Synthesis and secretion of high- and low-molecular-weight forms of the enzyme-releasing peptide (ERP) from the macrophage-like cell line THP-1.

Neutrophil enzymes have been implicated as a source of lung injury in patients with the adult respiratory distress syndrome (ARDS) and with emphysema. We studied a human alveolar macrophage-derived peptide messenger, the enzyme-releasing peptide (ERP), which causes neutrophils to secrete their enzymes. The secretion and synthesis of ERP was studied in human alveolar macrophages and in the macrophage-like cell lines THP-1, HL-60, and U937. All four cell types secrete an ERP-like peptide. THP-1 cells secrete a higher concentration of the peptide than do macrophages. The secretion of ERP by THP-1

Cellular retention, toxicity and carcinogenic potential of seafood arsenic. I. Lack of cytotoxicity and transforming activity of arsenobetaine in the BALB/3T3 cell line.

Cytotoxicity, morphological neoplastic transformation, intracellular retention and metabolic behaviour have been investigated in BALB/3T3 Cl A 31-1-1 cells for arsenobetaine, the main form of arsenic in certain seafoods, in comparison to inorganic sodium arsenite. In order to avoid false results, particular attention was paid to the purity, checking for the presence of any trace amounts of inorganic arsenic as well as methylated contaminants in the chemically synthesized arsenobetaine. Cytotoxicity and morphological transformation assays gave obvious positive results for sodium arsenite at a d

Surface and cytoplasmic expression of CD2, CD13, and CD25 antigens in human T lymphotropic virus type I infected cell line. An immunoelectron microscopic study.

Immunoperoxidase electron microscopy revealed intracellular expression of CD2, CD13, and CD25 antigens in a unique human T lymphotropic virus type I infected cell line, TI-CL. This cell line is thought to be derived from cord mononuclear cells in the bifurcation stage of T cell and myelomonocytic lineage, based on cytochemical, immunologic, and molecular genetic analyses. Each antigen expression was observed on the plasma membranes, the cisternae of rough endoplasmic reticulum, and the perinuclear cisternae. Surface and cytoplasmic expression of CD25 antigen in adult T cell leukemia/lymphoma c

Characterization and affinity cross-linking of receptors for human recombinant lymphotoxin (tumor necrosis factor-beta) on a human histiocytic lymphoma cell line, U-937.

Recombinant human lymphotoxin (rhLT) expressed in a mammalian cell line was purified and used to examine its receptors on the human histiocytic lymphoma cell line U-937. rhLT was radioiodinated by the IODO-GEN method to a specific activity of 60 microCi/micrograms; the labeled protein was biologically active in the cytolytic assay, and displaceable binding to U-937 cells was observed. The specific binding reached a plateau within 10, 60, and 180 min at 37, 23, and 4 degrees C, respectively. Scatchard analysis of the binding data revealed the presence of a single class of high affinity receptor

Extensive esterification of adrenal C19-delta 5-sex steroids to long-chain fatty acids in the ZR-75-1 human breast cancer cell line.

Estrogen-sensitive human breast cancer cells (ZR-75-1) were incubated with the 3H-labeled adrenal C19-delta 5-steroids dehydroepiandrosterone (DHEA) and its fully estrogenic derivative, androst-5-ene-3 beta,17 beta-diol (delta 5-diol) for various time intervals. When fractionated by solvent partition, Sephadex LH-20 column chromatography and silica gel TLC, the labeled cell components were largely present (40-75%) in three highly nonpolar, lipoidal fractions. Mild alkaline hydrolysis of these lipoidal derivatives yielded either free 3H-labeled DHEA or delta 5-diol. The three lipoidal fractions

The t(8;14) chromosome translocation of the Burkitt lymphoma cell line Daudi occurred during immunoglobulin gene rearrangement and involved the heavy chain diversity region.

Recent molecular analyses of Burkitt lymphomas carrying the t(8;14) chromosome translocation have indicated that a dichotomy exists regarding the molecular mechanisms by which the translocations occur. Most sporadic Burkitt tumors carry translocations that apparently arise due to mistakes in the immunoglobulin isotype-switching process. In contrast, there is evidence that the translocations of most endemic Burkitt lymphomas occur as a consequence of aberrant V-D-J recombination of variable, diversity, and joining gene segments, catalyzed by the recombinase enzymes. This phenomenon was first no

Production of human hematopoietic survival and growth factor by a myeloid leukemia cell line (KPB-M15) and placenta as detected by a monoclonal antibody.

Fifty-five hematopoietic cell lines, including 19 T-, 16 B-, 5 pre-B-, 5 non-T non-B-, 1 erythroid, and 9 myeloid-monocytoid cells, were screened for production of human hematopoietic survival and stem cell growth factor (SCGF) by enzyme immunoassay using anti-SCGF monoclonal antibody. The KPB-M15 myeloid cell line constitutionally secreted a considerable quantity of SCGF, while other T- or myeloid-monocytoid cell lines did not secrete SCGF. Other biomaterials investigated were fetal calf, horse, and human serum; granulocyte-macrophage colony-stimulating factor and erythropoietin preparations;

Real-World Comparative Survival Outcomes of First-Line Serplulimab Plus Chemotherapy in Extensive-Stage Small-Cell Lung Cancer: A Single-Center Retrospective Cohort Study.

BACKGROUND: Extensive-stage small-cell lung cancer (ES-SCLC) remains an aggressive malignancy with poor survival despite platinum-etoposide chemotherapy. Although serplulimab plus chemotherapy has demonstrated a survival benefit in randomized trials, comparative real-world evidence remains limited. We therefore evaluated survival outcomes with first-line serplulimab plus chemotherapy versus chemotherapy alone in patients with ES-SCLC. METHODS: We conducted a single-center retrospective cohort study at Tianjin Chest Hospital. Patients with ES-SCLC who initiated first-line platinum-etoposide-ba

Generation of an Induced Pluripotent Stem Cell Line from a Patient with Infantile Liver Failure Syndrome 2 Carrying a Compound Heterozygous Mutation in NBAS.

Infantile liver failure syndrome 2 (ILFS2, OMIM 616483) is a rare autosomal recessive disorder caused by neuroblastoma amplified sequence (NBAS) gene mutation, resulting in recurrent episodes of acute liver failure (ALF). The first onset of ALF is more common in infancy or early childhood. Recently, we identified a compound heterozygous mutation, c.3596G>A (p.C1199Y)/ex.9del (p.216-248del), in the NBAS gene in a patient with ILFS2. The human dermal fibroblasts (HDFs) of the patient were obtained, and a line of induced pluripotent stem cells (iPSCs) was successfully generated. The iPSC line wil

First-Line Lorlatinib for ALK-Rearranged Pulmonary Large-Cell Neuroendocrine Carcinoma with Brain Metastases and Suspected Leptomeningeal Involvement: A Case Report and Literature Review.

We report a case of ALK-rearranged pulmonary large-cell neuroendocrine carcinoma (LCNEC) with brain metastases and radiologically suspected leptomeningeal involvement, successfully treated with first-line lorlatinib. A 58-year-old woman was diagnosed with LCNEC, and molecular testing revealed an EML4-ALK fusion. Lorlatinib achieved a partial response, with regression of the primary lesion, brain metastases, and suspected leptomeningeal lesions. Our literature review showed that ALK-positive LCNEC occurs more often in younger, female, and never- or light-smoking patients. In such cases, molecul

Cell line-dependent effects of β‑estradiol and testosterone on aurora kinases, GATA3, and associated miRNAs in hepatocellular carcinoma models HepG2 and Huh‑7.

BACKGROUND: Higher prevalence of hepatocellular carcinoma (HCC) in men is largely attributed to the sex hormones and behavioral risk factors. This study investigated the effects of male (testosterone) and female (estradiol) hormones on Aurora-A/B kinases, GATA-3, and microRNAs (miR-3941, miR-4500, miR-4742) in HepG2 and Huh-7 cell lines. METHODS: In this study, two human hepatocellular carcinoma cell lines, HepG2 (with a differentiated phenotype) and Huh-7 (with more aggressive tumorigenic behavior), were treated with the sex hormones β-estradiol and testosterone. Hormonal effects were evalua

Osmolality-Independent Impact of Sodium on Glycosylation of an Fc-Fusion Protein and the Hexosamine Biosynthesis Pathway in a Chinese Hamster Ovary Cell Line.

This study investigates the osmolality-independent effects of sodium and potassium on the glycosylation of an Fc-fusion protein and the activity of the hexosamine biosynthesis pathway in a Chinese Hamster Ovary cell line. Previous research linked low molecular weight proteoforms of this Fc-fusion protein to reduced N-glycan complexity and O-glycan site occupancy. Through a series of batch experiments, we demonstrated that increased concentrations of sodium or potassium ions led to a reduction of these proteoforms. Our findings suggest that ion availability impacts the activity of the hexosamin

CDK8 Inhibition Increases E2F1 Transcriptional Activity and Promotes STAT3-Dependent Suppression of Mcl-1 in Triple-Negative Breast Cancer Cell Line MDA-MB-468.

The targeting of cyclin dependent kinase 8 (CDK8) as a potential strategy for cancer treatment has been of interest since the identification of CDK8 as an oncogene product. In this report, we communicate the results of our continuing investigation into the effects of CDK8 inhibitor on triple-negative breast cancer cell line MDA-MB-468. Here, we demonstrate that inhibition of CDK8 decreases phosphorylation of CDK8 substrates E2 promoter binding factor 1 (E2F1) at serine 375 and signal transducer and activator of transcription 3 (STAT3) at serine 727 in these cells. Additionally, luciferase expr
