbreaks and high mortality, is a major global health threat. Rapid and scalable molecular diagnostics is essential; however, few automated platforms can accommodate both commercial assays and laboratory-developed tests (LDTs). We evaluated the analytical and clinical performance of two real-time PCR assays for C. auris detection using the AltoStar Automation System AM16. METHODS: One assay was an in-house LDT (targeting the internal transcribed spacer region), while the other was AltoSta

Droplet digital PCR assay for simultaneous detection of the ERG11 mutation and copy number variation associated with azole resistance in Candida tropicalis.

BACKGROUND: Azole resistance in Candida tropicalis has increased worldwide and is largely driven by an emerging population previously reported Cluster AZR/X. This cluster exhibits high-level azole resistance driven by ERG11 A395T/W mutations accompanied by amplification of mutant ERG11 alleles, underscoring the clinical need for a quantitative, mechanism-informed molecular assay. METHODS: We developed and validated a droplet digital PCR (ddPCR)-based assay enabling simultaneous detection of the ERG11 mutation and quantitative assessment of ERG11 copy number variations (CNVs). The assay employ

Environmental and genetic regulation of mannan architecture in Candidozyma auris (formerly Candida auris), a comparison to other related yeast species: A review.

Candidozyma auris (formerly Candida auris) is a globally emerging, multidrug-resistant fungal pathogen characterized by its unique ability to colonize human skin and persist on abiotic surfaces. Central to its survival and pathogenicity is the fungal cell wall, a highly dynamic structure that mediates host-pathogen interactions and immune evasion through the regulation of its outermost mannan layer. Unlike the primary model Candida albicans, C. auris possesses a denser mannan shield and structurally distinct glycans characterized by an exceptionally low molecular mass and unique phosphomannan

Candidozyma auris (formerly Candida auris) in the acute care environment: Identifying and controlling transmission through a systematic investigation in an academic hospital.

A nosocomial Candidozyma auris (formerly Candida auris) outbreak affecting cardiac surgery patients prompted implementation of a multifaceted infection prevention strategy in a large academic hospital. Interventions included an expanded epidemiological line listing, enhanced surveillance, environmental culturing, whole-genome sequencing, and implementation of an in-house polymerase chain reaction assay. This integrated approach achieved sustained interruption of transmission in a high-risk cardiac population and provides an adaptable model for outbreak investigation in high-acuity settings.

Discovery and evaluation of a marine-derived chlorinated flavone derivative CHNQD-02204 as a potent and selective antifungal agent against Candida albicans.

Fungal infections are a significant contributor to global morbidity and mortality, among which Candida albicans infections, a major cause of systemic candidiasis, have a mortality rate of approximately 40%. Developing effective and safe antifungal drugs with novel chemical scaffolds is urgently needed. In this study, more than 200 fungi from a deep-sea-derived fungal library were screened for anti-C. albicans activity, and a fungus Aspergillus candidus (CHNSCLM-1227) strain with strong activity was discovered. Using anti-C. albicans activity screening combined with LC-MS/MS-based molecular net

In silico identification of isosilybin a targeting squalene epoxidase as an antifungal adjuvant with in vitro validation against azole-resistant Candida and emerging yeasts.

The rising incidence of antifungal resistance in Candida species and emerging non-albicans yeasts has markedly reduced the efficacy of azole antifungal therapy, highlighting the need for resistance modifying strategies. Natural products represent a valuable reservoir of bioactive scaffolds, and computational prioritization facilitates the systematic identification of pharmacologically relevant lead molecules. In this study, we employed an integrated in silico-in vitro antimicrobial pharmacology approach to identify Isosilybin A as a potential ERG1-targeting antifungal adjuvant and to assess it
