Cystathionine γ-lyase deficiency exacerbates high-fat diet-induced kidney and liver injury associated with disrupted glutathione homeostasis.

Obesity is a major risk factor for chronic kidney disease (CKD), partly mediated by oxidative stress. The liver is the primary source of glutathione (GSH), a key antioxidant that maintains tissue redox balance. Cystathionine γ-lyase (CSE) regulates cysteine availability for GSH synthesis; however, whether disruption of CSE-dependent GSH metabolism contributes to kidney injury under high-fat diet (HFD)-induced metabolic stress remains unclear. To investigate the role of CSE in HFD-induced kidney injury, CSE-deficient (Cse-/-) and wild-type (Cse+/+) mice were fed a normal-fat diet (NFD) or HFD f

Serpinb2 is Upregulated in Dorsal Root Ganglia and Associated With Neuroinflammatory Changes in a Mouse Model of Neuropathic Pain.

BACKGROUND: Neuropathic pain (NP) is a refractory chronic pain condition that severely impairs quality of life, while currently available treatments often provide insufficient relief. Emerging evidence indicates that neuroinflammation in the dorsal root ganglion (DRG) is critically involved in the development and maintenance of NP; however, the molecular mechanisms responsible for this process remain to be elucidated. The aim of this study was to identify and validate key genes involved in NP and to investigate their potential as therapeutic targets. METHODS: Integrated bioinformatic analyses

Autologous in vitreo generated plasmin assisted vitrectomy in the pediatric population.

BACKGROUND: Inducing posterior vitreous detachment (PVD) is a crucial step in vitreoretinal surgery. However, compared with older patients, induction of PVD in pediatric and young adult populations is considerably more difficult. Recent studies have shown the utility of autologous plasmin enzyme in inducing PVD in pediatric patients. AIM: To evaluate the anatomical and functional outcomes of patients with retinal detachment, treated with in vivo generated autologous plasmin, as an adjunct to vitreoretinal surgery. METHODS: This is a retrospective observational case series including ten eyes

The Role of the VWF/ADAMTS13 Axis in the Thromboinflammatory Response in Ischemic Stroke After SARS-CoV2 Infection.

BACKGROUND: SARS-CoV2 infections increase the risk of ischemic stroke (IS), potentially through a thromboinflammatory cascade driven by an imbalance in the ratio of Von Willebrand Factor (VWF) and a disintegrin and metalloproteinase with a thrombospondin type 1 motif, member 13 (ADAMTS13), leading to the formation of ultra-large VWF (UL-VWF). However, the SARS-CoV2 infection's contribution to any VWF/ADAMTS13 axis imbalance and the subsequent thromboinflammatory response post-stroke remain poorly understood. METHODS: We performed a detailed thromboinflammatory profile of the plasma samples fr

Stem Cell-Based Therapies via Different Administration Route for Stroke: A Meta-analysis of Comparative Studies.

Stroke, a neurological condition from compromised cerebral blood perfusion, remains a major global cause of mortality and disability. Conventional therapies like tissue plasminogen activator are limited by narrow therapeutic windows and potential adverse effects, highlighting the urgency for novel treatments. Stem cell-based therapies, with their neuroprotective and regenerative properties, present a promising yet highly diverse alternative. By conducting literature search and data extraction from the PubMed, Embase, and Cochrane databases, this meta-analysis assessed the clinical efficacy and

Intravenous thrombolysis prevents neurological deterioration in patients with acute pontine infarction.

OBJECTIVES: Neurological deterioration (ND) is common after acute isolated pontine infarction, and no evidence-based treatment is available to prevent this. We determined whether intravenous thrombolysis (IVT) with tissue plasminogen activator soon after pontine infarction prevents ND. METHODS: We retrospectively enrolled consecutive patients admitted to our hospital within 4.5 h after the onset of isolated pontine infarction identified using diffusion-weighted imaging. Patients were divided into the IVT and non-IVT groups. ND was defined as any ≥2-point increase in the National Institutes of

Therapeutic effects of intracerebral transplantation of human modified bone marrow-derived stromal cells (SB623) with voluntary and forced exercise in a rat model of ischemic stroke.

Ischemic stroke results in significant long-term disability and mortality worldwide. Although existing therapies, such as recombinant tissue plasminogen activator and mechanical thrombectomy, have shown promise, their application is limited by stringent conditions. Mesenchymal stem cell (MSC) transplantation, especially using SB623 cells (modified human bone marrow-derived MSCs), has emerged as a promising alternative, promoting neurogenesis and recovery. This study evaluated the effects of voluntary and forced exercise, alone and in combination with SB623 cell transplantation, on neurological

Novel potential neuroprotective targets for DengZhanXiXin injection in middle cerebral artery occlusion rats recommended by quantitative proteomics and simulated docking.

Stroke, which leads to death and disability in high proportions globally, is one of the most deleterious neurological diseases. Ischemic stroke (IS) is the major cause of disease attack and accounts for ~70% of all incident stroke cases in China. Up to now, only two therapies for IS were officially approved, which are intravenous administration of recombinant tissue-plasminogen activator (rt-PA) and endovascular mechanical thrombectomy to rapidly recanalize the occluded artery, which both recanalize the occluded artery rapidly to reduce disability, but are limited in a fixed time window. Understanding the influence of transfusion and blood loss on tranexamic acid concentration in scoliosis surgery with blood loss.

AIMS: Tranexamic acid (TXA) stabilizes clot formation by inhibiting fibrin degradation and improves postoperative outcomes. However, rare adverse events (e.g., thrombosis, seizures) warrant dose-risk evaluation. This study examines how perioperative blood loss and transfusion practices affect TXA concentrations during paediatric scoliosis surgery. METHODS: Forty-three patients undergoing scoliosis surgery with TXA were retrospectively analysed. The study assessed the impact of packed red blood cell (PRBC) transfusion on plasma TXA levels and whether maintaining concentrations ≥10 μg/mL correl

Isolated Aphasia From a Left Parietotemporal Infarct Secondary to a Patent Foramen Ovale: A Case Report.

Isolated aphasia (IA) as the sole manifestation of stroke is rare and is often attributed to metabolic, toxic, or psychiatric disorders. Without typical stroke symptoms, such as hemiplegia or cranial nerve deficits (CND), IA can be easily overlooked. We present the case of a 41-year-old male accountant with a history of hyperlipidemia who presented to the emergency department with the sudden onset of an inability to read or speak, accompanied by confusion and anxiety. On examination, he had no CNDs, and his strength, sensation, reflexes, and cerebellar function were normal. His vital signs, bl

PAI-1 as a critical factor in the resolution of sepsis and acute kidney injury in old age.

Elevated plasma levels of plasminogen activator inhibitor type 1 (PAI-1) are documented in patients with sepsis and levels positively correlate with disease severity and mortality. Our prior work demonstrated that PAI-1 in plasma is positively associated with acute kidney injury (AKI) in septic patients and mice. The objective of this study was to determine if PAI-1 is causally related to AKI and worse sepsis outcomes using a clinically-relevant and age-appropriate murine model of sepsis. Sepsis was induced by cecal slurry (CS)-injection to wild-type (WT, C57BL/6) and PAI-1 knockout (KO) mice
