High-Performance Regenerated Silk Fibers as Building Blocks of Tissue Scaffolds: The European THOR Project.

The European Pathfinder THOR project envisages the creation of a vascularized fragment of tissue that can be implanted in a patient using regenerated silk fibers as its building blocks. The selection of regenerated silk as the main building block of the scaffold relies heavily on its outstanding biocompatibility in comparison with either other artificial polymeric fibers or even natural silk fibers. Additionally, regenerated fibers produced through the Dynamic Dope Destabilization Spinning (D3STM) process are shown to exhibit high mechanical performance as reflected in values of strain at brea

Isoform-Selective Targeting of Akt Through Covalent Allosteric Inhibition.

The Akt family of serine/threonine kinases plays a crucial role in various cellular processes, including proliferation, survival, and metabolism. Three Akt isoforms (Akt1, Akt2, and Akt3) have distinct physiological roles, and while individual isoform dysregulation is disease-linked, unselective Akt inhibition leads to side effects. Here, we report the development of selective covalent-allosteric Akt inhibitors (CAAIs) targeting Akt2 and Akt3 while sparing Akt1. Guided by protein x-ray crystallography and molecular modeling, key structural differences within the allosteric pockets of the isofo

Dual-Switchable Nanoparticles Resolving the Safety-Efficacy Paradox of Sonodynamic Immunotherapy for Hepatocellular Carcinoma.

Clinical translation of sonodynamic therapy for hepatocellular carcinoma is limited by a safety-efficacy paradox: sonosensitizer activation in healthy tissues poses risks, while tumor efficacy is constrained by hypoxia and redundant antioxidant defenses. To resolve this, dual-switchable core-shell nanoparticles were engineered. Under physiological conditions, a carboxymethyl chitosan shell and a peptide radical scavenger keep the nanoparticles inert, ensuring systemic safety. In the acidic tumor microenvironment, the shell disassembles, and ultrasound exposure triggers a coordinated therapeuti

A Topology-Programmed Circular DNAzyme Enables High-Precision Diagnosis of Metastatic Triple-Negative Breast Cancer.

DNAzymes capable of RNA-cleaving are attractive, protein-independent candidates for biosensing and gene regulation. However, precise modulation of their activity remains a significant obstacle to biomedical applications. Herein, we uncovered a cyclization-provoked, size-reliant topological barrier that suppresses the catalytic core folding of DNAzymes, thereby governing their catalytic activity. Leveraging this underlying topological insight, we engineered a catalytically inert, minimal-sequence circular DNAzyme bearing a cleavable apurinic/apyrimidinic (AP) site and revealed that its substrat

Advancing MSC-EV Therapies: Harnessing Preconditioning and Mito-EVs to Tackle Neuroinflammation and Neurodegeneration.

Neuroinflammation plays a central role in the onset and progression of neurodegenerative disorders. Several disease-modifying therapies have been developed to target neuroinflammatory pathways in specific disorders. However, their ability to stop disease progression or restore neuronal and mitochondrial homeostasis remains limited. This is still a major unmet clinical need. In this context, mesenchymal stromal cell (MSC)-derived Extracellular Vesicles (EVs) have emerged as a promising cell-free therapeutic strategy due to their ability to modulate immune responses and promote neuroprotection t

Locoregional Immune Checkpoint Blockade and Remodeling of Lymph Nodes by Engineered Dendritic Cell-Derived Exosomes for Suppressing Tumor Progression and Metastasis.

Tumor-draining lymph nodes (TDLNs) are the primary sites of eliciting anti-tumor immunity, which play an important role in controlling tumor progression and metastasis. However, the immunosuppressive microenvironment of TDLNs propels the formation of pre-metastatic niche, in which the immunocytes are dysfunctional, and the high expression of programmed death-ligand 1 (PD-L1) on dendritic cells (DCs) restricts the activation of cytotoxic T lymphocytes. Herein, engineered exosomes (EmDEX@GA) are developed for locoregional immunomodulation of TDLNs. EmDEX@GA possess CC-chemokine receptor 7 (CCR7)

A T-cell receptor targeting RUNX1 frameshift mutations in acute myeloid leukemia.

Runt-related transcription factor 1 (RUNX1) is a key regulator of hematopoietic differentiation. RUNX1 mutations in acute myeloid leukemia (AML) are associated with poor prognosis. One-third are frameshift mutations encoding an oncogenic protein with an elongated C-terminus translated in an alternative reading frame. Here, we investigated whether the alternative reading frame of oncogenic RUNX1 can be targeted by immunotherapy. We introduced a construct with a RUNX1 frameshift mutation into B-cell lines with common HLA class I alleles and identified 13 neopeptides by immunopeptidomics. To inve

Clinically Translatable Injectable Hydrogel Platform for Dual Intraoperative Elevation and Postoperative Healing in Endoscopic Submucosal Dissection.

Endoscopic submucosal dissection (ESD) enables en bloc resection of early gastrointestinal neoplasms but remains constrained by two critical limitations: rapid dissipation of submucosal injectates during surgery and insufficient wound management postresection. To overcome this clinical bottleneck, we develop a multifunctional, clinically translatable injectable hydrogel engineered to unify intraoperative mucosal elevation and postoperative tissue repair. This hydrogel system is synthesized via dynamic cross-linking between oxidized sodium alginate and carboxymethyl chitosan, reinforced with Ca

Interface Cleaning Strategy of Carbon Nanotube Field-Effect Transistor Biosensors for Ultrasensitive Detection of Ophthalmic Biomarkers.

Carbon nanotube (CNT) field-effect transistor (FET) biosensors combine the molecular recognition capabilities of bioreceptors with the signal amplification and transduction functions of nano-FET devices, enabling highly specific and ultrasensitive biomolecule detection. However, interface degradation and contamination introduced in the micronanofabrication process of batch manufacturing led to inhomogeneity in biofunctionalization of device and sensing performance, thus limiting the reproducible preparation and large-scale application of CNT FET biosensors. In this study, we proposed a compreh

Slippery Behavior of PEGylated Surfaces.

Slippery hydrophilic surfaces have gained significant attention due to their hydrophilicity and ultralow liquid adhesion, offering excellent biofouling resistance as well as enhanced condensation efficiency. Although polyethylene glycol (PEG) is widely used for such surfaces, the detailed molecular-level mechanism by which PEG chain length governs surface slipperiness remains elusive. Herein, we present a systematic approach to address this challenge by grafting silane-terminated PEGs of different molecular weights (0.3k, 5k, and 20k) onto smooth, nontextured substrates to unveil the role of P

Knockout of Perilipin-2 in Microglia Alters Lipid Droplet Accumulation and Response to Alzheimer's Disease Stimuli.

Lipid droplets (LDs) are emerging as key regulators of metabolism and inflammation, with their buildup in microglia linked to aging and neurodegeneration. Perilipin-2 (Plin2) is a ubiquitously expressed LD-associated protein that stabilizes lipid stores; in peripheral tissues, its upregulation promotes lipid retention, inflammation, and metabolic dysfunction. Yet, its role in microglia remains unclear. Using CRISPR-engineered Plin2 knockout (KO) BV2 microglia, we examined how Plin2 contributes to lipid accumulation, bioenergetics, and immune function. Compared to wild-type (WT) cells, Plin2 KO

Cellulophaga algicola alginate lyase and Pseudomonas aeruginosa Psl glycoside hydrolase inhibit biofilm formation by Pseudomonas aeruginosa CF2843 on three-dimensional aggregates of lung epithelial cells.

Pseudomonas aeruginosa is an opportunistic pathogen that produces a biofilm containing the polysaccharides, alginate, Psl, and Pel, and causes chronic lung infection in cystic fibrosis patients. Others and we have previously explored the use of alginate lyases in inhibiting P. aeruginosa biofilm formation on plastic and lung epithelial cell monolayers. We now employ a more physiologically representative model system, i.e., three-dimensional aggregates of A549 lung epithelial cells cultured under conditions of microgravity in a rotary cell culture system to mimic the natural lung environment, a

Closed-loop cascade nanozyme strategy for mutually reinforced catalytic and mild-temperature photothermal therapeutic effects.

Nanocatalysis coupled with photothermal therapy is a potent anti-cancer approach, yet its clinical utility is limited by low concentration of tumor substrate, redox interference, and risks of overheating normal tissues. Herein, we propose an innovative closed-loop nanozyme approach that leverages the synergistic effects of catalytic and mild photothermal therapy (mPTT) to address aforementioned challenges. The strategy features a folic acid-functionalized iron single-atom catalyst (FeNC-FA), designed to exhibit exceptional multienzymatic capabilities and an optimal photothermal response. In th
