Rhein-Emodin Combination Activates UQCRC1 to Attenuate MPP+-Induced Ferroptosis in Dopaminergic Neurons.

Ferroptosis plays a critical role in Parkinson's disease (PD). This study investigates the neuroprotective effect of a combination of two natural anthraquinones, rhein and emodin, against ferroptosis, focusing on the mitochondrial complex III subunit UQCRC1. Using network pharmacology and experimental validation in erastin/MPP+-induced PC12 cells, we found that the optimal rhein-emodin combination potently inhibited ferroptotic cell death. This effect was associated with restored mitochondrial function, reduced lipid peroxidation, and the coordinated regulation of ferroptosis-related proteins,

Dual Metabolic Blockade in Pancreatic Cancer: Potent Anticancer Activity of Mitochondria-Targeted Glycolysis and OXPHOS Inhibitors.

Simultaneous targeting of glycolysis and oxidative phosphorylation (OXPHOS) is an effective strategy for overcoming the metabolic plasticity of pancreatic ductal adenocarcinoma (PDAC). In this study, we present compound 14c, a rationally designed, mitochondria-targeted small molecule that disrupts PDAC energy metabolism. Compound 14c markedly inhibited PDAC cell glycolysis and mitochondrial function, as evidenced by the PDKs inhibition and the downregulation of OXPHOS-associated proteins, including SDHB and SIRT3, in vitro and in vivo. Mechanistically, 14c induced ferroptotic cell death, accom

Immunotherapy and ferroptosis regulation in thyroid cancer: Novel strategies and future perspectives.

Thyroid cancer (TC) is a prevalent malignancy of the endocrine system, and therapeutic strategies for this disease have continued to evolve. As an emerging therapeutic approach, immunotherapy has demonstrated promising efficacy and a favorable safety profile in select patients with refractory, poorly differentiated, or advanced TC; however, the overall response rate remains suboptimal and requires further improvement. Ferroptosis, characterized as an iron-dependent and lipid peroxidation-driven form of programmed cell death, has recently been shown to engage in complex and synergistic interact

Targeted extracellular degradation of LRP8 promotes ferroptosis in cancer cells.

Tumor reliance on antioxidant defenses creates a vulnerability to ferroptosis, yet strategies to therapeutically disable these systems remain limited. Here, we identify targeted degradation of the selenium uptake receptor lipoprotein receptor-related protein 8 (LRP8) as an effective approach to decrease the abundance of the ferroptosis-protective enzyme glutathione peroxidase 4 (GPX4). Using bispecific cytokine receptor-targeting chimeras (KineTACs) that couple LRP8 to cytokine receptor internalization pathways, we selectively direct LRP8 to the lysosome for degradation. LRP8 degradation reduc

Qualitative and flow cytometric evaluations of goat's frozen semen in Mito-TEMPO supplemented plant-based extender.

BACKGROUND: Cryopreservation of buck spermatozoa is an efficient strategy to improve qualified sperm cells for artificial insemination; however, this process potentially decreases the qualitative characteristics of post-thawed sperm. Therefore, this research aimed to evaluate the influence of the mitochondria-targeted antioxidant "Mito-TEMPO" on buck sperm quality during the cryopreservation process in a plant-based extender. RESULTS: This study indicated that the dosages 10 and 100 µM Mito-TEMPO presented higher (P ≤ 0.05) percentages of total motility, progressive motility, membrane integri

Bisphenol A-induced toxicity in Drosophila melanogaster: Mitigation by vanillic acid and modulation of CncC/Keap1 pathway gene expression.

Bisphenol A (BPA) is a pervasive endocrine-disrupting chemical with documented toxic effects in living organisms. This study evaluated the protective potential of vanillic acid (VA) against BPA-induced toxicity in Drosophila melanogaster. BPA exposure elevated lipid peroxidation (MDA), reduced antioxidant defenses (SOD and CAT activities and GSH content), increased DNA damage (Comet assay), lowered acetylcholinesterase levels, and impaired developmental parameters (pupation, climbing, crawling, lifespan, and food intake). BPA also upregulated SOD, CAT, gclc, hsp70, CncC, and Keap1 mRNA express

Physico-chemical properties of edible starch-based films with herbal extracts: cistus and white mulberry and their application as active sachets for oily products.

The aim of the work was to prepare films based on potato thermoplastic starch (TPS) with herbal leaf extracts: from cistus (Cistus incanus L.- C) and white mulberry (Morus alba L.- M). The influence of the extracts on physico-chemical and bioactive properties of the films obtained via casting method was tested and compared. Starch gelatinization in the presence of the herbal infusion (5, 10 and 20% in the gelatinizing media) led to better mechanical and barrier (towards moisture, oxygen and UV) properties of the final TPS films. The modified films exhibited high antioxidative (DPPH scavenging

Mechanistic investigation of sucrose monostearate-mediated antifungal effects against Penicillium oxalicum.

Penicillium oxalicum is a major postharvest pathogen of kiwifruit, causing substantial economic losses. This study investigated the antifungal activity and mechanism of sucrose monostearate (SMS) against P. oxalicum. The minimum inhibitory concentration (MIC) of SMS was determined to be 1.25 g L- 1, which significantly inhibited spore germination by 88.28% and reduced mycelial growth by 65.94% (p < 0.05). Mechanistically, SMS at 1.25 g L- 1 induced intracellular ROS accumulation by 1.7-fold, suppressed catalase (CAT) and peroxidase (POD) activities by 54% and 60%, and increased malondialdehyde

Mesenchymal Stem Cells Attenuate Diabetic Nephropathy by Suppressing the ERK-Ferroptosis-ROS Axis.

Diabetic nephropathy (DN) is a leading cause of end-stage renal disease with limited therapeutic options. Ferroptosis contributes to renal tubular injury in DN. This study investigates whether mesenchymal stem cells (MSCs) ameliorate DN by inhibiting ferroptosis and elucidates the underlying mechanism. In a rat model of type 2 DN, MSCs transplantation improved renal function and histopathology, while reducing mitochondrial dysfunction, iron overload, and ROS-driven ferroptosis. In vitro, MSCs reversed high glucose-induced ferroptosis hallmarks in tubular epithelial cells. Mechanistically, RNA

Subchronic Exposure to Nonylphenol Ethoxylate at Environmentally Relevant Level Promotes Mutagenic, Neurotoxic, and Pro-oxidant Effects in Bullfrog Tadpoles.

Nonylphenol compounds released into aquatic environments have been shown to exert deleterious effects on aquatic organisms. Amphibians, particularly during larval stages, are highly vulnerable to chemical stressors and are chronically exposed to pollutants such as nonylphenol ethoxylate (NPE). This study evaluated the sublethal effects of environmentally relevant concentrations of NPE (30 µg L−1) under sub-chronic exposure in Aquarana catesbeiana tadpoles (Gosner stage 25), with emphasis on mutagenic, neurotoxic, and oxidative responses. The analyses included blood cell composition, frequency

Pathogenesis and role of ferroptosis mediated by Nrf2 signaling pathway in ischemic stroke.

Ferroptosis, a novel mode of programmed cell death, plays a significant role in neurological injury following ischemic stroke (IS). Nuclear factor E2-related factor 2 (Nrf2), a central transcription factor that combats oxidative stress, is a critical target for inhibiting ferroptosis by modulating iron metabolism, lipid peroxidation, and the glutathione system. In this paper, we systematically review the molecular mechanisms, signaling pathways, and potential therapeutic strategies, including pharmacological activators like dimethyl fumarate (DMF), natural compounds, and nanotechnology-based d

Protection by maslinic acid against cisplatin-induced ototoxicity: rescue of ferroptosis by targeting the SLC7A11-GSH-GPX4 pathway.

Cisplatin (CP) chemotherapy often causes ototoxicity, a major side effect. Maslinic acid (MA), a pentacyclic triterpenoid with known anti-inflammatory and antioxidant properties, was investigated for its protection against CP-induced hearing loss. In vivo, MA maintained the hearing threshold of CP-treated mice at 50-60 dB SPL and prevented damage to the stria vascularis and spiral ganglion. Proteomic analysis indicated that MA reduces ototoxicity by inhibiting oxidative stress and ferroptosis pathways. In vitro, MA counteracted the loss of HEI-OC1 cell viability induced by CP or the ferroptosi

Schwann Cell TRPA1, a Proalgesic Ion Channel, Mediates Neuroinflammation and Fibromyalgia-Associated Behaviours in Mice.

Transient receptor potential ankyrin 1 (TRPA1) is an ion channel that integrates the somatosensory system and is specialised in detecting thermal, mechanical, and chemical stimuli. It acts as both a sensor and an amplifier of reactive oxygen and nitrogen species, carbonylic species and lipid peroxidation products, which are overproduced in several painful conditions, including fibromyalgia. Studies have linked TRPA1 to heightened sensitivity to mechanical and cold pain in fibromyalgia patients. In a preclinical mouse model of fibromyalgia induced by reserpine administration, activated Schwann

BMAL1-GPX3 axis in the choroid plexus mitigates Aβ pathology in an amyloid mouse model.

Alzheimer’s disease (AD) is the most common neurodegenerative disorder, with circadian rhythm disturbances strongly linked to its pathogenesis. The choroid plexus (ChP) is a circadian-regulated structure in the brain ventricles, but the role of the core clock gene brain and muscle ARNT-like protein 1 (BMAL1) in ChP in relation to AD pathology remains unclear. Here, we report that knockdown of Bmal1 in ChP epithelial cells of 5xFAD mice alleviates amyloid-β (Aβ) pathology, primarily by improving the function of astrocytes and border-associated macrophages (BAMs), with the latter potentially med

MircoRNAs as regulators of regulated cell death in cancer progression: a focus on ferroptosis, anoikis and pyroptosis.

Cancer progression is shaped not only by genetic alterations but also by dynamic remodeling of regulated cell death. MicroRNAs (miRNAs) have emerged as central regulators of these processes by coordinating gene expression, metabolic balance, and stress responses. This review summarizes current understanding of how miRNAs modulate ferroptosis, anoikis, and pyroptosis across liver, lung, gastric, breast, and other cancers. They influence ferroptosis through lipid peroxidation and antioxidant systems, regulate anoikis via integrin signaling and cytoskeletal remodeling, and tune pyroptosis through

Peroxynitrite: A potential therapeutic target for Alzheimer's disease.

The pathogenesis of Alzheimer's disease (AD) involves multiple pathological processes, including β-amyloid (Aβ) deposition, aberrant tau modification, oxidative stress, and neuroinflammation. The molecular interaction mechanism among them remains to be further clarified. Peroxynitrite (ONOO⁻) is a potent oxidizing and nitrating reactive nitrogen species formed from the rapid reaction between nitric oxide and superoxide. It mediates protein nitration, lipid peroxidation, and mitochondrial damage, thereby contributing to multiple pathological events in AD. This review outlines the physicochemica

A Review of the Potential Use of Antioxidants in Spinal Cord Injuries.

Spinal cord injury (SCI) is a debilitating neurological condition marked by primary mechanical damage followed by a complex secondary injury cascade, in which oxidative stress plays a central role. Mitochondrial dysfunction, ionic imbalance, and inflammatory responses drive excessive generation of reactive oxygen and nitrogen species, leading to lipid peroxidation, protein and DNA damage, apoptosis, and progressive neurological impairment. Antioxidant-based therapies have emerged as promising neuroprotective strategies, with compounds such as A91 peptide, curcumin, edaravone, ginsenosides, and

Silica-induced ferroptosis activates retinoic acid signaling in dendritic cells to drive inflammation and fibrosis in silicosis.

Silicosis, a chronic lung disease caused by inhalation of silica (SiO2) particles from environmental contamination or industrial exposure, is characterized by persistent inflammation and fibrosis. This study elucidates a novel mechanism where SiO2 exposure triggers ferroptosis, a lipid peroxidation-dependent form of cell death, in dendritic cells (DCs), thereby activating retinoic acid (RA) signaling. The RA response amplifies inflammatory pathways, including cGAS-STING-IFN-I and IL-1β signaling, exacerbating lung inflammation and fibrosis. The study uses murine models to demonstrate that ferr
