BDCD: a comprehensive Brain Disease Cell-cell communication Database.

Dysregulated cell-cell communication (CCC) is increasingly recognized as a driver of brain disease pathology, contributing to neuroinflammation, synaptic dysfunction, and neurodegeneration. Nevertheless, existing resources remain limited in brain specificity, regional coverage, and functional annotation. To address this gap, we develop the Brain Disease Cell-cell communication Database (BDCD), the first comprehensive resource focused on CCC networks across major brain diseases. BDCD integrates 38 manually curated datasets, comprising 8 519 425 single cells from single-cell RNA-seq studies and

Neural regulation of cancer: from microenvironmental hijacking to systemic circuit co-option.

The nervous system is increasingly recognized as an active regulator of tumor biology rather than a passive mediator of cancer-associated symptoms. Growing evidence supports a hierarchical framework of neuro-tumor interactions encompassing chemical, structural, and electrical levels, through which bidirectional communication influences tumor initiation, progression, metastasis, and therapeutic response. At the chemical level, neurotransmitters and neuropeptides regulate tumor cell proliferation, invasion, immune modulation, and stemness. At the structural level, neurogenesis and tumor-nerve in

From Microbial Homeostasis to Systemic Pathogenesis: A Narrative Review on Gut Flora's Role in Neuropsychiatric, Metabolic, and Cancer Disorders.

As a pivotal ecological regulator in humans, the gut microbiota profoundly participates in the pathological processes of neurodegenerative diseases, psychiatric disorders, metabolic syndromes, and cancers through metabolite exchange, epigenetic regulation, and gut-brain axis signaling. This review focuses on analyzing relationships between gut microbial communities and four major disease spectra: neurodegenerative diseases (Alzheimer's disease, Parkinson's disease, Huntington's disease)-revealing microbiota-derived lipopolysaccharide activation of microglia and gut-brain transmission pathways

Multiple subcortical and subcortico-cortico dynamic network reconfigurations characterize focal-to-bilateral tonic-clonic seizures.

Temporal lobe epilepsy (TLE) is the most common focal epilepsy, with focal to bilateral tonic-clonic seizures (FBTCS+) a more severe form of the disorder. Evidence has underscored the critical role of the thalamus, mesial temporal region, basal ganglia and cerebellum, along with the cortex, in the propagation, termination, and modulation of seizure activity. We examined time variant patterns of interaction within and between 7 cortical and 4 subcortical systems in 55 healthy controls and 56 patients with TLE (n = 40 with FBTCS+ and 15 without), isolating those patterns most distinctive of FBTC

Considerable variation in the 95-95-95 targets accomplishment between children and adults might delay achievement of set targets.

Despite a significant reduction in the global HIV disease prevalence in recent years, children under 15 years of age still account for 3% of people living with HIV, 9% of new incidence, and 12% of AIDS-related deaths. Although there is increased access and use of antiretroviral drugs, children under 1 year in resource-poor settings with a high HIV disease burden remain vulnerable due to poor initiation of these critical interventions impeding progress to meet the 95-95-95 targets. There are renewed efforts to ensure that exposed children under 15 years are not left behind by scaling diagnostic

Host Immune Cell Membrane Deformability Governs the Uptake Route of Malaria-Derived Extracellular Vesicles.

The malaria parasite, Plasmodium falciparum, secretes extracellular vesicles (EVs) to facilitate its growth and to communicate with the external microenvironment, primarily targeting the host's immune cells. How parasitic EVs enter specific immune cell types within the highly heterogeneous pool of immune cells remains largely unknown. Using a combination of imaging flow cytometry and advanced fluorescence analysis, we demonstrated that the route of uptake of parasite-derived EVs differs markedly between host T cells and monocytes. T cells, which are components of the adaptive immune system, in

Stable brain PET metabolic networks using a multiple sampling scheme.

Interregional communication within the human brain is essential for maintaining functional integrity. A promising approach for investigating how brain regions communicate relies on the assumption that the brain operates as a complex network. In this context, positron emission tomography (PET) images have been suggested as a valuable source for understanding brain networks. However, such networks are typically assembled through direct computation without accounting for outliers, impacting the reliability of group representative networks. In this study, we used brain [18F]fluoro-2-deoxyglucose P

Microbial quality of beef tripe (mogodu) sampled from butcheries around Gaborone, Botswana.

Beef tripe is a traditional delicacy in Botswana and many African communities. Despite its popularity and nutritional value, tripe remains highly perishable and prone to microbial contamination due to its origin in the bovine gastrointestinal system and potential exposure during processing. This study assessed the microbial quality of beef tripe samples collected from 18 butcheries in Gaborone, Botswana, including both microbial enumeration and species-level identification. The results revealed a high microbial load across all samples, with aerobic plate counts exceeding 5 log colony forming u

Perspectives on tuberculosis in migrants, refugees, and displaced populations in Europe.

Finding and treating all forms of tuberculosis (TB) (latent, drug-susceptible, drug-resistant, multidrug-resistant, and extensively drug-resistant tuberculosis) among migrants, displaced populations, and refugees are important challenges facing TB control programs in Europe. Many of these populations live in poor conditions, with limited access to healthcare and TB services. Ever-increasing armed conflicts in Europe and other parts of the world continue to exacerbate rates of migration to and within Europe, with considerable implications for health services. TB in Europe is more prevalent in m

"We Don't Get Drugs Targeted for Us:" Applying the Integrated Behavioral Model to Understand Why Black Women Chose to Participate in a Breast Cancer Clinical Trial.

Guided by the integrated behavioral model, the authors interviewed 14 Black breast cancer survivors (N = 14) who had participated in a breast cancer clinical trial. This study aimed to better understand what may motivate Black women to engage in medical research and decide to participate in medical research. Findings revealed that Black women's altruistic desires to serve others and their communities are greatly influenced by the need to leave a "legacy" of better treatment for other Black women. The participants mostly learned about clinical trials through communicating with friends, family,

Characteristics and advances in signaling pathways, cellular communication, cell junctions, and oxidative stress in lymphedema.

Lymphedema is a chronic inflammatory disease without an effective treatment method, and it results in a high disease burden and psychological distress in patients. Although there have been significant advances in targeted therapies, there are still no effective options to treat this refractory disease. In recent years, new advances and breakthroughs have been made in signaling pathways, including RAS/MAPK, PI3K/AKT, VEGF-C/VEGFR-3, HGF/MET, and TGF-β1, which are important for understanding the pathogenesis and disease progression of lymphedema. Mutations in genes encoding cell junctions affect

Extracellular Vesicles and Purinergic Signaling in Alzheimer's Disease-Joining Forces for Novel Therapeutic Approach.

Neurodegenerative diseases, including Alzheimer's disease (AD), are a global problem affecting millions of people. Thanks to years of research and huge efforts, it has been possible to discover the pathophysiological changes accompanying Alzheimer's disease at the cellular level. It turns out that the formation of amyloid-beta plaques and hyperphosphorylation of tau protein in the brain play a key role in disease development. Purinergic signaling (PS) is implicated in the pathophysiology of several disorders in the central nervous system, and recent findings link some disturbances in PS with A

Process Regulation of Microbial-Driven Aldehyde Metabolism in Sauce-Flavor Baijiu Fermentation.

Aldehyde compounds are crucial for the flavor profile of Baijiu; however, their metabolic interplay with microbial communities remains inadequately understood. This study demonstrates that the dynamics of aldehydes during Sauce-Flavor Baijiu (SFB) fermentation are primarily driven by stage-specific microbial activities. Based on microbial succession patterns, the fermentation process was divided into early (0-7 days) and late (20-30 days) stages. Seven major aldehydes were identified, with furfural being the dominant component, accounting for over 70% of the total aldehyde content. An integrat
