A novel therapeutic agent in the form of beta galactosidase immobilized on the surface of concanavalin A layered calcium alginate-starch beads has been developed. Immobilized beta galactosidase exhibited significantly very high stability against conditions of digestive system such as pH, salivary amylase, pepsin and trypsin. Soluble and immobilized beta galactosidase exhibited same pH-optima. However, the immobilized enzyme retained greater fraction of catalytic activity at higher and lower pH to pH-optima as compared to soluble enzyme. Immobilized enzyme preparation was quite stable under con

The phototrophic bacterium Rhodobacter capsulatus E1F1 photoreduced 2,4-dinitrophenol to 2-amino-4-nitrophenol by a nitrophenol reductase activity which was induced in the presence of nitrophenols and was repressed in ammonium-grown cells. The enzyme was located in the cytosol, required NAD(P)H as an electron donor, and used several nitrophenol derivatives as alternative substrates. The nitrophenol reductase was purified to electrophoretic homogeneity by a simple method. The enzyme was composed of two 27-kDa subunits, was inhibited by metal chelators, mercurial compounds, and Cu2+, and contain

Bile salt sulphation in primary biliary cirrhosis was studied by measurements of the liver bile salt sulphotransferase levels in 16 patients. Although the enzyme activity varied among the patients it did not correlate with the severity of cholestasis. Furthermore, the mean bile salt sulphotransferase magnitude in patients with primary biliary cirrhosis did not differ significantly from corresponding enzyme activity in patients with non-cholestatic, alcohol induced liver disease. The present data indicates that chronic cholestasis, as evidenced in patients with primary biliary cirrhosis, does n

The authors describe two cases of clinical Fabry's disease. The first patient presents a deficiency of alpha galactosidase and a urinary excretion of ceramide trihexosides and dihexosides ; the second patient had a normal alpha galactosidase and normal excretion of urinary lipids. In this latter case the Km and the activity of the enzyme measured at different pH were similar to those of normal enzyme. The other lysosomal enzymes, beta galactosidase, beta glucosidase, hexosaminidases A and B, alpha fucosidase, arylsulfatases, phosphatase acids were also measured in patient 2 and all have normal

Arylsulfatase A was isolated from urine and human liver. The enzyme was homogeneous with respect to charge and had high specific activity--64 U/mg and 34 U/mg for arylsulfatase A from urine and liver respectively. The enzyme from urine as well as the liver one contained two nonidentical subunits with molecular weights varying about 5 kDa. Treatment of the enzyme from urine, liver and from placenta with endo-beta-N-acetylglucosaminidase F did not remove all carbohydrate from any subunit even in denaturing conditions. Deglycosylation of the enzyme with this one and other glycosidases under vario

Previous investigations suggested that a mechanism independent of cAMP may be associated with the action of some retinoids. An alternative pathway involving calcium, phospholipid-dependent protein kinase (C-kinase), was therefore studied. In order to demonstrate this, C-kinase was partially purified from skin of hairless, Balb/c normal and Balb/c nude mouse. Interaction and effects of various response modifiers such as phospholipids, retinoids and phorbol ester tumor promoters showed both major and minor differences among these enzymes. In general, retinal, retinoic acid, 13-cis-retinoic acid

Background: Small-cell lung cancer (SCLC) has a poor prognosis because of the development of drug resistance. Our previous studies showed that the plant steroid hormone, epibrassinolide (EB), interacts with human SCLC cells through inhibition of GSK3β-mediated signaling and overcomes drug resistance. However, the latter does not occur via membrane-mediated drug efflux, a commonly reported mechanism. Methods: We investigated a novel mechanism, ferroptosis, for drug resistance and the effects of EB. Pharmacological, cellular and molecular investigations were performed. We used drug-sensitive (NC

Inflammation and immune activation are crucially involved in the pathogenesis of atherosclerosis and cardiovascular disease. Accordingly, markers of inflammation such as fibrinogen, ferritin, C-reactive protein or neopterin are found in patients with vascular diseases, correlating strongly with the extent of disease and predicting disease progression. Neopterin formation by human monocyte-derived macrophages and dendritic cells is induced by the pro-inflammatory cytokine interferon-gamma, which is released by activated T-lymphocytes. Human macrophages are centrally involved in plaque formation

Metals are frequently exploited in the active sites of enzymes; however, the involvement of rare earth elements (REEs) in biocatalysis has been less commonly observed. Here, we show that the alcohol dehydrogenase D (AdhD) from Pyrococcus furiosus can bind REE ions, and the forward activity (alcohol oxidation) is accelerated by the addition of REEs, while, unexpectedly, the reverse reaction is inhibited. In the companion paper, we discovered a REE binding site in the AdhD structure that is distal to the active site. Here, we identified a second REE binding interaction that is only observed in t

The demand for d-amino acids is increasing as their unique physiological role continues to be elucidated. meso-Diaminopimelate dehydrogenase catalyzes the reversible NADP+-dependent oxidative deamination of meso-diaminopimelate to produce l-2-amino-6-oxopimelate. Moreover, a few enzymes show a broad range of reductive amination activity toward 2-oxo acids, synthesizing d-amino acids. Here, we report the identification and characterization of a novel NADP+-dependent meso-diaminopimelate dehydrogenase from Candidatus Syntrophocurvum alkaliphilum (CSaDAPDH). The purified enzyme exhibited oxidativ

Safflower, a traditional Chinese medicine, is renowned for its efficacy in promoting blood circulation and alleviating blood stasis. Its principal bioactive components are flavonoids, which predominantly exist as flavonoid glycosides. Glycosyltransferases, as downstream post-modification enzymes in the biosynthesis of these active glycosides, are of considerable research interest. This study leverages transcriptome data from safflower to identify a glycosyltransferase gene, UGT95A2, which was subjected to comprehensive bioinformatics and enzymatic property analyses. In vitro enzymatic assays d

Vitamin D hydroxylation in the liver/kidney results in conversion to its physiologically active form of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3]. 1,25(OH)2D3 controls gene expression through the nuclear vitamin D receptor (VDR) mainly expressed in intestinal epithelial cells. Cytochrome P450 (CYP) 24A1 is a catabolic enzyme expressed in the kidneys. Interestingly, a recently identified mutation in another CYP enzyme, CYP3A4 (gain-of-function), caused type III vitamin D-dependent rickets. CYP3A are also expressed in the intestine, but their hydroxylation activities towards vitamin D substrates ar

Nitrite and nitric oxide (NO), two active and critical nitrogen oxides linking nitrate to dinitrogen gas in the broad nitrogen biogeochemical cycle, are capable of interacting with redox-sensitive proteins. The interactions of both with heme-copper oxidases (HCOs) serve as the foundation not only for the enzymatic interconversion of nitrogen oxides but also for the inhibitory activity. From extensive studies, we now know that NO interacts with HCOs in a rapid and reversible manner, either competing with oxygen or not. During interconversion, a partially reduced heme/copper center reduces the n

Many drugs, which are used in clinical treatments, show pharmacological effects on enzyme activity with an essential role in the pathogenesis of various diseases. Paraoxonase-I (PON1) is a member of the mammalian lactonase enzyme family, serves to the prevention of blood vessel plaque formation by protecting high-density lipoprotein and low-density lipoprotein against oxidation. In the current study, we aimed to contribute to drug discovery and to determine the interaction of some calcium channel blockers (CCBs) with PON1. For this purpose, first, the PON1 enzyme was purified from fresh human

Cocaine addiction is a severe psychiatric condition for which currently no effective pharmacotherapy is available. Brain mechanisms for the establishment of addiction-related behaviors are still not fully understood, and specific biomarkers for cocaine use are not available. Sphingolipids are major membrane lipids, which shape neuronal membrane composition and dynamics in the brain. Here, we investigated how chronic cocaine exposure during establishment of addiction-related behaviors affects the activity of the sphingolipid rheostat controlling enzymes in the brain of rats. As we detected spec

In this study, an attempt has been made to evaluate the effect of products of β-galactosidase (βGS) catalyzed reaction i.e. glucose and galactose and their structurally related compound vitamin C (VC) on the catalytic activity of native and PANI-CS-NC and PANI-CS-Ag-NC adsorbed βGS. Results indicated a decline in catalytic activity of soluble enzyme in the presence of all investigated compounds. The order of inhibition was found to be VC < glucose < galactose. However, the immobilized preparations were found more resistant to inactivation caused by the added compounds. About 48% activity was r

The protective effect of the extracts of the plants Silybum marianum and Taraxacum officinale by carbon tetrachloride (CCl4) was researched. Sixty-six female Wistar albino rats were divided into six groups: Control, Silybum marianum, Taraxacum officinale, CCl4, Silybum marianum+ CCl4, Taraxacum officinale+CCl4. The Silybum marianum and Taraxacum officinale extracts were administered as 100 mg/kg/day by gavage. The CCl4 was administered as 1.5 mL/kg (i.p.). At the end of the trial period, in the serums obtained from the animals, in the CCl4 group it was found that the MDA level increased in the

Activity of the aminoglycoside phosphotransferase APH(3')-Ia leads to resistance to aminoglycoside antibiotics in pathogenic Gram-negative bacteria, and contributes to the clinical obsolescence of this class of antibiotics. One strategy to rescue compromised antibiotics such as aminoglycosides is targeting the enzymes that confer resistance with small molecules. We demonstrated previously that ePK (eukaryotic protein kinase) inhibitors could inhibit APH enzymes, owing to the structural similarity between these two enzyme families. However, limited structural information of enzyme-inhibitor com

The biosynthesis of nonribosomally formed peptides (NRPs), which include important antibiotics such as vancomycin, requires the activation of amino acids through adenylate formation. The biosynthetic gene clusters of NRPs frequently contain genes for small, so-called MbtH-like proteins. Recently, it was discovered that these MbtH-like proteins are required for some of the adenylation reactions in NRP biosynthesis, but the mechanism of their interaction with the adenylating enzymes has remained unknown. In this study, we determined the structure of SlgN1, a 3-methylaspartate-adenylating enzyme
