Activation of T-cell protein-tyrosine phosphatase suppresses keratinocyte survival and proliferation following UVB irradiation.

Chronic exposure to UV radiation can contribute to the development of skin cancer by promoting protein-tyrosine kinase (PTK) signaling. Studies show that exposure to UV radiation increases the ligand-independent activation of PTKs and induces protein-tyrosine phosphatase (PTP) inactivation. In the present work, we report that T-cell PTP (TC-PTP) activity is stimulated during the initial response to UVB irradiation, which leads to suppression of keratinocyte cell survival and proliferation via the down-regulation of STAT3 signaling. Our results show that TC-PTP-deficient keratinocyte cell lines

Proteasome inhibition upregulates Bim and induces caspase-3-dependent apoptosis in human mast cells expressing the Kit D816V mutation.

The majority of patients with systemic mastocytosis exhibit a D816V mutation in the activating loop of the Kit receptor expressed on mast cells. The Kit ligand regulates mast cell survival by transcriptional repression of the proapoptotic BH3-only protein Bim and by promoting Bim phosphorylation that makes it vulnerable for proteasomal-dependent degradation. We investigated here whether prevention of Bim degradation by a proteasomal inhibitor, MG132, would induce apoptosis in mast cells with the D816V mutation. Human umbilical cord blood-derived mast cells (CBMCs) with wild-type (wt) Kit and t

Platelet-activating factor in vasoobliteration of oxygen-induced retinopathy.

PURPOSE: To test whether platelet-activating factor (PAF) directly causes retinovascular endothelial cell (EC) death. METHODS: Retinovascular density was calculated in rat pups exposed to 80% O(2) from postnatal days (P)6 to P14 (to produce oxygen-induced retinopathy [OIR]), using the adenosine diphosphatase (ADPase) technique, in animals treated with distinct PAF receptor blockers (PCA-4248, BN52021, or THG315). PAF levels were then measured in the retinas. Viability of ECs from piglets and humans in response to C-PAF (a stable PAF analogue) was determined by the reduction of the tetrazolium

Spermidine attenuates cisplatin-induced dorsal lingual injury through modulation of oxidative stress, inflammation, apoptosis, and epithelial proliferation in rats.

Cisplatin (CIS)-induced oral mucosal injury is a common complication of chemotherapy that is largely mediated by oxidative stress, inflammation, apoptosis, and impaired epithelial regeneration. Spermidine (SP), an endogenous polyamine, contributes to cellular homeostasis, tissue repair, and epithelial regeneration and has well-documented antioxidant, anti-inflammatory, and cytoprotective activities. The present investigation evaluated the protective impact of SP against CIS-induced dorsal tongue injury in adult albino rats. Four groups were created: control, SP, CIS, and SP-CIS. Tongue tissues

Correlating the C-5 Induction Effect on Uracil-Based Nucleoside Lipid Nanoparticles for Enhanced Redox-Mediated Therapeutic Efficacy in Hepatocellular Carcinoma.

The clinical effectiveness of doxorubicin (DOX) in hepatocellular carcinoma (HCC) remains constrained by poor tumor selectivity and the emergence of multidrug resistance. In this work, we examine how the electronic nature of the C-5 substituent on uracil-based nucleoside lipids shapes the physicochemical and biological behavior of DOX-loaded lipid nanoparticles (LNPs). Four ionizable lipids were designed, combining either thymidine (methyl, electron-donating) or 5-fluorouridine (FURD; fluorine, electron-withdrawing) head groups with α-tocopherol or cholesterol tails through a triazole linker,

Hyperoside Stabilizes Redox-Mitochondrial-Inflammatory Networks in a Haloperidol-Induced Tardive Dyskinesia-Like Model.

Tardive dyskinesia (TD) is a persistent hyperkinetic movement disorder associated with prolonged dopamine D2 receptor blockade, particularly during chronic haloperidol (HP) exposure. Emerging evidence suggests that TD-like pathology is sustained by an interconnected redox-mitochondrial-inflammatory network within striatal circuits; however, the regulatory architecture of this network remains incompletely defined. Hyperoside (HS), a flavonol glycoside with cytoprotective properties, has been implicated in cellular stress-response modulation, yet its role in antipsychotic-induced motor dysfuncti

Isoform-specific expression of peroxiredoxins in human cancer cell lines with various radioresistance.

To investigate the role of peroxiredoxin isoforms (Prdx) in the radioresistance of cancer cells, the expression of Prdx1-6, DNA repair genes, and apoptosis regulators was studied in human cancer cell lines with varying radiosensitivities (A549, Caco-2, and MCF-7) after exposure to ionizing radiation. A correlation was found between high constitutive Prdx1-6 expression levels and increased radioresistance. Predominantly cytosolic isoforms Prdx2 and Prdx6 demonstrated pronounced induction after irradiation, indicating their critical role in protecting against radiation-induced oxidative stress.

Cannabidiol potentiates olaparib-induced cytotoxicity through cell cycle arrest and DNA damage modulation in breast cancer cells.

Triple-negative breast cancer (TNBC) remains a major clinical challenge due to its aggressive nature and limited treatment options, while therapeutic resistance in estrogen receptor-positive (ER+) breast cancer continues to limit treatment efficacy. Although olaparib is primarily effective in BRCA-mutated cancers, its activity in BRCA-wild type (BRCA-wt) tumors is limited. Therefore, this study aimed to investigate whether cannabidiol (CBD) can enhance the response of BRCA-wt breast cancer cells to PARP inhibition. The effects of olaparib (OLAP) and CBD, alone and in combination, were evaluate

Honokiol antagonizes cadmium-induced ultrastructural nuclear variation and mitochondrial dysfunction of hepatocytes through targeting Bax protein.

Cadmium (Cd) induces oxidative stress and inflammation, leading to hepatotoxicity in animals. Honokiol (HNK) has gained much attention owing to its anti-inflammatory and antioxidant properties, and may offer protection against liver diseases. However, whether HNK can improve Cd-induced ultrastructural and functional variation in hepatocytes is largely unknown. In this study, day-old broiler were divided into four treatment groups including control/untreated group Cd (50mg/L), HNK (50mg/kg), and Cd+HNK (50mg/L+50mg/kg) for 42 days, respectively. Copper complexes Cu1 triggers synergistic immunogenic cell death and pyroptosis for chemoimmunotherapy of triple-negative breast cancer.

Metal complexes possess unique spatial coordination frameworks and tunable biological functions, rendering them promising candidates for antitumor drug development. Herein, we rationally designed and synthesized a novel copper(II) complex Cu1 with the molecular formula Cu1(Cu(L)₂(ClO₄)₂·2H₂O, C₃₆H₂₆Br₂Cl₂CuN₈O₁₀S₂, where the ligand L corresponds to 2-([2,2':6',2″-terpyridin]-4'-yl)-4-bromothiazole. We systematically characterized its antitumor potency toward triple-negative breast cancer (TNBC) and dissected the underlying immunomodulatory mechanisms. UV-vis absorption titration and viscosity

In vitro tolerability of soluble silicic acid and tetraethyl orthosilicate in ocular epithelial cells.

Biodegradable silica microparticles (SiMPs) are a promising carrier matrix for controlled ocular drug delivery. In tissues, SiMPs degrade into silicic acid (SiA), a soluble inorganic weak acid. This study evaluated the in vitro toxicity and tolerability of SiA and SiMPs' precursor, tetraethyl orthosilicate (TEOS), in human corneal epithelial cells (HCE-2) and retinal pigment epithelial cells (ARPE-19). Cells were exposed to serial dilutions of SiA and TEOS. Cell viability was assessed after 24, 48, and 72 h using the alamarBlue assay, while membrane integrity was analyzed through LDH release.

Nitroxide Polymer Encapsulating Triptolide for Safe and Effective Small Cell Lung Cancer Therapy by Targeting Energy Metabolism and Pyroptosis.

Small cell lung cancer (SCLC) is highly aggressive neuroendocrine tumor with a paucity of effective therapeutic option. Triptolide (TP), a natural diterpenoid triepoxide, exhibits potent antitumor activities, yet its efficacy and underlying mechanisms in SCLC remain largely undefined. Here, we demonstrate that TP effectively inhibits cell viability across all molecular subtypes of SCLC, with greater potency than in nonsmall cell lung cancer. Mechanistically, TP transcriptionally represses hexokinase 1 and 2 (HK1/2), thereby disrupting the glycolysis-TCA-OXPHOS axis and resulting in ATP depleti
