PDCD1 gene polymorphisms linked with altered risk of female-specific hormone-dependent cancers: a systematic review and meta-analysis.

BACKGROUND: The programmed cell death-1 (PDCD1) gene plays a critical role in immune regulation and tumor immune evasion, making it a key target in cancer immunotherapy. Genetic variants in PDCD1, including rs2227981, rs2227982, rs7421861, and rs11568821, may influence susceptibility to female-specific cancers. This systematic review and meta-analysis aimed to evaluate the association of these variants with the risk of breast, cervical, ovarian, and endometrial cancers across ethnic populations. METHODS: A systematic literature search was conducted in PubMed, Scopus, Web of Science, and Googl

Incorporating dietary information to enhance polygenic prediction models with applications to body mass index and type 2 diabetes.

BACKGROUND: Polygenic predictors can enhance screening for metabolism-related traits such as body mass index (BMI) and type 2 diabetes (T2D). However, these predictors explain limited phenotypic variance and face implementation challenges in non-European populations. Dietary patterns are well-established metabolic risk factors that remain under-investigated in quantitative risk stratification models. METHODS: We developed and evaluated risk stratification models combining polygenic predictors and data-driven dietary scores (DDS) for BMI and T2D in 14,346 Native Hawaiians from the Multiethnic

Association between leptin gene polymorphisms and hematological metabolic parameters in obese Saudi adults.

BACKGROUND: Obesity represents a major global public health challenge. However, among the Middle Eastern countries and members of the Gulf Cooperation Council, the Kingdom of Saudi Arabia (K.S.A) has been particularly affected, with prevalence rates exceeding 24.7%. The impact of obesity on hematological parameters has been previously established; nevertheless, the relationship between genetic variants and obesity-related hematological changes in the Saudi population remains unexplored. AIM: This study aimed to investigate the hematological and metabolic profiles of obese and non-obese Saudi

Interplay between CARD9 genetic variants and fungal colonization patterns among patients with diabetes.

BACKGROUND: Diabetes mellitus significantly increases susceptibility to opportunistic fungal colonization; however, the genetic architecture underlying this vulnerability in Middle Eastern populations remains underexplored. Caspase recruitment domain-containing protein 9 (CARD9) is a fundamental adaptor in fungal sensing. While CARD9 deficiency is traditionally viewed as a rare pediatric immunodeficiency, we hypothesize that common CARD9 variants serve as significant predisposing factors for fungal burden in the adult diabetic population. METHODS: A cohort of 107 diabetic participants (49.5%

Boron/sodium co-doping enhances phase transformation in TiO2 nanofibers and amplifies photoelectrochemical nucleic acid oxidation detection.

BACKGROUND: Oxidative damage in genetic molecules disrupts cellular function and poses long-term risks to organismal health, underscoring the need for early, rapid, and sensitive detection methods. Early and precise detection of this oxidative damage in genetic molecules can enable necessary precautions to be taken and initiate the treatment process early, thereby saving the lives of patients. Oxidation detection and analysis using electrochemical methods are frequently employed. The properties of the electrodes used in electrochemical measurements are significant for sensor sensitivity. Clinical-genetic predisposition and medium-term outcomes after bariatric surgery: A 5-year validation of the OBEGEN score.

BACKGROUND: Weight-loss outcomes after bariatric surgery (BS) show interindividual variability that is not fully explained by clinical factors. In the OBEGEN study we previously developed a clinical-genetic score combining three clinical variables with nine single nucleotide polymorphisms to predict weight-loss success at 12-18 months after BS. In the present study we evaluated the usefulness of this clinical-genetic score to predict longitudinal weight-loss trajectories up to five years after BS. METHODS: Multicenter retrospective cohort study including 342 adults who underwent Roux-en-Y gas

Infant respiratory syncytial virus and childhood asthma: a nationwide registry-based, sibling-controlled, and genome-wide association study.

BACKGROUND: Early-life respiratory syncytial virus (RSV) infection is associated with childhood recurrent wheeze or asthma (RW/A), but causality and shared genetic liability remain unclear. METHODS: We combined Finnish nationwide registries and Nordic genetic cohorts. First, in 965,312 Finnish children born between 1998 and 2014, we defined severe RSV as RSV hospitalisation before age 1 year, and recurrent wheezing or asthma (RW/A) as inhaled medication reimbursement between ages 1 and 7 years, and compared medication and eosinophil trajectories by RSV history. Second, we assessed familial co

Genetic and environmental contributions to dental fluorosis in the primary dentition: A census-based twin study.

BACKGROUND: Although environmental exposure is the main determinant of dental fluorosis (DF), the contribution of genetic factors remains unclear. Twin studies offer an effective approach to distinguish genetic from environmental influences on this condition. METHODS: This was a cross-sectional census-based study involving twin preschoolers aged 3 through 5 years in Teresina, Brazil. Children with complete primary dentition were included, and children who did not cooperate were excluded. The children's guardians answered questionnaires regarding socioeconomic-, health-, and zygosity-related a

The very rare association between T-cell acute lymphoblastic leukemia and down syndrome: a case report and review of the literature.

BACKGROUND: Children with Down syndrome (DS) show a higher incidence of acute lymphoblastic leukemia (ALL) compared to general population. They are almost entirely affected by B-cell precursor ALL, linked to the constitutional chromosome 21 trisomy and associated with recurrent mutational patterns. The occurrence of T-cell ALL (T-ALL) is extremely rare in DS patients, with only few reports described. This entity remains poorly characterized either genetically and clinically, and there are no standardized guidelines for the treatment of these rare and fragile patients. Here we describe the case

Polygenic risk and air pollution trends in relation to type 2 diabetes: evidence from the Taiwan Biobank.

BACKGROUND: Both genetic predisposition and air pollution may lead to type 2 diabetes mellitus (T2D). However, the synergistic effects of gene by environment interactions between air pollution exposure and genetic susceptibility to T2D remain underexplored in Asian populations. This study assessed the association between air pollutants, including fine particulate matter (PM2.5), nitrogen dioxide (NO2), sulfur dioxide (SO2), and ozone (O3), and T2D, while considering polygenic risk scores (PRS). METHODS: Data were obtained from 104,554 participants in the Taiwan Biobank. Air pollutant concentr

Genome-wide association analysis of treatment response to onabotulinumtoxinA in Han Chinese patients with chronic migraine: a pilot study.

BACKGROUND/OBJECTIVES: OnabotulinumtoxinA (BoNT-A) use is an established preventive treatment for chronic migraine (CM); however, individual response rates vary. Although genetic factors may contribute to this variability, the underlying genetic determinants remain largely undefined, especially in Asian populations. Therefore, we aimed to identify genetic variants associated with the response to BoNT-A treatment in Han Chinese patients with CM. METHODS: We conducted a genome-wide quantitative trait locus (QTL) analysis using the Taiwan Precision Medicine (TPM) array in Han Chinese patients wit
