Considerations in the choice of a skin donor site for harvesting keratinocytes containing a high proportion of stem cells for culture in vitro.

The treatment of severely burned patients has benefited from the grafting of skin substitutes obtained by expansion of epithelial cells in culture. The aim of this study was to evaluate whether the anatomic site chosen for harvesting skin had an impact on the quality of the derived cell cultures. Considering that hair follicles contain epithelial stem cells, we compared hairy skin sites featuring different densities and sizes of hair follicles for their capacity to generate high quality keratinocyte cultures. Three anatomic sites from adult subjects were compared: scalp, chest skin and p-auric

A Single-Step Method for the Simultaneous Preparation of DNA, RNA, and Protein from Cells and Tissues.

This protocol allows the simultaneous recovery of RNA, DNA, and protein from an aliquot of tissue or cells by lysis of cells with a monophasic solution of guanidine isothiocyanate and phenol. Addition of chloroform generates a second (organic) phase into which DNA and proteins are extracted, leaving RNA in the aqueous supernatant. The DNA and proteins can be isolated from the organic phase by sequential precipitation with ethanol and isopropanol, respectively. The DNA recovered from the organic phase is ∼20 kb in size and is a suitable template for PCRs. The proteins, however, remain denatured

Fractionation of mouse bone-marrow cells limits functional efficacy in non-reperfused mouse model of acute myocardial infarction.

BACKGROUND AND OBJECTIVES: Clinical trials of bone-marrow (BM)-derived cells for therapy after acute myocardial infarct (MI) have been controversial. The most commonly used cells for these trials have been mononuclear cells (MNC), obtained by fractionation of BM cells (BMCs) via different protocols. In this study, we performed a head-to-head comparison of: 1) whole BMC; 2) fractionated BM (fBM) using the commonly used Ficoll protocol; 3) the extract derived from the fBM (fBM extract) versus 4) saline (HBSS) control for treatment of acute MI. METHODS: In total, 155 male C57BL/6J (10-12-week ol

Deltins: immunochemical evidence for a novel population of peptides of the D cells of the gastro-entero-pancreatic endocrine system.

Differences between the immunocytochemical behaviour of antisera to partially purified porcine gastrins and antisera to either synthetic human gastrin-17-I or highly purified porcine gastrin-17-I raised the hypothesis that hog antral gastrin extracts contain peptides different from somatostatin and gastrin that are responsible for the immunocytochemical reaction of the former antisera in the D (delta) cells of the gastro-entero-pancreatic (GEP) endocrine system. This study was performed to prove this hypothesis. A discard fraction obtained after gel filtration of hog antral gastrin extracts on

Immunochemical study and cellular localization of human pepsinogens during ontogenesis and in gastric cancers.

Gastric pepsinogens were studied by immunoenzymologic and immunohistochemical methods in non-cancerous adult gastric mucosa, in fetal stomach, and in gastric carcinomas. In noncancerous adult mucosa, immunoenzymologic methods showed that pepsinogen II (previously called Pg I-II) was found mostly in fundic or mediogastric extracts, whereas Pg IV was predominant in antropyloric extracts. Pg II was localized by immunofluorescence techniques in the chief cells of deep glands found near the muscularis. Pg II was localized by immunofluorescence techniques in the chief cells of deep glands found near

Extraction and Preparation of Listeria monocytogenes Subproteomes for Mass Spectrometry Analysis.

Proteomics has become an essential tool to answer biologists' questions. For bacteriologists, the proteome of bacteria is much less complex than that of eukaryotic organisms. However, not all the different cell "compartments" are easily accessible, and the analysis of cell envelope proteins is particularly challenging. For the Gram-positive bacterium Listeria monocytogenes, one of the main foodborne pathogen microorganisms, the study of surface proteins is crucial to better understand the mechanisms of pathogenicity, as well as adaptation/resistance to and persistence in hostile environments.

BioFlow: a non-invasive, image-based method to measure speed, pressure and forces inside living cells.

Cell motility is governed by a complex molecular machinery that converts physico-chemical cues into whole-cell movement. Understanding the underlying biophysical mechanisms requires the ability to measure physical quantities inside the cell in a simple, reproducible and preferably non-invasive manner. To this end, we developed BioFlow, a computational mechano-imaging method and associated software able to extract intracellular measurements including pressure, forces and velocity everywhere inside freely moving cells in two and three dimensions with high spatial resolution in a non-invasive man

Identification of a specific protein factor defective in group A xeroderma pigmentosum cells.

A protein factor which corrects the defect in xeroderma pigmentosum cells belonging to complementation group A (XP-A cells) was detected in a cell extract prepared from calf thymus. The activity of this factor was measured as the amount of unscheduled DNA synthesis (UDS) reappearing in UV-irradiated XP-A cells after microinjection of the extract. The native molecular mass of this factor was estimated to be 80 kDa by gel-filtration and 25 kDa by glycerol gradient centrifugation. The activity was, however, recovered at a position corresponding to 43 kDa after renaturation on an SDS-PAGE gel. The
