Deep sequencing-based analysis of the anaerobic stimulon in Neisseria gonorrhoeae.

BACKGROUND: Maintenance of an anaerobic denitrification system in the obligate human pathogen, Neisseria gonorrhoeae, suggests that an anaerobic lifestyle may be important during the course of infection. Furthermore, mounting evidence suggests that reduction of host-produced nitric oxide has several immunomodulary effects on the host. However, at this point there have been no studies analyzing the complete gonococcal transcriptome response to anaerobiosis. Here we performed deep sequencing to compare the gonococcal transcriptomes of aerobically and anaerobically grown cells. Using the informat

Conjugative IncFI plasmids carrying CTX-M-15 among Escherichia coli ESBL producing isolates at a University hospital in Germany.

BACKGROUND: Multi-drug-resistant, extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae, constitute an emerging public-health concern. Little data on the molecular epidemiology of ESBL producing Escherichia coli is available in Germany. Here we describe the prevalence and molecular epidemiology of ESBL producing-Escherichia coli isolates at a German University hospital. METHODS: We analysed 63 non-duplicate clinical ESBL isolates obtained over an 8-month period using PCR and sequence-based ESBL allele typing, plasmid replicon typing, phylogenetic group typing. Identification and evolution of drug efflux pump in clinical Enterobacter aerogenes strains isolated in 1995 and 2003.

BACKGROUND: The high mortality impact of infectious diseases will increase due to accelerated evolution of antibiotic resistance in important human pathogens. Development of antibiotic resistance is a evolutionary process inducing the erosion of the effectiveness of our arsenal of antibiotics. Resistance is not necessarily limited to a single class of antibacterial agents but may affect many unrelated compounds; this is termed 'multidrug resistance' (MDR). The major mechanism of MDR is the active expulsion of drugs by bacterial pumps; the treatment of gram negative bacterial infections is comp

In vitro activity of temocillin against extended spectrum beta-lactamase-producing Escherichia coli.

OBJECTIVES: Temocillin is a semi-synthetic 6-alpha-methoxy derivative of ticarcillin. It is highly stable to most bacterial beta-lactamases. However, data concerning its activity against extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae are limited. We have analysed the in vitro activity of temocillin against clinical isolates of ESBL-producing Escherichia coli over the past 4 years in a university hospital in Brussels, Belgium. METHODS: Strains were screened for ESBL production using the double-disc synergy test. The MIC of 12 antimicrobial agents was determined for ESBL-p

The microbiology of ventilator-associated pneumonia.

Ventilator-associated pneumonia (VAP) is a common complication of ventilatory support for patients with acute respiratory failure and is associated with increased morbidity, mortality, and costs. Awareness of the microbiology of VAP is essential for selecting optimal antibiotic therapy and improving these outcomes. The specific microbial causes of VAP are many and varied. Most cases of VAP are caused by bacterial pathogens that normally colonize the oropharynx and gut, or that are acquired via transmission by health-care workers from environmental surfaces or from other patients. Common pathog

Application of an rRNA probe matrix for rapid identification of bacteria and fungi from routine blood cultures.

One of the most important functions of the clinical microbiology laboratory is the identification of the etiology of sepsis. For this study, aliquots from 405 positive blood cultures were tested against a unique array of DNA probes directed against rRNA subsequences of bacteria and fungi for identification. Another 280 samples that were negative after 5 days of incubation were also tested. Blood culture bottles were incubated in a BacT/Alert3D instrument. For the rRNA assay, a 0.4-ml aliquot was removed, and the cells were pelleted by centrifugation. The pellet was washed and frozen at -70 deg

Recent developments in carbapenems.

Carbapenems are beta-lactam antibiotics characterised by the presence of a beta-lactam ring with a carbon instead of sulfone in the 4-position of the thyazolidinic moiety. The first carbapenem to be utilised in therapy was imipenem, the N-formimidoyl derivative of thienamycin. Imipenem is coadministered with cilastatin, an inhibitor of human renal dehydropeptidase I, as imipenem is hydrolysed by this enzyme. Meropenem was the first carbapenem with a 1-beta-methyl group and 2-thio pyrrolidinyl moiety, which renders this antibiotic stable to renal dehydropeptidase I. Other carbapenems for parent

Pharmacodynamic (kinetic) considerations in the treatment of moderately severe infections with cefotaxime.

Information about the pharmacodynamics of beta-lactams has accumulated rapidly over the last 20 years, and their application to cefotaxime are discussed in this review. Application of pharmacodynamics requires an integration of the pharmacokinetic and in vitro properties of the agent. Cefotaxime is similar to other beta-lactams in that it has little concentration-dependent killing and produces no postantibiotic effect against Gram-negative bacteria. However, it has a microbiologically active metabolite, deascetylcefotaxime, which can show synergy, partial synergy, or an additive effect in comb

Pediatric blood cultures.

The clinical and laboratory issues important in pediatric blood cultures are similar to those in adult blood cultures with a few noteworthy exceptions. The collection of an uncontaminated specimen and an ample volume of blood is more difficult, especially in neonates. In addition, children often have previously received oral antibiotics or a broad-spectrum parenteral antibiotic. The relative frequencies of the pathogens causing bacteremia in children are different in important ways from in adults. Haemophilus influenzae b, although much less common than in the past, is still an important pedia

Antibiotic resistance in adult female patients hospitalized for acute pyelonephritis: rates and predicting factors.

INTRODUCTION: The empiric treatment of acute pyelonephritis (APN) with third generation cephalosporins (3GC) or fluoroquinolones (FQ) has been challenged by Escherichia coli resistance reported by community surveillance networks. But these could overestimate resistance because they do not discriminate between uncomplicated and complicated, or between community and care-related infections. OBJECTIVES: We had for aim to: quantify resistance rates in hospitalized patients presenting with APN; identify subgroups with resistance <10% that could still be treated empirically with FQ or 3GC. PATIENT

Gut microbiota profiles in Korean children with growth hormone deficiency: a case-control study.

BACKGROUND: The gut microbiota plays an important role in childhood growth and metabolic regulation and may interact with the growth hormone (GH)-insulin-like growth factor-1 axis. However, the gut microbiota characteristics in children with GH deficiency (GHD) are poorly understood. PURPOSE: This study aimed to compare the gut microbiota composition and predict the functional profiles of Korean children with GHD and those growing normally. METHODS: In this pilot case-control study, fecal samples were collected from children with biochemically confirmed GHD and age- and sex-matched healthy c

Mallard super-shedders of avian influenza exhibit distinct cloacal microbial abundance profiles.

In many infectious disease systems, a small fraction of hosts accounts for a disproportionate share of transmission, but the biological basis of this heterogeneity remains poorly resolved. In avian influenza, mallards show substantial variation in viral shedding, raising the possibility that only a subset of individuals contributes strongly to environmental contamination and onward spread. To examine whether host microbiome structure is associated with this variation, we experimentally infected wild, captive mallards with low-pathogenic avian influenza virus. We characterized cloacal microbiom

The gut microbiota dysbiosis in ischemic stroke: a systematic review and meta-analysis of microbial diversity and taxonomic alterations.

OBJECTIVE: Clinical and epidemiological evidence suggests a link between gut microbiota and ischemic stroke severity as well as outcomes, yet inconsistent findings and unclear mechanisms underscore the need for a systematic synthesis. METHODS: We systematically reviewed and meta-analyzed observational studies linking gut microbiota to ischemic stroke, following PRISMA guidelines. The search covered PubMed, Embase, and the Cochrane Library, with animal studies and in vitro studies excluded. Two reviewers independently screened the literature, extracted data, and assessed study quality via the

Xu Chunfu's Modified Xianglian Pill Regulates the NOX2/ROS/Mitochondria/NLRP3 Axis to Treat Ulcerative Colitis.

Background/Objectives: Xu Chunfu's Modified Xianglian Pill (XXLP) has been used for centuries in Chinese medicine to treat "diarrhea" and "dysentery," conditions analogous to modern ulcerative colitis (UC). However, the scientific basis for its efficacy and mechanisms remains unclear. Methods: The chemical composition of XXLP was analyzed via UPLC-ESI-MS/MS. A colitis mouse model was established using DSS, and the therapeutic effects were assessed based on body weight, disease activity index (DAI), colon length, and histopathology. Inflammatory cytokines were measured using ELISA. Proteomic an

Laetiporus sulphureus polysaccharides mitigate colitis by reshaping the gut microbiota and regulating immune responses.

BACKGROUND: Inflammatory bowel disease (IBD) involves epithelial barrier disruption, immune dysregulation, and microbial imbalance. The present study investigated the protective mechanisms of Laetiporus sulphureus polysaccharides (LSP) in dextran sulfate sodium (DSS)-induced colitis, focusing on intestinal barrier restoration, immunomodulation, and gut microbiota remodeling. METHODS: LSP was structurally characterized using HPLC, FTIR, and SEM analyses, revealing a heteropolysaccharide primarily composed of glucose (55.16%), galactose (16.55%), and mannose (13.52%). Evaluating Co-Ensiling Strategies to Valorise Duckweed as a Sustainable Feed Ingredient.

Duckweed (Lemnaceae) is a promising alternative feed crop, particularly in regions with nutrient surpluses and protein deficits, as it grows efficiently on nutrient-rich agricultural wastewater and provides protein-rich biomass. However, its high moisture content and rapid post-harvest spoilage pose major storage challenges. This study evaluated (co-)ensiling as a cost-effective preservation strategy for duckweed. Three separate experiments were conducted to assess the ensilability of duckweed alone and in combination with various agricultural co-substrates and additives, including corn silage
