Carrier-Free Glycyrrhizic Acid and Baicalin Co-Assembled Hydrogel for Methicillin-Resistant Staphylococcus aureus Infected Wound Repair.

Methicillin-resistant Staphylococcus aureus (MRSA) and its biofilms severely impair wound healing, necessitating therapies that integrate antibacterial activity with tissue repair functions. In this study, we developed a carrier-free injectable supramolecular hydrogel, referred to as the GB gel, through the co-assembly of two natural small molecules, glycyrrhizic acid (GA) and baicalin (BA). The principal design logic of this system lies in the cooperative integration of GA-mediated amphiphilic assembly and BA-mediated antibacterial activity: GA improves the aqueous solubility of BA, drives na

Solithromycin mitigates Prevotella intermedia-induced methicillin-resistant Staphylococcus aureus ventilator-associated pneumonia by enhancing alveolar macrophage function.

BACKGROUND: Ventilator-associated pneumonia (VAP) is a fatal intensive care infection. VAP caused by methicillin-resistant Staphylococcus aureus (MRSA) can be exacerbated by Prevotella intermedia culture supernatant (P. int. sup.). Solithromycin (SOL), a fourth-generation macrolide, inhibits bacterial protein synthesis and modulates immunity; however, its effects on exacerbation of MRSA-VAP by P. int. sup. remain unclear. This study examined whether SOL inhibits bacterial protein synthesis by binding to the 50S ribosomal subunits in P. int. sup. and subsequently reduces the worsening of MRSA-V

Biogenic Silver Nanoparticles Synthesised Using Staphylococcus saprophyticus Cell-Free Supernatant Exhibit Potent Antimicrobial Activities via Oxidative Stress.

In this study, we report the eco-friendly synthesis of silver nanoparticles (AgNPs) using cell-free supernatant of Staphylococcus saprophyticus as a biological reducing and stabilising agent. Nanoparticle formation was confirmed by visible colour change and UV-visible spectroscopy (λmax at 418 nm). Structural characterisation by SEM, XRD and FT-IR indicated predominantly spherical, crystalline, silver-containing nanoparticles associated with extracellular biomolecules that may contribute to stabilisation; XRD analysis indicated a surface phase dominated by silver halides (AgBr/AgCl) rather tha

Companion animals as potentially important reservoir of methicillin-resistant and virulent strains of coagulase-positive Staphylococcus spp.

BACKGROUND: Microorganisms such as Staphylococcus spp., in particular coagulase-positive strains, continue to pose a serious challenge to human and veterinary medicine. These bacteria can be a component of the natural biota of the body but can also cause infections in companion animals, including dogs and cats. Therefore, the aim of this study was to analyze the occurrence of Staphylococcus bacteria in companion animals, with particular emphasis on predilection sites, and to characterize these strains in the context of their epidemiological significance and threat to human health. RESULTS: We

Conserved and context-dependent changes in pathogen and host cell gene expression during Staphylococcus aureus intracellular infection.

BACKGROUND: The opportunistic intracellular lifestyle of Staphylococcus aureus contributes to its ability to maintain chronic infective states. However, knowledge of pathogen-encoded factors that enable S. aureus to invade and persist within human cells, host cell responses to infection, and how such features vary across different bacterial strains and host cell types remains incomplete. Here, we used dual RNA-seq to examine alterations in S. aureus and host cell transcription following bacterial invasion, comparing results from two different S. aureus strains (JE2 and ATCC29213) and human cel

Sinomenine protects mice against Staphylococcus aureus-induced endometritis through inhibition of P2X7 receptor-mediated ferroptosis and inflammation.

Endometritis is a major cause of infertility that adversely impacts both human reproductive health and livestock productivity. This study investigated the protective effects and underlying mechanisms of sinomenine (SIN) against Staphylococcus aureus (S. aureus)-induced endometritis. Seventy-two adult female mice were randomly divided into six groups: control group, SIN group (100 mg/kg), S. aureus group (1 × 107 CFU per 200 μL PBS), and SIN (25 mg/kg, 50 mg/kg, and 100 mg/kg, respectively) + S. aureus groups. Uterine tissues were collected for analysis. Inflammatory factors, ferroptosis, and P

A secreted AIP-Like peptide from Helcococcus kunzii inhibits the Agr quorum sensing system of Staphylococcus aureus.

Staphylococcus aureus is a major human pathogen whose virulence is tightly regulated by the Agr quorum sensing system. In this study, we investigated the impact of Adh2, a secreted protein from the commensal bacterium Helcococcus kunzii, on S. aureus physiology and pathogenicity. Adh2 shares structural similarity with native auto-inducing peptides (AIPs), including the conserved CDFIM motif characteristic of Agr group I. We hypothesized that Adh2 interferes with Agr signaling by competitively binding the AgrC receptor. Exposure to Adh2 significantly repressed agrA and its downstream α-hemolysi

Lauric Acid- and BDSF-Derived N-Acyl Sulfonamides as Potent Antibiofilm and Antivirulence Agents against Staphylococcus aureus.

Biofilm-associated Staphylococcus aureus infections remain difficult to treat using conventional antibiotics. Herein, we report the synthesis and biological evaluation of lauric acid- and BDSF-derived N-acyl sulfonamides as antibiofilm and antivirulence agents. Structure-activity relationship (SAR) analysis identified 4-tert-butylphenyl sulfonyldodecenamide (59) as a lead compound with a minimum inhibitory concentration of 5 μg/mL and >60% inhibition of MSSA and MRSA biofilm formation at sub-minimum inhibitory concentration levels. Microscopy confirmed marked reductions in biofilm biomass and

Characterization of bacteriophage JD929 targeting Staphylococcus aureus from chronic otitis media and its ability to inhibit biofilm.

BACKGROUND: Staphylococcus aureus (S. aureus) is a key pathogen involved in chronic suppurative otitis media (CSOM). Antibiotic resistance and biofilm formation complicate CSOM treatment. Therefore, alternative therapies are urgently needed. Bacteriophage therapy, which specifically targets and destroys methicillin-resistant S. aureus (MRSA) and degrades biofilms, presents a promising option. METHODS: In this study, a lytic bacteriophage targeting S. aureus was isolated and named JD929. Its biological properties were analyzed, including its stability across different pH levels and temperature

Computational and experimental evaluation of Pisolithus arhizus metabolites targeting major efflux pumps of mastitis-associated Staphylococcus aureus.

Antimicrobial resistance (AMR) in pathogenic bacteria, particularly Staphylococcus aureus, is an increasing global concern in veterinary medicine. The present study evaluated bioactive metabolites from P. arhizus as potential inhibitors of major S. aureus efflux pumps. The methanolic extracts of P. arhizus were profiled using GC-MS to identify the main constituents, and their antibacterial activity against S. aureus was evaluated using a well diffusion assay. The experimental results showed that the crude methanolic extract exhibited a 20 mm zone of inhibition with a MIC of 30 µg/mL. Furthermo

Pre-colored Prussian blue: A substrate-free dual-mode immunosensor for visible-to-thermal Staphylococcus aureus quantification.

Rapid identification of postoperative Staphylococcus aureus (S. aureus) infections is essential for timely treatment and improved clinical outcomes. Herein, we developed a substrate-free dual-mode lateral flow immunoassay (LFIA) using intrinsically colored Prussian Blue nanoparticles (PB NPs) as multifunctional signal reporters. The intrinsic blue color of PB NPs enables direct colorimetric visualization, while their excellent near-infrared photothermal conversion provides quantitative temperature readout. This self-signaling strategy eliminates enzymatic amplification and substrate incubation

Astrakurkurone targets multidrug resistant Staphylococcus aureus biofilms through ROS-mediated cell membrane damage and β-lactamase inhibition.

Methicillin-resistant Staphylococcus aureus (MRSA) pose a formidable health threat owing to their multidrug resistance (MDR) nature, robust biofilms and persistence. Natural product-based medications are a highly viable strategy for challenging MDR and biofilm persistence with diverse therapeutic benefits. In this study, the triterpenoid Astrakurkurone, previously purified from a wild edible mushroom Astraeus hygrometricus, showed antibacterial efficacy against MRSA BAA1717 and a clinical isolate (MRSA CI-1) with MICs ranging from 150 to 300 μg/mL. 1/2 MIC concentration of Astrakurkurone also

Unraveling Resistance and Thresholds in Phage Therapy Using In Vitro Staphylococcus aureus experiments and Pharmacokinetic/Pharmacodynamic Models.

BACKGROUND: Phage therapy is a promising approach for infections caused by multidrug-resistant Staphylococcus aureus, but its clinical development is limited by knowledge gaps in pharmacokinetic/pharmacodynamic (PK/PD) and potential emergence of resistance. METHODS: We performed in vitro time-kill assays to study the effect of three anti-staphylococcal phages (V1SA019, V1SA020, V1SA022) against two S. aureus strains (SH1000 and USA300). Resistance emergence was monitored through phenotyping and whole-genome sequencing of resistant clones.Bacteria and phage counts were analyzed by pharmacokine

NIR- and pH-responsive double-crosslinked inulin-tannic acid hydrogel with antioxidant and Anti-Staphylococcus aureus Activity.

Oxidative stress and bacterial infections can lead to compromised tissue regeneration in chronic wounds. Since traditional treatments are often ineffective, in this study a hydrogel based on two inulin derivatives, called dcINU-TA-NS, was design as multifunctional wound dressing platform with in vitro Anti-Staphylococcus aureus and antioxidant performance. Thanks to double crosslinking the hydrogel is characterized by modulable viscoelastic properties: dynamic interactions, based on the boronic ester bonds between inulin functionalized with 3-aminophenyl boronic acid (INU-APBA) and tannic acid

Methicillin-Resistant Staphylococcus aureus Bacteremia Originating From a Ureteral Stent-Associated Urinary Tract Infection: A Case Report.

Methicillin-resistant Staphylococcus aureus (MRSA) is an uncommon cause of urinary tract infection but can result in severe systemic complications, particularly in patients with indwelling urinary devices. We report a case of MRSA bacteremia associated with a ureteral stent in an 83-year-old woman who presented with a high-grade fever. Computed tomography demonstrated left hydronephrosis despite stent placement. Emergency stent exchange revealed whitish purulent debris in the bladder and draining from the stent side holes. Blood, bladder urine, and renal pelvic urine cultures all yielded MRSA.

From prevention to treatment: Comprehensive strategies for the management of Staphylococcus aureus induced catheter-related bloodstream infections.

Staphylococcus aureus (SA) is a prominent pathogen that frequently causes catheter-related bloodstream infections (CRBSIs) in hemodialysis patients with tunneled central lines. The guidelines' key recommendation for managing SA-induced CRBSIs is to remove the infected catheter. In real life, however, this may be hindered by logistical or clinical problems (e.g. unavailability of surgical rooms, incarcerated catheters, catheter-associated thrombosis). Furthermore, an interventional approach may expose patients to complications, such as those related to the procedure itself or the development of
