A new simple method for isolation of microvascular endothelial cells avoiding both chemical and mechanical injuries.

Our study indicates that when small pieces of lung or muscles of chest wall are cultured, erythrocytes and leukocytes (PMNs) leave the tissues first, followed by vascular endothelial cells (ECs). Fibroblasts and other mixed cells grow after 72 hr culture. The ECs can then be isolated avoiding mechanical and chemical injuries. The lung tissue is obtained from the peripheral surface and muscles from the chest. It is then cut into pieces and cultured with DMEM containing 20% fetal bovine serum. After 60 hr culture, the tissues are discarded. The flask contains only ECs and blood cells. Blood cell

Altered pulmonary microvascular reactivity to norepinephrine in canine pacing-induced heart failure.

Pulmonary hypertension in congestive heart failure causes medial hypertrophy in pulmonary vessels and thickening of the endothelial basement membrane. In this study, the functional consequences of such pulmonary vascular adaptations were evaluated. Heart failure was induced in dogs by rapid ventricular pacing (240 beats per minute) for 28 days, at which time left ventricular shortening fraction was decreased by 57% compared with that at baseline. Lung lobes from paced (n = 56) and control dogs (n = 68) were isolated and perfused with autologous blood. Total, arterial (Ra), and venous (Rv) vasc

Evidence that portal tract microvascular destruction precedes bile duct loss in human liver allograft rejection.

In liver allograft rejection, interlobular bile ducts are thought to be the main target of rejection. In contrast, in other organ allografts the capillary bed of the graft appears to be the primary target. To determine whether portal tract microvasculature is destroyed in liver allografts during rejection, we have identified portal tract microvasculature in 11 normal livers and 38 liver allograft biopsy specimens using monoclonal antibodies to capillary endothelium in immunohistochemical staining. E1.5, CD31 and EL-4 antibodies identified portal microvascular endothelium in normal liver that h

Hyperoxic sheep pulmonary microvascular endothelial cells generate free radicals via mitochondrial electron transport.

Free radical generation by hyperoxic endothelial cells was studied using electron paramagnetic resonance (EPR) spectroscopy and the spin trap 5,5-dimethyl-1-pyrroline-N-oxide (DMPO). Studies were performed to determine the radical species produced, whether mitochondrial electron transport was involved, and the effect of the radical generation on cell mortality. Sheep pulmonary microvascular endothelial cell suspensions exposed to 100% O2 for 30 min exhibited prominent DMPO-OH and, occasionally, additional smaller DMPO-R signals thought to arise from the trapping of superoxide anion (O2-.), hyd

Diabetic cutaneous microangiopathy.

PURPOSE: To determine a potential relationship between skin blood flow changes and the duration of diabetes and the presence of other microvascular complications. PATIENTS AND METHODS: Skin blood flow was measured by laser Doppler techniques at the finger and toe pulps, areas of predominant arteriovenous anastomotic (AVA) flow, and on the finger and toe dorsums, which have a greater nutritive microvascular contribution, in 83 diabetic patients and 39 nondiabetic control subjects. The average duration of diabetes was 14 +/- 1 years. Thirty-four patients had retinopathy. Eighteen patients had p

Possible mechanisms of type I collagen-induced vascular tube formation.

UNLABELLED: We investigated the effect of type I collagen on endothelial behaviour following its contact with the apical versus basal surface of cultured human endothelial cells. When endothelial cells were plated onto type I collagen they attached via their basal surface and formed a confluent monolayer. However, when type I collagen (100 micrograms/ml) was added directly to the growth medium, so that it made contact with the apical surface of endothelial cells, it induced rapid capillary-like tube formation. Possible mechanisms were assessed using a) polyclonal (anti-VLA-2) and monoclonal (A

Esculine, ranitidine and carbenoxolone: different modes of action on gastric mucosa.

1. This study was designed to determine the antiulcerogenicity of esculine in various types of experimentally induced gastric ulcers in which the appearance of lesions is due to an ischemic process: cold-restraint stress and pylorus-ligated induced ulcers. 2. In the first experimental model, esculine (50 mg/kg) produced a significant diminution not only in the number of haemorrhagic stomachs (21.5% by 37.5% of the controls) but also in the ulcer index, U.I. (1.00 +/- 0.63, P less than 0.05). 3. When the mucosal damage was induced as a consequence of the pylorus-ligated gastric secretion, pretr

[Diagnostic contribution of the new imaging systems in arterial and nerve disorders of the posterior fossa].

Giant aneurysms of the vertebrobasilar system are rare, representing only 5% of intracranial aneurysms and are the cause of severe pseudo-tumoral of hemorrhagic syndromes. Their endovascular or surgical treatment is always very dangerous. The basilar megadolicho-artery, of congenital origin, is frequently expressed by a hemispasm or facial neuralgias. The diagnosis is made easily by TDM and angiography. Abstaining from therapy is the rule. The much debated vascular rings are often held responsible for the microvascular compression syndrome, leading to facial neuralgias, hemispasm, Ménièrie-for

Effects of barbiturates, phencyclidine, ketamine and analogs on cerebral circulation and cerebrovascular muscle.

Although barbiturates are often effective as therapeutic agents in several types of brain ischemia, there is no consensus as to their mechanisms of action. Exactly why other intravenous anesthetics such as ketamine are not effective therapies in brain ischemia is not known. Structural analogs of ketamine such as phencyclidine (PCP) not only exert potent hallucinogenic properties and are widely abused drugs, but often result in hypertensive encephalopathies and death. In view of the paucity of information on the cerebral circulatory actions of barbiturates, ketamine and PCP (and analogs), in-vi

Intraneural tissue reactions induced by internal neurolysis. An experimental study on the blood-nerve barrier, connective tissues and nerve fibres of rabbit tibial nerve.

Surgical treatment of peripheral nerve lesions associated with intraneural fibrosis is sometimes extended to include internal neurolysis. This procedure is performed in order to release the individual nerve fascicles from interfascicular scar tissue which is believed to constrict nerve fibres and thereby interfere with their function and regenerative capacity. However, an internal neurolysis per se implies a significant trauma to the nerve and may induce microvascular damage and formation of new intraneural scar tissue. Considering this, such a procedure appears justified only when the preoper

Intramyocardial small-vessel disease in chronic alcoholism.

A morphologic study of the small (50 to 200 micron) intramyocardial coronary arteries was performed. The cases chosen for study were selected from a relatively young group of patients without clinical evidence of alcoholic cardiomyopathy or pathologic evidence of large coronary artery disease, in order to evaluate alterations in the small vessels which could possibly be attributed to the chronic alcoholic state. Five basic vascular abnormalities were described. The most common alteration found in all nine cases was vascular wall edema (48 per cent), followed by perivascular fibrosis (42 per ce

Clinical applications of free flap transfer in the burn patient.

Free microvascular flap transfer is indicated in situations in which flap coverage is required and no local flap is available. The technique of transfer of a groin flap based on the superficial circumflex iliac vessels was described. The procedure has been applied to two patients with acute electrical burns with extensive soft-tissue loss, three patients with scar contractures, and one with osteoradionecrosis. Satisfactory results were achieved in all patients. The procedure is contraindicated if recipient or donor vessels are abnormal or inadequate, and in the face of fulminating infection, g

Insulin resistance and coronary microvascular dysfunction: a complex interplay.

Up to 40-60% of patients undergoing coronary angiography because of angina and/or evidence of inducible ischaemia on non-invasive stress testing are diagnosed with ischaemia and non-obstructive coronary artery disease (INOCA). The pathogenesis of this condition is primarily attributed to two mechanisms: coronary microvascular dysfunction (CMD) and coronary vasospasm. Notably, ischaemic heart disease is the leading cause of death in patients with type 2 diabetes mellitus (T2DM). Insulin resistance (IR), affecting 10-25% of the general adult population, plays a major role in the pathophysiology

Early detection of glycocalyx and microvascular damage in suspected sepsis in the emergency department: the EDGE study.

BACKGROUND: The prompt identification of clinical deterioration in emergency department (ED) patients presenting with infection is crucial yet challenging. Microvascular dysfunction has been linked to poor clinical outcome in critically ill patients, but it remains unclear whether its detection can predict clinical deterioration in early sepsis. This study aims to evaluate the utility of quantitative microvascular videomicroscopy for predicting clinical deterioration in patients with suspected sepsis. METHODS: In this prospective observational study, 299 ED patients with suspected infection o

Chronic Kidney Disease, Circulating Ceramides, and Coronary Microvascular Dysfunction: Insights from the CRUISE-MET Study.

INTRODUCTION: Ceramides and chronic kidney disease (CKD) independently associate with coronary microvascular dysfunction (CMD), but their combined association remains unestablished, warranting investigation. METHODS: This cross-sectional analysis included coronary artery disease (CAD) patients from the CRUISE-MET trial (NCT06383208) who underwent coronary angiography and ceramide profiling measurement. The microvascular resistance was assessed in all patients using the angiography-derived microcirculatory resistance (AMR). CKD was classified based on both estimated glomerular filtration rate

Percutaneous coronary interventions improve phasic cardiac-coronary interaction in chronic coronary syndromes as assessed by wave intensity analysis.

BACKGROUND: Clinical benefit of percutaneous coronary intervention (PCI) in chronic coronary syndromes (CCS) remains controversial. Coronary wave intensity analysis (WIA) provides mechanistic insights into cardiac-coronary coupling. OBJECTIVES: To assess peri-PCI WIA changes at rest and during hyperaemia, and to identify features associated with improvements in epicardial and microvascular resistance. METHODS: Intracoronary Doppler and pressure measurements were analysed in 27 CCS patients undergoing elective PCI. Haemodynamics (fractional flow reserve (FFR), basal and hyperaemic stenosis re

Association of the TyG index and inflammatory burden index with AMR-defined coronary microvascular dysfunction and 12-month MACE after STEMI.

BACKGROUND: Metabolic dysfunction and inflammation may jointly contribute to coronary microvascular injury and adverse outcomes after primary percutaneous coronary intervention (PPCI) for ST-segment elevation myocardial infarction (STEMI). We examined the independent and combined associations of the triglyceride-glucose (TyG) index and inflammatory burden index (IBI) with angiographic microvascular resistance (AMR)-defined coronary microvascular dysfunction (CMD), their incremental value beyond conventional factors, and their associations with 12-month major adverse cardiovascular events (MACE
