Factors influencing the incidence of recurrent glomerulonephritis following renal transplantation are poorly understood. Bilateral pretransplant native nephrectomy has been advocated to reduce the likelihood of recurrence after renal transplant. However, there is significant morbidity of native nephrectomy in the uremic population. Therefore, we sought to determine the effect of pretransplant native nephrectomy on the incidence of recurrent primary glomerulonephritis and the attendant risk of graft failure due to recurrent disease. Three hundred sixty-four consecutive cadaveric (n = 214), livi

The production of hydrofluoric acid (HF) and its use have increased remarkably due to the rapid development of the electronic and petrochemical industries. HF has a highly corrosive and penetrating action when it comes in contact with organic material, including body tissue. When burns are caused by a high concentration of HF, fatal cases have been reported, even when the contact with HF is only for a short time or on a small area of the body. The causes of death have been reported to be lung edema, mineral disturbance and systemic injury. But no pathological experiments have been reported. In

Within 20 min after i.v. injection or unilateral renal perfusion of rabbit anti-rat proximal tubular brush border antigens (RARFAXIA) into rats, fluorescence microscopy (FM) demonstrated rabbit IgG (RIgG) in a linear fashion along the endothelial region of the glomerular capillary walls. This finding was confirmed by immuno-electron microscopy (IEM) which revealed the presence of reaction product on the plasma membranes of the endothelial cells. Between 8 h and 26 days following i.v. injection of RARFXIA, granular subepithelial deposits of RIgG were demonstrated by FM and IEM, and the endothel

BACKGROUND: Late-phase thrombolysis within the 4.5-hour window for acute ischemic stroke exacerbates blood-brain barrier (BBB) disruption, increasing the risk of hemorrhagic transformation. Recombinant human prourokinase (rhPro-UK) is an emerging thrombolytic agent, but its impact on BBB integrity compared to alteplase (recombinant tissue plasminogen activator, rt-PA) and its potential additive effect with adjuvant neuroprotectants like Total Salvianolic Acid Injection (TSI) remain unclear.
METHODS: A mouse thromboembolic stroke model was established, and treatments were administered intraven

Mucous Membrane Pemphigoid is an orphan multi-system autoimmune scarring disease involving mucosal sites, including the ocular surface (OcMMP) and gut. Loss of tolerance to epithelial basement membrane proteins and generation of autoreactive T cell and/or autoantibodies are central to the disease process. The gut microbiome plays a critical role in the development of the immune system. Alteration in the gut microbiome (gut dysbiosis) affects the generation of autoreactive T cells and B cell autoantibody repertoire in several autoimmune conditions. This study examines the relationship between g

Hemidesmosomes mediate stable anchorage of epithelial cells to laminin-5 in the basement membrane zone and have been likened to spot-welds. Indeed, it has been assumed that hemidesmosomes are not dynamic, at least when compared to other matrix adhesion sites including focal contacts. We tested this notion by monitoring the fate of green fluorescent protein (GFP)-tagged human integrin beta4 subunit (GFP-hbeta4) and GFP-tagged 180-kD human bullous pemphigoid (BP) autoantigen (GFP-BP180) in live cultures of 804G cells that assemble numerous mature hemidesmosomes. In subconfluent 804G cells, both

BACKGROUND: More than 50 human placental proteins were isolated and physico-chemically characterized in the 70-80s by Hans Bohn and co-workers. Many of these proteins turned to have important role in placental functions and diagnostic significance in pregnancy complications. Among these proteins was membrane-associated placental protein 4 (MP4), for which identity or function has not been identified yet. Our aim was to analyze the sequence and placental expression of this protein in normal and complicated pregnancies including miscarriage, preeclampsia and HELLP syndrome.

The eye of the laughing dove (Spilopelia senegalensis) is uniquely adapted to thrive in arid environments, withstanding airborne particles and harsh conditions, demonstrating its remarkable structural and functional resilience. This study aimed to explore the cellular composition of the laughing dove's cornea, focusing on telocytes-a cell type previously unreported in avian corneas. This work represents the first demonstration of telocytes in the cornea of any bird species. We used hematoxylin and eosin (H&E) staining to comprehensively visualize the corneal layers, while silver staining, meth

It has been proposed that oxidative tissue damage is involved in the development of diabetic angiopathies. To evaluate this hypothesis, experiments were conducted to identify the retinal vessel changes induced by the oxidative stress related to alpha-tocopherol deficiency and examine possible similarities with the lesions characteristic of diabetic retinopathy. Twenty-one-day-old male Fisher 344 albino rats were divided randomly to receive a basal, chemically defined diet either with (adequate group) or without (deficient group) alpha-tocopherol. After 6 and 8 months, some rats (n = 3 per grou

Podocyte depletion is a common mechanism driving progression in glomerular diseases. Alport Syndrome glomerulopathy, caused by defective α3α4α5 (IV) collagen heterotrimer production by podocytes, is associated with an increased rate of podocyte detachment detectable in urine and reduced glomerular podocyte number suggesting that defective podocyte adherence to the glomerular basement membrane might play a role in driving progression. Here a genetically phenotyped Alport Syndrome cohort of 95 individuals [urine study] and 41 archived biopsies [biopsy study] were used to test this hypothesis. Po

Teneurins are type II transmembrane proteins that play important roles in pattern formation in Drosophila, axon fasciculation and organogenesis in Caenorhabidits elegans, and neuronal pathfinding in the visual system of the mouse. There is evidence that a peptide derived from a proteolytic event near the C-terminus of teneurins leads to formation of an active neuropeptide, while processing at and near the transmembrane domain leads to shedding of the extracellular domain into the extracellular matrix and the generation of an intracellular fragment that is transported to the nucleus. In vertebr

Horseradish peroxidase (HRP) tracer was applied to the gingival sulcus of implants or natural teeth at 5, 25, or 50 mg/ml to investigate the sealing capacities of the peri-implant epithelium (PIE) and junctional epithelium (JE); the extent of HRP penetration was observed under electron microscopy. A Ti-6Al-4V implant was inserted either immediately (immediate implantation) or 2 weeks (delayed implantation) after extraction of the maxillary left first molar of rats. The JE of the right molar was used as a control. Although the whole PIE of undecalcified sections appeared to be attached to the i

PURPOSE: Activation of epidermal growth factor receptor (EGFR) signaling in airway epithelial cells leads to epithelial-mesenchymal transformation (EMT) and airway remodeling in asthma. However, the mechanism regulating EGFR expression is not fully understood. We hypothesized that cadherin-26 (CDH26), a highly expressed cadherin in airway epithelium, promotes airway remodeling via regulating EGFR signaling in asthma.
METHODS: The expression of CDH26 in airway brush cells of asthmatic patients was examined using quantitative polymerase chain reaction. Airway hyperresponsiveness and remodeling

BACKGROUND: Human papillomaviruses (HPVs) utilize specifically modified proteoglycans (PGs) within the basement membrane as attachment factors prior to cell entry and infection. Similarly modified PGs are also found on cancer cells, which enables HPV with tumor-selective binding properties and highlights the potential development of HPV capsid-based anti-cancer therapies. In this study, we sought to investigate cancer-specific gene signatures and genetic factors that give rise to selective binding by HPV capsids.
METHODS: Using belzupacap sarotalocan (bel-sar; AU-011), an HPV virus-like drug

The extracellular matrix (ECM) provides key biochemical and biomechanical cues that govern fundamental cellular processes, including growth and migration. ECM dysregulation and altered cell-matrix interactions are drivers of cancer progression, exemplified by pancreatic ductal adenocarcinoma (PDAC), where an abnormally dense, collagen-rich, and stiff ECM correlates with poor patient outcomes. The PDAC microenvironment is poorly perfused, resulting in altered nutrient availability, yet how this metabolic stress shapes the ECM and its biological activity remains largely unknown. Herein, using mu

Patient-derived in vitro systems are powerful preclinical models that replicate key features of human tumors and enable investigation of cancer biology and drug response. They are particularly promising for pancreatic cancer research. We examined whether culture dimensionality and basement membrane extract (BME) composition affect establishment rates, morphology, proliferation, marker expression, and drug response in patient-derived models of pancreatic ductal adenocarcinoma (PDAC). From 12 PDAC samples, matched two-dimensional primary cell lines (PDCLs) and three-dimensional organoids were es

Liver fibrosis is a serious yet reversible intermediate stage in the progression of liver disease, which can ultimately advance to cirrhosis and hepatocellular carcinoma. Targeted and selective inhibition of activated hepatic stellate cells (aHSCs) has emerged as a promising therapeutic strategy for the treatment of liver fibrosis. However, the capillarization of liver sinusoidal endothelial cells (LSECs) characterized by the loss of fenestrae and continuous formation of basement membrane presents a significant barrier to effective delivery of anti-fibrotic agents. In this study, we propose a

INTRODUCTION: Pediatric rapidly progressive glomerulonephritis (RPGN) encompasses heterogeneous serology-based subtypes, including anti-glomerular basement membrane (anti-GBM), immune-complex-associated, and ANCA-associated (pauci-immune) RPGN. Although these subtypes differ in their underlying immune mechanisms, their ability to predict early renal recovery in children remains uncertain. This study aimed to evaluate the association between serology-based RPGN subtypes and early changes in renal function and tubular injury markers.
METHODS: This prospective cohort study included 30 children w

Diabetic kidney disease (DKD) is the main cause of end-stage renal disease (ESRD) worldwide. Podocytes are important components of the glomerular filtration barrier, which is located between glomerular capillary endothelial cells and the basement membrane, and is essential for maintaining normal renal function. Its injury is a key factor in the progression of DKD. Lipid homeostasis plays an important role in maintaining the normal physiological function of cells. In recent years, many studies have revealed that lipid metabolism disorders play a core role in driving podocyte injury, but its spe
