Isolated Limb Perfusion on Nonmelanoma Skin Cancer for Limb Salvage: A Series of Four Cases.

Nonmelanoma skin cancer (NMSC) is more prevalent than all the other cancers combined together. The most common regions affected by skin cancer are the head and neck, but there is a large proportion of the cases located on the limbs. They could be bulky, very extensive and/or located in specialized regions like the hands or near to a joint. Most of those cases should be amputated, with several compromises of the function and a negative impact on the patients' quality of life. Isolated limb perfusion is a proven alternative to limb salvage on soft tissue sarcomas, but there are just a few report

An unusual presentation of gastric mucosa-associated lymphoid tissue (MALT)-type lymphoma.

MALT: Mucosa-associated lymphoid tissue (MALT)-type lymphoma is a relatively rare disease; nevertheless, it is the third most common lymphoma type, accounting for 5-7% of all non-Hodgkin lymphomas. Case series and retrospective analysis published in the literature have suggested that extra gastrointestinal (GI) MALT-type lymphoma can occur simultaneously with MALT-type lymphoma involving the GI tract. We report the case of a healthy, 64-year-old Caucasian male who presented with progressive fatigue, non-productive cough, and worsening exertional shortness of breath for 3 months who was subsequ

Improvement of wound healing by water-filtered infrared-A (wIRA) in patients with chronic venous stasis ulcers of the lower legs including evaluation using infrared thermography.

BACKGROUND: Water-filtered infrared-A (wIRA) is a special form of heat radiation with a high tissue-penetration and with a low thermal burden to the surface of the skin. wIRA is able to improve essential and energetically meaningful factors of wound healing by thermal and non-thermal effects. AIM OF THE STUDY: prospective study (primarily planned randomised, controlled, blinded, de facto with one exception only one cohort possible) using wIRA in the treatment of patients with recalcitrant chronic venous stasis ulcers of the lower legs with thermographic follow-up. METHODS: 10 patients (5 mal

Repurposing Masitinib Mesylate as a Novel FOXM1 Inhibitor for the Treatment of Aggressive Solid Tumors: Preclinical Validation in Human Breast and Oral Cancer Cells and Organotypic Tumor Slice Culture.

The Forkhead box M1 (FOXM1) transcription factor, recognized as a prominent oncogene, facilitates cell cycle progression and mitotic fidelity in aggressive cancers. Overexpression of FOXM1 is correlated with uncontrolled cell proliferation, metastasis, and poor prognosis. Despite its pivotal role in promoting genomic instability, cancer stemness, and metastasis, FOXM1 remains a clinically undruggable target. In this study, we identified masitinib mesylate, a small-molecule inhibitor known for its multitargeted tyrosine kinase inhibition, targeting c-Kit, PDGFRα, and PDGFRβ, as well as Lyn, whi

Animal-free matrix alternative for three-dimensional in vitro angiogenesis models.

All metabolically active tissues have a dense vascular network to supply oxygen and nutrients. Angiogenesis, the formation of new blood vessels from existing ones, can be activated by the lack of oxygen in cells, disease, trauma, or tumor growth. Due to its essential role in cell survival, angiogenesis has been extensively studied, and therapeutic control of this process is of growing interest. Simultaneously, research aims to find and standardize non-animal testing methods for future use. However, the establishment of reproducible and physiologically relevant vascular networks in vitro is cha

The Application of 2d Mxene Nanosheet -Based Thermosensitive Gel Delivery System Loaded with Cisplatin and Imiquimod for Lung Cancer.

INTRODUCTION: Lung cancer's high incidence and dismal prognosis with traditional treatments like surgery and radiotherapy necessitate innovative approaches. Despite advancements in nanotherapy, the limitations of single-treatment modalities and significant side effects persist. To tackle lung cancer effectively, we devised a temperature-sensitive hydrogel-based local injection system with near-infrared triggered drug release. Utilizing 2D MXene nanosheets as carriers loaded with R837 and cisplatin (DDP), encapsulated within a temperature-sensitive hydrogel-forming PEG-MXene@DDP@R837@SHDS (MDR@

Stromal Barrier-Dismantled Nanodrill-Like and Cancer Cell-Targeted pH-Responsive Polymeric Micelles for Further Enhancing the Anticancer Efficacy of Doxorubicin.

Cancer-associated fibroblasts (CAFs) were believed to establish a tight physical barrier and a dense scaffold for tumor cells to make them maintain immunosuppression and drug resistance, strongly hindering nanoparticles to penetrate into the core of tumor tissues and limiting the performance of tumor cell-targeted nanoparticles. Here, we fabricated the substrate Z-Gly-Pro of fibroblast activation protein α (FAPα) and folic acid-codecorated pH-responsive polymeric micelles (dual ligand-modified PEOz-PLA polymeric micelles, DL-PP-PMs) that possessed nanodrill and tumor cell-targeted functions ba

Epstein-Barr Virus-Positive T/NK-Cell Lymphoproliferative Disorders Manifested as Gastrointestinal Perforations and Skin Lesions: A Case Report.

Systemic Epstein-Barr virus (EBV)-positive T-cell lymphoproliferative disorders (LPDs) of childhood is a highly aggressive EBV-positive T/natural killer (NK)-cell LPD, which emerges in the background of chronic active EBV infection (CAEBV) or shortly after primary acute EBV infection. The clinical presentations of CAEBV are varied; patients with atypical manifestations are easily misdiagnosed. We described a 14-year-old boy suffering from digestive disorders and intermittent fever for 1 year and 9 months, whose conditions worsened and skin lesions occurred 2 months before hospitalization. Harnessing exosomes in dry eye disease: a triple threat approach.

BACKGROUND: Dry eye disease is a chronic ocular surface disorder primarily driven by tear film instability and hyperosmolarity, which propagates inflammatory cascades leading to ocular tissue damage. Exosomes facilitate the transfer of proteins, nucleic acids, and lipids between donor and recipient cells, mediating the exchange of bioactive molecules. These vesicles demonstrate transformative potential as carriers of bioactive molecules, drug delivery vehicles, and disease biomarkers. Although exosome research in posterior segment eye diseases is extensive, studies investigating their triple f

Evaluation of PRG4 mRNA-Loaded Polymeric Nanoparticles in In Vitro and In Vivo Models of Osteoarthritis.

BACKGROUND: Osteoarthritis (OA) is a highly prevalent degenerative joint disease for which no disease-modifying treatments are available. Proteoglycan 4 (PRG4) is a glycoprotein with lubricating and immunomodulatory properties, making it a promising therapeutic target for mRNA-based therapies. Therefore, this study evaluated the effects of PRG4 mRNA delivered via polymeric nanoparticles in both in vitro and in vivo OA models. METHODS: Human chondrocytes from OA patients were cultured in 3D pellet models and transfected with PRG4 mRNA-loaded polymeric nanoparticles at low or high dose, once or

Strategies to promote liver fibrosis amelioration with involvement of restorative macrophages.

Chronic liver diseases represent a major global health burden, with fibrosis as the common pathological outcome of sustained hepatic injury. Once considered irreversible, liver fibrosis is now recognized as a dynamic and potentially reversible process driven by complex cellular and molecular interactions within the hepatic microenvironment. Among these, hepatic macrophages have emerged as central regulators of both fibrogenesis and fibrosis resolution due to their remarkable phenotypic and functional plasticity. This review integrate experimental, translational, and emerging clinical evidence

Non-Coding RNA in Type 2 Diabetes Cardio-Renal Complications and SGLT2 Inhibitor Response.

Type 2 diabetes mellitus (T2DM) is characterized by an uncontrolled increase in blood glucose levels and insulin resistance in cells of various tissues. Vascular complications in T2DM have an inflammatory nature. Drugs with different mechanisms of action have been developed and used to treat T2DM, initially aimed at controlling blood glucose levels. Among them, sodium-glucose cotransporter 2 inhibitors (SGLT2-i) were developed as specific inhibitors of glucose reabsorption in the kidneys, but along with lowering blood glucose levels, they demonstrated multiple (including non-glycemic) positive

A potential therapeutic strategy of an innovative probiotic formulation toward topical treatment of diabetic ulcer: an in vivo study.

BACKGROUND: The probiotic potential of Lacticacid bacteria has been studied in various medical complications, from gastrointestinal diseases to antibiotic resistance infections recently. Moreover, diabetic ulcer (DU) is known as one of the most significant global healthcare concerns, which comprehensively impacts the quality of life for these patients. Given that the conventional treatments of DUs have failed to prevent later complications completely, developing alternative therapies seems to be crucial. METHODS: We designed the stable oleogel-based formulation of viable probiotic cells, incl

Bioprinting of anisotropic functional corneal stroma using mechanically robust multi-material bioink based on decellularized cornea matrix.

Corneal scarring is a common cause of blindness, affecting millions globally each year. A huge gap between the demand and supply of donor tissue currently limits corneal transplantation, the only definitive therapy for patients with corneal scarring. To overcome this challenge, researchers have harnessed the efficacy of 3D bioprinting to fabricate artificial corneal stromal constructs. With all the different bioinks available, the decellularized corneal matrix-based bioprinted construct can fulfill the required biological functionality but is limited by the lack of mechanical stiffness. FLASH and minibeams in radiation therapy: the effect of microstructures on time and space and their potential application to protontherapy.

After years of lethargy, studies on two non-conventional microstructures in time and space of the beams used in radiation therapy are enjoying a huge revival. The first effect called "FLASH" is based on very high dose-rate irradiation (pulse amplitude ≥106 Gy/s), short beam-on times (≤100 ms) and large single doses (≥10 Gy) as experimental parameters established so far to give biological and potential clinical effects. The second effect relies on the use of arrays of minibeams (e.g., 0.5-1 mm, spaced 1-3.5 mm). Both approaches have been shown to protect healthy tissues as an endpoint that must

Ferroptosis Promotes Photodynamic Therapy: Supramolecular Photosensitizer-Inducer Nanodrug for Enhanced Cancer Treatment.

The noninvasive nature of photodynamic therapy (PDT) enables the preservation of organ function in cancer patients. However, PDT is impeded by hypoxia in the tumor microenvironment (TME) caused by high intracellular oxygen (O2) consumption and distorted tumor blood vessels. Therefore, increasing oxygen generation in the TME would be a promising methodology for enhancing PDT. Herein, we proposed a concept of ferroptosis-promoted PDT based on the biochemical characteristics of cellular ferroptosis, which improved the PDT efficacy significantly by producing reactive oxygen species (ROS) and suppl

In vitro and ex vivo immunomodulatory effects of human placental mesenchymal stem cells in hidradenitis suppurativa.

BACKGROUND: Hidradenitis suppurativa is a chronic inflammatory skin disease marked by immune dysregulation and elevated pro-inflammatory cytokines. While biologics like adalimumab target specific pathways, their limited efficacy highlights the need for broader immunomodulatory treatments. Mesenchymal stem/stromal cells (MSCs) have shown promise due to their immunosuppressive properties and ability to modulate both innate and adaptive immunity. This study investigates the effects of naïve (n-MSCs) and cytokine-preactivated (a-MSCs) placental MSCs on the immune responses in HS. METHODS: MSCs we

Evaluation of PET Brain Radioligands for Imaging Pancreatic β-Cell Mass: Potential Utility of 11C-(+)-PHNO.

Type 1 diabetes mellitus (T1DM) is characterized by a loss of β-cells in the islets of Langerhans of the pancreas and subsequent deficient insulin secretion in response to hyperglycemia. Development of an in vivo test to measure β-cell mass (BCM) would greatly enhance the ability to track diabetes therapies. β-cells and neurologic tissues have common cellular receptors and transporters, therefore, we screened brain radioligands for their ability to identify β-cells. Methods: We examined a β-cell gene atlas for endocrine pancreas receptor targets and cross-referenced these targets with brain ra
