FishFace: interactive atlas of zebrafish craniofacial development at cellular resolution.

BACKGROUND: The vertebrate craniofacial skeleton may exhibit anatomical complexity and diversity, but its genesis and evolution can be understood through careful dissection of developmental programs at cellular resolution. Resources are lacking that include introductory overviews of skeletal anatomy coupled with descriptions of craniofacial development at cellular resolution. In addition to providing analytical guidelines for other studies, such an atlas would suggest cellular mechanisms underlying development. DESCRIPTION: We present the Fish Face Atlas, an online, 3D-interactive atlas of cr

Molecular regulation of striatal development: a review.

The central nervous system is composed of the brain and the spinal cord. The brain is a complex organ that processes and coordinates activities of the body in bilaterian, higher-order animals. The development of the brain mirrors its complex function as it requires intricate genetic signalling at specific times, and deviations from this can lead to brain malformations such as anencephaly. Research into how the CNS is specified and patterned has been studied extensively in chick, fish, frog, and mice, but findings from the latter will be emphasised here as higher-order mammals show most similar

DNA microarray profiling of developing PS1-deficient mouse brain reveals complex and coregulated expression changes.

Presenilin 1 (PS1) plays a critical role in the nervous system development and PS1 mutations have been associated with familial Alzheimer's disease. PS1-deficient mice exhibit alterations in neural and vascular development and die in late embryogenesis. The present study was aimed at uncovering transcript networks that depend on intact PS1 function in the developing brain. To achieve this, we analyzed the brains of PS1-deficient and control animals at embryonic ages E12.5 and E14.5 using MG_U74Av2 oligonucleotide microarrays by Affymetrix. Based on the microarray data, overall molecular brain

Embryologic and Fetal Development of the Human Lacrimal System.

PURPOSE: To systematically describe the morphogenesis of the lacrimal system and its associated adnexal structures along the developmental timeline, from their earliest identifiable embryonic stages through fetal maturation, and to characterize postnatal anatomical and functional changes. METHODS: The authors conducted an extensive review of classical and contemporary studies of the last 200 years (1837-2025) detailing the morphogenesis of the human lacrimal excretory system-including the puncta, canaliculi, lacrimal sac, and nasolacrimal duct-across embryonic, fetal, and postnatal stages. New classification of spinal lipomas based on embryonic stage.

OBJECTIVE Spinal lipomas are generally thought to occur as a result of failed primary neurulation. However, some clinical features cannot be explained by this theory. The authors propose a novel classification of spinal lipomas based on embryonic changes seen during primary and secondary neurulation. METHODS A total of 677 patients with occult spinal dysraphism underwent 699 surgeries between August 2002 and May 2015 at the National Center for Child Health and Development and Tokyo Metropolitan Children's Medical Center. This group of patients had 378 spinal lipomas, including 119 conus spinal

A rare case of bilateral cystic nephroma associated with embryonal rhabdomyosarcoma of the penile urethra.

Bilateral cystic nephroma is an extremely rare benign renal neoplasm. Here we present a case of bilateral cystic nephroma in a four month old boy who subsequently developed embryonal rhabdomyosarcoma of the urethra. Both tumors were successfully treated. To our knowledge this is the first reported case of this association which could be related to Dicer-1 mutation.

Anastomosis between internal carotid artery and anterior cerebral artery with other anomalies of the circle of Willis in a fetal brain.

A rare vascular anomaly was found in a 21-week-old fetal brain. It consisted of an anastomotic loop between the internal carotid artery and the anterior cerebral artery. Other variations and anomalies were also found. Anomalies and variations of the circle of Willis are probably genetically determined and develop at an early embryonic stage. They are frequently found in association with other cerebrovascular diseases, such as aneurysms.

MicroRNA expression patterns and signalling pathways in the development and progression of childhood solid tumours.

The development of childhood solid tumours is tied to early developmental processes. These tumours may be complex and heterogeneous, and elucidating the aberrant mechanisms that alter the early embryonic environment and lead to disease is essential to our understanding of how these tumours function. MicroRNAs (miRNAs) are vital regulators of gene expression at all stages of development, and their crosstalk via developmental signalling pathways is essential for orchestrating regulatory control in processes such as proliferation, differentiation and apoptosis of cells. Oncogenesis, from aberrant

The increased activity of a transcription factor inhibits autophagy in diabetic embryopathy.

BACKGROUND: Maternal diabetes induces neural tube defects and stimulates the activity of the forkhead box O3 (Fox)O3a in the embryonic neuroepithelium. We previously demonstrated that deleting the FOXO3a gene ameliorates maternal diabetes-induced neural tube defects. Macroautophagy (hereafter referred to as "autophagy") is essential for neurulation. Rescuing autophagy suppressed by maternal diabetes in the developing neuroepithelium inhibits neural tube defect formation in diabetic pregnancy. This evidence suggests a possible link between FoxO3a and impaired autophagy in diabetic embryopathy.

Cyclic AMP-dependent protein kinase in human embryonic palate mesenchymal cells.

Growth and differentiation of cells derived from the embryonic palate are critically dependent on the intracellular cAMP-mediated signal transduction pathway. Human embryonic palate mesenchymal (HEPM) cells have been widely used to examine the effect of teratogens on palatal tissue growth and differentiation, as well as a prescreen for environmental teratogens. This study examined responsiveness of HEPM cells to agents known to stimulate adenylate cyclase, characterized cAMP-dependent protein kinases (cAMP-dPK) (EC 2.7.1.37) and investigated to what extent HEPM cells reveal adaptational respon

Embryonic exposure to environmental levels of roundup affects neurodevelopment in zebrafish.

N-(phosphonomethyl)glycine (glyphosate) is the most sprayed herbicidal chemical in the world. Although glyphosate is considered safe in humans and higher animals due to its targeting of the shikimate pathway found only in plants and microorganisms, recent in vivo and in vitro studies show evidence of toxicity across a range of systems, including the vertebrate brain. Given that developing brains with immature blood-brain barriers can be especially sensitive to environmental contaminants, here we test the effects of embryonic exposure to environmental levels of glyphosate or its commercial form

Clinicopathological Characteristics and Outcomes of Genitourinary Rhabdomyosarcoma in Two Girls.

Rhabdomyosarcoma (RMS) is a soft tissue neoplasm accounting for about 8% of solid tumors in children. There are mainly four histologic subtypes of RMS: embryonal, alveolar, spindle cell/sclerosing, and pleomorphic. The genitourinary tract is the second most affected primary site of RMS. Genitourinary RMS is more common in young boys than girls. Among the subtypes of RMS, embryonal RMS is the primary histologic subtype identified in the bladder and female genitourinary tract of children. We present two cases of genitourinary RMS in two girls, one affecting the bladder/urethra and the other affe

Rescue of Proteus syndrome lethality in mice with prenatal miransertib treatment.

PURPOSE: Proteus syndrome is caused by constitutive AKT1 activation, resulting in severe overgrowth and tumor susceptibility. We developed a mouse model, featuring endogenously regulated, mosaic expression of the Akt1 c.49G>A, p.(Glu17Lys) (Akt1 E17K) variant. Prior results showed that ubiquitous Akt1E17K expression causes embryonic lethality. We hypothesized that a therapeutic effect of a small molecule could rescue the embryonic lethality and serve as a screen for the therapeutic potential of an agent. METHODS: We first tested whether miransertib, an allosteric pan-AKT inhibitor can cross t

The basic helix-loop-helix transcription factor TCF4 recruits the Mediator Complex to activate gonadal genes and drive ovarian development.

The bipotential gonad is the precursor organ to both the ovary and testis and develops as part of the embryonic urogenital system. In mice, gonadogenesis initiates around embryonic day 9.5 (E9.5), when coelomic epithelial (CE) cells overlaying the mesonephric ducts proliferate and acquire competence to differentiate into the somatic cell types of the embryonic gonad, the pre-supporting cell (Sertoli cells in the testis and granulosa cells in the ovary) and the interstitial cell lineages. While some transcription factors that drive gonadal cell fate are known, we found that basic helix-loop-hel

Embryonic thermal challenge is associated with increased stressor resiliency later in life: Molecular and morphological mechanisms in the small intestine.

Developing chick embryos that are subjected to increased incubation temperature are more stressor-resilient later in life, but the underlying process is poorly understood. The potential mechanism may involve changes in small intestine function. In this study, we determined behavioral, morphological, and molecular effects of increased embryonic incubation temperatures and post-hatch heat challenge in order to understand how embryonic heat conditioning (EHC) affects gut function. At 4 days post-hatch, duodenum, jejunum, and ileum samples were collected at 0, 2, and 12 h relative to the start of

Microenvironment derived from metanephros transplantation inhibits the progression of acute kidney injury in glycerol-induced rat models.

Background: Embryonic metanephros is the mammalian renal anlagen, which is considered as a potential source for the regeneration of functional whole kidneys. Some studies reported that metanephros implanted into unilateral nephrectomized animals can develop into kidney tissue. However, kidneys are nephrotoxic in renal failure patients, and whether metanephros can grow in nephrotoxic has not been reported. This study aims to investigate the growth of metanephros in acute nephrotoxic environment and analyze the therapeutic effect of metanephros microenvironment on acute kidney injury (AKI).Metho

The proboscipedia locus of the Antennapedia complex: a molecular and genetic analysis.

The homeotic proboscipedia (pb) locus of the Antennapedia Complex (ANT-C) directs the differentiation of adult labial and maxillary structures. Loss-of-function pb alleles show a transformation of adult mouth parts to legs and affect maxillary palp morphology. We have identified the pb transcription unit by inducing and analyzing a series of pb null chromosomal breakpoints. In addition, we describe a variegating pb phenotype associated with two rearrangement breakpoints. Having identified the pb locus, we describe the expression of its RNA and protein products. Unlike the other homeotic genes
