Functional Assessment of Hypertrophic Phenotype Cardiomyopathies Using Combined Cardiopulmonary Exercise Testing and Echocardiography: A Pilot Single-Centre Study.

Background: In patients with left ventricular hypertrophy, resting structural parameters alone may not explain exertional symptoms. Hence, we investigate whether combined Cardiopulmonary Exercise Testing- Exercise Stress Echocardiography (CPET-ESE) can provide an integrated functional characterisation of hypertrophic phenotypes. Methods: As a preliminary investigation, this prospective single-centre pilot study enrols 43 patients, categorised into: obstructive hypertrophic cardiomyopathy (n = 19), transthyretin cardiac amyloidosis (n = 15), or preserved-ejection-fraction hypertrophic phenotype

Recent Progress in Hydrogen-mediated Neuroprotection via Modulation of Mitochondrial Quality Control Mechanisms in Brain Injury.

Brain injury is a leading cause of mortality and long-term disability worldwide, characterized by energy metabolism dysfunction, oxidative stress, inflammatory responses, and programmed cell death, with mitochondrial dysfunction serving as a central pathological nexus. In recent years, hydrogen, as an emerging gaseous signaling molecule, has demonstrated remarkable neuroprotective effects in various experimental models of brain injury owing to its unique biological properties, including selective antioxidant, anti-inflammatory, anti-apoptotic, and mitochondrial- protective activities. Neurocardiac consequences of traumatic brain injury: integrating neuroimmune, autonomic, and cerebrovascular mechanisms.

Traumatic brain injury (TBI) is increasingly recognized as a systemic disorder with consequences that extend beyond the central nervous system (CNS) to include clinically relevant cardiac dysfunction. Clinical and experimental evidence indicate that TBI is associated with arrhythmias, myocardial injury, autonomic imbalance, and impaired cardiac performance, even in the absence of primary cardiac disease. These observations support the concept of a brain-heart axis through which neural injury influences cardiovascular regulation. Current evidence shows that multiple interacting mechanisms contr

Ultrasound-activated RuO2 amplified oxidative stress therapy.

Malignant tumors have limited therapeutic options, primarily due to the lack of actionable molecular targets and the redox-adaptive tumor microenvironment. Herein, a RuO2 sonosensitizer is developed to enable cascade-amplified oxidative stress therapy. Upon US stimulation, RuO2 generates a measurable sonoelectric response (∼12 nA cm-2), which promotes charge separation and accelerates catalytic electron transfer, thereby enhancing endogenous H2O2 decomposition and oxygen evolution. Notably, this process triggers excessive generation of reactive oxygen species (ROS), including ˙OH and ˙O2 -, wh

Cellular hitchhiking of selenium carbon dots for targeted macrophage polarization in acute lung injury.

Acute lung injury (ALI) is a life-threatening pulmonary disorder characterized by dysregulated inflammatory responses, excessive activation of pro-inflammatory M1 macrophages, severe oxidative stress, and potentially fatal cytokine storms. Conventional therapies are severely limited by poor targeting of the inflammatory microenvironment and insufficient cellular uptake by hyperactivated immune cells. To address these translational barriers, we developed a "cellular hitchhiking" nanoplatform by coating selenium-doped carbon quantum dots (SC) with bacterial outer membrane vesicles (OMVs) derived

A Perfect Storm of Pollutants: Environmental Mixtures in the Pathogenesis of Atherosclerosis.

Atherosclerosis is a complex, multifactorial disease and the leading underlying cause of cardiovascular morbidity and mortality worldwide. Beyond traditional risk factors, growing evidence highlights the critical role of environmental exposures in modulating atherogenesis. In real-world scenarios, individuals are exposed to complex mixtures of contaminants, including particulate matter, toxic metals, pesticides, polycyclic aromatic hydrocarbons, and endocrine-disrupting chemicals. These combined exposures may exert additive, synergistic or antagonistic effects, resulting in biological response

Endoplasmic reticulum stress in Hashimoto's thyroiditis: a candidate amplification node linking thyroid-specific vulnerability and immune dysregulation.

Hashimoto's thyroiditis (HT) is the most prevalent autoimmune thyroid disease, characterized by lymphocytic infiltration and destruction of thyroid follicular architecture. Prior research has predominantly focused on aberrant adaptive immune activation, whereas the contribution of the thyroid follicular cell itself to this process remains incompletely elucidated. This narrative review synthesizes evidence implicating endoplasmic reticulum stress (ERS) in HT pathogenesis, stratifying the evidence by its proximity to HT pathology into three tiers: direct evidence, thyroid-cell evidence, and extr

Exploration of the efficacy of eucalyptus oil (micro-capsules) and mangosteen extract against Eimeria tenella infection in chickens.

Due to the increasing prevalence of drug-resistant Eimeria strains, there is an urgent need to develop novel, eco-friendly, and safe alternatives for controlling chicken coccidiosis. This study aimed to investigate the combined anticoccidial efficacy of eucalyptus oil (EO) and mangosteen extract (ME) and develop an advanced delivery system via microencapsulation to enhance their therapeutic potential. In a broiler chick model of Eimeria tenella (E. tenella) infection, the combination of EO (100 mg/kg body weight, oral gavage) and ME (250 mg/kg feed) exerted the optimal effect against E. tenell

Therapeutic transformation of a rhodium nanophotosensitizer into a tamoxifen-loaded targeted nano-drug for dual chemo-photodynamic therapy of ER-positive breast cancer.

Rhodium-based nanophotosensitizers have recently emerged as promising platforms for near-infrared (NIR)-activated photodynamic therapy (PDT). However, their therapeutic potential remains largely restricted to reactive oxygen species-mediated cytotoxicity, limiting their ability to address the complex molecular drivers of tumor progression. Here, we report the therapeutic transformation of a previously validated transferrin-targeted rhodium nanophotosensitizer into a dual-action nano-therapeutic by incorporating the selective estrogen receptor modulator tamoxifen within the mesoporous silica ma

Research progress on the therapeutic effects of effective components of traditional Chinese medicine in the treatment of gastric cancer precursors through modulation of multiple signaling pathways.

Precancerous lesions of the stomach (PLGC), a critical stage in gastric cancer development, have garnered significant attention for its prevention and treatment. PLGC refers to a series of pathological and histological changes preceding cancer. Due to its complex pathogenesis and multi-factor influence, no currently existing drug can effectively prevent or delay this process. Therefore, identifying a safe and effective treatment method remains an important research focus. In recent years, traditional Chinese medicine has demonstrated unique advantages and potential in PLGC treatment. Many stud

Multi-functionalized probiotics through layer-by-layer coating with tannic acid-Mg2+ and casein phosphopeptide complexes for preventing ulcerative colitis.

Gut microbiota imbalance-induced inflammatory response and oxidative stress are two of the main reasons causing ulcerative colitis (UC). Probiotics show potent modulating effects on microbiota imbalance and have been considered as an optimal substitute of antibiotics for preventing UC. However, the harsh environment of the gastrointestinal tract is not conducive to the survival and persistence of probiotics. Herein, we developed an efficient surface coating strategy to overcome the delivery challenges of probiotics and also endow them with multiple functions through layer-by-layer coating with

Microfluidics-enabled polydopamine-coated selenium nanoparticles in hyaluronic hydrogel microspheres for targeted antioxidant and immunomodulatory therapy of ulcerative colitis.

Ulcerative colitis (UC) is manifested by excessive oxidative stress and immune dysregulation in the colonic microenvironment. To address these pathological features, we developed a microfluidics-engineered oral delivery system comprising methacrylated hyaluronic acid (HAMA) hydrogel microspheres encapsulating polydopamine-coated selenium nanoparticles (CS-Se@PDA). The uniform microspheres featured a microfluidics-enabled structure that enabled stable CS-Se@PDA nanoparticles anchoring via electrostatic and covalent interactions. The PDA coating enhanced the stability, mucosal adhesion, and scav

Pathogenic OTUD3 Mutations Predispose to Ulcerative Colitis Due to Barrier Dysfunction.

BACKGROUND & AIMS: The contribution of common genetic polymorphisms to ulcerative colitis (UC) pathogenesis is modest; however, families with severe colitis may harbor rare variants with large effect sizes that highlight unrecognized pathways. METHODS: A multigenerational family with UC necessitating colectomy was identified. Whole exome sequencing of this kindred was performed, implicating a rare variant in OTUD3. Constitutive knock-out and intestinal specific Otud3 deficient and heterozygous mice were generated. OTUD3 expression in human colonic biopsies and intestinal organoids was assesse

Unraveling the Role of LRP1 in Alzheimer's Disease: A Focus on Aβ Clearance and the Liver-Brain Axis.

Alzheimer's disease (AD) is the most prevalent form of dementia, significantly contributing to the global health burden. The progressive accumulation of amyloid-beta (Aβ) plaques and tau tangles triggers neuroinflammation, oxidative stress, and neuronal damage, highlighting the critical need for effective clearance mechanisms. Recent research has identified low-density lipoprotein receptor-related protein 1 (LRP1) as a key factor in the regulation of Aβ clearance, neuroinflammation, and blood-brain barrier integrity, particularly in relation to the liver-brain axis. This review provides a comp
