During an ENU (N-ethyl-N-nitrosourea) mutagenesis screen, we observed a dominant small-eye mutant mouse with viable homozygotes. A corresponding mutant line was established and referred to as Aey69 (abnormality of the eye #69). Comprehensive phenotyping of the homozygous Aey69 mutants in the German Mouse Clinic revealed only a subset of statistically significant alterations between wild types and homozygous mutants. The mutation causes microphthalmia without a lens but with retinal hyperproliferation. Linkage was demonstrated to mouse chromosome 3 between the markers D3Mit188 and D3Mit11. Sequ

Neural crest stem/progenitor cells (NCSCs) populate a variety of tissues, and their dysregulation is implicated in several human diseases including craniosynostosis and neuroblastoma. We hypothesised that small molecules that inhibit NCSC induction or differentiation may represent potential therapeutically relevant drugs in these disorders. We screened 640 FDA-approved compounds currently in clinical use for other conditions to identify those which disrupt development of NCSC-derived skeletal elements that form the zebrafish jaw. In the primary screen, we used heterozygous transgenic sox10:gfp

Ciliopathies are a group of genetic multi-systemic disorders related to dysfunction of the primary cilium, a sensory organelle present at the cell surface that regulates key signaling pathways during development and tissue homeostasis. In order to identify novel genes whose mutations would cause severe developmental ciliopathies, >500 patients/fetuses were analyzed by a targeted high throughput sequencing approach allowing exome sequencing of >1200 ciliary genes. NEK8/NPHP9 mutations were identified in five cases with severe overlapping phenotypes including renal cystic dysplasia/hypodysplasia

Alcohol exposure during embryogenesis results in a variety of developmental disorders. Here, we demonstrate that continuous exposure to 1.5% ethanol causes substantial apoptosis and abrogated retinal and CNS development in zebrafish embryos. Chronic exposure to ethanol for 24h before hatching also induces apoptosis and retinal disorder. After the 2-day post-fertilization (dpf) stage, chronic exposure to ethanol continued to induce apoptosis, but did not block retinal differentiation. Although continuous ethanol exposure induces substantial accumulation of reactive oxygen species (ROS) and incr

Lateral zebrafish hypoblast cells initiate dorsal convergence near mid-gastrulation and exhibit non-polarized morphologies, limited cell-cell contact and indirect migration trajectories. By late gastrulation, mesodermal cells become packed as they engage in planar cell polarity (PCP)-dependent movement. Here, we aimed to understand this transition in cell behavior by examining the relationship between protrusion dynamics and establishment of PCP and directed migration. We found that wild-type cells undergo a reduction in bleb protrusions near late gastrulation accompanied by a VANGL planar cel

Fetal alcohol spectrum disorder (FASD) is a leading cause of neurodevelopmental disability. Individuals with FASD may exhibit a characteristic facial appearance that has diagnostic utility. The mechanism by which alcohol disrupts craniofacial development is incompletely understood, as are the genetic factors that can modify individual alcohol vulnerability. Using an established avian model, we characterized the cranial transcriptome in response to alcohol to inform the mechanism underlying these cells' vulnerability. Gallus gallus embryos having 3-6 somites were exposed to 52 mM alcohol and th

Haematopoietic stem cells (HSCs) are a self-renewing population of cells that continuously replenish all blood and immune cells during the lifetime of an individual. HSCs are used clinically to treat a wide array of diseases, including acute leukaemias and congenital blood disorders, but obtaining suitable numbers of cells and finding immune-compatible donors remain serious problems. These difficulties have led to an interest in the conversion of embryonic stem cells or induced pluripotent stem cells into HSCs, which is not possible using current methodologies. To accomplish this goal, it is c

The adult pancreas possesses limited capacity for regeneration. Evidence suggests conversion between terminally differentiated pancreatic cells, either through transdifferentiation or dedifferentiation, but the extent, cellular origins and physiological relevance of these processes are not fully understood. Key insights have emerged from studies of pancreatic injury and stress, which induce cellular reprogramming and expansion of the beta cell compartment. Additionally, model organisms such as zebrafish and axolotls exhibit a greater capacity for pancreatic regeneration, providing evolutionary

Hydrazine (N2H4) is a highly toxic industrial chemical that threatens food safety and public health via food chain accumulation. To address the scarcity of reliable detection tools for N2H4 in food and biological matrices, a chitosan (CTS)-based fluorescent probe (CTS-NI) was fabricated by covalently conjugating a naphthalimide fluorophore to modified CTS. The probe exhibits significant fluorescence enhancement at 558 nm toward N2H4, with a large Stokes shift (118 nm) and a low detection limit (2.03 × 10-7 M). Its biocompatibility and CTS-derived structural advantages enable sensitive and quan

Achieving isotropic high-resolution three-dimensional imaging across a large field of view (FOV) remains a key challenge in two-photon light-sheet microscopy (TPLSM). To address this limitation, we developed a TPLSM system that stitches multiple short Bessel beams into a large, ultrathin illumination light sheet and employs a dedicated control protocol to suppress stray light. The system delivers isotropic near-diffraction-limited resolution across a 332.8 μm FOV, enabling uniform visualization of fine structural details in biological samples and avoiding morphological misinterpretation. Understanding morphogenesis increasingly requires attention to physical aspects. In complex embryonic structures comprising multiple adjacent tissues, morphogenetic change in one tissue can impose geometric constraints that shape another tissue. However, such cross-tissue physical coordination remains largely unexplored. Using zebrafish jaw development, we show that dynamic folding of the oral ectoderm drives the elongation and fusion of mandibular primordia by constraining and guiding mesenchymal cell motion. This folding is initiated by Shh signaling, mainly from the adjacent neural tube, wh

Inflammatory bowel disease (IBD) is a chronic gastrointestinal disorder characterized by oxidative stress, impaired antioxidant defense, and gut microbiota dysbiosis. Despite advances in treatment, current therapies remain limited by insufficient efficacy and systemic side effects. Here, we developed a multifunctional nanoplatform (BG/SOD@ZIF-zc) by encapsulating superoxide dismutase (SOD) within Cu-doped ZIF-zc and coating it with bacterial ghosts for targeted delivery. This system preserved enzymatic activity under gastrointestinal conditions, scavenged reactive oxygen species, and optimized

Cholesterol immunometabolism is a critical controller of immunopathology in respiratory infections such as tuberculosis. Smith-Lemli-Opitz syndrome (SLOS) patients are affected by a loss of 7-dehydrocholesterol reductase (DHCR7) function and have elevated 7-dehydrocholesterol (7DHC) and reduced cholesterol. Increased 7DHC has been found to be protective against viral infections in a range of infection models however SLOS patients have a higher susceptibility to respiratory infection. Here we use the zebrafish-Mycobacterium marinum infection model to demonstrate a compromised innate immune resp

Genetic testing in amyotrophic lateral sclerosis (ALS) often reveals variants of uncertain significance (VUS), which are frequently omitted from diagnostic reports or reported with limited clinical interpretation. To address this gap, we developed a rapid functional assessment pipeline in collaboration with FILSLAN, the French ALS care network, combining in vitro and in vivo neurogenetic assays. We illustrate this approach through the reclassification of the SOD1 p.Val120Leu variant, identified in an ALS patient, as pathogenic. Functional studies demonstrated that this variant leads to cytopla

Nephropathic cystinosis is a severe monogenic kidney disorder caused by mutations in CTNS, encoding the lysosomal transporter cystinosin, resulting in lysosomal cystine accumulation. The sole treatment, cysteamine, slows down the disease progression, but does not correct the established renal proximal tubulopathy. Here, we developed a new therapeutic strategy by applying omics to expand our knowledge on the complexity of the disease and prioritize drug targets in cystinosis. We identified alpha-ketoglutarate as a potential metabolite to bridge cystinosin loss to autophagy, apoptosis and kidney

Despite the wide range of skin pigmentation in humans, little is known about its genetic basis in global populations. Examining ethnically diverse African genomes, we identify variants in or near SLC24A5, MFSD12, DDB1, TMEM138, OCA2, and HERC2 that are significantly associated with skin pigmentation. Genetic evidence indicates that the light pigmentation variant at SLC24A5 was introduced into East Africa by gene flow from non-Africans. At all other loci, variants associated with dark pigmentation in Africans are identical by descent in South Asian and Australo-Melanesian populations. Functiona

Although manganese is an essential trace metal, little is known about its transport and homeostatic regulation. Here we have identified a cohort of patients with a novel autosomal recessive manganese transporter defect caused by mutations in SLC39A14. Excessive accumulation of manganese in these patients results in rapidly progressive childhood-onset parkinsonism-dystonia with distinctive brain magnetic resonance imaging appearances and neurodegenerative features on post-mortem examination. We show that mutations in SLC39A14 impair manganese transport in vitro and lead to manganese dyshomeosta

The gene networks underlying closure of the optic fissure during vertebrate eye development are poorly understood. Here, we profile global gene expression during optic fissure closure using laser capture microdissected (LCM) tissue from the margins of the fissure. From these data, we identify a unique role for the C(2)H(2) zinc finger proteins Nlz1 and Nlz2 in normal fissure closure. Gene knockdown of nlz1 and/or nlz2 in zebrafish leads to a failure of the optic fissure to close, a phenotype which closely resembles that seen in human uveal coloboma. We also identify misregulation of pax2 in th

Wnt signaling regulates some aspect of development of nearly all endoderm-derived organs and Wnts mediate both differentiation and proliferation at different steps during visceral organogenesis. Wnt2b induces liver formation in zebrafish 1 and may combine with other inducers, Fibroblast Growth Factors 1 & 4 and Bone Morphogenetic Protein 4, to specify the mammalian liver.2-5 Later in development, Wnts are critical for liver expansion and, finally, for terminal hepatocyte differentiation,6-12 as reviewed elsewhere in this issue (Monga). Likewise, in the pancreas, Wnts drive proliferation of exo

Key actors of mammalian immunity originated from bacterial antiphage systems. The full extent of immune system conservation between bacteria and eukaryotes is unknown. Here, we show that the silent information regulator 2 (SIR2) protein domain, present in antiphage systems, plays a role in eukaryotic innate immunity. We identified SIRal, a human protein with a SIRim domain (subtype of SIR2) that plays a pivotal role in the animal Toll-like receptor pathway of innate immunity and protects against bacterial and viral infections. Proteins containing a SIRim domain are found across 19% of eukaryot
