Delineating the autoimmune mechanisms in Graves' disease.

The immunologic processes involved in autoimmune thyroid disease (AITD), particularly Graves' disease (GD), are similar to other autoimmune diseases with the emphasis on the antibodies as the most unique aspect. These characteristics include a lymphocytic infiltrate at the target organs, the presence of antigen-reactive T and B cells and antibodies, and the establishment of animal models of GD by antibody transfer or immunization with antigen. Similar to other autoimmune diseases, risk factors for GD include the presence of multiple susceptibility genes, including certain HLA alleles, and the

Thymoma and autoimmunity.

The thymus is a central lymphatic organ that is responsible for many immunological functions, including the production of mature, functional T cells and the induction of self-tolerance. Benign or malignant tumors may originate from the thymus gland, with thymoma being the most common and accounting for 50% of anterior mediastinal tumors. Malignancies linked to thymoma include the loss of self-tolerance and the presence of autoimmunity. In this review, we compiled the current scientific evidence detailing the various interactions between thymoma and autoimmune diseases, including myasthenia gra

Lichen sclerosus in adult men: a study of HLA associations and susceptibility to autoimmune disease.

Lichen sclerosus is a skin disorder of unknown prevalence affecting both men and women, and several studies have established HLA associations in women with this disease. Autoimmune disease associations in the form of a personal and/or family history of autoimmune disease have also been shown to be related to lichen sclerosus. In this study, we examined 58 men (mean age 38 years) with lichen sclerosus, 39 of whom had histologically proven disease. HLA tissue typing by phototyping was performed on these patients and contrasted with that of 602 control subjects. There was no difference in antigen

Association of raised levels of IL-4 and anti-TPO with hyperprolactinemia.

BACKGROUND AND OBJECTIVE: The modulatory role of prolactin in autoimmune regulation is well established. Hyperprolactinemia is often associated with autoimmune disease like systemic lupus erythematosus and autoimmune thyroid diseases. The objective was to compare levels of direct and indirect autoimmune factors in different categories of hyperprolactinemia cases and predict the direction of association between hyperprolactinemia and autoimmune factors, if any. METHODS: A total of 102 hyperprolactinemia cases (>100 ng/mL serum prolactin level) were included along with 24 controls. Among 102 hy

Selenium supplementation in thyroid associated ophthalmopathy: an update.

The therapeutic effect of selenium (Se) has already been proven in thyroid disease and thyroid associated ophthalmopathy (TAO). In spite of clear scientific proof of its benefits in TAO, there appears to be no clear agreement among the clinicians regarding its optimum dose, duration of the treatment, efficacy and safety to date. In this review, the author summarises the findings of 135 English language articles published on this subject over the past four decades from 1973 to 2013. The regulation and metabolism of thyroid hormones require a steady supply of Se and recent studies have revealed

Role of FNA in the medical management of minimally enlarged thyroid.

Fine-needle aspiration (FNA) of thyroid is a cost-effective and simple diagnostic tool in the initial screening of patients with thyroid nodules. But, its role in a clinically normal thyroid or a minimally enlarged thyroid in a symptomatic patient suspected of having thyroid dysfunction is unknown. With our 2-yr experience in a setup of a tertiary health care hospital in a developing country, we have aimed to implement a management protocol using FNA thyroid done without ultrasound guidance and TSH estimation done during the same visit in symptomatic patients suspected of having thyroid pathol

Adverse effects of iodides on thyroid function.

The administration of pharmacologic quantities of iodine such as iodides for the treatment of pulmonary disease, organic iodine present in medications and x-ray contrast dyes, and the ingestion of iodine-rich natural foods, may result in goiter, hypothyroidism, or hyperthyroidism, especially in patients with underlying thyroid disease. Medications containing iodide may induce hypothroidism in euthyroid patients with Hashimoto's thyroiditis, 131I or surgically treated Graves' disease, or following hemithyroidectomy for nodules; and they may induce hyperthyroidism in patients with endemic iodine

Clinical Characteristics and Thyroid Findings in Children with Isolated Premature Adrenarche.

Objectives: Premature adrenarche (PA) is associated with accelerated adrenal maturation, but data regarding thyroid characteristics in children with isolated PA are limited. We aimed to describe the clinical features of children with idiopathic PA and investigate the relationships between thyroid function, thyroid autoimmunity, and adrenal androgen secretion. Methods: This retrospective single-center study included 435 children (378 girls, 57 boys) with idiopathic PA evaluated between 2014 and 2024. Clinical, anthropometric, biochemical, and thyroid data were analyzed. Associations between adr

Maternal thyroid peroxidase antibody positivity and its association with incidence of low birth weight in infants.

BACKGROUND: Autoimmune thyroid disease is a prevalent condition affecting women of reproductive age, leading to thyroid dysfunction and impacting pregnancy outcomes. While the critical role of thyroid hormone in pregnancy outcomes is well-established, the potential association between positive anti-thyroid peroxidase antibodies (TPOAb) and adverse pregnancy outcomes in pregnant women with normal thyroid function remains unclear. OBJECTIVE: This study aims to investigate the relationship between maternal TPOAb positivity and adverse pregnancy outcomes with normal thyroid function. METHODS: We

Abnormal Cardiac Repolarization in Thyroid Diseases: Results of an Observational Study.

Background: The relationship between thyroid function and cardiac disease is complex. Both hypothyroidism and thyrotoxicosis can predispose to ventricular arrhythmia and other major adverse cardiovascular events (MACE), so that a U-shaped relationship between thyroid signaling and the incidence of MACE has been postulated. Moreover, recently published data suggest an association between thyroid hormone concentration and the risk of sudden cardiac death (SCD) even in euthyroid populations with high-normal FT4 levels. In this study, we investigated markers of repolarization in ECGs, as predictor

Mouse Model of Graves' Orbitopathy in Current Research and Future Perspectives.

Graves' orbitopathy (GO) is a representative extrathyroidal manifestation of autoimmune thyroid disease. Despite extensive research, the precise pathogenic mechanisms remain incompletely understood. Moreover, the lack of an ideal animal model has limited in vivo investigation, with most studies relying on in vitro systems using orbital fibroblasts. In this review, we discuss currently available mouse models of GO and recent advances in experimental modeling strategies, aiming to provide an integrated framework for future research.

Progressive systemic sclerosis and Graves' disease. Report of three cases.

Progressive systemic sclerosis (PSS) is associated with a broad spectrum of autoimmune thyroid diseases. While an association between PSS and hypothyroidism is well established, a relationship between PSS and hyperthyroidism is less well defined. We treated three patients with PSS whose course was complicated by Graves' disease. Because hyperthyroidism can simulate many of the symptoms of PSS progression and treatment of hyperthyroidism can lead to resolution of clinical deterioration in such patients, it is important to recognize the simultaneous occurrence of these diseases.

The role of microRNA-16 in the pathogenesis of autoimmune diseases: A comprehensive review.

MicroRNAs (miRNAs) are a class of small noncoding RNAs that are only 21-25 nt long. Many studies have shown that miRNA dysfunction is closely related to the occurrence and development of diseases. By combining with the 3' noncoding region of target gene mRNA, miRNAs can mediate the degradation or translation inhibition of mRNA and exert a powerful regulation effect on gene expression at the posttranscriptional level, mainly inhibiting the translation or degradation of targets. Therefore, they are a class of molecules that play a negative regulatory role. Current studies have found that miR-16

Emergence of rheumatoid arthritis following exposure to natalizumab.

We report a patient with relapsing-remitting multiple sclerosis, who developed rheumatoid arthritis after exposure to natalizumab. While some multiple sclerosis therapies are known to unmask autoimmune conditions, natalizumab is rarely implicated as a cause of alternative autoimmunity. This case illustrates an unusual clinical scenario which may support recent scientific work suggesting that, when natalizumab blocks T helper 1 cells from entering the central nervous system, T helper 17 cells may continue to migrate into immune-privileged spaces and cause pathologic inflammation. BRIEF BACKGROU

[Extrahepatic manifestations in hepatitis C. Are they overlooked?].

BACKGROUND: There are accumulating documentation of autoimmune mediated extrahepatic manifestations of hepatitis C virus infection. The virus is hepatotrophic and lymphotrophic. It mutates frequently with subsequent inadequate immune response and chronic stimulation of T and B cells. This may be one explanation for the increased frequency of the autoimmune diseases associated with hepatitis C virus infection. MATERIAL AND METHODS: In this review of the literature published in the period of 1990 to 2000, we present the most common extrahepatic manifestations of the hepatitis C virus infection.

[Cytofluorometric analysis of chosen markers of apoptosis CD95/CD95L (Fas/FasL) in thyroid tissues from young patients with Graves' disease and Hashimoto's thyroiditis].

BACKGROUND: Apoptosis, one of the forms of programmed cell death, is a physiologic process of cell death that is central to normal development and occurs in response to a variety of physiologic and pathophysiologic stimuli. In the thyroid, abnormal apoptotic activity may be involved in a variety of diseases such as Hashimoto thyroiditis and Graves disease. The aim of this study was to estimate the expression of chosen apoptotic molecules CD95 (Fas) and CD95L (FasL) on the surface of thyroid follicular cells in application of mouse monoclonal antibodies #64 which recognized B antigen regions of
