BACKGROUND: Intestinal epithelial barrier dysfunction is intricately linked to the pathogenesis of ulcerative colitis (UC). Dietary interventions that bolster intestinal epithelial barrier function can effectively thwart UC onset. Our prior research revealed that p-Hydroxy benzaldehyde (HD), a phenolic compound from Nostoc commune (an edible cyanobacterium), markedly upregulated the expression of E-cadherin, a pivotal protein in intestinal mucosa, thereby mitigating mucosal damage in mice afflicted with dextran sulfate sodium (DSS)-induced colitis. Nevertheless, the precise molecular mechanism

Diabetic foot ulcer (DFU), which is characterised by damage to minute blood vessels or capillaries around wounds, is one of the most serious and dreaded complications of diabetes. It is challenging to repair chronic non-healing DFU wounds. Vascular endothelial growth factor (VEGF) plays an important role in angiogenesis and promotes wound healing in DFU. However, it is difficult to sustainably deliver VEGF to the wound site owing to its poor stability and easy degradation. To overcome this challenge, lipid nanoparticles (LNP) encapsulating circular RNA (circRNA) encoding VEGF-A have been devel

The third pandemic of coronavirus infection, called COVID-19 disease, was first detected in November 2019th. Various determinants of disease progression such as age, sex, virus mutations, comorbidity, lifestyle, host immune response, and genetic background variation have caused clinical variability of COVID-19. The causative agent of COVID-19 is an enveloped coronavirus named severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) that invades host cells using an endocytic pathway. The SARS-CoV-2 spike protein is the main viral protein that contributes to the fusion of the virus particle

BACKGROUND: One of the most fascinating discoveries in biology in recent years is unquestionably the identification of the family of small, noncoding RNAs known as microRNAs (miRNAs). Each miRNA targets multiple mRNA species through recognition of complementary sequences, typically located at multiple sites within the 3 untranslated region. In animals, single-stranded miRNA binds specific messenger RNA (mRNA) by a mechanism that is yet to be fully characterized. The bound mRNA remains untranslated resulting in reduced levels of the corresponding protein; however, if the sequence match between

Sustained postoperative anti-inflammatory therapy following intraocular surgery remains an unmet clinical need, as conventional topical corticosteroid eye drops suffer from poor bioavailability, patient non-compliance, and an inability to maintain therapeutic drug levels over the multi-week inflammatory resolution period. Here we report the development and characterization of alkaline phosphatase (ALP)-responsive enzyme-instructed self-assembling (EISA) peptide hydrogels based on the ultrashort amphiphilic sequence NapFFKY(p)-OH, covalently conjugated with dexamethasone (DEX) via an ester-clea

Research in functional foods faces two major challenges due to the complexity of natural food components: accurate and rapid identification of bioactive compounds, and their precise extraction. To address these challenges, this study developed an integrated strategy to identify and isolate acetylcholinesterase inhibitors (AChEIs) from Gardenia jasminoides Ellis (GJE)-a plant with both medicinal and edible value-while evaluating their potential against Alzheimer's disease (AD). First, affinity ultrafiltration-liquid chromatography-mass spectrometry (AUF-LC-MS) (a technique that uses AChE as a t

Purpose: The increase in butyrylcholinesterase (BChE) activity in the brain of Alzheimer disease (AD) patients and animal models of AD position this enzyme as a potential biomarker of the disease. However, the information on the ability of BChE to serve as AD biomarker is contradicting, also due to scarce longitudinal studies of BChE activity abundance. Here, we report 11C-labeling, in vivo stability, biodistribution, and longitudinal study on BChE abundance in the brains of control and 5xFAD (AD model) animals, using a potent BChE selective inhibitor, [11C]4, and positron emission tomography

RATIONALE: Activation of p38 mitogen-activated protein kinase (MAPK) has a significant impact on cardiac gene expression, contractility, extracellular matrix remodeling, and inflammatory response in heart. The p38 kinase pathway also has a controversial role in cardiac hypertrophy. MAPK-activated protein kinase-2 (MK2) is a well-established p38 downstream kinase, yet its contribution to p38-mediated pathological response in heart has not been investigated.
OBJECTIVE: We examined the specific contribution of MK2 to the pathological remodeling induced by p38.
METHODS AND RESULTS: We used a car

ETHNOPHARMACOLOGY RELEVANCE: Alhagi honey is a light yellow sugar granule formed by concentrating the liquid secreted by Alhagi branches and leaves. It is a traditional Uygur medicine often used to treat abdominal pain, diarrhea, dysentery, and other conditions. Modern research has indicated that the main active components of Alhagi honey are oligosaccharides and polysaccharides. Our previous research had identified that the extract of Alhagi honey exhibits good anti-inflammatory pharmacological activity, however, its efficacy against Crohn's disease (CD) remains to be elucidated.

Pseudomonas aeruginosa has been implicated in a wide range of post-operation wound and lung infections. A wide range of acquired resistance and virulence markers indicate surviving strategy of P. aeruginosa. Complete-genome analysis has been identified as efficient approach towards understanding the pathogenicity of this organism. This study was designed to sequence the entire genome of P. aeruginosa UY1PSABAL and UY1PSABAL2; determine drug-resistance profiles and virulence factors of the isolates; assess factors that contribute toward stability of the genomes; and thereafter determine evoluti

PURPOSE: Exocrine pancreatic insufficiency (EPI) is a highly prevalent, yet modifiable determinant of treatment tolerance in pancreatic cancer (PC). Despite its clinical significance, management remains inconsistent due to a lack of oncology-specific guidelines. This project aimed to establish the first U.S. multidisciplinary consensus to standardize pancreatic enzyme replacement therapy (PERT) dosing, education, and longitudinal management in pancreatic oncology.
METHODS: A mixed-methods design was employed, integrating a parallel literature review, six virtual focus groups with patients and

SARS-CoV-2 infection causes diverse clinical outcomes, with men facing higher risks of severe disease. Adenosine-to-inosine (A-to-I) RNA editing mediated by ADAR enzymes governs innate antiviral immunity, yet its contribution to COVID-19 severity and sex-biased disease remains unclear. We performed epitranscriptomic profiling on 126 blood samples from non-COVID controls and COVID-19 patients spanning different disease severities. We detected 1,466 differentially RNA-edited sites in 514 genes linked to clinical outcomes. Although ADAR expression increased in COVID-19 patients overall, it was st

Chloramphenicol resistance in Riemerella anatipestifer is primarily mediated by chloramphenicol acetyltransferases (CATs). Inconsistent nomenclature of the CatB superfamily in public databases currently complicates the genomic surveillance of these resistance determinants. In this study, we tested the subtype scheme recorded in the NCBI Reference Gene Catalog against a complete pairwise amino acid identity matrix, applying a ≥ 80% identity criterion. The analysis supports the existing type B-1 to type B-5 subtypes, assigns two previously unassigned sequences to Type B-2, and recommends retirem

Microbial pesticides offer a sustainable alternative for crop protection due to their target specificity, low resistance risk, and environmental compatibility. However, their practical application is hindered by poor environmental tolerance to ultraviolet radiation, high temperature, and desiccation, as well as short shelf life and inadequate foliar retention. Traditional formulations such as wettable powders and suspension concentrates often provide limited protection or controlled release. Biomimetic materials inspired by natural structures including biofilms, plant cell walls, and mussel ad

BACKGROUND: The most common cause of endogenous hyperinsulinemic hypoglycemia in neonates [congenital hyperinsulinemic hypoglycemia (CHH)] is different monogenic forms of gene mutations. About 50% of the mutations are known. We present a new mutation within the short-chain L-3-hydroxyacyl-CoA dehydrogenase (HADH) gene causing CHH in 2 related patients.
METHODS: The course of 2 consanguineous patients with CHH are presented with a follow-up of >30 years. NextSeq 500 sequencing was performed and confined on known genes with autosomal recessive inheritance, namely ABCC8 (MANE Select: NM_00352.6)

Hepatocellular carcinoma (HCC) is sustained by coordinated interactions among malignant hepatocytes, immune cells, and stromal populations that collectively drive tumor growth, immune evasion, and vascular remodeling. Using integrative single-cell transcriptomics on 93,032 cells from tumor and healthy human liver, we characterized cell-type-specific transcriptional programs underlying immunometabolic reprogramming and reconstructed the intercellular communication circuits that maintain the tumor microenvironment. Malignant hepatocytes displayed upregulation of genes encoding both glycolytic an

Halogens are utilized in drug discovery and development due to their ability to precisely tune the molecular properties of drug candidates. The halogen substituents play a crucial role in determining the stability, lipophilicity, and receptor binding, as well as significantly influencing the metabolic fate of the drug candidates. Literature insights reveal that 30% of all small molecules approved by the USFDA carry a halogen as a substituent. Considering the vital importance of halogen-containing substituents, we have compiled concise information on halogen-containing US FDA-approved drugs ove

This study aims to investigate the molecular mechanism by which sulfurized garlic (SAC) inhibits ferroptosis through targeting Keap1/Nrf2/GPX4 signaling pathway, thereby restoring intestinal epithelial cell barrier function and alleviating ulcerative colitis (UC). Oxidative stress was induced in cells using H₂O₂ and detected by DCFH-DA. CETSA and Western blotting were employed to assess SAC-target protein binding and antioxidant capacity using DPPH and total antioxidant assays. In vivo, C57BL/6 mice were divided into control, DSS, and SAC groups. DAI was scored based on weight loss, stool cons

Pomegranate and carob extracts have been characterized as potential bioprotectors against Verticillium wilt of olive caused by Verticillium dahliae, suggesting they may act as host resistance inducers. This study aimed to elucidate whether these extracts activate metabolic pathways involved in olive defense against V. dahliae infection through marker gene expression analysis. The relative expression of seven resistance-related marker genes was evaluated by RT-qPCR in noninoculated and treated olive plants. Subsequently, the effect of the extracts as inducers of resistance against Verticillium
