The moderate affinity of clozapine at H3 receptors is not shared by its two major metabolites and by structurally related and unrelated atypical neuroleptics.

We determined the affinity and/or potency of two metabolites of clozapine (clozapine-N-oxide and N-desmethylclozapine) and of five atypical neuroleptics, chemically related (olanzapine) or unrelated to clozapine (remoxipride, risperidone, thioridazine, zotepine), at H3 receptors. The specific binding of 3H-N alpha-methylhistamine to rat brain cortex homogenates was inhibited by the seven compounds; the pKi values were: N-desmethylclozapine (5.33); clozapine-N-oxide (4.18); olanzapine (5.45); thioridazine (4.91); zotepine (4.75); remoxipride (4.51) and risperidone (4.43). Three compounds were e

[Phencarol--an antihistaminic preparation from the quinuclidylcarbinol group].

A high antihistaminic activity of quinuclidyl-3-diphenylcarbinol (phencarol) has been ascertained. In pharmacological experiments phencarol is superior to dimedrol (diphenhydramine), both as concerns its antihistaminic activity and the duration of its action. Unlike dimedrol and diprazine (promethazine) it has no inhibitory effect on the central nervous system. With its clinical use phencarol is effective in treating urticaria, Quincke's edema and allergic skin reactions. The drug is well tolerated by the patients and does not produce any inhibitory or soporific action. Phencarol has an author

Enabling artificial placenta cannulation: mechanisms of postnatal umbilical vessel closure and strategies to prolong patency.

Umbilical vessel closure after birth prevents hemorrhage but limits the time window for large-bore umbilical cannulation required for artificial placenta support. This review synthesizes physiologic triggers, vasoactive pathways, and vessel-wall specializations that drive rapid postnatal occlusion and highlights engineering-relevant targets to prolong functional patency. Umbilical vessel closure is promoted by the transition to extrauterine life (cooling, rising oxygen tension, oxidative stress, loss of placental vasodilator influence), altered hemodynamics after lung aeration, and mechanical

Glycosidase-Derived Arabinoxylan Hydrolyzates Attenuate Intestinal Inflammation via Restructuring the Gut Microbiota-Metabolite Axis.

The highly polymeric structure and complex branching of arabinoxylan (AX) limit its immunomodulatory potential. This study investigated whether enzymatic hydrolysis using xylanase (XYN) and α-l-arabinofuranosidase (ARF) could enhance anti-inflammatory efficacy in DSS-induced colitis. Synergistic ARF-XYN treatment, yielding low-polymerization and debranched oligosaccharides, exhibited superior efficacy in ameliorating colitis symptoms, suppressing pro-inflammatory cytokines (IL-6, TNF-α, IL-1β), and restoring intestinal barrier integrity compared to native AX. Multiomics analyses revealed that

Discovery of 3-indolealkylamines as novel dual-target σ1R/H3R ligands with potent analgesia.

Dual-acting ligands targeting the sigma-1 receptor (σ1R) and histamine H3 receptor (H3R) are emerging as promising candidates for novel and safe analgesics. In this work, we designed, synthesized, and evaluated twenty-nine 3-indolealkylamines as dual σ1R/H3R ligands. In vitro radioligand receptor binding assay or surface plasmon resonance assay were performed to determine their affinities toward σ1R or H3R. Among them, compound 67 demonstrated high binding affinity for both σ1R (Kᵢ = 8.8 nM) and H3R (KD = 31.2 nM). Further in vivo pharmacological evaluations confirmed its antagonistic activity

Non-targeted metabolomics reveals the metabolic atlas of macrophages remodeled by SVPLA2 in Naja atra venom.

Snake venom phospholipase A2 (SVPLA2) is a key toxic component of Naja atra (N. atra) venom, yet its systematic impact on macrophage metabolism remains elusive. Using non-targeted metabolomics, combined with multivariate statistics and KEGG pathway enrichment, this study delineated the metabolic reprogramming of RAW 264.7 macrophages induced by SVPLA2 in N. atra venom and assessed the reversal effects of the specific inhibitor varespladib. SVPLA2 was associated with a a triple metabolic phenotype in macrophages: massive production of histamine and arachidonic acid derivatives, activating infla

[Interpretation of standard technical specifications for methacholine chloride (Methacholine) bronchial challenge test (2023)].

Bronchial challenge tests are important clinical examinations for detecting airway hyperresponsiveness, with methacholine being a commonly used provocative agent. However, the previous guidelines had not been updated in a timely manner, leading to some problems in clinical application. The "Standard technical specifications for methacholine chloride (Methacholine) bronchial challenge test (2023)" is an updated and refined version of 2014 "Guidelines for Pulmonary Function Testing (Part Ⅲ)-Histamine and Methacholine Bronchial Challenge Tests". In the 2023 version, the scope of MCT applicability

Neural and metabolic mechanisms of emetogenic foodborne toxins via the brain-gut axis in ageing and cachexia.

Emetic foodborne toxins (bacterial enterotoxins, mycotoxins like DON/ZEA, marine toxins, cereulide, T-2 toxin, domoic acid, and biogenic amines such as histamine) cause acute gastrointestinal reactions and chronic systemic pathologies. This review outlines their activation of the brainstem's area postrema (AP) via intestinal barrier disruption, dysbiosis, vagus nerve stimulation, or direct central penetration. Acute exposure induces anorexia/defense responses through AP/NTS receptor pathways (GLP-1R, CCK, PYY, ghrelin, GDF15-GFRAL, 5-HT3). Chronic exposure impairs the AP-hypothalamus-locus coe

Zingerone Targets LKB1/AMPK to Block FcεRI-Dependent Mast Cell Degranulation and Anaphylaxis.

AMP-activated protein kinase (AMPK) acts as a cellular energy sensor and a central regulator of metabolism. Recent studies indicate that pharmacological AMPK activation can simultaneously ameliorate metabolic disorders (e.g., type II diabetes, obesity) and allergic diseases. Zingerone, a primary bioactive compound in ginger, demonstrates protective effects in vascular calcification, non-alcoholic fatty liver disease, and asthma via AMPK activation. This study aimed to evaluate the anti-allergic activity of Zingerone and elucidate its AMPK-dependent mechanisms. In vitro, Zingerone suppressed Fc

Microbiota-brain interaction: The role of gut-derived proteins in addressing various neurological disorders including Parkinson's (PD) and Alzheimer's diseases (AD).

The microbiota-brain interaction is a complex network connecting gut microbiota to the enteric nervous system (ENS) and the central nervous system (CNS) that are crucial for neurotransmission and neurological health. Metabolites such as short-chain fatty acids (SCFA), neurotransmitters, and neuromodulators from the gut microbiota influence behavior and brain function. This review focuses on the study related with gut bacteria, including Bifidobacterium infantis and Lactobacillus species, producing various metabolites in gut including bile acids, SCFA, histamine, and others to communicate with

Pathophysiological insights into delayed pressure urticaria: A comprehensive review.

Delayed pressure urticaria (DPU) is a chronic inducible subtype of urticaria, characterized by painful erythematous swelling triggered by sustained mechanical pressure. After pressure stimuli, lesions often appear within 4 to 6 hours, peak around 6 hours later, and persist for 8 hours to 3 days. More than half of patients may have associated symptoms, most often malaise and fatigue. The diagnosis of DPU is based on clinical history and confirmation through provocation testing. Dermographic tester and weighted rod testing are recommended as diagnostic tools by several current guidelines due to

L. acidophilus/L. johnsonii ratio affects slow transit constipation in rats.

Slow Transit Constipation (STC) is characterized by impaired colonic motility, but its relationship with gut microbiota remains unclear. This study investigated the correlation between specific gut microbial populations and STC, focusing on the Lactobacillus acidophilus to Lactobacillus johnsonii (A/J) ratio. We used four rat groups: Control (CON), Loperamide-induced STC (LOP), antibiotic-treated (ABX), and antibiotic plus Loperamide (ABX + LOP). Fecal samples were analyzed using 16S rRNA gene sequencing, and serum metabolites were examined through LC-MS. The LOP group showed an increased A/J

Novel applications of sleep pharmacology as delirium therapeutics.

Sleep-wake and circadian disruption (SCD) is a core feature of delirium. It has been hypothesized that SCD contributes to delirium pathogenesis; therefore, interventions that prevent or reverse SCD represent an array of promising opportunities in relation to delirium. This review explores the relationship between sleep-wake/circadian physiology and delirium pathophysiology with a focus on neurotransmitter systems. Across potential targets aimed at preventing or treating delirium, three broad approaches are considered: 1. Pharmacological mechanisms that contribute to physiological sleep may pre

Unraveling Complexities: Rhabdomyolysis, Acute Renal Injury, and Compartment Syndrome Following a Wasp's Sting.

This study delves into the rare occurrence of rhabdomyolysis induced by wasp stings, emphasizing its toxic systemic repercussions. Drawing parallels with documented instances of insect bites worldwide, including those by honey bees and Africanized bees, the research explores the correlation between multiple wasp stings and acute renal failure associated with rhabdomyolysis. The venom's active components, such as amines, kinins, and histamine-releasing peptides, underpin toxic systemic reactions, leading to hemolysis, coagulopathy, and severe cytotoxicity-induced acute renal failure. Roles of Mast Cells in Cutaneous Diseases.

Mast cells are present in all vascularized tissues of the body. They are especially abundant in tissues that are in frequent contact with the surrounding environment and act as potential sources of inflammatory and/or regulatory mediators during development of various infections and diseases. Mature mast cells' cytoplasm contains numerous granules that store a variety of chemical mediators, cytokines, proteoglycans, and proteases. Mast cells are activated via various cell surface receptors, including FcϵRI, toll-like receptors (TLR), Mas-related G-protein-coupled receptor X2 (MRGPRX2), and cyt
