African Genetic Similarity Is Associated with Reduced Inflammation and Enhanced Adaptive Immunity in Multiple Myeloma.

The bone marrow immune microenvironment (BMME) is a critical factor in multiple myeloma (MM) pathogenesis. Despite higher incidence of MM among self-identified African Americans, the relationship between African genetic similarity and the BMME remains unclear. We present the largest analysis to date assessing the impact of Ancestry on the BMME in myeloma (N=320, including 64 with ≥50% African genetic similarity, AFR-High) leveraging our recently generated Immune Atlas. AFR-High was associated with enrichment of late T-effector cells, driven by rare cytotoxic CD4⁺ T cells, suggesting a distinct

Hepatitis B Screening and Immunity Gaps in Cambodia: Should Vaccination Be Mandated for Healthcare Workers and Health Sciences Students?

Hepatitis B virus (HBV) infection remains a major public health challenge globally, particularly in low- and middle-income countries (LMICs), where diagnosis, treatment, and vaccination coverage remain suboptimal. In Cambodia, despite more than two decades of inclusion of HBV vaccination in the national immunization program, protective immunity among healthcare workers and health sciences students is not routinely assessed. This policy-oriented perspective examines gaps in HBV immunity among health sciences students. Recent screening data from a Cambodian medical university (2025) reveal that

BCAT1 inhibits crotonate-related epigenetic modulation of metabolic genes in tumor-associated macrophages to counter immunosuppression.

The heterogeneous and immunosuppressive tumor microenvironment (TME) in hepatocellular carcinoma (HCC) contributes to poor immunotherapy responses. Tumor-associated macrophages (TAM) are central to the immunosuppressive TME, but how metabolic programs regulate TAM pro-tumorigenic functions remain incompletely understood. Here, we identify branched-chain amino acid transaminase 1 (BCAT1) as a metabolic checkpoint in TAMs constraining tumor progression. Compared with wild-type TAMs, BCAT1-deficient TAMs have increased intracellular crotonate, as well as enhanced histone H3 lysine 14 crotonylatio

Ablation of the adhesion G protein-coupled receptor ADGRL1 activates anti-tumor immune response via cDC1-T cell hub formation.

Immune-like coordinated cell death (CCD) indirectly eliminates target cells via immune activation, bypassing intrinsic programmed cell death. Using RNAi screening in a Drosophila tumor model, we identified a series of genes capable of inducing CCD. Here, we characterize adhesion G protein-coupled receptor ADGRL1 as a key mediator that activates anti-tumor immunity by facilitating cDC1-CD8+ T cell hub formation, as revealed by single-cell transcriptomics. Mechanistically, ADGRL1 ablation activates type I interferon signaling and promotes JAK/STAT1-dependent Decorin secretion, mediated by disrup

High-resolution promoter interaction analysis implicates genes involved in activation of type 3 innate lymphoid cells in immune disease risk.

Innate lymphoid cells (ILCs) are rare tissue-resident lymphocytes that functionally mirror cells of CD4+ T helper lineage but lack antigen receptors. Type 3 ILCs (ILC3s) are enriched at barrier sites, regulating inflammation and promoting tissue integrity. Here we profile the promoter-anchored chromosomal contacts of primary human ILC3s using low-input, high-resolution targeted chromosome conformation capture and compare them with those in CD4+ T cells. We use these data to link Crohn's disease genome-wide association study variants with target genes, implicating both known and unanticipated c

Inhaled Virus-Mimicking Particles Elicit Dose-Dependent Immune Response against SARS-CoV-2 with Sex-Bias ACE2 Activity in Mice.

There is an urgent need to develop antigen presentation platforms that can induce broadly systemic immunity against multiple severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) subvariants. Here, virus-mimicking particles (VMP) are functionalized with substantial receptor binding domain protrusions from Delta variant SARS-CoV-2 on particle surface, programmed by biotin-streptavidin interaction. The fabricated VMP reveal superior pulmonary biodistribution and retention for at least 1 day in C57BL/6 female mice after oropharyngeal aspiration exposure. VMP at higher dosage elicit a robus

Targeted delivery of mRNA to immune cells for in vivo cell therapy.

Messenger RNA (mRNA) therapeutics have revolutionized biomedicine by enabling direct in vivo programming of immune cells. This strategy bypasses the complex manufacturing and high costs associated with ex vivo cell therapies. However, efficient and specific systemic delivery of mRNA to target immune cell subsets remains a major translational hurdle. This review systematically examines engineering strategies that address this challenge. We first outline the key biological barriers to mRNA delivery, such as serum instability, nonspecific biodistribution, cellular uptake heterogeneity, and ineffi

Time-staggered chemo-immunotherapy via engineered nanofiber resists postoperative dynamic immunosuppression in glioblastoma.

Glioblastoma (GBM) almost inevitably recurs after surgical resection owing to residual infiltrative tumor cells and postoperative immunosuppression. However, delayed initiation of adjuvant therapy fails to restrain rapid tumor regrowth during the early postoperative period. Here, we elucidate postoperative dynamic immune pathology characterized by early explosive tumor proliferation (Ki67⁺ > 38.5%) and progressive protumoral macrophage polarization, and provide a time-staggered chemo-immunotherapy strategy to promptly remedy the postoperative therapeutic gap. To implement the adaptive interven

Molecular Mechanisms and Therapeutic Strategies for Immune Checkpoint Inhibitors in Breast Cancer: From Pathogenesis to Precision Medicine.

The advent of immunotherapy, and particularly the development of immune checkpoint inhibitors (ICIs), has revolutionized the landscape of breast cancer treatment, especially for triple-negative breast cancer. However, some patients still do not benefit from immunotherapy. Breast cancer is inherently an immunotherapy "cold" tumor, which results in suboptimal clinical outcomes when ICIs are used as monotherapy. Identifying additional immunotherapeutic targets and drugs, along with developing novel strategies for combination therapy, is crucial for addressing the challenges posed by immunotherapy

CMPK2 restricts Mycobacterium tuberculosis replication and regulates macrophage gene expression.

Host cell metabolic pathways influence innate immune responses to intracellular pathogens, but the contribution of nucleotide metabolism to antimicrobial defense remains incompletely defined. Here, we identify the mitochondrial nucleoside monophosphate kinase CMPK2 as a regulator of macrophage responses to Mycobacterium tuberculosis (Mtb). Using a targeted genetic screen of candidate host factors, we found that depletion of CMPK2 enhances intracellular Mtb replication in human macrophages. This phenotype was confirmed using both shRNA-mediated knockdown and CRISPR-Cas9-mediated knockout approa

Programmed cell death of keratinocytes: an active driver in psoriasis.

Psoriasis is a common, chronic inflammatory skin disorder with a high prevalence across all age groups, imposing a substantial burden on both individuals and society. It is characterized by epidermal hyperplasia and infiltration of immune cells into the dermis. Within the psoriatic microenvironment, epidermal keratinocytes contribute to a self-amplifying inflammatory response through the generation of "feed-forward" inflammation. Contrary to the earlier view of keratinocytes as passive "victims" in psoriasis pathogenesis, growing evidence supports their role as "active drivers" of disease prog

T-bet-expressing Tr1 cells driven by dietary signals dominate the small intestinal immune landscape.

Intestinal immunity defends against enteric pathogens, mediates symbiotic relationships with the resident microbiota, and provides tolerance to food antigens, safeguarding critical nutrient absorption and barrier functions of this mucosal tissue. Despite the abundance of tissue resident activated T cells, their contributions to these various roles remain poorly understood. Here, we identify a dominant population of IL-10 producing, T-bet-expressing Tr1 T cells, residing in the small intestinal lamina propria at homeostasis. Remarkably, these intestinal Tr1 cells emerge at the time of weaning a

A tissue-intrinsic mechanism sensitizes HIV-1 particles for TLR-triggered innate immune responses.

In vivo, HIV-1 replicates within tissues, yet the impact of three-dimensional (3D) environments on viral spread remains unclear. Our laboratory previously showed that collagen-rich 3D extracellular matrix (ECM) imposes an Environmental Restriction to cell-free Virus Infectivity (ERVI). Here, we demonstrate that ERVI is mediated by adhesive ECM components assembled into tissue-like scaffolds. Transient interactions with collagen fibers rapidly diminish virion infectivity across diverse primary strains by impairing virus fusogenicity. Notably, collagen-experienced particles also induce a distinc

The Role of Autophagy in HIV Infection and Immunological Recovery of ART-Treated PLWH.

Human immunodeficiency virus (HIV) is responsible for acquired immunodeficiency syndrome (AIDS), a condition characterized by the depletion of CD4+ T lymphocytes, which predisposes individuals to opportunistic infections and, ultimately, death. Although antiretroviral therapy (ART) has substantially improved clinical outcomes, certain limitations persist. Notably, 15-30% of individuals undergoing ART achieve viral suppression but fail to restore adequate CD4+ T cell counts, being defined as immunological non-responders (INR) and remaining at increased risk of disease progression to AIDS. Functional and Metabolic Heterogeneity of Dendritic Cells in Self-Tolerance and Autoimmunity.

Dendritic cells (DCs) demonstrate remarkable functional and metabolic heterogeneity that governs the balance between immune tolerance and autoimmune pathogenesis. Under homeostatic conditions, tolerogenic DC subsets maintain immunological equilibrium through distinct metabolic programs and the production of immunoregulatory metabolites, promoting T cell anergy and regulatory T cell (Treg) differentiation. In contrast, autoimmune conditions trigger pathogenic metabolic rewiring, shifting DCs toward glycolysis and enhanced lipid synthesis, which drives DC hyperactivation and breakdown of self-to

Human brain cell types shape host-rabies virus transcriptional interactions revealing a preexisting pro-viral astrocyte subpopulation.

How virus-host cell interactions and innate immune antagonism shape neurotropic infection dynamics across diverse brain cell types are largely unknown. To "unmask" and study how innate immune inhibition affects cell type-specific transcriptional regulation of the human and viral genome, we performed single-cell RNA sequencing of human brain cell co-cultures, comparing an isolate of rabies virus (RABV) to its mutant incapable of antagonizing interferon- and NF-κB-dependent responses. RABV gene expression was shaped by host cell type. RABV induced small-scale, cell-type conserved transcriptional

The temporal progression of lung immune remodeling during breast cancer metastasis.

Tumor metastasis requires systemic remodeling of distant organ microenvironments that impacts immune cell phenotypes, population structure, and intercellular communication. However, our understanding of immune phenotypic dynamics in the metastatic niche remains incomplete. Here, we longitudinally assayed lung immune transcriptional profiles in the polyomavirus middle T antigen (PyMT) and 4T1 metastatic breast cancer models from primary tumorigenesis, through pre-metastatic niche formation, to the final stages of metastatic outgrowth at single-cell resolution. Computational analyses of these da

High-grade serous ovarian cancer development and anti-PD-1 resistance is driven by IRE1α activity in neutrophils.

High-grade serious ovarian cancer (HGSOC) is an aggressive malignancy that remains refractory to current immunotherapies. While advanced stage disease has been extensively studied, the cellular and molecular mechanisms that promote early immune escape in HGSOC remain largely unexplored. Here, we report that primary HGSO tumors program neutrophils to inhibit T cell anti-tumor function by activating the endoplasmic reticulum (ER) stress sensor IRE1α. We found that intratumoral neutrophils exhibited overactivation of ER stress response markers compared with their counterparts at non-tumor sites.

Mitochondrial defects in sporadic inclusion body myositis-causes and consequences.

Sporadic inclusion body myositis (sIBM) is a distinct subcategory of Idiopathic Inflammatory Myopathies (IIM), characterized by unique pathological features such as muscle inflammation, rimmed vacuoles, and protein aggregation within the myofibers. Although hyperactivation of the immune system is widely believed as the primary cause of IIM, it is debated whether non-immune tissue dysfunction might contribute to the disease's onset as patients with sIBM are refractory to conventional immunosuppressant treatment. Moreover, the findings that mitochondrial dysfunction can elicit non-apoptotic prog

Comparative Investigation of Coincident Single Nucleotide Polymorphisms Underlying Avian Influenza Viruses in Chickens and Ducks.

Avian influenza is a severe viral infection that has the potential to cause human pandemics. In particular, chickens are susceptible to many highly pathogenic strains of the virus, resulting in significant losses. In contrast, ducks have been reported to exhibit rapid and effective innate immune responses to most avian influenza virus (AIV) infections. To explore the distinct genetic programs that potentially distinguish the susceptibility/resistance of both species to AIV, the investigation of coincident SNPs (coSNPs) and their differing causal effects on gene functions in both species is imp
