Sensitivity of Ablation Targets Prediction to Electrophysiological Parameter Variability in Image-Based Computational Models of Ventricular Tachycardia in Post-infarction Patients.

Ventricular tachycardia (VT), which could lead to sudden cardiac death, occurs frequently in patients with myocardial infarction. Computational modeling has emerged as a powerful platform for the non-invasive investigation of lethal heart rhythm disorders in post-infarction patients and for guiding patient VT ablation. However, it remains unclear how VT dynamics and predicted ablation targets are influenced by inter-patient variability in action potential duration (APD) and conduction velocity (CV). The goal of this study was to systematically assess the effect of changes in the electrophysiol

Development of a Functionally Equivalent Model of the Mitral Valve Chordae Tendineae Through Topology Optimization.

Ischemic mitral regurgitation (IMR) is a currently prevalent disease in the US that is projected to become increasingly common as the aging population grows. In recent years, image-based simulations of mitral valve (MV) function have improved significantly, providing new tools to refine IMR treatment. However, clinical implementation of MV simulations has long been hindered as the in vivo MV chordae tendineae (MVCT) geometry cannot be captured with sufficient fidelity for computational modeling. In the current study, we addressed this challenge by developing a method to produce functionally eq

Research on Load Reverse Engineering and Vibration Fatigue Analysis Technology of Rapid Box Wagon.

The overall stiffness and modal frequency of the car body of a rapid box car are reduced by the design of the full-side open movable side door structure. The vibration fatigue performance of the welded structure in this car body needs to be verified. The rigid-flexible coupling model of the rapid box wagon was established first, and the model was verified by modal test data. By the application of the virtual iteration method on this model, the displacement excitation loads of this vehicle were acquired. The effectiveness of the load reverse obtaining technology was verified through the compari

Ryanodine receptor cluster fragmentation and redistribution in persistent atrial fibrillation enhance calcium release.

AIMS: In atrial fibrillation (AF), abnormalities in Ca(2+) release contribute to arrhythmia generation and contractile dysfunction. We explore whether ryanodine receptor (RyR) cluster ultrastructure is altered and is associated with functional abnormalities in AF. METHODS AND RESULTS: Using high-resolution confocal microscopy (STED), we examined RyR cluster morphology in fixed atrial myocytes from sheep with persistent AF (N = 6) and control (Ctrl; N = 6) animals. RyR clusters on average contained 15 contiguous RyRs; this did not differ between AF and Ctrl. However, the distance between clust

Examining Competing Factor Analysis Models of the PCL-5 Applied to the COVID-19 Pandemic.

This investigation scrutinized various competing factor structures and psychometric properties for the COVID-19 version of the Posttraumatic Stress Disorder Checklist for DSM-5 (COVID-19 PCL-5) to understand stress reactions to the pandemic. Participants (N = 985) completed this measure with instructions modified to exclusively refer to the COVID-19 pandemic as the stressful index event. Descriptive data showed that 76% of participants experienced at least one COVID-19 traumatic stress symptom. Confirmatory factor analysis evaluated various competing factor structures of the COVID-19 PCL-5: (a

One Size Fits All? Development of the CPOSS209 Data Set of Experimental and Hypothetical Polymorphs for Testing Computational Modeling Methods.

Organic crystal structure prediction (CSP) studies have led to the rapid development of methods for predicting the relative energies of known and computer-generated crystal structures. There is a compromise between the level of theoretical treatment, its reliability across different types of organic systems, how its accuracy depends on the size and shape of the unit cell, and the size and the number of structures that can be modeled at an affordable computational cost. We have used our database of crystal structure prediction studies, often performed as a complement to experimental screening,

A comparison between block and smooth modeling in finite element simulations of tDCS.

Current density distributions in five selected structures, namely, anterior superior temporal gyrus (ASTG), hippocampus (HIP), inferior frontal gyrus (IFG), occipital lobe (OCC) and pre-central gyrus (PRC) were investigated as part of a comparison between electrostatic finite element models constructed directly from MRI-resolution data (block models), and smoothed tetrahedral finite element models (smooth models). Three electrode configurations were applied, mimicking different tDCS therapies. Smooth model simulations were found to require three times longer to complete. The percentage differe

Modeling spike-wave discharges by a complex network of neuronal oscillators.

PURPOSE: The organization of neural networks and the mechanisms, which generate the highly stereotypical for absence epilepsy spike-wave discharges (SWDs) is heavily debated. Here we describe such a model which can both reproduce the characteristics of SWDs and dynamics of coupling between brain regions, relying mainly on properties of hierarchically organized networks of a large number of neuronal oscillators. MODEL: We used a two level mesoscale model. The first level consists of three structures: the nervus trigeminus serving as an input, the thalamus and the somatosensory cortex; the seco

Revisiting the continuum model of tendon pathology: what is its merit in clinical practice and research?

The pathogenesis of tendinopathy and the primary biological change in the tendon that precipitates pathology have generated several pathoaetiological models in the literature. The continuum model of tendon pathology, proposed in 2009, synthesised clinical and laboratory-based research to guide treatment choices for the clinical presentations of tendinopathy. While the continuum has been cited extensively in the literature, its clinical utility has yet to be fully elucidated. The continuum model proposed a model for staging tendinopathy based on the changes and distribution of disorganisation w

Follistatin lowers blood pressure and improves vascular structure and function in essential and secondary hypertension.

Hypertension is characterized by resistance artery remodeling driven by oxidative stress and fibrosis. We previously showed that an activin A antagonist, follistatin, inhibited renal oxidative stress and fibrosis in a model of hypertensive chronic kidney disease. Here, we investigate the effects of follistatin on blood pressure and vascular structure and function in models of essential and secondary hypertension. 5/6 nephrectomised mice, a model of secondary hypertension, were treated with either exogenous follistatin or with a follistatin miRNA inhibitor to increase endogenous follistatin for

Comparison of model predictions of typhoid conjugate vaccine public health impact and cost-effectiveness.

Models are useful to inform policy decisions on typhoid conjugate vaccine (TCV) deployment in endemic settings. However, methodological choices can influence model-predicted outcomes. To provide robust estimates for the potential public health impact of TCVs that account for structural model differences, we compared four dynamic and one static mathematical model of typhoid transmission and vaccine impact. All models were fitted to a common dataset of age-specific typhoid fever cases in Kolkata, India. We evaluated three TCV strategies: no vaccination, routine vaccination at 9 months of age, an

GraphReco: Probabilistic Structure Recognition for Chemical Molecules.

In scientific documents, molecule structure information is usually delivered by chemical structure images. Although convenient for human interpretation, the image-based molecule structure depiction is not a machine-readable format, blocking productivity improvements in many fields including chemical data mining and drug discovery. Molecule structures can be modeled as graphs with atoms as nodes and bonds as edges. Following this intuitive modeling, optical chemical structure recognition (OCSR) can be achieved by first detecting individual atoms and bonds and then assembling into a graph. Howev

Screening potential inhibitors of lactate dehydrogenase from Plasmodium knowlesi via virtual screening approaches.

Lactate dehydrogenase from Plasmodium knowlesi (Pk-LDH) has been suggested as a potential therapeutic target for the development of drugs against malaria disease. This paper reported the screening of compounds which have potentials to be developed as drugs specific for malaria caused by P. knowlesi via in silico screening. Due to the unavailability of Pk-LDH crystal structure, a protein model was built based on the crystal structure of the closest similar protein, lactate dehydrogenase from Plasmodium falciparum (Pf-LDH) with 91% sequence identity between the two enzymes. The model was develop

Utility of three-dimensional computed tomography for anatomical assistance in endoscopic endonasal transsphenoidal surgery.

Endoscopic endonasal transsphenoidal surgery (ETSS) has been widely applied to pituitary adenomas. However, anatomical orientation is difficult when structures of the sphenoidal sinus are complicated. This study investigated the usefulness of three-dimensional computed tomography (3D-CT) modeling in planning surgical procedures for ETSS and providing anatomical guidance during surgery. CT data from 99 consecutive patients with pituitary adenoma treated between January 2008 and March 2014 were used to reconstruct 3D-CT models. Based on these images, the architecture of sphenoidal sinus, particu
