SINEUP-mediated enhancement of HNF4α improves metabolic function in Huh7-based bioengineered hepatic microtissues.

BACKGROUND/AIMS: Human liver cell-based drug-screening platforms require stable hepatocyte identity and metabolic competence, but Huh7 cells show reduced hepatic function. We investigated whether SINEUP-mediated enhancement of hepatocyte nuclear factor 4 alpha (HNF4α) could improve the metabolic performance of Huh7-based three-dimensional hepatic microtissues. METHODS: Decellularized liver extracellular matrix-derived microparticles were incorporated into three-dimensional microtissues containing Huh7 cells, human umbilical vein endothelial cells, and Wharton's jelly mesenchymal stem cells us

Polystyrene Microplastic Exposure Adversely Affects Oocyte Quality and Ovary Health Status in Mytilus galloprovincialis.

Microplastics pose a growing threat to marine ecosystems as they can accumulate in aquatic organisms, interfering with essential physiological processes including reproduction. This study analyzed the effects of short-term exposure (48 h) to two concentrations (0.5 and 1 µg/mL) of 5 µm diameter polystyrene microplastic bead particles in female Mytilus galloprovincialis mussels, a bioindicator species of the Mediterranean Sea. Histological analyses revealed progressive oocyte degeneration and the development of hypertrophic PAS-positive mucous cells, indicative of a stress response, in a dose-d

FBXO28 targets SNAI1 for ubiquitin-proteasomal degradation in non-small cell lung cancer.

SNAI1 is a central regulator of the epithelial-mesenchymal transition, which is highly unstable and degraded through the ubiquitin-proteasome system. Although a panel of E3 ubiquitin ligases responsible for SNAI1 degradation have been discovered, whether there exists other E3 ubiquitin ligase remains unclarified. In this study, we demonstrate that FBXO28 expression is downregulated in non-small cell lung cancer (NSCLC) tissues compared to adjacent non-cancerous tissues, correlating with beneficial patient prognosis. Specifically, FBXO28 physically interacts with SNAI1, promoting its polyubiqui

Genomic and Morphological Insights Into the Adaptive Evolution of Honeybees in Rocky Desertification Habitats.

Unlike traditional desertification, rocky desertification is a distinct form of land degradation characterized by low vegetation coverage and a high proportion of rock exposure. Honeybees are crucial pollinators and ecological restorers and have coevolved closely with plants as their primary habitat and food resource to naturally inhabit rocky desertification habitats. In this study, classical morphological analyses showed that under mild desertification conditions, honeybee body size increases, while in severely degraded habitats, their morphology resembles that of normal individuals. Whole-g

Tramiprosate, the Active Agent of ALZ-801, Modulates Amyloidogenic APP Processing and Tau Phosphorylation in a Cellular Model of Alzheimer's Disease.

ALZ-801, a valine-conjugated prodrug of tramiprosate, has recently been evaluated in a Phase III trial as a potential Alzheimer's disease (AD)-modifying therapy. Tramiprosate reduces misfolding of amyloid beta (Aβ) monomers and inhibits oligomer formation. In the present study, we investigated tramiprosate's effects on amyloidogenic processing of amyloid precursor protein (APP) and tau phosphorylation using the N2a-APPSwe cell line as a cellular model of AD. Tramiprosate decreased levels of secreted Aβ40 and Aβ42 in a dose-dependent manner without affecting cell viability. It also reduced the

Navigating the Complexity: A Comprehensive Review of GSK-3 Inhibition in Regenerative Medicine.

Glycogen synthase kinase-3 (GSK-3) is a central regulator of numerous cellular signaling pathways, with critical roles in metabolism, proliferation, differentiation, and tissue regeneration. This review explores the multifaceted effects of pharmacological GSK-3 inhibition across multiple body districts, focusing on its highly context-dependent impact. It has been proven that blocking GSK-3 promotes embryonic stem cell self-renewal, neuronal differentiation, osteogenesis, and dentinogenesis, while it has an inhibitory effect on adipogenesis. GSK-3 inhibition effects on muscle, heart, liver, and

Fueling Performance of Female Soccer Players: Appraisal of the Likelihood of Between-Sex Differences in Carbohydrate Requirements.

Skeletal muscle glycogen depletion is considered one of the key contributing factors toward fatigue and associated performance decrements in soccer. Ensuring adequate carbohydrate availability before and during training and matches is widely advised as a priority for players, and for that reason, carbohydrate-based fueling guidelines for soccer have been developed. However, the lack of female-specific research used to inform the development of these guidelines raises important questions regarding their applicability for female soccer players. This review critically appraises the likelihood of

Ganoderic Acid A Enhances Glycometabolism in Mouse Gastrocnemius Muscle During Exercise-Induced Fatigue via AMPK/PGC-1α Pathway Activation.

Exercise-induced fatigue (EIF) is closely associated with impaired glycometabolism in skeletal muscle. This study investigated the protective effects of Ganoderic acid A (GAA) on glycometabolism in EIF mice and explored its underlying mechanisms. 60 KM mice were divided into five groups: a blank control (BC), a model control (MC), and three GAA-treated groups (20, 40, and 60 mg/kg/d). After a 7-week intervention, exhaustive treadmill tests and biochemical analyses were conducted to assess fatigue resistance, metabolic parameters, and molecular pathways. GAA administration significantly prolong

Protein persulfidation: The missing link in Alzheimer's disease defense mechanisms.

Despite decades of research dominated by the amyloid-beta hypothesis, clinical treatment of Alzheimer's disease (AD) has yet to achieve a decisive breakthrough. This editorial advances an alternative pathological paradigm: the collapse of endogenous hydrogen sulfide (H₂S) signaling represents a central failure point in the brain's intrinsic defense mechanisms against AD. We dissect the molecular cascade triggered by cystathionine γ-lyase (CSE) deficiency, focusing on how reduced persulfidation of glycogen synthase kinase 3β (GSK3β) directly promotes Tau hyperphosphorylation and subsequent neur

Ghost-rocks' microbiota: metagenomic insights into their influence on the biogeochemistry of karstic cave and groundwater.

Microbial communities in the critical zone drive key geochemical processes, but many subsurface habitats remain poorly characterized. Ghost-rock karst systems in particular represent unexplored microbial niches. Here, we provide the first genome-resolved metagenomic comparison of ghost-rock and groundwater microbial communities from the Sterkfontein karst system (South Africa). Ghost-rock and groundwater communities host distinct taxonomic and metabolic assemblages. Groundwater communities are dominated by chemolithotrophs capable of oxidizing sulfur- and nitrogen-bearing compounds, and by het

Gut microbial H2S promotes metabolic dysfunction in mice via hepatic PPARα suppression.

This study investigates the role of gut microbiota-derived hydrogen sulfide (H2S) in obesity and glucose metabolism disorders. By integrating human gut metagenomic data, intervention experiments in mouse models, and in vitro cellular assays, we identified a signature of microbial sulfur metabolism in human cohorts and provided experimental evidence for its causal role and underlying metabolic mechanisms in mice. In clinical cohorts with obesity and glucose metabolism disorders, we observed a notable enrichment of genes involved in sulfur transport and H2S production. In mouse models, administr

A novel covalent inhibitor alleviates type 2 diabetes mellitus by restraining GSK-3β.

Type 2 diabetes (T2DM), which is characterized by insulin resistance and diminished sensitivity, accounts for more than 90 % of the incidence rate of diabetes. At present, the drugs used to treat T2DM in clinic are restricted due to their poor compliance or serious adverse reactions. The abnormally elevated activity of GSK-3β plays an important regulatory role in the occurrence and development of T2DM. Herein, we report the therapeutic effect of a GSK-3β covalent inhibitor N-(3-(4-benzyl-3,5-dioxo-1,2,4-thiadiazol-2-yl) phenyl) acrylamide (GL10a), which was discovered by our group with excelle

Metabolic and proteomic profiles provide insights on mechanism of late onset Pompe disease.

Late onset Pompe disease (LOPD) is caused by a deficiency of the enzyme acid α-glucosidase, resulting in glycogen accumulation in lysosomes. The mechanism of LOPD has been less explored. In this study, we used an integrative analysis of the proteomics and metabolomics of LOPD muscle samples to reveal the potential mechanisms. Proteomic analysis identified 635 upregulated proteins and 89 downregulated proteins in the LOPD group. Similarly, metabolomic analysis revealed 15 upregulated and 143 downregulated metabolites; notably, L-arginine levels were significantly decreased in the LOPD group. Ly
