RNA Promotes Synapsin Condensates That Organize Synaptic Vesicles in Live Neurons and Enable Localized Translation in Reconstituted System.

Condensates at synapses organize synaptic vesicle (SV) clusters and are essential for efficient neurotransmitter release. While it is established that RNA granules traffic along axons, the function of RNA at the presynapse remains unclear. Here, we uncover a direct structural role of coding RNAs in organizing presynaptic condensates by focusing on SV clusters, condensates between synapsin-1 and lipid vesicles. Using in vitro reconstitution systems, we show that RNA drives synapsin-1 coacervation, with structured RNAs being more effective at promoting phase transitions. The importance of RNA wa

Chlorinated bis-4-hydroxycoumarins suppress flavivirus replication by inhibiting dengue virus type 2 translation and replication.

Dengue virus (DENV) remains a major global health threat, and no clinically approved antiviral therapy is currently available. Halogenated biscoumarins have been reported as versatile antimicrobial and antiviral agents. Here, we screened eleven halogenated biscoumarin derivatives against DENV and identified compounds 3 and 4-bearing chlorine substitutions at the 3- or 4-position of the phenyl moiety-as the most potent inhibitors of DENV2, with EC50 values of 3.62-9.72 µM and 4.62-10.88 µM, respectively, and selectivity indices (SI) up to 20.97. Both compounds also inhibited all four DENV serot

Artificial intelligence and big data for precision regenerative medicine in knee osteoarthritis: endotyping, responder prediction, and clinical translation.

Knee osteoarthritis (KOA) is a heterogeneous whole-joint disease, and regenerative and orthobiologic therapies such as platelet-rich plasma (PRP), mesenchymal stem cells (MSCs), bone marrow aspirate concentrate (BMAC), microfragmented adipose tissue (MFAT), and extracellular vesicles (EVs) show variable clinical effects. This variability reflects a dual heterogeneity: patients differ in structural damage, inflammation, metabolism, biomechanics, pain mechanisms, and molecular endotypes, while therapeutic products differ in composition, dose, viability, secretome, and manufacturing protocols. Translational insights into miR-126 and miR-423: biomarkers and therapeutic targets in cancer, cardiovascular, metabolic and kidney diseases.

MicroRNAs (miRNAs) are key post-transcriptional regulators that orchestrate complex gene regulatory networks controlling endothelial function, metabolic adaptation, inflammation, and tissue remodeling. Among them, miR-126-3p, miR-126-5p, and miR-423-5p have emerged as context-dependent modulators linking vascular biology with cardiometabolic and oncologic disorders. MiR-126, through its 3p and 5p strands, plays a central role in maintaining endothelial integrity and angiogenic homeostasis. By modulating phosphoinositide 3-kinase/protein kinase B (PI3K/AKT), mitogen-activated protein kinase (MA

Next-Generation Vaccine Design for Porcine Enteric Coronaviruses: Aligning Antigenic Breadth, Mucosal Immunity, and Translational Evaluation.

Porcine enteric coronaviruses (PECs), including porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (TGEV), porcine deltacoronavirus (PDCoV), and swine acute diarrhea syndrome coronavirus (SADS-CoV), remain major causes of neonatal diarrhea, dehydration, mortality, and economic loss in swine production. Despite substantial progress in vaccine development, durable field protection is still inconsistent. In this narrative review, this narrative review synthesizes current knowledge on PEC vaccine design from three connected perspectives: antigenic breadth, mucosal immunity

A strategy for scalable antibody production: the combination of Escherichia coli expression, translation-enhancing peptide and automated refolding system.

Monoclonal antibodies are essential tools in biological research, diagnostics, and therapeutics. While mammalian expression systems are widely used, microbial hosts such as Escherichia coli offer cost-effective and scalable production of antibody fragments but often yield inclusion bodies requiring refolding. In this study, we report a combined strategy using a translation-enhancing SKIK peptide tag and a leucine zipper-fused Fab format to improve expression in Escherichia coli, together with a continuous, dialysis-based automated refolding system. Compared with the conventional stepwise metho

Post-translational modification crosstalk in acute pancreatitis: linking acinar stress to innate immune amplification and tissue repair.

Acute pancreatitis (AP) is a sterile inflammatory disease initiated by acinar cell stress and amplified through innate immune, vascular, and inter-organ responses, yet effective disease-modifying therapies remain unavailable. Although premature digestive enzyme activation, organelle stress, sterile inflammation, and microvascular failure are well-recognized features of AP, the mechanisms linking early acinar stress to coordinated injury across epithelial, immune, stromal, and vascular compartments remain insufficiently integrated. Emerging evidence suggests that post-translational modification

Targeting NR4A1 (Nur77) in Macrophage Polarization: Molecular Mechanisms and Translational Prospects for Cardiac Repair After Myocardial Infarction.

Sterile inflammation in the course of myocardial infarction can be a factor contributing to cardiac remodeling and heart failure. To break this pathological vicious circle, the polarization of macrophages to a reparative M2 phenotype, to start the tissue repair process and alleviate the inflammation, is the key. Recent studies have shown that the nuclear receptor subfamily 4 group A (NR4A) orphan nuclear receptor family, particularly nuclear receptor subfamily 4 group A member 1 (NR4A1/NUR77), plays a role in macrophage reprogramming in relation to the modulation of macrophage polarization. Health equity-oriented design for dissemination: products and impact of rapid community translation for COVID-19 vaccine promotion.

BACKGROUND: Public health messaging often falls short of the needs of underserved and minority communities, missing the mark on translating information that captures cultural, linguistic, or community relevance to influence health behaviors. Rapid community translation (rapid-CT) is an adaptation of the Boot Camp Translational method for community engagement in translating medical evidence into community health promotion messages and interventions. This paper describes the dissemination of materials from a rapid-CT of messages promoting COVID-19 vaccination among children and adults, boosters,

Full-scale translation of methane-enhancement strategies in sewage sludge anaerobic digestion: evidence, constraints, and decision support.

Reported methane gains in sewage-sludge anaerobic digestion do not consistently translate into sustained full-scale benefits. This critical review links process constraints to intervention selection by assessing translational directness and attribution confidence independently, while distinguishing digital prediction from operational deployment. Eighteen full-scale facility cases covering pre-treatment, temperature-driven operation, staged digestion, and co-digestion were evaluated using comparator design, reporting basis, operating duration, and plant-wide burdens. Only five cases provided in

Artificial intelligence-enabled multi-omics biomarkers for immune checkpoint blockade: mechanisms, predictive modeling, and clinical translation.

Immune checkpoint inhibitors (ICIs) have transformed oncology, yet durable benefit remains confined to a minority of patients, revealing the limitations of single biomarkers such as PD-L1 expression, tumor mutational burden, and microsatellite instability. Multi-omics profiling, spanning genomics, transcriptomics, epigenomics, proteomics, metabolomics, microbiomics, and imaging-derived radiomics/pathomics, enables a systems-level interrogation of tumor-immune interactions. It captures lineage plasticity, antigen-presentation defects, metabolic and epigenetic suppression, stromal remodeling, an

Anti-PD-1 single-chain variable fragments in cancer immunotherapy: from molecular engineering to clinical translation.

The programmed death-1 (PD-1) immune checkpoint is a core regulator of T-cell exhaustion and immune evasion in cancer. While PD-1/PD-L1 monoclonal antibodies have changed the landscape of cancer immunotherapy, there are barriers to broader application due to their large size, potential for systemic toxicities, and high production costs. In this review article, we will focus on therapeutic development, functional applications, and translational advances of anti-PD-1 single-chain variable fragments (scFvs). Advances in phage-display libraries, immunization methods, and antibody engineering have

Fast tracking and imaging of translational moving objects via single-pixel imaging leveraging Fourier conjugate symmetry.

In single-pixel imaging (SPI), the need to sequentially project numerous encoded patterns leads to low frame rates and pronounced motion blur when capturing moving targets, which severely limits its practical applications. Most existing approaches address this by inserting additional reference frames for motion estimation, but this inevitably reduces imaging efficiency and degrades the signal-to-noise ratio(SNR). To overcome these limitations, we propose a method that enables simultaneous high-precision motion tracking and image reconstruction for arbitrary translational motion without requiri

Porous Polymeric Materials for Cell Delivery and Regenerative Therapy: Design Principles, AI-Assisted Optimization, and Clinical Translation.

Cell therapy is increasingly recognized as a cornerstone of 21st-century medical innovation, but its success heavily relies on the effective delivery of therapeutic cells to the target site while maintaining their viability and functionality. Traditional intravenous injection methods are typically limited by poor targeting and reduced cell survival rates due to mechanical stress during injection. Recent advancements have introduced alternative delivery vehicles, such as microspheres (MSs), microneedles (MNs), and scaffolds, to enhance targeting and efficacy. Among these, porous polymeric mater
