Characterizing trajectories of innate immune cells in larval zebrafish.

It is well established from in vitro studies of immune cells that stimulation by a wide range of potential signals leads to motility and morphology changes. How these physical behaviors manifest inside a living animal remains unclear due to limitations of conventional imaging and analysis approaches. Here, we establish a quantitative framework for imaging and tracking neutrophil and macrophage dynamics in larval zebrafish, spanning a large fraction of the animal for multi-hour timescales with few-minute temporal resolution. We focus especially on the gut, examining innate immune responses to d

Artemisitene Ameliorates Diabetic Wounds by Inhibiting Ferroptosis Through Activation of the Nrf2/GPX4 Pathway.

Diabetic wounds are hard to heal due to ferroptosis, a specific type of cell death triggered by high blood sugar in human umbilical vein endothelial cells (HUVECs) in the lining of blood vessels. Ferroptosis occurs as a result of excessive iron accumulation and lipid peroxidation. The resulting impairment in endothelial function and tissue scaffolding creates a persistent barrier against effective wound healing. Our study found that artemisitene (ATT), a bioactive compound derived from the herb Artemisia annua, speeded up diabetic wound healing by blocking ferroptosis through the Nrf2 (NFE2 li

Randomized, crossover clinical trial on the safety, feasibility, and usability of the ABLE exoskeleton: A comparative study with knee-ankle-foot orthoses.

Wearable exoskeletons are emerging as a new tool for gait training. However, comparisons between exoskeletons and conventional orthoses in terms of safety and feasibility are scarce. This study assessed the safety, feasibility, usability, and learning process of using the ABLE Exoskeleton in people with spinal cord injury (SCI) while comparing it with knee-ankle-foot orthoses (KAFOs). In this randomized, crossover clinical trial, 10 patients with chronic complete SCI (T4-T12) conducted a 10-session training and assessment protocol with each device: KAFOs and the ABLE Exoskeleton. Outcomes on s

Intensive weight loss intervention versus usual care in adults with obesity: a protocol for the LightCARE randomised clinical trial.

INTRODUCTION: Total diet replacements (TDRs) and weight loss medications (WLMs) have proven effective in producing substantial weight loss for individuals with obesity. Evidence is lacking on whether combining these treatments is effective and cost-effective in primary care for adults with obesity class I (body mass index (BMI) 30-34.9) or uncomplicated obesity class II or higher (BMI≥35 without obesity-related disease). METHODS AND ANALYSIS: LightCARE is a 2-year 1:1 randomised, parallel-group, clinical superiority trial with blinded outcome assessment evaluating the benefits and harms of an

Pre-implementation findings on the barriers and potential solutions to initiate same-day antiretroviral therapy among people who inject drugs in Malaysia.

BACKGROUND: Rapid-start antiretroviral therapy (RS-ART), including same-day ART (SD-ART), effectively improves individual and public health by speeding up treatment initiation but is underutilized among people who inject drugs (PWID). As a pre-implementation activity, we assessed barriers and facilitators to SD-ART for PWID in Malaysia using insights from multiple stakeholders. METHODS: We conducted focus groups (n = 4) using nominal group technique (NGT), a mixed-methods research strategy, with PWID, HIV specialists, and community health workers (CHWs) in Greater Kuala Lumpur, Malaysia. Transferable and data efficient metamodeling of storm water system nodal depths using auto-regressive graph neural networks.

Storm water systems (SWSs) are essential infrastructure providing multiple services including environmental protection and flood prevention. Typically, utility companies rely on computer simulators to properly design, operate, and manage SWSs. However, multiple applications in SWSs are highly time-consuming. Researchers have resorted to cheaper-to-run models, i.e. metamodels, as alternatives of computationally expensive models. With the recent surge in artificial intelligence applications, machine learning has become a key approach for metamodelling urban water networks. Specifically, deep lea

Perceptual Response Training for Reduction of Injury Risk Among High School Girls' Soccer Players.

Background/Objectives: Neural processes involved in visual detection, decision-making, and motor plan execution are believed to play a key role in the avoidance of sport-related injuries, but very little evidence exists to guide the development of training activities for the optimization of brain function. Immersive virtual reality provides a means to precisely measure the amount of time that elapses from visual stimulus presentation to the initiation of a motor response (i.e., perceptual latency) or its completion (i.e., response time). Methods: The median value of a metric quantifying both t

A cell abundance analysis based on efficient PAM clustering for a better understanding of the dynamics of endometrial remodelling.

BACKGROUND: Single-cell RNA sequencing (scRNA-seq) is a powerful tool for investigating cell abundance changes during tissue regeneration and remodeling processes. Differential cell abundance supports the initial clustering of all cells; then, the number of cells per cluster and sample are evaluated, and the dependence of these counts concerning the phenotypic covariates of the samples is studied. Analysis heavily depends on the clustering method. Partitioning Around Medoids (PAM or k-medoids) represents a well-established clustering procedure that leverages the downstream interpretation of cl

Modulating preferences during intertemporal choices through exogenous midfrontal transcranial alternating current stimulation: A registered report.

Decision conflicts may arise when the costs and benefits of choices are evaluated as a function of outcomes predicted along a temporal dimension. Electrophysiology studies suggest that during performance monitoring a typical oscillatory activity in the theta rhythm, named midfrontal theta, may index conflict processing and resolution. In the present within-subject, sham controlled, cross-over preregistered study, we delivered online midfrontal transcranial Alternating Current Stimulation (tACS) to modulate electrocortical activity during intertemporal decisions. Research on Safety Helmet Detection Algorithm Based on Improved YOLOv5s.

Safety helmets are essential in various indoor and outdoor workplaces, such as metallurgical high-temperature operations and high-rise building construction, to avoid injuries and ensure safety in production. However, manual supervision is costly and prone to lack of enforcement and interference from other human factors. Moreover, small target object detection frequently lacks precision. Improving safety helmets based on the helmet detection algorithm can address these issues and is a promising approach. In this study, we proposed a modified version of the YOLOv5s network, a lightweight deep l

Water-based medium-expansion foam depopulation of adult cattle.

Current options for depopulation of adult cattle are limited, have logistic constraints, and may not be practical on a large scale. Aspirated water-based foam (WBF) has been shown to be successful in depopulating poultry and swine but has yet to be tested in cattle. WBF is advantageous because necessary equipment can be readily available, easy to use, and presents minimal personnel risk. With the use of a modified rendering trailer in a field setting, we evaluated the efficacy of aspirated WBF for depopulation of adult cattle. Water-based medium-expansion foam was added to the trailer holding

Exploration of the mechanism of NORAD activation of TGF-β1/Smad3 through miR-136-5p and promotion of tacrolimus-induced renal fibrosis.

BACKGROUND: Tacrolimus is a potent immunosuppressant, but has various side effects, with nephrotoxicity being the most common. Renal fibrosis is an important process of tacrolimus nephrotoxicity. Therefore, it is important to identify the factors that contribute to renal fibrosis after tacrolimus nephrotoxicity, and control its development. METHODS: The present study aims to determine whether tacrolimus may speed up the course of renal fibrosis by upregulating noncoding RNA activated by DNA damage (NORAD) to compete with miR-136-5p, and activating the TGF-β1/Smad3 pathway. Furthermore, in viv

Absorption rate governs cell transduction in dry macroporous scaffolds.

Developing the next generation of cellular therapies will depend on fast, versatile, and efficient cellular reprogramming. Novel biomaterials will play a central role in this process by providing scaffolding and bioactive signals that shape cell fate and function. Previously, our lab reported that dry macroporous alginate scaffolds mediate retroviral transduction of primary T cells with efficiencies that rival the gold-standard clinical spinoculation procedures, which involve centrifugation on Retronectin-coated plates. This scaffold transduction required the scaffolds to be both macroporous a

Live-cell fluorescence imaging of microgametogenesis in the human malaria parasite Plasmodium falciparum.

Formation of gametes in the malaria parasite occurs in the midgut of the mosquito and is critical to onward parasite transmission. Transformation of the male gametocyte into microgametes, called microgametogenesis, is an explosive cellular event and one of the fastest eukaryotic DNA replication events known. The transformation of one microgametocyte into eight flagellated microgametes requires reorganisation of the parasite cytoskeleton, replication of the 22.9 Mb genome, axoneme formation and host erythrocyte egress, all of which occur simultaneously in <20 minutes. Whilst high-resolution ima
