Probing single ventricle heart defects with patient-derived induced pluripotent stem cells and emerging technologies.

Single ventricle heart defects (SVHDs) are a severe type of congenital heart disease with poorly understood pathogenic mechanisms. New research using patient-specific induced pluripotent stem cells (iPSCs) as a cellular model is beginning to uncover genetic and cellular etiologies of SVHDs. Hypoplastic left heart syndrome (HLHS) is a type of SVHD that is characterized by an underdeveloped left ventricle and other malformations in the left side of the heart. Hypoplastic right heart syndrome (HRHS), the second type of SVHD, is characterized by an underdeveloped right heart, including malformed t

Efficient computed tomography-based image segmentation for predicting lateral cervical lymph node metastasis in papillary thyroid carcinoma.

PURPOSE: Papillary thyroid carcinoma (PTC) is a common thyroid cancer, and accurate preoperative assessment of lateral cervical lymph node metastasis is critical for surgical planning. Current methods are often subjective and prone to misdiagnosis. This study aims to improve the accuracy of metastasis evaluation using a deep learning-based segmentation method on enhanced computed tomography (CT) images. APPROACH: We propose a YOLOv8-based deep learning model integrated with a deformable self-attention module to enhance metastatic lymph node segmentation. The model was trained on a large datas

Incidence and Risk Factors of Thromboembolism with Multiple Myeloma in the Presence of Death as a Competing Risk: An Empirical Comparison of Statistical Methodologies.

Multiple myeloma (MM) has an inherent high risk of thromboembolic events associated with patient as well as disease- and treatment-related factors. Previous studies have assessed the association of MM-related thromboembolism using "traditional" Kaplan-Meier (KM) and/or Cox proportional hazard (PH) regression. In the presence of high incidence of death, as would be the case in cancer patients with advanced age, these statistical models will produce bias estimates. Instead, a competing risk framework should be used. This study assessed the baseline patient demographic and clinical characteristic

Multicenter Evaluation of Large Language Models Versus Hepatologists for Prognostic Prediction in Drug-Induced Liver Injury.

BACKGROUND & AIMS: Drug-induced liver injury (DILI) would progress to chronicity or death. Large language models (LLMs) may enhance clinical decision-making, yet their utility relative to physicians in DILI remains unclear. Therefore, we evaluated their performance in predicting DILI outcomes. METHODS: We enrolled 943 DILI patients from three centers as internal and external cohorts. Based on 12-month follow-up, outcomes were classified as recovery, 6/12-month chronicity, and death. LLMs (Gemini-2.5 Pro, GPT-5.1, DeepSeek-3.2), hepatologists (Junior, middle, senior), and models (Hy's Law, nHy

Deep learning approach to predict autism spectrum disorder: a systematic review and meta-analysis.

Источник пометил публикацию как отозванную

BACKGROUND: The use of the deep learning (DL) approach has been suggested or applied to identify childhood autism spectrum disorder (ASD). The capacity to predict ASD, however, differs across investigations. Our study's objective was to conduct a meta-analysis to determine the DL for ASD in children's classification accuracy. METHODS: Eligibility criteria were designed according to the purpose of the meta-analysis; PubMed, EMBASE, Cochrane Library, and Web of Science Database were searched for articles published up to April 16, 2023, on the accuracy of DL methods for ASD classification. A case study of an individual participant data meta-analysis of diagnostic accuracy showed that prediction regions represented heterogeneity well.

The diagnostic accuracy of a screening tool is often characterized by its sensitivity and specificity. An analysis of these measures must consider their intrinsic correlation. In the context of an individual participant data meta-analysis, heterogeneity is one of the main components of the analysis. When using a random-effects meta-analytic model, prediction regions provide deeper insight into the effect of heterogeneity on the variability of estimated accuracy measures across the entire studied population, not just the average. This study aimed to investigate heterogeneity via prediction regi

Using ROPScore and CHOP ROP for early prediction of retinopathy of prematurity in a Chinese population.

PURPOSE: Retinopathy of prematurity (ROP) is a disease that causes vision loss, vision impairment, and blindness, most frequently manifesting among preterm infants. ROPScore and CHOP ROP (Children's Hospital of Philadelphia ROP) are similar scoring models to predict ROP using risk factors such as postnatal weight gain, birth weight (BW), and gestation age (GA). The purpose of this study was to compare the accuracy and difference between using ROPScore and CHOP ROP for the early prediction of ROP. METHODS: A retrospective study was conducted from January 2009 to December 2019 in China. Patient

TB-SERS analyzer: Analysis tool for tuberculosis prediction based on Raman spectroscopy with machine learning and convolutional neural network.

Raman spectroscopy (RS) and surface-enhanced Raman spectroscopy (SERS) are promising technologies that have been applied across various fields, including clinical diagnostics. In the context of tuberculosis (TB) diagnosis, RS/SERS offers significant potential for rapid, non-invasive, and sensitive biomolecular detection. However, no software currently exists that is specifically designed to analyze RS/SERS data for TB diagnosis. Our goal is to develop such a tool by integrating machine learning (ML) and a one-dimensional convolutional neural network (1D-CNN) into a user-friendly graphical user

Peritumoral Fat Radiomics for Dual Prediction of TNM Stage and Histological Grade in Clear Cell Renal Cell Carcinoma: Discovery of Target-Specific Optimal Imaging Distances.

Background/Objectives: Perirenal fat (PRF) constitutes a critical yet understudied component of the tumor microenvironment in clear cell renal cell carcinoma (ccRCC). While radiomics enables non-invasive tissue characterization, whether PRF-derived features can simultaneously predict both TNM stage and histological grade, and whether optimal peritumoral distances differ between these distinct biological targets, remains unexplored in the literature. Methods: This multi-cohort retrospective study included 474 histopathologically confirmed ccRCC patients from three independent datasets (2007-202

Glypican-3-Targeted Paired Radiotheranostics in Orthotopic Liver Cancer Models.

PURPOSE: Glypican-3 (GPC3), highly expressed in the most common liver cancer hepatocellular carcinoma (HCC) but absent from normal liver, represents an attractive tumor-selective target. We evaluated GPC3-targeted paired PET diagnostic and alpha particle therapeutic (radiotheranostic) agents in orthotopic models of liver cancer. EXPERIMENTAL DESIGN: Using the GPC3-specific antibody codrituzumab (GC33), we engineered an immunoPET agent, [89Zr]Zr-DFO-GC33 (89Zr-GC33), and a therapeutic agent, [225Ac]Ac-Macropa-GC33 (225Ac-GC33). Binding specificity and biodistribution were assessed in orthotopi

Neuromuscular Plasticity in a Mouse Neurotoxic Model of Spinal Motoneuronal Loss.

Despite the relevant research efforts, the causes of amyotrophic lateral sclerosis (ALS) are still unknown and no effective cure is available. Many authors suggest that ALS is a multi-system disease caused by a network failure instead of a cell-autonomous pathology restricted to motoneurons. Although motoneuronal loss is the critical hallmark of ALS given their specific vulnerability, other cell populations, including muscle and glial cells, are involved in disease onset and progression, but unraveling their specific role and crosstalk requires further investigation. In particular, little is k

[Discriminant analysis to predict the clinical diagnosis of primary immunodeficiencies: a preliminary report].

BACKGROUND: The features in a clinical history from a patient with suspected primary immunodeficiency (PID) direct the differential diagnosis through pattern recognition. PIDs are a heterogeneous group of more than 250 congenital diseases with increased susceptibility to infection, inflammation, autoimmunity, allergy and malignancy. Linear discriminant analysis (LDA) is a multivariate supervised classification method to sort objects of study into groups by finding linear combinations of a number of variables. OBJECTIVE: To identify the features that best explain membership of pediatric PID pa

Age-related white matter changes revealed by a whole-brain fiber-tracking method in bipolar disorder compared to major depressive disorder and healthy controls.

AIM: Several studies have reported altered age-associated changes in white matter integrity in bipolar disorder (BD). However, little is known as to whether these age-related changes are illness-specific. We assessed disease-specific effects by controlling for age and investigated age-associated changes and Group × Age interactions in white matter integrity among major depressive disorder (MDD) patients, BD patients, and healthy controls. METHODS: Healthy controls (n = 96; age range, 20-77 years), MDD patients (n = 101; age range, 25-78 years), and BD patients (n = 58; age range, 22-76 years)

Depression in small-vessel disease relates to white matter ultrastructural damage, not disability.

OBJECTIVE: To determine whether cerebral small-vessel disease (SVD) is a specific risk factor for depression, whether any association is mediated via white matter damage, and to study the role of depressive symptoms and disability on quality of life (QoL) in this patient group. METHODS: Using path analyses in cross-sectional data, we modeled the relationships among depression, disability, and QoL in patients with SVD presenting with radiologically confirmed lacunar stroke (n = 100), and replicated results in a second SVD cohort (n = 100). We then compared the same model in a non-SVD stroke co
