Neuronal cell death in alphavirus encephalomyelitis.

Alphaviruses are mosquito-borne, enveloped, plus-strand RNA viruses that cause a spectrum of diseases in humans that include fever, rash, arthritis, meningitis, and encephalomyelitis. Sindbis virus (SINV) is the prototype alphavirus, causes encephalomyelitis in mice, and provides a model system for studying the pathogenesis of alphavirus-induced neurological disease. Major target cells for SINV infection in the central nervous system (CNS) are neurons, and both host and viral factors determine the fate of infected neurons. Young animals are most susceptible to fatal disease. This correlates wi

Dynamic pelvic-pudendal reflex plasticity mediated by glutamate in anesthetized rats.

Reflex plasticity between pelvic afferent nerve (PAN) and pudendal efferent nerve (PEN), as well as external-urethral sphincter (EUS) activity was examined in anesthetized rats. A progressive increase in the number of evoked action potentials per stimulus occurred in PEN and EUS activity when PAN was repetitively stimulated (1 Hz). This potentiation in pelvic-pudendal reflex (PPR) activity induced by repetitive stimulation was abolished by APV (D-2-amino-5-phosphonoraleric acid, i.t. 100 microM, 2-5 microl) and attenuated by the NBQX (2, 3-dihydroxy-6-nitro-7-sulfamoyl-benzo (F) quinoxaline, i

Ultrastructural characteristics and conduction velocity of olfactory receptor neuron axons in the olfactory marker protein-null mouse.

Olfactory receptor neuron (ORN) axon diameters and the conduction velocity of the compound action potential along ORN axons were studied in olfactory marker protein (OMP)-null mice and genotypically matched controls. The compound action potential was distinguished from postsynaptic field potentials by its shorter latency, its persistence following application of cobalt or kynurenic acid that blocked postsynaptic responses, and its ability to follow paired-pulse stimulation at 300 Hz. Blockade of the postsynaptic field responses by kynurenic acid indicates that in the mouse, as in the rat, glut

Kynurenic acid in brain and cerebrospinal fluid of fetal, newborn, and adult sheep and effects of placental embolization.

Concentrations of the endogenous glutamate receptor antagonist kynurenic acid (KA) were measured in various brain regions and in cisternal cerebrospinal fluid of fetal, newborn, and adult sheep. KA concentrations were significantly higher in the fetal brain and cerebrospinal fluid at 90 and 140 d gestation compared with postnatal ages. In fetuses of 132-139 d gestation, KA concentrations in cerebrospinal fluid collected by drainage from an indwelling cisternal catheter increased significantly after infusion of the organic acid transport inhibitor probenecid (100 or 200 mg/kg, i.v.) indicating

Secondary excitotoxicity contributes to dopamine-induced apoptosis of dopaminergic neuronal cultures.

Dopamine (DA) and related catechols may contribute to selective degeneration of dopaminergic neurons in the substantia nigra in Parkinson's disease. To investigate whether DA induces apoptosis of dopaminergic neurons, we characterized the effects of various concentrations of exogenous DA on a substantia nigra/neuroblastoma hybrid cell line (MES 23.5 or MES). The hybrid MES cells were maintained in the presence of 50 microM glutamate in logarithmic growth on poly-D-lysine-precoated T-75 flasks and plated either onto petri dishes with glass coverslips for morphological studies or onto 6-well pla

The neurotoxicity of sulfur-containing amino acids in energy-deprived rat hippocampal slices.

The rat hippocampal slice preparation and its electrophysiology were used to assess the toxicity of two sulfur-containing amino acids, L-cysteate (CA) and L-cysteine (CYS). Both compounds were innocuous under normal conditions but became toxic in energy-deprived (lack of oxygen or glucose) slices. CA and CYS toxicity was apparent as both reduced the number of slices that normally recover their neuronal function (evoked CA1 population spike) after a standardized period of hypoxia or glucose deprivation (GD). The competitive N-methyl-D-aspartate (NMDA) antagonist DL-2-amino-5-phosphonovalerate b

Regional distribution of alpha-[3H]amino-3-hydroxy-5-methylisoxazole-4-propionic acid binding sites in rat brain: effect of chemical modification of SH- groups in tissue sections.

Previous studies have shown that chemical modifications of sulfhydryl (SH-) groups with mercurial compounds in rat brain membrane preparations increase the binding of alpha-[3H]amino-3-hydroxy-5-methylisoxazole-4-propionic acid [(3H]AMPA), a ligand for the quisqualate/AMPA type of glutamate receptors. In the present study we investigated the regional distribution of SH- group modification by quantitative analysis of autoradiographic images of [3H]AMPA binding in tissue sections. We also compared the effect of SH- group modification to that of the chaotropic ion thiocyanate (SCN-) which has bee

Amino acid-specific ADP-ribosylation. Identification of an arginine-dependent ADP-ribosyltransferase in rat liver.

A partially purified protein preparation from rat liver catalyzed the ADP-ribosylation of low molecular weight guanidino compounds and proteins. Agmatine and arginine, previously shown to be effective acceptors for the guanidine-dependent erythrocyte ADP-ribosyltransferase, were used as acceptors by the rat liver enzyme; lysine, histidine, and serine were inactive. The product of the reaction between [adenine-U-14C]NAD and agmatine catalyzed by the rat liver enzyme co-chromatographed with [adenine-U-14C]ADP-ribose-agmatine which was synthesized by the erythrocyte transferase; in parallel assay

Catabolism and protein binding of Tc-99m pyridoxylideneglutamate.

Various Tc-99m-labeled compounds have been suggested as replacements for [I-131] rose bengal for imaging of the hepatobiliary system. Among such compounds are Schiff's bases, which are tin-free Tc-chelates easily prepared by 30-min autoclaving of an equimolar mixture of pyridoxal and an amino acid at pH 8.5. We have compared the properties of several Schiff's bases, including Tc-99m pyridoxylideneglutamate (Tc PyG) with [I-131] rose bengal. Under conditions described, Tc-PyG can be prepared free of Tc-pyridoxal and with less than 2% TcO4- radiochemical impurity. Blood clearance and biliary exc

Multimode MALDI-MSI deciphers matrine-induced metabolic reprogramming in prostate cancer xenografts: spatial mapping of low-molecular-weight compound alterations.

BACKGROUND: Matrine, a bioactive isoquinoline alkaloid, exhibits antitumor efficacy by modulating multiple signaling pathways to suppress cancer cell proliferation, migration and invasion. However, its metabolic regulatory mechanisms in prostate cancer intervention require systematic characterization. METHODS: We implemented matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) for spatial metabolomic profiling of prostate tissues, integrated with multivariate analytical approaches including principal component analysis, Pearson correlation-based clustering heatmap

Determining the influence of tree age on tea quality-related metabolites in Camellia tachangensis var. remotiserrata by integrating metabolomic and transcriptomic analyses.

While tea tree age is generally considered to influence the quality of black tea, the relationship between tree age and tea quality is unclear. We examine relationships between tree age and key metabolite contents related to tea quality in new leaves of the wild arbor-type tea Camellia tachangensis var. remotiserrata. We report tree age to directly affect metabolic divergence in leaves in spring. Compared with 10-y-old trees, the leaves of 80- and 800-y-old trees have higher flavonoid contents; total soluble sugar, polyphenol, and total amino acid contents are largely unaffected by tree age, b

Untargeted serum metabolomics and air pollution in Parkinson's disease.

Mechanisms linking air pollution to Parkinson's disease (PD) remain poorly understood despite consistent toxicological and epidemiological evidence. We investigated metabolic disturbances associated with air pollution in PD using untargeted metabolomics. Serum samples were collected from 616 PD patients and 271 controls residing in central California, a region with high air pollution. Exposures to particulate matter (PM2.5), carbon monoxide, and nitrogen dioxide were estimated for the 1-, 3-, and 5-year periods prior to serum collection using residential addresses and three modeling approaches

Elucidating nutrient composition and biosynthesis in Macadamia integrifolia through multi-omics profiling across developmental stages.

Macadamia (Macadamia integrifolia), a high-value cash crop, has garnered significant attention due to its rich nutritional profile and unique health benefits. Previous studies have primarily focused on analyzing nutritional components, without systematically elucidating the mechanisms underlying their synthesis during kernel development. To elucidate the molecular and metabolic basis of kernel quality formation, we integrated morphological, nutritional, transcriptomic, and metabolomic analyses across five developmental stages (T1-T5, 60-190 days after fruit setting). Rapid kernel expansion occ

MUC2 shifts the metabolic profiles of commensal bacteria in defined microbial communities.

Background: The intestinal mucus layer is comprised of heavily glycosylated mucins, including mucin 2 (MUC2), that serve as a nutrient source for certain bacterial members of the gut microbiota. Only a subset of gut commensals encode the glycoside hydrolases required to degrade mucin glycans. However, mucin-degrading microbes can release glycans and generate compounds that can cross-fed non-mucin degrading microbes, creating complex microbial networks. While pairwise studies have shown that mucin degradation drives cross-feeding and metabolite exchange, the broader impact of mucins on communit

Characterization of Lactiplantibacillus paraplantarum HK-1 and GABA Synthesis Under Simulated Gastrointestinal Conditions.

Gamma-aminobutyric acid (GABA) is a bioactive amino acid with anti-inflammatory and neurotransmitter properties, yet limited information exists regarding its production by Lactiplantibacillus paraplantarum. We evaluated factors that influence GABA synthesis by L. paraplantarum HK-1 and assessed its production in vitro and under simulated gastrointestinal conditions. GABA production was analyzed using HPLC with pre-column derivatization, gene expression was assessed through RT-qPCR, and probiotic characteristics were evaluated using standard microbiological methods. L. paraplantarum HK-1 demons

Ferroptosis: when metabolism meets cell death.

We present here a comprehensive update on recent advancements in the field of ferroptosis, with a particular emphasis on its metabolic underpinnings and physiological impacts. After briefly introducing landmark studies that have helped to shape the concept of ferroptosis as a distinct form of cell death, we critically evaluate the key metabolic determinants involved in its regulation. These include the metabolism of essential trace elements such as selenium and iron; amino acids such as cyst(e)ine, methionine, glutamine/glutamate, and tryptophan; and carbohydrates, covering glycolysis, the cit

Christensenella minuta interacts with multiple gut bacteria.

INTRODUCTION: Gut microbes form complex networks that significantly influence host health and disease treatment. Interventions with the probiotic bacteria on the gut microbiota have been demonstrated to improve host well-being. As a representative of next-generation probiotics, Christensenella minuta (C. minuta) plays a critical role in regulating energy balance and metabolic homeostasis in human bodies, showing potential in treating metabolic disorders and reducing inflammation. However, interactions of C. minuta with the members of the networked gut microbiota have rarely been explored. MET
