Does Zn-Mediated Regulation of the Kynurenine Pathway Provide the Link Between Periodontal Disease and Diabetes?

It has long been known that there is a relationship between periodontal diseases and diabetes. The present study aimed to assess the effect of pancreatic zinc (Zn) levels on Kynurenin pathways (KP) and glucose homeostasis and the impact of Thymoquinone (TQ) in the periodontal disease animal model. Ten microliters of Porphyromonas gingivalis-lipopolysaccharide (P. gingivalis-Lps) (1 mg/ml) was injected at 48-h intervals into the palatal gingiva of rats, and TQ was given by oral gavage (10 mg/kg per day) for 2 weeks. Glucose homeostasis was assessed using the Homeostatic Model Assessment (HOMA-I

Comparative Transcriptome Profiling of Ileal and Cecal Tissues Between Pekin Ducks and Shaoxing Ducks.

Background: Pekin ducks are well-known meat-type ducks, whereas Shaoxing ducks are bred for their egg-laying abilities. Growth and development of poultry species is well studied; however, very little is known regarding differences in intestinal gene expression between Pekin and Shaoxing ducks. Methods: To investigate intestinal differences between Pekin and Shaoxing ducks, we conducted transcriptome analysis on ileal and cecal tissues from five 42-day-old ducks per breed, raised under identical housing and feeding conditions to minimize environmental influences. Results: The results showed tha

Acute and chronic effects of physical exercise on atherosclerosis, kynurenine pathway, endothelial function and inflammation in patients with coronary artery disease: a clinical trial protocol.

Coronary artery disease (CAD) is the leading cause of death worldwide. Patients with CAD exhibit increased levels of kynurenine (KYN) pathway (KP) metabolites, which are key products of tryptophan (TRP) catabolism and have been identified as predictive markers for cardiovascular events and atherosclerosis progression. However, the effects of exercise on KP metabolites and their influence on atherosclerosis and inflammation in these patients remain poorly understood. We aim to investigate exercise's acute and chronic effects on carotid intima-media thickness, KP metabolites, inflammatory marker

Dominant myosin storage myopathy mutations disrupt striated muscles in Drosophila and the myosin tail-tail interactome of human cardiac thick filaments.

Myosin storage myopathy (MSM) is a rare skeletal muscle disorder caused by mutations in the slow muscle/β-cardiac myosin heavy chain (MHC) gene. MSM missense mutations frequently disrupt the tail's stabilizing heptad repeat motif. Disease hallmarks include subsarcolemmal hyaline-like β-MHC aggregates, muscle weakness, and, occasionally, cardiomyopathy. We generated transgenic, heterozygous Drosophila to examine the dominant physiological and structural effects of the L1793P, R1845W, and E1883K MHC MSM mutations on diverse muscles. The MHC variants reduced lifespan and flight and jump abilities

Microbial translocation and gut damage is associated with hepatic fibrosis but not steatosis in women with and without HIV.

OBJECTIVE: Metabolic dysfunction-associated steatotic liver disease (MASLD) is highly prevalent in people living with HIV (PWH). Gut microbial translocation and gut damage may play a role in MASLD pathogenesis. We determined associations of circulating biomarkers of translocation and gut damage with hepatic steatosis and fibrosis in a large U.S. cohort of women with HIV (WWH) and without HIV. DESIGN: Vibration controlled transient elastography (VCTE) was conducted from 2013 to 2018 among 854 WWH and 349 women without HIV. Serum biomarkers were measured within 6 months of the VCTE: kynurenine

Synbiotic laden with Faecalibacterium prausnitzii, fructooligosaccharides, and galactooligosaccharides mitigates quinpirole-induced OCD-like symptoms in rats.

Obsessive-compulsive disorder (OCD) is a mental illness of unwanted recurrent thoughts and compulsive behaviors. The significance of the microbiota-gut-brain axis has been increasingly documented in the manifestation of neuropsychiatric disorders. Herein, we investigated the impact of synbiotic containing Faecalibacterium prausnitzii ATCC 27,766 (F. prausnitzii) and prebiotics [fructooligosaccharides (FOS) and galactooligosaccharides (GOS)] on quinpirole-induced OCD-like symptoms in rats. Repeated quinpirole injections led to compulsive- and anxiety-like behaviors, as rats showed reduced head-

Nutritional intake of micronutrient and macronutrient and type 2 diabetes: machine learning schemes.

BACKGROUND: Diabetes mellitus, an endocrine system disease, is a common disease involving many patients worldwide. Many studies are performed to evaluate the correlation between micronutrients/macronutrients on diabetes but few of them have a high statistical population and a long follow-up period. We aimed to investigate the relationship between intake of macro/micronutrients and the incidence of type 2 diabetes (T2D) using logistic regression (LR) and a decision tree (DT) algorithm for machine learning. METHOD: Our research explores supervised machine learning models to identify T2D patient

Baseline gut microbiome and metabolites are correlated with changes in alcohol consumption in participants in a randomized Zonisamide clinical trial.

Development and severity of alcohol use disorder (AUD) has been linked to variations in gut microbiota and their associated metabolites in both animal and human studies. However, the involvement of the gut microbiome in alcohol consumption of individuals with AUD undergoing treatment remains unclear. To address this, stool samples (n = 32) were collected at screening (baseline) and trial completion from a double-blind, placebo-controlled trial of zonisamide in individuals with AUD. Alcohol consumption was measured both at baseline and endpoint of 16-week trial period. Fecal microbiome was anal

Kv1.3 knockdown attenuates alcohol-related liver injury in mice through induction of tryptamine.

Alcohol-related liver disease (ALD) refers to a spectrum of liver diseases caused by chronic or acute excessive alcohol consumption. Excess alcohol consumption and dysregulated immunometabolism are the major contributors to the disease progression. The pathogenesis of ALD remains unclear with macrophage activity playing a key role in disease development. Kv1.3 potassium channel is essential for the regulation of macrophage function. In this study, we investigated whether Kv1.3 channels influenced the progression of ALD by modulating macrophage function. A murine model of ALD was established in

UPF 648, a Selective KMO Inhibitor, Attenuates Psychomotor and Cognitive Impairment in Chronic Kidney Disease.

Kynurenine-3-monooxygenase (KMO), a key enzyme in the kynurenine pathway (KP) of tryptophan metabolism, converts kynurenine into the neurotoxic intermediate quinolinic acid (QA). QA, an N-methyl-d-aspartate (NMDA) receptor agonist, increases glutamate release and inhibits its reuptake, resulting in excitotoxic cell death in the hippocampus and striatum. Plasma metabolomics study exhibited KP metabolites as the most altered pathway in patients with chronic kidney disease (CKD). Recently, QA was linked to the kidney-brain axis as one of the major neurotoxins responsible for cognitive impairment

Polysaccharides from Hippophae rhamnoides L. protected against ethanol-induced gastric ulcer in mice and its impact on metabolites and gut microbiota.

Gastric ulcer (GU) is a common gastrointestinal disorder, and alcohol overdose is one of the main causes of GU. Polysaccharide is considered a promising prebiotic agent to protect against alcohol-induced GU, but its exact mechanisms remain unclear. In this study, Hippophae rhamnoides L. polysaccharides (SP) was exacted and characterized for its structural properties. The gastroprotective effects of SP against ethanol-induced GU were evaluated in a mouse model by administering 200 mg/kg/day SP orally for 14 days. Histological analysis showed that SP significantly decreased gastric lesion areas

Crystal structure of leishmanial glucose-6-phosphate dehydrogenase and its implications for selective inhibitor development.

Glucose-6-phosphate dehydrogenase (G6PDH) is a vital enzyme in trypanosomatid parasites, including Leishmania, where it generates NADPH to combat oxidative stress. In this study, Leishmania donovani G6PDH (LdG6PDH) was cloned, expressed, and purified to homogeneity using chromatographic methods. The purified enzyme existed as a dimer in solution, exhibited optimal activity near physiological pH, and displayed higher affinity for its cofactor (NADP) than for its substrate, glucose-6-phosphate (G6P). The enzymatic activity assay of leishmanial G6PDH revealed that ellagic acid (EA) and epigalloca

In Vitro Anti-Glioblastoma Activity of Echinocereus engelmannii- and Echinocereus pectinatus-Associated Bacterial Endophyte Extracts.

Glioblastoma is the most common and aggressive brain tumor in adults. However, due to the limitations of conventional treatments, as well as their side effects, there is a need to develop more effective and less harmful therapy strategies. There is evidence that plants endemic to northern Mexico possess biological activities that positively impact human health, particularly against cancer. Echinocereus engelmannii and Echinocereus pectinatus are cacti from the north of Mexico that produce bioactive compounds with antitumor activity. We obtained methanol extracts from previously isolated and fe

Fate of Dissolved Organic Matter and Cooperation Behavior of Coagulation: Fenton Combined with MBR Treatment for Pharmaceutical Tail Water.

In this study, the treatment of pharmaceutical tail water (PTW) by coagulation, Fenton combined with membrane bioreactor (MBR), was studied. Optimal parameters were obtained according to batch experiment and central composite design (CCD). Results showed that Polymeric Ferric Sulfate (PFS) was the best coagulant for original pharmaceutical tailwater due to less dosage and higher removal efficiency to TOC, COD, NH4+-N and UV254m, with the optimized pH = 7.25 and 0.53 g/L PFS dosage. The best coagulation performance was achieved when the mixer was stirred at 250 rpm for 3 min, 60 rpm for 10 min,

Clostridium leptum attenuates allergic airway inflammation via tDC‒Treg axis activation in a murine model of gut dysbiosis-associated asthma.

Allergic pulmonary inflammation is driven by allergen-specific Th2/Th17 responses counterregulated by tolerogenic dendritic cells (tDCs) and regulatory T cells (Tregs). While early-life colonization with commensal Clostridia induces the generation of colonic Tregs, the therapeutic potential of Clostridium leptum (CL) monotherapy to treat asthma patients with comorbid gut dysbiosis remains unexplored. Therefore, this study aimed to investigate whether 14 days of oral CL administration restores gut-lung immune homeostasis and suppresses allergic inflammation via tDC/Treg induction in a dysbiosis

Indoxyl sulfate promoted glutamine metabolism and cell proliferation in urothelial carcinoma involving Runx2.

Patients with Chronic Kidney Disease (CKD) have an increased risk and poor prognosis of becoming diagnosed with urothelial carcinoma. Currently, the linking mechanisms underlying the connection between CKD and urothelial carcinoma are not well understood. CKD with declined renal function is associated with the accumulation of circulating indoxyl sulfate, a metabolite synthesized from tryptophan by gut microbes. We investigated the roles of CKD-related uremic toxins in regards to their linking with urothelial carcinoma by delivering indoxyl sulfate to urothelial carcinoma cells and tumor-bearin
