Do GM1 antibodies induce demyelination?

We review clinical, neurophysiological, immunological, and experimental data concerning multifocal motor neuropathy (MMN), a newly recognized disorder that mimics MND. It is separated from MND by the presence of multifocal conduction block (CB) demonstrated electrophysiologically, and in some instances by the association of high titers of GM1 antibodies. The possible immunopathogenetic effect of GM1 antibodies is discussed. However, 70% of patients with MMNCB do not have elevated titers of GM1 antibodies, but may respond nevertheless to immunosuppressive treatment. Thus, so far unrecognized an

Mutations in the gamma 2 chain gene (LAMC2) of kalinin/laminin 5 in the junctional forms of epidermolysis bullosa.

Junctional epidermolysis bullosa (JEB) is an autosomal recessive disorder characterized by blister formation within the dermal-epidermal basement membrane. Genes for the lamina lucida protein, kalinin/laminin 5, have been proposed as candidates for some forms of JEB, based on immunofluorescence analysis recognizing kalinin epitopes. We studied the cDNA of laminin gamma 2 chain for mutations in JEB using heteroduplex analysis. One patient showed a homozygous splice site mutation while another was heterozygous for a deletion-insertion, resulting in a premature termination codon in one allele. Ou

[The antigenic structure of glycoprotein E2 in the Venezuelan equine encephalomyelitis virus studied using rat monoclonal antibodies].

A collection of 21 rat hybridomas secreting high-affinity monoclonal antibodies to Venezuelan equine encephalomyelitis (VEE) virus was generated. Using a panel of 15 monoclonal antibodies to glycoprotein E2, the antigenic structure of this protein of VEE strains TC-83 and 230 was studied. A competitive radioimmunoassay suggested a new map of the antigenic structure of glycoprotein E2 in which 5 sites including 11 epitopes of monoclonal antibody binding were distinguished. Antibody to E2-2 site neutralized virus infectivity and blocked hemagglutination test and antibody to E2-3 site could only

Vaccination for disease.

Recombinant virus vaccines that express a limited number of epitopes are currently being developed to prevent disease by changing the relative balance between viral spread and the immune response. Some circumstances, however, were found in infections with a noncytopathic virus in which vaccination caused disease; sensitive parameters included the genetic background of the host, the time or dose of infection, and the constituents of the vaccine. Thus, immunopathologic damage by T cells may be an unwanted consequence of vaccination with the new types of peptide or recombinant vaccines that are b

The prolamin antibody reactivity against hordein polypeptides in sera from patients with coeliac disease.

The antibody reactivity against the barley prolamin, hordein, was investigated by an immunoblotting technique, in sera from patients with untreated coeliac disease, patients with other gastrointestinal diseases and healthy controls. No characteristic hordein polypeptide antibody pattern could be connected to coeliac disease, as observed in a similar study using different fractions of the wheat prolamin, gliadin. Gliadin- and hordein-immunoadsorbent column experiments demonstrated that the prolamin reactivity originates from the same population of antibodies. It is speculated that distinct anti

Reaction pattern of mitochondrial antibodies of primary biliary cirrhosis (PBC) is species specific but not organ specific.

The cross reactivity of a well-characterized PBC serum was studied with mitochondria from a number of sources. These studies were to establish the nonorgan, species specificity of the reaction of PBC sera. As well as confirming some previously reported data, we have strong evidence suggesting that, in spite of species differences in Mr of the major antigenic bands, all mitochondria contain a set of common cross-reactive epitopes. The multiplicity of antigenic bands seen for mitochondria from some sources are shown to arise, in part, as proteolytically derived artefacts of bands of higher Mr, r

Production of murine monoclonal antibodies to the major axial filament polypeptide of Treponema pallidum.

A suitable immunization protocol for the stimulation of a murine antibody response to the axial filament polypeptides of Treponema pallidum was established. A range of monoclonal antibodies (Mabs) specific for different epitopes of the major axial filament polypeptide (37 kDa) was generated which demonstrated diversity in their ability to react with other treponemal species. Immunogold electron microscopy located the 37 kDa antigen on the surface of the axial filament structure. The early appearance of specific antibody to this polypeptide in infected man and rabbit indicates that such Mabs ar

Polymyositis and molecular mimicry, a mechanism of autoimmunity.

The amino acid sequences of Escherichia coli histidyl-tRNA synthetase and alanyl-tRNA synthetase, two proteins recently identified as autoantigens in polymyositis, were compared by a computer alignment procedure with those of the 3600 proteins tabulated in the National Biomedical Research Foundation protein sequence database. Both proteins contain sequences long enough to function as epitopes that match sequences on viral and muscle proteins. The homology thus revealed not only lends strong support to mechanisms of autoimmunity that invoke the theory of molecular mimicry of viral proteins, but

Isolation of dinitrophenyl-skin protein conjugates by immunoadsorbent chromatography.

Immunoadsorbent columns charged with rabbit anti-dinitrophenyl (DNP) antibody were used to isolate and concentrate DNP-protein conjugates from the 105,000 g supernate of extracts prepared from guinea pig epidermis previously painted with dinitrofluorobenzene (DNFB). Conjugates isolated in this manner elicited positive delayed hypersensitivity skin tests when injected intradermally into DNFB-sensitized animals. Dinitrophenylated epidermal protein conjugates prepared in vitro were capable of specifically inhibiting the migration of peritoneal exudate cells obtained from DNFB-sensitized guinea pi

Predictive markers of SARS-CoV-2 spike-specific cytotoxic T cell activity following Omicron breakthrough infection.

At a population level, evaluation of vaccine efficacy and of immunity in general focuses on measuring humoral immunity, especially antibody-mediated neutralization. However, antibody-dependent cell-mediated cytotoxicity (ADCC) and cellular immunity are key processes for pathogen clearance and often provide broader protection against emerging variants. More information on vaccine and infection-related determinants or markers of cell-mediated cytotoxicity is needed to inform design of standardized, high throughput tests assessing these processes. We previously showed that persons infected with S

Immunoinformatics-based design of artificial chimeric proteins as universal vaccine candidates against foot-and-mouth disease virus serotypes A, O, and SAT2.

Foot-and-mouth disease virus (FMDV) remains a major constraint to livestock health due to its high mutation rate and serotype diversity. Currently, FMDV vaccines, primarily inactivated whole-virus formulations, have significant limitations, including limited cross-protection, high production costs, and potential biosafety risks. To address the need for broad-spectrum protection, this study aimed to design a universal vaccine candidate by rationally constructing artificial chimeric proteins (ACPs) integrating conserved structural (VP1-VP3) and non-structural (3 A, 3 C) proteins from the predomi

Unveiling a putative class 3 L-asparaginase in Glutamicibacter sp.: from soil isolation to in silico characterization.

BACKGROUND: Type II L-asparaginases are important therapeutic agents for treating childhood leukemia, but numerous side effects of these enzymes, besides neutralization by antibodies due to repeated injection, prompt extensive research to identify alternatives in other classes of this enzyme. RESULTS: Among the six asparaginase-producing strains, the best-producing isolate was identified as Glutamicibacter. The asparaginase and glutaminase activities in culture supernatant were 15 U/ml and 1.5 U/ml, respectively. The full-length asparaginase gene was sequenced, and the phylogenetic tree revea

Evaluation of the contribution of the minor envelope complex glycoprotein 3 ectodomain to the porcine reproductive and respiratory syndrome virus 1 neutralizing antibody response.

Porcine reproductive and respiratory syndrome viruses (PRRSVs) cause significant economic losses to the global swine industry. Current live attenuated vaccines are unable to effectively control PRRSV infection, and strain variability, driven by rapid mutation, complicates the development of improved vaccines. Identification of the targets of broadly neutralizing antibodies is crucial for guiding next-generation vaccine design, yet PRRSV glycoproteins bearing conserved neutralizing epitopes remain poorly defined. To address this, recombinant soluble forms of PRRSV-1 envelope glycoproteins, GP2,

Integrative bioinformatics approach to the mRNA vaccine development for the human papillomavirus using the capsid protein.

Human papillomavirus (HPV) is associated with several serious diseases, consisting of neck-related, lung, oropharyngeal, vaginal, penile, vulvar, and anal tumors. Potential candidates for mRNA vaccination include the capsid structural peptides L1 and L2, which are prominently recognized by the immune system. This recognition has resulted in the formation of mRNA vaccines targeting these capsid proteins. We employed various bioinformatics methods to predict epitopes for helper CD4 + lymphocytes, CD8 + lymphocytes, and B cells within these proteins, and to evaluate their allergenicity, toxicity,

Digestion supplemented with commercial proteases: Evaluation of the fate of gluten immunogenic peptides in pizza.

Enzymatic supplements designed to aid in gluten digestion can be found in the market, also in combination with enzymes targeting other macronutrients. Their impact on digestion of gluten sequences that are immunogenic and/or toxic for individuals with Celiac Disease (CeD), remains uncertain, especially within a complex food matrix. This study aims at applying a biochemical and immune-based approach for understanding the effect of such supplements on the degradation of gluten immunogenic peptides in pizza. Plane-pizza was digested using the INFOGEST model, with/without the addition of three ove

Interpretating SPR-Derived Reaction Kinetics via Self-Organizing Maps for Diagnostic Applications.

Biosensors emerge as promising, cost-effective infectious disease diagnostics in resource-limited settings, requiring neither laboratory infrastructure nor specialized personnel. Surface plasmon resonance (SPR)-based biosensors remain preeminent for label-free, real-time analysis of biological interactions and kinetic parameter determination. Integrating Artificial Intelligence (AI), particularly self-organizing maps (SOMs), automates infection screening by projecting high-dimensional data onto topology-preserving 2D maps, offering advantages in diagnostic strategies by enabling efficient clas

Robust polyfunctional CD8+ and CD4+ T cell responses in HLA-A*0201/DR1 transgenic mice following vaccination with modified vaccinia virus Ankara-based vaccines delivering Lassa virus glycoprotein or nucleoprotein.

Lassa virus (LASV) is circulating in rodents in several countries in West Africa and is the causative agent of the zoonotic disease Lassa fever. Several vaccine candidates have been successfully tested in preclinical and clinical research, while no LASV-specific vaccines or antiviral treatments have been licensed to date. Approximately 500,000 human cases of Lassa fever are estimated to occur every year. However, the high percentage (~80%) of asymptomatic cases and the low frequency of reporting systems in endemic regions demonstrate that Lassa fever cases are highly underreported. Given the f

Analysis of Wilms' tumor protein 1 specific TCR repertoire in AML patients uncovers higher diversity in patients in remission than in relapsed.

The Wilms' tumor protein 1 (WT1) is a well-known and prioritized tumor-associated antigen expressed in numerous solid and blood tumors. Its abundance and immunogenicity have led to the development of different WT1-specific immune therapies. The driving player in these therapies, the WT1-specific T-cell receptor (TCR) repertoire, has received much less attention. Importantly, T cells with high affinity against the WT1 self-antigen are normally eliminated after negative selection in the thymus and are thus rare in peripheral blood. Here, we developed computational models for the robust and fast

Tau maturation in the clinicopathological spectrum of Lewy body and Alzheimer's disease.

OBJECTIVE: Alzheimer's disease neuropathologic change and alpha-synucleinopathy commonly co-exist and contribute to the clinical heterogeneity of dementia. Here, we examined tau epitopes marking various stages of tangle maturation to test the hypotheses that tau maturation is more strongly associated with beta-amyloid compared to alpha-synuclein, and within the context of mixed pathology, mature tau is linked to Alzheimer's disease clinical phenotype and negatively associated with Lewy body dementia. METHODS: We used digital histology to measure percent area-occupied by pathology in cortical

Induction of islet autoimmunity to defective ribosomal product of the insulin gene as neoantigen after anti-cancer immunotherapy leading to autoimmune diabetes.

INTRODUCTION: The autoimmune response in type 1 diabetes (T1D), in which the beta cells expressing aberrant or modified proteins are killed, resembles an effective antitumor response. Defective ribosomal protein products in tumors are targets of the anti-tumor immune response that is unleashed by immune checkpoint inhibitor (ICI) treatment in cancer patients. We recently described a defective ribosomal product of the insulin gene (INS-DRiP) that is expressed in stressed beta cells and targeted by diabetogenic T cells. T1D patient-derived INS-DRiP specific T cells can kill beta cells and are pr
