Tuning the Affinity of Chimeric Antigen Receptors Enhances the Function of Human Engineered Regulatory T Cells.

Chimeric antigen receptors (CAR) incorporating single-chain variable fragments (scFv) exhibit much higher affinity for their cognate antigens than native T cell receptors (TCR). Reducing the affinity of the CAR has been shown to improve the function of effector CAR-T cells by limiting their activation-induced exhaustion while preserving their capacity for serial killing of antigen-rich tumor cells. CAR technologies are increasingly being applied to regulatory T cell (Treg)-based immunotherapies as well. However, the impact of CAR affinity on Treg biology and function remains poorly understood.

An interface engineered ZnO/rGO/Au NPs@MoS2 heterojunction for enhanced photoelectrochemical DNA sensing.

The conventional polymerase chain reaction (PCR) method for DNA detection suffers from limited sensitivity and linearity, restricting its application to samples with molar concentrations above 10-2 mmol. Here, we report a highly sensitive photoelectrochemical (PEC) biosensor for detecting the PML/RARA fusion gene, a key biomarker of acute promyelocytic leukemia (APL). A biosensing interface was constructed using an 18-mer single-stranded DNA probe immobilized on a ZnO/rGO/Au NPs@MoS2 heterojunction. The integration of three-dimensional ZnO nanoflowers with ultrathin MoS2 nanosheets not only su

Symmetry-breaking engineered for polarization-sensitive bulk photovoltaic effect in b-As/2H-TMDs heterostructures.

The bulk photovoltaic effect (BPVE) provides a junction-free route to spontaneous photocurrent and photovoltage beyond Shockley-Queisser limit. Current BPVE designs mainly rely on interfacial symmetry breaking induced by low-symmetry materials, yet the resulting interfacial polarity and symmetry-breaking strength are often constrained by the constituent symmetry, orbital polarizability and interfacial charge redistribution, restricting further enhancement of BPVE. Here, we realize a symmetry-engineered BPVE by vertically stacking twofold-symmetric black arsenic (b-As) with threefold-symmetric

Programmable engineered bacteria manipulate metabolism and remodel the TME in situ for enhancing adoptive cell therapy.

In solid tumors, the efficacy of adoptive T cell therapy (ACT) is limited by a metabolically constrained tumor environment that undermines T cell persistence, fitness, and infiltration. Here, we engineered a hypoxia-activated bacterial hybrid system (MM@TMV) to address these barriers. This hybrid system integrates metabolically engineered bacteria and tumor membrane vesicles (TMVs) to achieve tumor-restricted metabolic reprogramming and immune reinforcement within hypoxic tumors. Within hypoxic tumors, D-mannose is produced in situ to support stem-like phenotypes and limit exhaustion, while TM

Phytochemical engineered quantum dots as potential therapy to counter alpha synuclein aggregation in Parkinson's disease.

Parkinson's disease (PD) is a prevalent neurodegenerative disorder that critically impairs human health and presently lacks effective cellular-level therapeutic interventions. The disease is primarily characterized by pathological aggregation of misfolded α-synuclein in presynaptic neurons, leading to dopaminergic neuronal loss. The limited efficacy of current pharmacological treatments stems largely from challenges in crossing the blood-brain barrier. Recent studies suggest that nano-phytomedicine approaches offer promising alternatives for PD management. Specifically, phytochemical-engineere

Efficient targeting of human glial progenitor cells in vivo with engineered AAV vectors and glymphatic delivery.

Treatment of neurological diseases that involve oligodendrocytes or astrocytes would benefit from the selective delivery of viral vectors to their common parent, glial progenitor cells (GPCs). Here, we select adeno-associated virus (AAV) capsids with tropism for human GPCs and demonstrate efficient and widespread delivery of the resultant AAVs throughout the mouse brain through the glymphatic system. In vivo screening of a library of capsid-modified, recombination-reported AAVs in chimeric mice engrafted with PDGFRA-driven Cre recombinase-expressing human GPCs identified a set of AAV5-based ve

Microfluidic-mass spectrometry analysis of blood-brain barrier transport using engineered microparticle interfaces.

The blood-brain barrier (BBB) is vital for maintaining central nervous system (CNS) homeostasis but represents a formidable obstacle to drug delivery, underscoring the need for physiologically relevant in vitro models for CNS drug screening. However, existing in vitro BBB models remain limited in their ability to simultaneously recapitulate multicellular architecture, spatial anisotropy, and dynamic transport behavior while enabling real-time, quantitative analysis. Here, we present an integrated microfluidic-mass spectrometry platform that utilizes engineered BBB particles (BBBps) as a functi

An engineered M13 phage-rGO electrochemical biosensor for rapid detection of viral protein in complex matrices.

The demand for rapid and scalable biosensing technologies has motivated the development of antibody-free platforms capable of operating in complex sample environments. Here, we report an electrochemical biosensor based on engineered M13 bacteriophages displaying a SARS-CoV-2 spike S1-binding peptide immobilized on a reduced graphene oxide (rGO) transducer. The sensor employs a chemiresistive detection mechanism under a fixed low-voltage bias, enabling rapid electrical readout following target binding. Detection of S1 protein was achieved in buffer and in spiked complex matrices, including feta

Forecasting Thailand's mobility trends using Feature Engineered XGBoost for pandemic crisis movement management.

The COVID-19 pandemic significantly disrupted global mobility patterns, with widespread population movement playing a key role in the transmission of the virus. In such a situation, Google introduced the Community Mobility Reports, which use anonymized and aggregated location data to monitor changes in movement across various location categories. These mobility trends provide important insights that help inform timely public health interventions and support data-driven decisions during and after the pandemic. This study aims to forecast human mobility trends in Thailand during the COVID-19 pan

Twin engines of viral replication: Medicinal chemistry insights on SARS-CoV-2 nsp12 and nsp13.

The SARS-CoV-2 pandemic highlighted the urgent need for antivirals targeting essential viral enzymes. Herein, we critically examine the current landscape of small molecule inhibitors targeting the highly conserved non-structural protein 12 (nsp12), the RNA-dependent RNA polymerase, and non-structural protein 13 (nsp13), the helicase, both critical for viral genome replication. Structural and mechanistic features that inform rational inhibitor design, including active sites and cofactor interactions, are discussed. For nsp12, nucleoside analogues derived from a drug repurposing strategy, as wel

Integrated assessment of CI engine combustion and catalyst durability with graphene- infused Pongamia pinnata biodiesel.

The need for sustainable fuels in compression ignition (CI) engines is highlighted by the escalating energy crisis and stringent emissions regulations. This study examined diesel- Pongamia methyl Ester (PME) blends (B10, B20, B30) and their graphene-infused counterparts (30 ppm; B10Gr, B20Gr, and B30Gr) at 1500 rpm under various loads in a single cylinder CI engine. In addition to the comparatively unexplored topic of catalytic converter durability, the investigation concentrated on combustion, performance, and emission behaviour. Because of the improved atomisation and catalytic effects of gr

Engineered human iPSC-derived dendritic cells dressed with tumor MHC complexes as a cancer vaccine.

Autologous-derived dendritic cells (DCs) are a promising source for cell-based cancer vaccines. However, their therapeutic potential is challenged by the number and quality produced and the diversity of antigens presented. To address these limitations, we present an approach that involves differentiating universal MHC-deficient human-induced pluripotent stem cells (hiPSCs) into CCR7+ migratory DCs. These DCs are subsequently "dressed" with the full repertoire of MHC-antigen complexes derived from tumor cell membranes, transforming an allogeneic substrate into a personalized cancer vaccine prod

Integrative structural insights into the IgG-FcRn interactions revealed by engineered FcRn-immobilized affinity chromatography.

Molecular interactions between therapeutic antibodies and neonatal Fc receptor (FcRn) are major contributors to the long serum half-life of antibodies. Therefore, the in vitro affinity of IgG for FcRn is an important indicator of in vivo pharmacokinetic properties. However, the molecular basis of IgG-FcRn interactions is not fully understood from experimental and structural perspectives. Affinity evaluation using surface plasmon resonance is difficult. The molecular mechanism by which domains other than Fc or surface charges affect FcRn binding remains unclear. In this study, we developed an e

From Design to Bedside: Brain-Targeted Engineered Nanoparticles and Predictive Modeling for Neurodegenerative Diseases Therapeutics.

Nanoparticle (NP)-based drug delivery systems offer promising solutions for neurodegenerative diseases such as Alzheimer's, Parkinson's, and Huntington's, which have no cure due to the brain's structural complexity and the restrictive nature of the blood-brain barrier (BBB). Conventional therapeutics, despite strong in vitro efficacy, often fail clinically due to poor brain penetration. Recent advances in NP engineering, including optimization of size (10-100 nm), surface charge (-10 to -20 mV), and targeted ligand decoration, have improved BBB crossing and reduced premature clearance. No Environment, No Education: Honoring the Clinical Learning Environment as the Engine of Competency-Based Education - A Call to Action.

Competency-Based Health Professions Education (CBHPE) has transformed health professions training by emphasizing demonstrated competence over time-based progression. Yet the clinical learning environment (CLE), the social, cultural, and organizational ecosystem in which learning occurs, has not evolved in parallel. Persistent hierarchies, workload pressures, and inadequate faculty development limit CBHPE's full potential. Thus, the authors sought to identify priorities for CLE reform. A structured consensus process was conducted at the International Competency-Based Education Collaborators Sum

Reducing Specialist Wait Times: What Can We Learn from Highway Traffic Engineers?

Waiting times for specialist visits in the United States are increasing, and many organizations are responding by seeking to hire more physicians. However, regional data on the supply of physicians in various specialties and waiting times suggest that this approach alone is unlikely to be effective and may even worsen the problem. This observation is consistent with the dynamics observed when lanes are added to highways but fail to reduce congestion because drivers' thresholds for using the highways are lowered. Meeting this challenge will most likely require other tactics (analogous to those

Synergistic Thrombolysis-Antiplatelet Therapy of Engineered tPA Variant citPA5 and Tirofiban in a Mouse Stroke Model.

Acute ischemic stroke is a leading cause of global disability and mortality. Current therapies are limited by a narrow 4.5-hour treatment window, high risk of bleeding, frequent reocclusion post-thrombolysis, and poor neuroprotective/repair capacity. The thrombolytic activity of citPA5 with or without tirofiban was first assessed using in vitro clot lysis assays. Therapeutic efficacy and safety were then evaluated in a murine carotid embolism-induced stroke model. Mice received citPA5 plus tirofiban at 0.5 hours after occlusion. From biological surfaces to engineered systems: how synthetic and systems biology enable biomimetic design.

Biological surfaces perform a wide range of functions, including communication, molecular exchange, defense, and movement. While these architectures have long inspired biomimetic surface engineering, most existing reviews focus on fabrication methods, often overlooking the biological mechanisms underlying the functions. This review examines how the structural and functional properties of biological surfaces can inform the design of advanced biomimetic systems. Using an integrative framework combining systems and synthetic biology, we establish key design principles derived from biological surf
