A Scalable Organoid Model of Urothelial Aging for Metabolic Interrogation, Infection Modeling, and Reversal of Age-Associated Changes.

Aging leads to a progressive decline in overall bladder function resulting in lower urinary tract symptoms and increased susceptibility to infections. However, tissue-specific mechanisms of aging, specifically the contributions of the urothelium, remain elusive. Here, we introduce mouse bladder epithelium-derived organoids (mBEDOs) as a scalable platform to model urothelial aging. mBEDOs from aged mice recapitulate key features of age-associated cellular reprogramming, including oxidative stress, senescence, and DNA damage. We demonstrate the utility of mBEDOs for modeling Uropathogenic Escher

Study on the mechanism of radioactive salivary gland injury: a relatively comprehensive statement.

BACKGROUND: Radiation-induced salivary gland dysfunction (RSGD) is a common complication of radiotherapy in patients with head and neck cancers that leads to xerostomia and related oral complications. It reduces treatment tolerance and quality of life, while current clinical interventions only alleviate superficial symptoms and fail to restore salivary gland function. This highlights the necessity to explore the pathogenesis of RSGD and to identify targets; existing studies have focused on DNA damage, oxidative stress, and fibrosis, but lack insights into emerging mechanisms such as ferroptosi

Chronic Radiation-Induced Wounds: Pathogenesis, Current Therapeutic Strategies, and Emerging Regenerative Approaches.

Radiation-induced chronic wounds remain a major clinical challenge owing to persistent non-healing and progressive tissue degeneration. Rather than simply representing delayed wound repair, they arise from a self-perpetuating pathological cascade involving DNA damage, oxidative stress, chronic inflammation, microvascular dysfunction, cellular senescence, stem cell impairment, and fibrosis. Current clinical management remains largely supportive and symptomatic, with limited efficacy and no standardized therapeutic regimens targeting the core pathological mechanisms. Emerging strategies-antioxid

Modeling Chronic BaP Exposure in Bronchial Epithelial Cells Reveals Multi-Scale Drivers of Early Preneoplastic Reprogramming.

Chronic exposure to benzo[a]pyrene (BaP), a Group 1 IARC carcinogen, is a major driver of lung carcinogenesis; however, how sustained subcytotoxic exposure reprograms bronchial epithelium toward preneoplastic states remains poorly defined. Here, we subjected BEAS-2B human bronchial epithelial cells to 12 weeks of continuous BaP at environmentally relevant concentrations (0.1 and 1.0 µM) and interrogated the resulting phenotypes using an integrated multi-scale framework encompassing functional toxicology, RT-qPCR, RNA-seq, phospho-kinase/NF-κB arrays, and organotypic air-liquid interface (ALI)

Microplastics and nanoplastics: Characterizing potential health perturbations and mechanistic evidence.

Microplastics and nanoplastics (MNPs), as emerging environmental pollutants, have become the focus of global attention due to their widespread presence and potential health risks. Due to their unique physical and chemical properties, MNPs may trigger multiple cellular and molecular toxic responses, including lipid peroxidation, DNA damage, cell membrane rupture, and mitochondrial dysfunction, disrupt the balance of the gut microbiome, and induce chronic inflammation, increasing the risk of major chronic diseases such as cancer and cardiovascular diseases. This paper systematically elaborates o

Sensitizing Colorectal Cancer to PARP Inhibitors: Biomarkers, Mechanisms, and Combination Strategies.

Poly(ADP-ribose) polymerase inhibitors (PARPis) have transformed the treatment landscape of homologous recombination-deficient malignancies, particularly BRCA1/2-mutant breast, ovarian, pancreatic, and prostate cancers. In contrast, clinical studies evaluating PARPis in broadly selected colorectal cancer (CRC) patients have yielded disappointing outcomes despite evidence that a subset of CRCs harbor DNA damage response (DDR) defects and homologous recombination deficiency (HRD)-associated mutational signatures. These observations highlight the need for improved patient stratification and the d

Tannic acid protection against oxidative and inflammatory effects of monosodium glutamate on testis: Molecular and biochemical approach.

In this study, the toxic effects of monosodium glutamate (MSG), a widely used food additive, on rat testicular tissue were evaluated and the potential protective role of tannic acid (TA), a natural polyphenolic compound, against this toxicity was investigated. For this purpose, 24 rats were divided into four groups (n=6). They were administered 2 g/kg monosodium glutamate (MSG) for 21 days. Subsequently, they were treated with 50 mg/kg tannic acid (TA). At the end of this period, testicular tissues were collected for molecular and biochemical analyses. Antioxidant enzyme activities (SOD, CAT,

Comparison of different techniques for γH2AX/pH3 biomarkers quantification for chemical genotoxicity assessment.

Chemical risk assessment relies on in vitro genotoxicity tests. Histone modifications (γH2AX and pH3) have emerged as valuable biomarkers for genotoxicity detection. In this study, we compared three parameters (global intensity, nuclear intensity, and foci number) for the γH2AX biomarker and two parameters (global intensity and % cell in mitosis) for the pH3 biomarker. These analyzes were performed in three cell lines: human osteosarcoma U2OS cells, human hepatocellular carcinoma HepG2 cells and rat intestinal epithelial IEC-6 cells. Cells were exposed for 24 h to four well-characterized hazar

The effects of radiofrequency radiation on male reproductive health and potential mechanisms.

Recent studies have demonstrated that radiofrequency (RF) radiation emanating from devices such as mobile phones and Wi-Fi may have adverse effects on male reproductive health. This radiation can elevate testicular temperature, potentially compromising sperm quality and DNA integrity, and influence the specific absorption rate (SAR) across different body regions, leading to detrimental reproductive outcomes. Furthermore, exposure to RF radiation has been linked to conditions that could affect male reproductive function, such as oxidative stress, alterations in ion transitions across cell membr

cGAS-STING are responsible for premature aging of telomerase-deficient zebrafish.

Telomere shortening occurs in multiple tissues throughout aging. When telomeres become critically short, they trigger DNA-damage responses and p53 stabilization, leading to apoptosis or replicative senescence. In vitro, cells with short telomeres activate the cGAS-STING innate immune pathway resulting in type-I interferon-based inflammation and senescence. However, the consequences of these events for the organism are not yet understood. Here, we show that sting is responsible for premature aging of telomerase-deficient zebrafish. We generated sting-/- tert-/- double-mutant animals and observe

Mapping the non-coding RNA landscape in ataxia telangiectasia: a scoping review of ATM dependent miRNA and lncRNA dysregulation.

Ataxia-Telangiectasia (A-T) is caused by biallelic Ataxia-Telangiectasia Mutated (ATM) protein mutations and is hallmarked by neurodegeneration, immunodeficiency and cancer susceptibility. Emerging work suggests that non-coding RNAs are integral to the ATM driven DNA damage response, yet the scope of non-coding RNA perturbation in A-T has not systematically mapped. This study is done to chart and synthesize all primary evidence on ncRNA dysregulation in A-T and to identify mechanistic gaps and research priorities. The evidence base comprises a total of five studies focused exclusively on micro

Taurine ameliorates radiation-induced oxidative stress in bone marrow mesenchymal stromal cells and promotes osteogenesis.

Osteoradionecrosis of the jaw (ORNJ) is a severe complication following head and neck radiotherapy that significantly impacts the quality of life of patients. Currently, there is a lack of comprehensive understanding of the microenvironmental factors involved in ORNJ. In this study, we reveal the activation of taurine metabolism in irradiated mandibular stromal cells using scRNA-Seq and demonstrate a decrease in taurine levels in irradiated bone marrow mesenchymal stromal cells (BMSCs) through metabolomics. Compared with unirradiated BMSCs, taurine uptake in irradiated BMSCs increases. Taurine

Environmental factors ultraviolet a and ozone exacerbate the repeated inhalation toxicity of 6PPD in mice via accelerating the aging reaction.

The burden of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6PPD) and its oxidized products on human health can no longer be ignored due to the detection types and concentrations in the environment continue to increase. Environmental ozone (O3) and ultraviolet A (UVA) may induce ozonation and photoaging of 6PPD to produce toxic products. However, the impact of specific environmental conditions on the aging and toxic effects of 6PPD is unclear. This study investigated the aging effects of O3 and UVA on 6PPD, and compared the repeated inhalation toxicity of differently aging 6PPDs in C57BL

Exploring the nephrotoxicity and molecular mechanisms of Di-2-ethylhexyl phthalate: A comprehensive review.

Di-2-ethylhexyl phthalate (DEHP), a widely applied plasticizer in various products, can be absorbed into the human body through several channels and accumulate in the lungs, liver, testes, and kidneys, potentially impairing the function of these organs. Recently, the nephrotoxicity of DEHP has received heightened attention. Numerous epidemiologic findings have demonstrated that DEHP exposure may contribute to renal damage, leading to structural and functional abnormalities and exacerbating the progression of kidney disease. Recent research has discovered the mechanisms behind DEHP-induced neph

Assessment of the genotoxicity of tert-butyl alcohol in an in vivo thyroid comet assay.

Chronic exposure to high (20,000 ppm) concentrations of tert-butyl alcohol (TBA) in drinking water, equivalent to ~2100 mg/kg bodyweight per day, is associated with slight increases in the incidence of thyroid follicular cell adenomas and carcinomas in mice, with no other indications of carcinogenicity. In a recent toxicological review of TBA, the U.S. EPA determined that the genotoxic potential of TBA was inconclusive, largely based on non-standard studies such as in vitro comet assays. As such, the potential role of genotoxicity in the mode of action of thyroid tumors and therefore human rel

In vitro genotoxicity potential investigation of 7 oxy-PAHs.

Air pollutants include many compounds among them oxygenated polycyclic aromatic hydrocarbons (oxy-PAHs). As they are suspected to generate DNA damage and mutagenicity, an understanding of their mode of action could highlight a carcinogenic potential risk in exposed population. In this article, a prospective study on seven oxy-PAHs selected in terms of occurrence in the environment was conducted on mutagenicity, genotoxicity, and cytotoxicity potentials using in vitro assays including Ames test on five strains, kinetic analysis of cytotoxicity and apoptosis, phosphorylation of histone H2AX, and
