Failure of lysosomal acidification and endomembrane network in neurodegeneration.

Lysosomes have emerged as central hubs in the regulation of the endomembrane system, extending beyond degradation to coordinate organelle communication. Central to this regulatory role is vacuolar-type H+-ATPase (V-ATPase), a proton pump that acidifies the lysosomal lumen to enable hydrolase activity and support proteostasis. In addition to its lysosomal functions, V-ATPase influences the physiology of other organelles, including the endoplasmic reticulum (ER), Golgi apparatus and mitochondria, through both direct and indirect mechanisms involving acidification-dependent processes, such as pro

Macrophages drive a fibrogenic gene program of periductal fibroblasts in pediatric primary sclerosing cholangitis.

Primary sclerosing cholangitis (PSC) is an autoimmune, cholestatic liver disease characterized by inflammation and fibrosis surrounding bile ducts. The cellular crosstalk driving periductal fibrosis remains poorly defined. This study applied a multi-omics approach integrating MERSCOPE spatial transcriptomics, bulk RNA-seq, and SomaScan proteomics to characterize fibrotic periductal regions and their cell-cell communications. Macrophages (MP) expressing moderate-to-high CD163 were found co-localized with cholangiocytes, T cells, and collagen-producing hepatic stellate cells (HSC). Cell niche an

Early warning and response systems for respiratory disease outbreaks: lessons learnt from cluster-associated cases of acute respiratory illnesses in Gilgil subcounty, Nakuru County, Kenya, 2021.

Investigating acute respiratory illnesses (ARIs) is difficult due to non-specific symptoms, varied health-seeking behaviors, and resource limitations; yet early detection is critical to global health security. Kenya's Ministry of Health (MOH) uses the Integrated Disease Surveillance strategy for public health surveillance, incorporating event-based surveillance (EBS) and indicator-based surveillance (IBS) for early warning system. MOH, supported by the US-CDC, established Influenza Sentinel Surveillance (ISS) in 2006 and later launched community EBS (CEBS) and health facility EBS (HEBS) pilots

Perspectives of disabled adults on healthcare professionals role in promoting physical activity in China A reflexive thematic analysis.

Healthcare professionals (HCPs) have been identified as key messengers for disabled adults in promoting physical activity (PA). However, few studies revealed the disabled adults' views and preferences for HCPs in PA. This study aimed to understand disabled adults' perceptions and experiences of the role of HCPs in promoting PA in China. Forty-one Chinese disabled adults participated in one-to-one semi-structured interviews, recruited by three purposive sampling strategies: maximum variation, criterion-based and snowball sampling. Data from interviews were analysed using reflexive thematic anal

Core cooperative metabolism in low-complexity CO2-fixing anaerobic microbiota.

Biological conversion of carbon dioxide into methane has a crucial role in global carbon cycling and is operated by a specialised set of anaerobic archaea. Although it is known that this conversion is strictly linked with cooperative bacterial activity, such as through syntrophic acetate oxidation, there is also a limited understanding on how this cooperation is regulated and metabolically realised. In this work, we investigate the activity in a microbial community evolved to efficiently convert carbon dioxide into methane and predominantly populated by Methanothermobacter wolfeii. Through mul

Microenvironmental Gradients Drive Spatial Stratification of Saccharifying Microbial Communities and Enzyme Activity in Strong-Flavor Daqu Fermentation.

Daqu, a representative solid-state fermentation product, produces saccharifying enzymes to degrade sorghum starch into fermentable sugars for ethanol synthesis. Spatial heterogeneity in Daqu drives community assembly. However, its regulatory role in enzyme-driven saccharification remains unclear. By integrating metagenomics and PacBio full-length sequencing, this study investigated how microenvironmental gradients across distinct Daqu layers (QP (surface layer), HQ (middle layer), QX (center layer)) shape saccharifying microbiota and activity. Saccharifying activity exhibited a declining surfa

The treated wastewater enhances the biodegradation of sulfonamide antibiotics in biofilm-sediment downstream of the receiving river outlet.

Although the treated wastewater meets the discharge standards, it can still become a potential transmitted stressor that affects aquatic organisms in receiving rivers. Biofilms and sediments as the main solid-phase substances in natural aquatic environments can biodegrade micropollutants. However, most of the current studies have selected a single solid-phase material, and there are relatively few studies that comprehensively consider the effect of treated wastewater on the dissipation of micropollutants in a composite biofilm-sediment system. Therefore, this study investigated the dissipation

Biodegradable Implantable Electronics with Wireless Technology for Real-Time Clinical Applications.

Wireless biodegradable electronics offer a transformative approach to transient biomedical applications by combining fully implantable, resorbable architectures with untethered communication and power delivery. These systems address key limitations of conventional implants, including infection risk, foreign body response, and the need for surgical retrieval. As biodegradable implants are designed to disappear after fulfilling their function, wireless operation is essential to avoid permanent components such as transcutaneous wires. Advances in bioresorbable materials have enabled electronic co

The Role of Clostridioides difficile Within the One Health Framework: A Review.

Clostridioides difficile is the leading cause of antibiotic-associated diarrhea in hospitalized patients. In recent years, the incidence of C. difficile infection (CDI) has increased globally, with a notable rise in community-associated CDI (CA-CDI). The presence of the microorganism in animals, the environment, and food suggests that these sources may contribute to the spread of the infection in the community. This review applies a One Health approach, integrating human, animal, and environmental health, to provide a comprehensive strategy for understanding and managing this pathogen. Finding

A combined morphological and genetic survey of helminths in the European green toad Bufotes viridis (Laurenti, 1768) from eastern Slovakia.

Accurate characterization of helminth communities in amphibian hosts is essential for understanding host-parasite dynamics in changing environments. This study presents an integrative parasitological survey of Bufotes viridis populations in eastern Slovakia, using both morphological and molecular methods. A total of 61 road-killed individuals collected across 13 localities were examined for helminth presence. Only nematodes were detected, encompassing 3 families, Rhabdiasidae, Molineidae and Cosmocercidae. Four nematode species were recorded, Rhabdias rubrovenosa, Oswaldocruzia filiformis, O.

Mpox and the Ethics of Outbreak Management: Lessons for Future Public Health Crises.

Mpox, first identified in captive monkeys in 1958 and recognized in humans by 1970 in the Democratic Republic of Congo, was historically confined to sporadic zoonotic outbreaks in Central and West Africa. These outbreaks, often driven by rodent-to-human transmission in resource-limited settings, reflect persistent systemic health disparities. In recent years, mpox has also been reported in high-income countries (including the United States, United Kingdom and Europe), underscoring its global health implications beyond traditionally endemic regions. This article examines the mpox outbreak throu

A review of zoonotic chlamydiae species in Africa: assessing their burden and potential impact through One Health perspective.

Chlamydia species, a group of obligate intracellular Gram-negative bacteria, affect humans, livestock, companion animals, and wildlife, with infections ranging from asymptomatic to severe depending on host species and strain. Diagnosis can be difficult due to mild lesions or co-infections. Because Chlamydiaceae infect multiple hosts, a One Health approach, integrating human, animal, and environmental health is essential for effective control and prevention. C. trachomatis remains endemic in many regions, while Chlamydia pneumoniae is implicated in community-acquired pneumonia. C. abortus threa

Fungal diversity differences in the indoor dust microbiome from built environments on earth and in space.

Human occupied built environments are no longer confined to Earth. In fact, there have been humans living and working in low-Earth orbit on the International Space Station (ISS) since November 2000. With NASA's Artemis missions and the age of commercial space stations set to begin, more human-occupied spacecraft than ever will be in Earth's orbit and beyond. On Earth and in the ISS, microbes, especially fungi, can be found in dust and grow when unexpected, elevated moisture conditions occur. However, we do not yet know how indoor microbiomes in Earth-based homes and in the ISS differ due to th
