Socio-economic indicators, housing characteristics, site of residence, and substance use as household-level risk factors in GRT-INSPIRED communities.

INTRODUCTION: Substance use remains a major public health concern in South Africa and disproportionately affects urban communities experiencing socio-economic disadvantage, poor housing conditions, and spatial inequality. While existing research has largely focused on individual-level substance use, less is known about how household socio-economic characteristics, housing conditions, and place of residence shape household-level substance use in urban settings. This study examined the associations between socio-economic indicators, housing characteristics, site of residence, and household subst

"The weight of words": a phenomenological study of Chinese neonatologists' lived experiences of delivering bad news.

BACKGROUND: Delivering bad news in the Neonatal Intensive Care Unit (NICU) constitutes a profound ethical and emotional challenge. In China, this complexity is intensified by a defensive medical culture and strained physician-family dynamics. This study explores the lived experiences of Chinese neonatologists navigating these high-stakes interactions to uncover the phenomenon's essential structure. METHODS: A descriptive phenomenological study was conducted. Using a purposive sampling strategy, 19 neonatologists from a tertiary NICU in Western China were recruited. Data were collected through

Isoalantolactone targets NLRP3 to disrupt NLRP3-NEK7 interaction and suppress inflammasome activation.

NLRP3 inflammasome plays a critical role in innate immunity and has been implicated in the pathogenesis of multiple inflammatory diseases. However, pharmacological agents that directly target NLRP3 remain limited. In the present study, we investigated the inhibitory effect of isoalantolactone (IAL), a naturally occurring sesquiterpene lactone, on NLRP3 inflammasome activation. IAL markedly suppressed caspase-1 activation and IL-1β/IL-18 maturation induced by canonical NLRP3 stimuli in macrophages, while showing little effect on affecting AIM2 or NLRC4 inflammasomes, indicating selective inhibi

Family Members' Help-Seeking Behaviour for Their Relative Who Uses Substances: A Cross-Sectional National Study in Brazil.

The affected family members (AFM) of relatives with substance use problems (RSU) play an important role in supporting their relatives to enter substance use treatment. This study investigated the help-seeking behaviours for their relatives by AFM in Brazil, including the characteristics of those who sought help and the risk factors for delaying it. A secondary analysis from a national cross-sectional study of 3030 AFM was performed. Participants were recruited from a range of services focused on AFM across each of the five Brazilian regions (North, Northeast, Central-West, Southeast, South). W

Yogurt alleviates alcohol-induced hepatocyte pyroptosis and inflammation via autophagy/P62/Nrf2/ROS/NLRP3 pathway.

Alcohol, one of the most widely popular and consumed beverages worldwide, often leads to acute alcoholic liver injury (AALI) when excessively consumed within a short timeframe. It can further induce alcoholic hepatitis, posing a serious risk to human health and public health safety. Pyroptosis, a typical mode of programmed cell death, is closely associated with the body's inflammatory response and plays a central role in AALI. However, the role of yogurt in acute alcohol-induced pyroptosis and inflammation is unclear. In this study, we found that yogurt and its metabolite d-lactate effectively

Analysis of post-market adverse events of tafamidis base on the FDA adverse event reporting system.

Tafamidis is the world's first and only oral drug approved to treat the rare disease transthyretin amyloid cardiomyopathy (ATTR-CM). Medicines are known to have different adverse reactions during the course of treatment. However, the current limited clinical studies did not identify significant adverse drug reactions to tafamidis. Tafamidis has been on the market for 5 years now, a large number of adverse drug event (ADE) reports with tafamidis as the primary suspected drug have been reported in the United Food and Drug Administration's adverse event reporting system (FAERS). We retrieved 8170

"Speak from a place of love": The family dynamics surrounding Black father-daughter sexual health communication.

INTRODUCTION: Parent-child sexual health communication reduces sexual risk behaviors among youth, which in turn lowers the transmission rate of HIV and sexually transmitted infections. Despite being a known protective factor, gender differences persist with mothers being more likely to discuss sexual health with their children than fathers. Although individual characteristics, interpersonal familial factors and societal norms (e.g., communication self-efficacy, parent-child closeness, and cultural and gender norms) are associated with Black parents' likelihood to communicate about sex with the

Emotion socialization profiles in military parents: Associations with post-traumatic stress disorder.

Military families face many difficulties, including a parent deploying to a warzone and the subsequent risk of returning with symptoms of posttraumatic stress disorder (PTSD). Symptoms of PTSD are associated with parenting difficulties; however, little is known about how PTSD symptoms may be associated with emotion socialization (ES), a set of processes crucial to children's emotional well-being. This project investigated observed ES behaviors in deployed and non-deployed parents in a sample of 224 predominantly White, non-Hispanic National Guard/Reserve (NG/R) families with deployed fathers,

Protective effects of compound M01 on retinal ganglion cells in experimental anterior ischemic optic neuropathy by inhibiting TXNIP/NLRP3 inflammasome pathway.

Apoptotic death of retinal ganglion cells (RGCs) is a common pathologic feature in different types of optic neuropathy, including ischemic optic neuropathy and glaucoma, ultimately leading to irreversible visual function loss. Potent and effective protection against RGC death is determinative in developing a successful treatment for these optic neuropathies. This study evaluated the neuroprotective effect of a HECT domain-E3 ubiquitin ligase inhibitor, M01, on retinal ganglion cells after ischemic injury. Experimental anterior ischemic optic neuropathy (AION) was induced by photothrombotic occ

Homozygous Familial Hypercholesterolemia.

Familial hypercholesterolemia (FH) is an inherited disorder with retarded clearance of plasma LDL caused by mutations of the genes involved in the LDL receptor-mediated pathway and most of them exhibit autosomal dominant inheritance. Homozygotes of FH (HoFH) may have plasma LDL-C levels, which are at least twice as high as those of heterozygous FH (HeFH) and therefore four times higher than normal levels. Prevalence of HoFH had been estimated as 1 in 1,000,000 before but more recent genetic analysis surveys predict 1 in 170,000 to 300,000. Since LDL receptor activity is severely impaired, HoFH
