Expression of human immunodeficiency virus type 1 in the nervous system of transgenic mice leads to neurological disease.

Patients infected with the human immunodeficiency virus type 1 (HIV-1) frequently develop central and peripheral nervous system complications, some of which may reflect the effect of the virus itself. In order to elucidate the pathogenic mechanisms of HIV in neurological disease in a small animal model, we generated transgenic mice expressing the entire HIV genome under control of the promoter for the human neurofilament NF-L gene. The transgene was predominantly expressed in anterior thalamic and spinal motor neurons. Animals developed a neurological syndrome characterized by hypoactivity and

Inhibitory effect of a new androstenedione derivative, 14 alpha-hydroxy-4-androstene-3,6,17-trione (14 alpha-OHAT) on aromatase activity of human uterine tumors.

The development of human uterine estrogen-dependent tumors is considered to be closely related to estrogen biosynthesis. This study examined whether or not 14 alpha-hydroxy-4-androstene-3,6,17-trione (14 alpha-OHAT), a new 4-androstene-3, 17-dione derivative synthesized microbiologically, inhibits estrogen biosynthetase (aromatase) activities of human uterine tumors (i.e. uterine endometrial cancer, uterine leiomyoma and uterine adenomyosis tissues). 14 alpha-OHAT inhibited aromatase activity in all uterine tumors, dose-dependently (0.1-10 microM). Moreover, 14 alpha-OHAT did not show the bind

Biological characterization of 10-(2-propynyl) estr-4-ene-3, 17-dione (MDL 18,962), an enzyme-activated inhibitor of aromatase.

MDL 18,962 was shown to be a highly specific, potent (Ki = 3-4 nM), enzyme-activated inhibitor of aromatase with minimal intrinsic endocrine properties. The affinity of MDL 18,962 was higher for human and baboon placental aromatase than for rhesus placental or rodent ovarian aromatase. These species differences necessitated the development of a novel model of peripheral aromatase utilizing human enzyme. Human choriocarcinoma trophoblast xenografts in athymic nude mice were used for pharmacologic and pharmacokinetic evaluation of MDL 18,962. The ED50 for inhibition of aromatase activity in thes

Rabies immune globulin (human) in the prevention of rabies.

The development of rabies immune globulin of human origin is discussed. Antirabies serum of equine origin when used in conjunction with rabies vaccine, enhances significantly the chances of survival after a severe bite by a rabid animal. However, those who receive the equine serum may develop serum sickness or even anaphylaxis. To circumvent this risk, while preserving the benefit, many workers have attempted to develop rabies immune globulin of human origin. Success was impeded by the difficulty in obtaining rabies immune plasma with the needed antibody level. Despite this and other obstacles

Human SY5Y neuroblastoma cell interactions with laminin isoforms: neurite outgrowth on laminin-5 is mediated by integrin alpha 3 beta 1.

Laminin (Ln) isoforms may play important roles in neuronal development, particularly axon guidance, but neural receptors mediating interactions with Ln are not entirely understood. In this paper, we have compared the adhesive and process outgrowth activities of a human neuroblastoma cell line SY5Y on various laminin isoforms. Cell adhesion and process outgrowth were examined on murine Ln-1 (Englebreth-Holm-Swarm sarcoma laminin), human placental Ln-1 (human Ln-1[p]), human Ln-2 (merosin), human Ln-5 (kalinin/epiligrin/nicein), and human foreskin keratinocyte extracellular matrix extract (human

Serum and platelet-derived growth factor-induced expression of vascular permeability factor mRNA by human vascular smooth muscle cells in vitro.

1. Endothelial dysfunction and vascular smooth muscle cell (VSMC) proliferation are key events in the pathogenesis of atherosclerosis. Vascular permeability factor (VPF), an endothelial-cell-specific multifunctional cytokine, was recently described, and has the potential to contribute to the development of endothelial dysfunction. The present study determines whether cultured human VSMCs express mRNA for VPF and whether VPF mRNA expression is influenced by human VSMC proliferation. 2. A 204 bp cDNA fragment, specific for all known variants of VPF mRNA, was cloned and used to demonstrate that h

Investigation of the utility of the 1.1B4 cell as a model human beta cell line for study of persistent enteroviral infection.

The generation of a human pancreatic beta cell line which reproduces the responses seen in primary beta cells, but is amenable to propagation in culture, has long been an important goal in diabetes research. This is particularly true for studies focussing on the role of enteroviral infection as a potential cause of beta-cell autoimmunity in type 1 diabetes. In the present work we made use of a clonal beta cell line (1.1B4) available from the European Collection of Authenticated Cell Cultures, which had been generated by the fusion of primary human beta-cells with a pancreatic ductal carcinoma

Metabolic Dysfunction in Alzheimer's Disease: Brain Glucose Hypometabolism as an Early Precursor to Amyloid and Tau Pathology.

Objective: Alzheimer's disease (AD) is traditionally characterized by amyloid-β and tau pathology; however, accumulating evidence indicates that metabolic and inflammatory dysfunctions are early, central contributors to disease development. This narrative review explores how metabolic disturbances influence AD pathophysiology. Methods: A comprehensive literature search was performed on PubMed, Embase, and Scopus. Selected studies were original studies or reviews published in English within the past five years involving human subjects. Case reports, case series, editorials, and non-human studie

Reference-guided pseudotime inference across species and biological contexts.

Cells collected at the same chronological age can vary substantially in biological age due to the heterogeneity in the timing of differentiation, speed of maturation, and degeneration. However, existing pseudotime inference methods either disregard chronological time information, or rely on accurate time-series labels within similar species or biological conditions of interest. As a result, both types of strategies often fail to faithfully order cells from biological contexts without reliable time labels, along the desired axis of interest such as human embryonic development or disease progres

Drug Repurposing as a Broad-Spectrum Strategy Against Coronaviruses: Frontiers in Mechanisms and Clinical Translation.

Coronaviruses (family Coronaviridae) are enveloped, positive-sense, single-stranded RNA viruses with broad host adaptability. They transmit across species among humans, livestock, companion animals, and wildlife, eliciting a disease spectrum ranging from mild respiratory and gastrointestinal symptoms to fatal multi-organ failure, thereby posing significant challenges to global health. In this context, drug repurposing has emerged as a practical strategy for rapidly identifying broad-spectrum antivirals against emerging and re-emerging coronaviruses. Using coronaviruses as a paradigm, this revi

Hydrogels and microfluidics: toward more physiologically relevant organ-on-a-chip models.

Hydrogel-integrated organ-on-a-chip (OoC) models have emerged as a promising platform for recapitulating the complexity of native human tissues. This review highlights the role of hydrogels as tissue-specific extracellular matrix (ECM) mimics, focusing on the physicochemical properties that regulate cellular behaviour and tissue function in three-dimensional (3D) systems. We also explore the contribution of microfluidic systems in further enhancing physiological relevance by integrating multiple microenvironmental cues, including dynamic biochemical gradients, biophysical stimuli, tissue archi

The Recent Research Progress On Cadmium-Induced Kidney Injury.

Kidney diseases are a group of disorders that cause kidney damage, characterized by renal dysfunction, pathological changes in renal tissue, and abnormal components in blood or urine. A plethora of factors have been identified as causes of renal diseases, including cadmium exposure. Cadmium is an environmental pollutant, and excessive intake of cadmium in the human body can result in cadmium poisoning. Accumulating evidence has demonstrated that cadmium exposure is a contributing factor to the development of kidney diseases. The associated symptoms include damage to the renal tubules and glome

Serum exosomes protect heat-stressed mouse 2-cell embryos: Enhanced antioxidant defense and base excision repair of oxidative DNA damage.

High temperatures pose a significant threat to human health; however, there are limited measures to address infertility resulting from elevated temperatures. This study aimed to evaluate the protective effects of exosomes on heat-stressed mouse 2-cell Embryos. The results indicated that exposure to high environmental temperatures caused substantial damage to the developmental potential of embryos, while the inclusion of serum exosomes during early pregnancy demonstrated a beneficial impact. Specifically, high environmental temperatures lead to embryonic retardation and increase oxidative stres

NeMO Analytics: a compendium of transcriptomic data for the exploration of neocortical development.

Here, to enable researchers to more fully harness the collective discovery potential of multiomic data in the public domain, we have assembled gene-level transcriptomic data from ~200 studies of neocortical development and in vitro models. Applying joint matrix decomposition to mouse, macaque and human data, we define transcriptome dynamics that are conserved across neocortical neurogenesis and identify a program that emerges in ventricular progenitors, is later expressed in neurogenic outer, or basal, radial glia of primates, but is limited to gliogenic precursors in the rodent. Growth and nutrition in early life for very preterm infants - Implications for neonatal lung health.

Very preterm birth (< 32 weeks' gestation) disrupts a critical window of lung development, placing infants at substantial risk of bronchopulmonary dysplasia (BPD) and long-term respiratory morbidity. Nutrition is increasingly recognised as a modifiable determinant of neonatal lung health across three distinct but interconnected phases: (1) During the antenatal period, maternal macronutrient and micronutrient status, including vitamin D, omega-3 fatty acids, and folate, shapes placental function, fetal lung growth, and surfactant biosynthesis. Fetal growth restriction, a consequence of placenta

Longevity, Education, and Income: How large is the triangle?

While health affects economic development and wellbeing through a variety of pathways, one commonly suggested channel is a "horizon" mechanism in which increased longevity induces additional education. A recent literature devotes much attention to how much education responds to increasing longevity, while this study asks instead what impact this specific channel has on wellbeing (welfare). I note that death is like a tax on human-capital investments, which suggests using a standard tool of introductory economics: triangles. I estimate the (triangular) gain from reoptimization when education ad

ZAR1 pathogenic variants disrupt maternal mRNA storage and cause oocyte maturation defects in humans.

Zygote arrest 1 (Zar1) was one of the earliest identified maternal-effect genes that function during the maternal-to-zygotic transition (MZT) in mice. However, the role of human ZAR1 remains unclear. In this study, we investigated the association of ZAR1 gene variants with infertility in women, focusing on their impact on oocyte maturation and early embryonic development. Using whole-exome sequencing, we identified homozygous and compound heterozygous ZAR1 variants in two independent families with female patients diagnosed with primary infertility owing to oocyte maturation and early embryonic

mRNA Vaccine Development in the Fight Against Zoonotic Viral Diseases.

Zoonotic diseases are transmitted from animals to humans, and they impose a significant global burden by impacting both animal and human health. It can lead to substantial economic losses and cause millions of human deaths. The emergence and re-emergence of zoonotic diseases are heavily influenced by both anthropogenic and natural drivers such as climate change, rapid urbanization, and widespread travel. Over time, the unprecedented rise of new and re-emerging zoonotic diseases has prompted the need for rapid and effective vaccine development. Following the success of the COVID-19 mRNA vaccine
