Microbial communities populate the mucosal surfaces of all animals. Metazoans have co-evolved with these microorganisms, forming symbioses that affect the molecular and cellular underpinnings of animal physiology. These microorganisms, collectively referred to as the microbiota, are found on many distinct body sites (including the skin, nasal cavity, and urogenital tract), however the most densely colonized host tissue is the intestinal tract. Although spatially confined within the intestinal lumen, the microbiota and associated products shape the development and function of the host immune sy

Kinetoplastid parasites have caused human disease for millennia. Significant achievements have been made toward developing new treatments for leishmaniasis (particularly on the Indian subcontinent) and for human African trypanosomiasis (HAT). Moreover, the sustained decrease in the incidence of HAT has made the prospect of elimination a tantalizing reality. Despite the gains, no new chemical or biological entities to treat kinetoplastid diseases have been registered in more than three decades, and more work is needed to discover safe and effective therapies for patients with Chagas disease and

Vibrio cholerae, the agent of cholera, is a motile non-invasive pathogen that colonizes the small intestine (SI). Most of our knowledge of the processes required for V. cholerae intestinal colonization is derived from enumeration of wt and mutant V. cholerae recovered from orogastrically infected infant mice. There is limited knowledge of the distribution of V. cholerae within the SI, particularly its localization along the villous axis, or of the bacterial and host factors that account for this distribution. Here, using confocal and intravital two-photon microscopy to monitor the localization

BACKGROUND: Simultaneous quantification of liposoluble vitamins is not a new area of interest, since these compounds co-determine the nutritional quality of food and feed, a field widely explored in the human and animal diet. However, the development of appropriate methods is still a matter of concern, especially when the vitamin composition is highly complex, as is the case with feed designated for laboratory animals, representing a higher health and microbiological status.
RESULTS: A method combining microemulsion electrokinetic chromatography (MEEKC) with liquid-liquid extraction was devel

Herpes simplex encephalitis (HSE) is a rare but severe complication of frequent and mostly benign infection with herpes simplex virus (HSV). Although rapid and sensitive diagnosis tools and active antiviral drugs are available, HSE morbidity/mortality levels remain unsatisfactory. Molecular and cellular determinants of HSE are incompletely understood. The rarity and severity of the disease have suggested an increased susceptibility of some subjects to HSV infection. Numerous experimental studies have investigated the respective role of host and viral factors in HSE. The results of these studie

BACKGROUND: Shigella flexneri is the causative agent of bacillary dysentery and generates a significant global disease burden. The aim of this study was to analyze the pathogenesis and host immune response, at both the physiological and molecular level, using the model organism Caenorhabditis elegans, in response to S. flexneri. C. elegans is a nematode that responds to infection with a simple innate immune system, key aspects of which have been shown to be conserved.
METHODS: S. flexneri-mediated infection of C. elegans was performed in both solid and liquid assays. The expression and subseq

Staphylococcus aureus is a formidable pathogen and the leading cause of skin and soft tissue infections. We recently showed that the highly conserved sasF gene, encoding for an uncharacterized sortase-anchored surface protein, is up-regulated when S. aureus colonizes skin. Here, we demonstrate that SasF is an enzyme that has glycoside epimerase activity, which is required for S. aureus resistance to skin fatty acids. Mutants of sasF are highly impaired in skin colonization and acute infection models. The SasF enzyme is crucial for maintaining the structure of wall teichoic acids and proper S.

The human body harbors highly diverse and complex microbial communities across various regions. Among these, the oral cavity and stomach are directly connected and engage in dynamic microbial interactions. Microorganisms capable of migrating between these two distinct niches are referred to as trans-organ microbes, which constitute a core component of the oral-gastric microbiome axis. In this review, we elucidate the mechanisms underlying microbial translocation along the oral-gastric axis. We further discuss how these cross-organ microbes exert bidirectional influences on health and disease i

STAT3 is a central mediator of pro-survival signaling within the STAT family, functioning through both canonical transcriptional activity and noncanonical mechanisms. As a pleiotropic regulator, STAT3 integrates cytokine, growth factor, and stress signals to coordinate diverse cellular processes, including epithelial architecture, T cell development, macrophage polarization, apoptosis, metabolism, and inflammation-associated wound healing. This integrative capacity positions STAT3 as a critical signaling hub at the interface of tissue homeostasis and immune regulation. Dysregulated STAT3 activ

Viral receptors have traditionally been viewed as docking sites that mediate viral attachment and entry. However, this perspective no longer captures the full complexity of receptor function during infection. Receptor engagement links viral binding to intracellular trafficking, signal transduction, barrier integrity, and ultimately disease outcome. Rather than acting solely as entry determinants, receptors influence endocytic routing, cellular permissiveness, tissue tropism, immune responses, and systemic dissemination. A functional distinction between attachment factors, entry receptors, co-r

Valosin-containing protein (VCP), a conserved AAA ATPase hexamer, participates in multiple biological processes including ERAD, ubiquitin-dependent degradation by extracting misfolded proteins for proteasomal degradation. Although its interactions with cofactors are well-characterized, and its dysregulation is implicated in multisystem proteinopathy, amyotrophic lateral sclerosis, and cancer, the tissue-specific VCP interactomes underlying its functional versatility remain elusive. Here, we generated HA-N-tagged VCP knock-in mice via CRISPR/Cas9 strategy and performed affinity purification cou

Marine Microsporidia represent an increasingly important but historically underestimated group of aquatic intracellular parasites. Although traditionally associated with invertebrates and fishes, recent molecular and environmental studies have revealed extensive hidden diversity across marine ecosystems together with unexpected phylogenetic links between aquatic and human-associated lineages. This review synthesizes five decades of research (1975-2025) concerning the biology, evolution, taxonomy, host-pathogen interactions, ecological diversity, and zoonotic significance of marine Microsporidi

Lymphatic filariasis, caused by Brugia malayi, leads to chronic lymphatic dysfunction and lymphedema. The molecular mechanisms underlying interactions between filarial worms and their hosts remain poorly understood. Using untargeted metabolomics, we identify previously unknown acetylcholine analogs synthesized from leucine and isoleucine that are secreted by Brugia adult males. In the mammalian host, these metabolites selectively activate host M2 muscarinic acetylcholine receptors, alter lymphatic endothelial cell integrity, and trigger constriction of lymphatic vessels. These metabolites adde

The bimodality of amino-modified silica nanoparticles (SNs) manifested as an emission band with a maximum at 612 nm and relaxivity values r1(2) of 17.7(30.3) mM-1s-1, was achieved by co-doping SNs with Mn2+ oxalate and [Ru(dipy)3]2+ complex. Stepwise introduction of these dopants during the synthesis increases the manganese content and reduces the leaching of Mn2+ ions due to the electrostatic attraction of the cationic complex to oxalate. Conjugation of folate via amino groups provides both effective targeting and higher mobility of bimodal SNs during their uptake into cells. Formation of the

Cytochrome P450 (P450) enzymes play an important role in the biotransformation of both exogenous and endogenous compounds. Variability in P450 expression and activity impacts drug-metabolizing capacity and contributes to interindividual differences in drug exposure and therapeutic outcomes. Exogenous probe substrates have traditionally been used to assess the in vivo P450 enzyme activity. However, their application can be limited in vulnerable populations such as pregnancy, transplantation, pediatrics, and elderly. Endogenous biomarkers have emerged as minimally invasive potential alternatives

Tyrosinase is an enzyme involved in the early stages of melanin synthesis. Melanin, which gives skin its pigment, is the most targeted molecule in the development of skin-whitening cosmetic products. Consequential risks of chemically induced skin diseases are associated with the usage of skin-whitening products that contain tyrosinase-inhibiting compounds. Currently, products containing tyrosinase inhibitors on the market lack collective analysis of their toxicity on melanocytes. Moreover, national regulatory agencies (e.g., FDA) place the burden of ensuring the safety of whitening products on

The field of biomedical materials is constantly undergoing innovations to meet increasingly complex physiological needs. A particularly prominent area of research in this regard is natural active ingredient hydrogels, which are self-assembled entirely from natural active ingredients, do not involve any chemical cross-linking agents, and have unique physicochemical and biological properties that are believed to help adapt to the complex and variable in vivo microenvironment. Hydrogels have a wide range of applications, including in critical medical areas such as wound healing, cancer therapy an

Over 10 species of Rickettsia cause serious human disease across the globe, including deadly spotted fever and typhus. These arthropod-borne microbes and their close relatives are the only bacteria that obligately reside in the eukaryotic cytosol, making them distinctive tools to study innate immunity. Yet, the molecular details on how Rickettsia species interact with innate immunity have remained largely enigmatic. Recent advances are revealing that Rickettsia species are not only distinct from other bacterial pathogens that reside in the cytosol, but in some cases, they are highly distinct f

Based on a strategy integrating topological feature identification and path priority assessment, this study systematically explored the efficacy targets and differentiated mechanisms of Colquhounia Root Tablets(CRT) in the "homotherapy for heteropathy" of rheumatoid arthritis(RA) and diabetic kidney disease(DKD). Firstly, candidate targets of CRT and disease-specific genes were retrieved by integrating multi-source databases and transcriptomic data. Protein-protein interaction(PPI) networks were constructed to identify key targets shared by or specific to RA and DKD via topological f
