Human eastern equine encephalitis. Electron microscopic study of a brain biopsy.

An electron microscopic study was done on brain biopsy tissue from an eight-month-old female with acute eastern equine encephalitis diagnosed by indirect immunofluorescence. Rare clusters of round virions were found almost exclusively in the extracellular space. All virions observed had spiked envelopes, and their sizes averaged approximately 55 nm. Also found were rare enveloped virions along with degenerate organelles in a membrane-bound structure in the cytoplasm of macrophages. Intracytoplasmic development of virions was not found. Tubuloreticular complexes were seen in the endothelial cel

[The experimental study of the cerebral vasospasm--the micro-circulation on the hypothalamus and the brain stem (author's transl)].

The correlation between the cerebral vasospasm and the micro-circulation in the hypothalamus and brainstem was observed histologically and electronmicroscopically using the colloidal carbon infusion method through the right vertebral artery of dogs. The posterior communicating artery of the dog was ruptured with a fine needle, which resulted in subarachnoid hemorrhage. The colloidall infusion was done at 30 min., 48 hours, 1 week and 4 weeks after the subarachnoid hemorrhage. Results 1) The cerebral vasospasm was confirmed by vertebral angiography on all cases with 30 min., 48 hours, and 1 wee

Genetic susceptibility to heat identifies rare neurological diseases at particular risk from climate change impacts.

BACKGROUND: Climate change is one of the greatest contemporary challenges to human health, undermining human health through multiple mechanisms. Among relatively understudied mechanisms are those related to individual genomic variation. We aimed to examine this possibility. METHODS: Through a defined, agnostic literature review-based approach, we curated human genetic variants with functionally characterised temperature-dependent effects: we call these 'calortypic variants', some of which are linked to temperature-sensitive disease phenotypes. Next, we examined their occurrence in whole-genom

Spatiotemporal histogenesis of the developing human cerebellum reveals dynamic layering of Bergmann glia.

Bergmann glia (BG) are a specialized glial population essential for cerebellar development, yet their developmental timeline and molecular identity in the human cerebellum remain poorly understood. Here, we combined detailed histopathological analysis with spatial transcriptomics and single-nucleus RNA sequencing to generate a developmental atlas of human cerebellar BG. Histology revealed that BG emerge around 11 postconception weeks (PCW), initially serving as a scaffold for Purkinje cells (PCs) migrating into the PC layer of the cerebellar cortex. Following the establishment of a multilayere

Morphological Changes in the Rat Brain in the Early Post-Resuscitation Period: The Relationship Between Hypoxia-Sensitive Immunohistochemical Markers, Hemodynamic and Acid-Base Status.

The relevance of studying the molecular mechanisms of ischemia-reperfusion brain injury is due to the ongoing need to identify therapeutic targets to improve neurologic outcomes following cardiac arrest. This study aims to identify early morphological and molecular changes in the rat brain in the post-resuscitation period after asphyxial cardiac arrest, as well as to assess the relationship of these changes with hemodynamic and acid-base status. Morphological analysis using immunohistochemistry (IHC) revealed an increase in the intensity of the IHC reaction to S-100β protein in astrocytes of t

Host-cell dependent epigenetic profiles associated with survival outcomes in T. gondii infection.

Toxoplasmosis is a widespread parasitic disease affecting roughly one-third of the global population. In immunocompromised individuals or during pregnancy, infection can result in severe complications. Following primary infection, Toxoplasma gondii forms dormant bradyzoite cysts in tissues such as the brain and eyes. These cysts can rupture, particularly in immunocompromised hosts, releasing active parasites and triggering recrudescence. Efforts to experimentally induce and study bradyzoite cyst recrudescence have been hindered by the limited capacity of cell culture adapted strains to form ti

Age-Dependent Activation of Pannexin1 Function Contributes to the Development of Epileptogenesis in Autosomal Dominant Sleep-related Hypermotor Epilepsy Model Rats.

To explore the processes of epileptogenesis/ictogenesis, this study determined the age-dependent development of the functional abnormalities in astroglial transmission associated with pannexin1-hemichannel using a genetic rat model of autosomal dominant sleep-related hypermotor epilepsy (ADSHE) named 'S286L-TG'. Pannexin1 expression in the plasma membrane of primary cultured cortical astrocytes and the orbitofrontal cortex (OFC), which is an ADSHE focus region, were determined using capillary immunoblotting. Neuroinflammatory Proteins in Huntington's Disease: Insights into Mechanisms, Diagnosis, and Therapeutic Implications.

Huntington's disease (HD) is a hereditary neurodegenerative disorder caused by a CAG tract expansion in the huntingtin gene (HTT). HD is characterized by involuntary movements, cognitive decline, and behavioral changes. Pathologically, patients with HD show selective striatal neuronal vulnerability at the early disease stage, although the mutant protein is ubiquitously expressed. Activation of the immune system and glial cell-mediated neuroinflammatory responses are early pathological features and have been found in all neurodegenerative diseases (NDDs), including HD. However, the role of infl

Autophagy is a defense mechanism controlling Streptococcus suis serotype 2 infection in murine microglia cells.

Streptococcus suis (S. suis) is an important swine and human pathogen, causing severe meningitis with high morbidity and mortality worldwide. Microglial activation and inflammation are responsible for bacterial meningitis. S. suis has been identified to activate microglia, but the role of autophagy following S. suis infection in microglial cells remains elusive. In this study, using western blot, immunofluorescent staining and transmission electron microscopy (TEM), we demonstrated that S. suis serotype 2 (SS2) triggered autophagosome and enhanced autophagic flux in BV2 microglial cells. [Novel Autoantibody-Related Encephalitis: anti-mGluR1 encephalitis, anti-IgLON5 disease, autoimmune GFAP astrocytopathy].

Various neurological syndromes are associated with autoimmune encephalitis. Anti-mGluR1 antibody encephalitis presents mainly as subacute cerebellar ataxia, while behavioral changes and involuntary movements also occur. Anti-IgLON5 disease presents mainly as a sleep disorder. It is sometimes difficult to distinguish these diseases from other neurodegenerative diseases. Autoimmune glial fibrillary acidic protein (GFAP) astrocytopathy is seen in meningoencephalomyelitis of unknown origin. Because these new autoimmune encephalitis diseases might be treatable, early diagnosis and treatment are nec

Effect of electroacupuncture on relieving central post-stroke pain by inhibiting autophagy in the hippocampus.

INTRODUCTION: To explore the underlying mechanism of electroacupuncture (EA) treatment on central post-stroke pain (CPSP), and provide basic evidence for the EA treatment on CPSP. METHODS: Firstly, 40 male SD rats were successfully established with a model of CPSP, under the intervention of different EA frequencies (2 Hz and 15 Hz) and fluoxetine (5 ml/kg and 0.4 mg/ml), whose brain tissue was then removed for paraffin-embedded sectioning; secondly, LPS induced the primary brain cells in the hippocampus to cause inflammation model which were added NS398 (inhibitor of COX-2) and DKK-1 (inhibit

Evidence of hippocampal astrogliosis and antioxidant imbalance after L-tyrosine chronic administration in rats.

Tyrosinemia type II is a genetic disorder characterized by elevated blood levels of the amino acid tyrosine caused by the deficiency of tyrosine aminotransferase enzyme, resulting in neurologic and developmental difficulties in the patients. Although neurological sequelae are common in Tyrosinemia type II patients, the mechanisms involved are still poorly understood. The oxidative stress appears to be, at least in part, responsible for neurological complication in this inborn error metabolism. We observed that an acute injection of tyrosine in rats caused a massive oxidative stress in differen

Combining Ceftriaxone with Doxycycline and Daptomycin Reduces Mortality, Neuroinflammation, Brain Damage, and Hearing Loss in Infant Rat Pneumococcal Meningitis.

Despite appropriate antibiotic therapy, pneumococcal meningitis (PM) is associated with a case fatality rate of up to 30% in high-income countries. Survivors often suffer from severe lifelong disabilities. An excessive inflammatory reaction drives the pathophysiology, leading to brain damage and neurologic sequelae. We aimed to improve the outcome of experimental PM by simultaneously targeting different pathophysiological mechanisms with combined adjunctive therapies previously shown to be neuroprotective. In vitro, the anti-inflammatory effects of doxycycline and daptomycin were evaluated on

Astroglial water channel aquaporin 4-mediated glymphatic clearance function: A determined factor for time-sensitive treatment of aerobic exercise in patients with Alzheimer's disease.

Currently, there are no effective drug therapies for Alzheimer's disease (AD). Thus, exploring new non-pharmacological strategies, including the neuroprotective mechanisms of aerobic exercise, to enhance therapeutic treatment of AD are essential. Previous studies have shown that the beneficial efficiency of aerobic exercise in the prevention and treatment of AD is time-sensitive, but its mechanism is not clear. Recent studies revealed that the water channel protein aquaporin 4 (AQP4) mediates the glymphatic system to clear interstitial solutes, including β-amyloid, from the brain. More recentl

Involvement of the subplate zone in preterm infants with periventricular white matter injury.

Studies of periventricular white matter injury (PWMI) in preterm infants suggest the involvement of the transient cortical subplate zone. We studied the cortical wall of noncystic and cystic PWMI cases and controls. Non-cystic PWMI corresponded to diffuse white matter lesions, the predominant injury currently detected by imaging. Glial cell populations were analyzed in post-mortem human frontal lobes from very preterm [24–29 postconceptional weeks (pcw)] and preterm infants (30–34 pcw) using immunohistochemistry for glial fibrillary acidic protein (GFAP), monocarboxylate transporter 1(MCT1), i

Integrins in trabecular meshwork and optic nerve head: possible association with the pathogenesis of glaucoma.

Integrins are a family of membrane-spanning proteins that are important receptors for cell adhesion to extracellular matrix proteins. They also provide connections between the extracellular environment and intracellular cytoskeletons and are responsible for activation of many intracellular signaling pathways. In vitro and in vivo data strongly indicate that integrin-mediated signaling events can modulate the organization of the actin cytoskeleton in trabecular meshwork (TM) cells and are associated with astrocyte migration and microglia activation of the optic nerve head in patients with prima

Evolution of neuronal and astroglial disruption in the peri-contusional cortex of mice revealed by in vivo two-photon imaging.

In traumatic brain injury mechanical forces applied to the cranium and brain cause irreversible primary neuronal and astroglial damage associated with terminal dendritic beading and spine loss representing acute damage to synaptic circuitry. Oedema develops quickly after trauma, raising intracranial pressure that results in a decrease of blood flow and consequently in cerebral ischaemia, which can cause secondary injury in the peri-contusional cortex. Spreading depolarizations have also been shown to occur after traumatic brain injury in humans and in animal models and are thought to accelerat

Synaptic alterations in acoustic cortex in Creutzfeldt-Jacob disease.

The acoustic cortex was studied in electron microscope in 10 cases of Creutzfeldt-Jacob disease. The most prominent finding was the tremendous loss of neurons associated with marked reactive astrocytosis. The so called Cajal-Retzius cell had completely disappeared in the first cortical layer. The neuronal synapses were rare and most of those that were preserved demonstrated marked morphological alterations such as extensive dilatation of the synaptic terminals, synaptic polymorphism, dilatation of the cisternae of the smooth endoplasmic reticulum in the presynaptic terminals and accumulation o
