Comparison of donkey and stallion semen preservation: effects of egg yolk plasma, seminal plasma and extenders on sperm motility, MALDI profiles and fertility.

ABSTRACT: The conservation of donkey breeds at risk of extinction requires the development and optimisation of semen preservation and artificial insemination strategies aimed at improving reproductive efficiency, increasing genetic diversity and supporting sustainable population management. The effects of commercial extenders (INRA96, BotuSemen Gold, Beyond), seminal plasma (SP) removal and egg yolk plasma (EYP; 0-10%) supplementation on jack semen stored at 4°C for 24-48 h were evaluated, using stallion semen as a reference. Sperm motility was assessed by computer-assisted sperm analysis (CAS

Mobile genetic elements drive the evolution and multidrug resistance of Salmonella infantis along the United States poultry production line.

BACKGROUND: Salmonella is the primary enteric pathogen related to foodborne illnesses worldwide, posing significant public health concerns. Amongst the diversity and pathogenicity of over 2000 Salmonella serovars, Salmonella Infantis (S. Infantis) ranks among the top Salmonella serovars implicated in foodborne outbreaks, with thousands of reported cases annually in the United States. Moreover, the incidence of S. Infantis infections has spread rapidly worldwide from the U.S. to Europe, where more than 50% of isolated S. Infantis strains have developed antibiotic resistance. Previous studies ha

Virulence and resistance patterns of Vibrio cholerae non-O1/non-O139 acquired in Germany and other European countries.

Global warming has caused an increase in the emergence of Vibrio species in marine and estuarine environments as well as fresh water bodies. Over the past decades, antimicrobial resistance (AMR) has evolved among Vibrio species toward various antibiotics commonly used for the treatment of Vibrio infections. In this study, we assessed virulence and resistance patterns of Vibrio cholerae non-O1/non-O139 strains derived from Germany and other European countries. A total of 63 clinical and 24 environmental Vibrio cholerae non-O1/non-O139 strains, collected between 2011 and 2021, were analyzed. In

The Cryptococcus neoformans transcriptome at the site of human meningitis.

UNLABELLED: Cryptococcus neoformans is the leading cause of fungal meningitis worldwide. Previous studies have characterized the cryptococcal transcriptome under various stress conditions, but a comprehensive profile of the C. neoformans transcriptome in the human host has not been attempted. Here, we extracted RNA from yeast cells taken directly from the cerebrospinal fluid (CSF) of two AIDS patients with cryptococcal meningitis prior to antifungal therapy. The patients were infected with strains of C. neoformans var. grubii of molecular type VNI and VNII. Using RNA-seq, we compared the trans

Exercise in cardiomyopathies: updated evaluation, risk stratification, and clinical prescription.

Cardiomyopathies comprise a heterogeneous group of myocardial disorders that may increase the risk of potentially life-threatening arrhythmias, particularly during physical activity. Historically, this risk prompted conservative recommendations, often leading to broad restrictions on exercise participation. However, advances in the understanding of pathophysiological mechanisms, coupled with improved tools for phenotypic assessment, risk stratification, and genetic evaluation, have enabled a more nuanced and individualized approach. This narrative review provides an updated overview of the rel

Dissecting PGE2-driven inhibition of T cell activation using single-cell multi-omic and inflammatory bowel disease genetic association analysis.

Genetic and immunologic studies implicate the interleukin (IL)-23/T helper (Th)17 pathway in inflammatory bowel disease (IBD). IL-23 and IL-1β drive human Th17 differentiation, while prostaglandin E2 (PGE2) and transforming growth factor-β (TGF-β) further modulate Th17 development and plasticity. However, how these inflammatory mediators influence human T cell regulatory programs remains incompletely understood. We used single-cell multi-omics to profile 171,829 peripheral blood T cells from 25 healthy donors in 64 samples exposed to activation stimuli, IL-1β and IL-23 alone or in combination

Genetic dissection of protein content in cowpea using custom-made NIRS equations and GWAS as a model for nutritional breeding and undergraduate research training.

As the demand for plant-based nutrition increases, improving the protein profile of legumes like cowpea has become a breeding priority. Cowpea, a multiuse legume and staple in many low-income regions, provides important dietary protein that can help meet the demand in our growing population. Our research used genome-wide association studies (GWAS) and phenomic tools to investigate the genetic architecture of seed protein content in cowpea and integrated 4 cohorts of undergraduate researchers through a USDA-AFRI REEU program. Using wet chemistry and near-infrared spectroscopy (NIRS), we assesse

All-trans retinoic acid combined with the VAH regimen for EVI1-positive acute myeloid leukemia: a case report with a brief literature review.

EVI1 positivity is a well-recognized high-risk factor in acute myeloid leukemia (AML), typically characterized by complex molecular genetic abnormalities, suboptimal responses to conventional chemotherapy, and a poor prognosis. We report a case of a 42-year-old patient with EVI1-positive AML harboring the MLL-AF6 fusion gene, who failed to achieve remission after undergoing standard "IA" induction therapy and was then treated with VAH (venetoclax, azacitidine, and homoharringtonine) consolidation chemotherapy. The patient subsequently received all-trans retinoic acid (ATRA) (20 mg twice daily,

RNA-triggered cell killing with CRISPR-Cas12a2.

Selectively eradicating target cells on the basis of their genetic or transcriptional identity remains important in basic research, medicine, biotechnology and agriculture1-3. For applications involving bacteria, CRISPR nucleases offer promising options due to their ability to enact RNA-guided counterselection4-7; however, using these same nucleases for counterselection in eukaryotes has proven much more restrictive8-14. Here we show that Cas12a2, a recently discovered type V CRISPR nuclease, exhibits RNA-triggered DNA shredding15,16, and enables programmable and sequence-specific elimination

Galactosylated LNP-mediated hepatic Sam68 silencing improves glucose homeostasis in diabetes by suppressing gluconeogenesis.

Excessive hepatic glucose production is a key driver of the progression of type 2 diabetes (T2DM), a highly prevalent global metabolic disorder. We previously reported that Sam68 expression is upregulated in the livers of both diabetic patients and mouse models, and hepatocyte-specific knockdown of Sam68 greatly alleviates hyperglycemia and improves insulin sensitivity in diabetic mice. Here, we engineered a series of ligand-functionalized lipid nanoparticles (LNPs) and identified a galactose-decorated formulation (LNP-Gal) that enables efficient hepatocyte-selective delivery of Sam68 siRNA, t

A day sleep promoting role of phototransduction in Drosophila melanogaster.

The daily sleep-wake cycle is a conserved behaviour defined by locomotion quiescence and enhanced responsive threshold to sensory stimuli. Both the circadian clock and sleep-homeostasis determine the daily sleep profile. Environmental light is a major sensory input and also regulates circadian clock and the balance between sleep and wakefulness. In Drosophila, the cellular mechanism and neural circuitry underlying light-mediated circadian synchronization are well-established, yet the direct relationship between light/visual input and sleep remains unclear. To address this knowledge gap, we mea

Summer-pattern seasonal affective disorder: A case study and implications for primary care.

Seasonal affective disorder (SAD) is most often recognized as a fall-winter subtype characterized by hypersomnia, hyperphagia, and anergia. Less commonly, patients experience a reverse presentation with spring-summer onset marked by irritability, insomnia, appetite loss, and psychomotor agitation. Pathophysiologic contributors to reverse SAD include circadian misalignment, neuroendocrine dysregulation, environmental stressors, and genetic susceptibility. Management requires a multimodal approach incorporating psychoeducation, psychotherapy (notably cognitive behavioral therapy [CBT]-SAD), phar

Hereditary renal cell carcinoma surveillance protocols: a review of the literature and proposed recommendations.

Hereditary renal cell carcinoma (hRCC) syndromes represent a small but significant proportion of renal cancer cases, accounting for 5-8%. They are characterized by distinct genetic etiologies, early-onset presentations, and unique clinical features. Timely identification and surveillance of at-risk individuals are essential to improving outcomes, as early detection facilitates interventions at a localized stage. However, existing recommendations are highly variable and often lack robust evidence. This extensive review consolidates current knowledge on major hRCC syndromes, namely the von Hippe

CD38 Inhibition Ameliorates Age-Related CognitiveDecline via a Choroid Plexus-Cerebrospinal Fluid-Hippocampus Axis.

Age-related cognitive decline represents a major and unresolved challenge of human aging. Here, we identify the NAD+-consuming enzyme CD38 as a central regulator of cognitive aging acting through a choroid plexus-cerebrospinal fluid (CSF)-hippocampus axis. CD38 expression increases with age and localizes primarily to pericytes in the choroid plexus, where it depletes NAD+, impairs mitochondrial function, and promotes cellular senescence. Genetic ablation or pharmacological inhibition of CD38 restores NAD+ levels, suppresses senescence markers, and improves choroid plexus function, resulting in

Gastrointestinal digestion governs insect protein hydrolysis and predicted bioactive peptide release: Species-dependent implications for functional food applications.

This study investigates the digestion of insect proteins and the release of predicted bioactive peptides during human gastrointestinal digestion. Using the Infogest in vitro model, mealworm, cricket, and black soldier fly larvae (BSFL) proteins were digested and analyzed through discovery proteomics and bioinformatics to identify predicted bioactive peptides. Sequential windowed acquisition of all theoretical fragment ion mass spectra (SWATH-MS) quantified insect proteins including predicted bioactive peptide precursor proteins, the precursors of predicted bioactive peptides. Results indicated

Clinical Trial Design Considerations for the Development of Disease-Modifying Therapies for Type 1 Diabetes.

Monocyte-derived macrophages, the major source of pathogenic macrophages in COVID-19, are oppositely instructed by macrophage CSF (M-CSF) or granulocyte macrophage CSF (GM-CSF), which promote the generation of antiinflammatory/immunosuppressive MAFB+ (M-MØ) or proinflammatory macrophages (GM-MØ), respectively. The transcriptional profile of prevailing macrophage subsets in severe COVID-19 led us to hypothesize that MAFB shapes the transcriptome of pulmonary macrophages driving severe COVID-19 pathogenesis. We have now assessed the role of MAFB in the response of monocyte-derived macrophages to

Host iron deficiency protects against Plasmodium infection and drives parasite molecular reprofiling.

Iron deficiency, anemia, and Plasmodium infection are global health challenges with overlapping geographical distributions, particularly affecting pregnant women in Africa, yet the mechanisms underlying their interactions remain poorly understood. We used a multilayered approach combining clinical data from Malawian pregnant women (n = 711) in the REVAMP trial, a genetic mouse model [Tmprss6-knockout (KO)], and in vitro Plasmodium falciparum cultures to clarify iron-malaria associations. Iron deficiency was associated with 50% reduced P. falciparum parasitemia in pregnant women [95% CI (30 to
