Rhinovirus is the main pathogen causing common colds, acute exacerbations of asthma, chronic obstructive pulmonary disease and other respiratory diseases. Due to its numerous serotypes and the lack of common conserved viral targets, the development of traditional direct-acting antiviral drugs (DAAs) faces challenges. Host-targeting antivirals (HTAs), as an emerging strategy, provide a new approach to overcome this difficulty by interfering with the host cell factors or pathways necessary for viral replication. This review systematically presents the latest progress in host-targeted rhinovirus

Leishmaniasis is a complex parasitic disease marked by intricate interactions between Leishmania parasites and host immune responses. Recent evidence has shown the importance of microRNAs (miRNAs), small non-coding RNAs, in the modulation of immunopathogenesis of leishmaniasis. This review synthesizes current understanding of miRNA biogenesis and their dynamic regulation during Leishmania infection. We detailed the mechanisms by which host-derived miRNAs modulate key signaling pathways, cytokine expression, and immune cell functions, thereby influencing disease progression and resolution. Nota

The oocyst is the longest life stage of Plasmodium, the causative agent of malaria, one of the most persistent and devastating infectious diseases of humankind. Following ingestion during blood feeding, parasites reproduce sexually and traverse the mosquito midgut epithelium to differentiate into oocysts on the basal lamina, where they undergo prolonged development, ultimately giving rise to thousands of sporozoites capable of infecting a new human host. Oocyst formation represents a severe population bottleneck, resulting in the lowest parasite numbers observed across the parasite life cycle.

Toxoplasmosis is a widespread parasitic disease affecting roughly one-third of the global population. In immunocompromised individuals or during pregnancy, infection can result in severe complications. Following primary infection, Toxoplasma gondii forms dormant bradyzoite cysts in tissues such as the brain and eyes. These cysts can rupture, particularly in immunocompromised hosts, releasing active parasites and triggering recrudescence. Efforts to experimentally induce and study bradyzoite cyst recrudescence have been hindered by the limited capacity of cell culture adapted strains to form ti

Parasites have traditionally been viewed primarily as pathogenic organisms associated with disease and economic burden. However, growing evidence indicates that parasitofauna-the diverse assemblage of parasitic organisms across ecosystems-plays fundamental roles in ecological regulation, host evolution, and biomedical innovation. Despite increasing recognition of these functions, existing literature largely treats ecological, immunological, and translational aspects of parasitism in isolation, lacking an integrated mechanistic framework that links these domains across biological scales. 3'-nucleotidases/nucleases, distinct class I nucleases of protozoan parasites, play a pivotal role in extracellular purine salvage. As Leishmania are purine auxotrophs and lack de novo synthesis, ectoenzymes facilitating nucleotide and nucleic acid cleavage are indispensable for subsequent uptake. Employing quantitative proteomics, we characterized a class I nuclease p1/s1 cluster in L. major that comprises enzymes exhibiting dual 3'-nucleotidase and endonuclease activity. Expression of these enzymes is induced upon miltefosine or staurosporine treatment and was specifically detected in statio

The mechanisms governing host-parasite interactions in human toxoplasmosis remain insufficiently characterized, as research has relied on murine models that fail to capture human cellular responses. Murine resistance to Toxoplasma gondii depends on the IL-12/IFN-γ axis and immunity-related GTPases (IRGs); however, these differ in humans due to the absence of TLR11/12 and a distinct IRG repertoire. We implemented the EXMOWS (ex vivo model without supplements) to investigate early human host-parasite interactions without the biological artifacts induced by fetal bovine serum (FBS) or cryopreserv

Chagas disease, caused by Trypanosoma cruzi, remains an important neglected infection due to underdiagnosis, global spread resulting from increased migration of people infected with T. cruzi and concomitant organ transplantation and blood transfusion, and limited availability of chemotherapy. The parasite's surface is covered by a dense glycocalyx enriched in GPI-anchored mucins, trans-sialidases, and glycoinositolphospholipids (GIPLs), which collectively shape host-parasite interactions. Here we synthesize structural and biosynthetic features of key glycoconjugates and discuss how they modula

Tick-borne encephalitis virus (TBEV) is a major arthropod-borne flavivirus responsible for severe neurological disease in humans across Europe and Asia. It is maintained in nature through complex interactions within ticks and between tick vectors, vertebrate hosts and environmental factors. This review summarizes current knowledge on TBEV-tick interactions, focusing on virus acquisition, dissemination, vector competence, and long-term persistence within tick vectors. TBEV is acquired by ticks during blood feeding on viremic hosts or through co-feeding transmission under experimental conditions

Environmental risk factors and gene-environment interactions play critical roles in Parkinson's disease (PD) pathogenesis, and approximately two-thirds of the phenotypic variance of sporadic PD appears to be attributed to non-heritable factors. The majority of research on environmental risk factors in PD is focused on adult exposures, largely reliant on recall from occupational or lifestyle factors. However, because PD is a progressive disorder with prolonged preclinical and prodromal phases, exposures that contribute to PD risk are likely to occur long before symptom onset, including during e

Helicobacter pylori (H. pylori) is a globally prevalent bacterium that causes chronic infection and, while often asymptomatic, is a major cause of gastritis, peptic ulcer disease, and a risk factor for gastric adenocarcinoma and mucosa-associated lymphoid tissue (MALT) lymphoma. Infection triggers robust innate and adaptive immune responses in the gastric mucosa, including immune cell recruitment and pro-inflammatory cytokine production; however, the bacterium frequently persists by evading host immunity through mechanisms such as CagA-mediated epithelial damage and modulation of heat shock pr

Enterohemorrhagic Escherichia coli (EHEC) is a major foodborne pathogen that causes hemorrhagic colitis and hemolytic uremic syndrome. Increasing evidence indicates that the gut microbiota plays a central role in modulating EHEC pathogenesis through complex metabolic and signaling networks. Beneficial commensals, including Bifidobacterium, Lactobacillus, and segmented filamentous bacteria, contribute to colonization resistance by competing for nutrients and adhesion sites, producing antimicrobial metabolites, and enhancing epithelial barrier integrity. In contrast, certain species such as Bact

The pathogenic potential of Mycobacterium tuberculosis (Mtb), the etiological agent of tuberculosis, is attributable primarily to its complex and highly specialized repertoire of molecular determinants, comprising both proteins and lipids. These virulence factors operate synergistically to facilitate evasion of host immune defenses, establish persistent infection, and sustain prolonged survival within the host environment, often for months or even years. These factors can be systematically classified according to their functional roles, molecular composition, and spatial localization within or

Mycobacterium tuberculosis (Mtb) reshapes host immunity not only by rewiring signaling pathways but also by perturbing post-transcriptional control. Alternative splicing (AS) provides a switch-like layer that can reconfigure protein function without changing gene abundance, making it a plausible driver of infection-induced immune state transitions. Here, we synthesize emerging evidence that Mtb effectors promote isoform remodeling in macrophages and lymphocytes, with the resulting splicing programs further shaped by host inflammatory context and damage-associated signals. Selected AS events di

BACKGROUND: The pathogenesis of polycystic ovary syndrome (PCOS) is still unknown, so finding the molecular mechanisms of pathogenesis is crucial in PCOS.
METHODS: The GSE34526 dataset from the Gene Expression Omnibus (GEO) database was used to screen biomarkers in this study. KEGG enrichment analysis of GSE34526 was performed using Gene Set Enrichment Analysis (GSEA). The differentially expressed genes(DEGs) were screened and analyzed for lysosome-related genes. Subsequently, further KEGG and GO analyses were performed, and it was found that it was enriched in the ferroptosis pathway, and th

DDX17 (DEAD-box RNA helicase 17) is an essential RNA helicase and regulatory ATPase in host cells, extensively involved in various cellular processes during viral infections, such as RNA splicing, transcriptional regulation, and post-transcriptional modification. DDX17 exhibits dual functionality in viral infections: it enhances the stability, packaging, and replication of viral RNA through interactions with viral ribonucleoprotein complexes, as evidenced in infections caused by influenza viruses and Hantaan virus (HTNV). Conversely, DDX17 can inhibit viral proliferation by disrupting viral RN

Inflammatory bowel disease (IBD), including Crohn's disease and ulcerative colitis, is considered significant global health concerns worldwide. Many studies have demonstrated that environmental and dietary factors influence the gut microbiota, which in turn orchestrates the host immune responses. These interactions are also involved in complex metabolic processes that contribute to the pathogenesis of IBD. Furthermore, recent studies in genomics and metabolomics have unveiled the intricate relationship between microbial influencers and host epigenetics. The dynamics of gut microbiota and its m

Anxiety and depressive disorders constitute a public health concern with a high negative impact on patients' quality of life. These disorders are among the prevalent neuropsychiatric conditions significantly contributing to the global burden. Although the precise mechanisms underlying the development of anxiety- and depressive-like behaviors remain incompletely understood, increasing evidence indicates that these disorders arise from complex and multifactorial processes involving dysfunction across multiple body organs. The gut microbiota (GM) seem to play certain role in developing of these c

SEARCH RESULTS: Reactive oxygen species (ROS) play a dual role in leishmaniasis by contributing to both host defence and parasite survival mechanisms. In the host, ROS promote parasite clearance through induction of apoptosis, activation of pro-inflammatory signalling pathways (e.g., MAPK, JNK), inflammasome assembly, and M1 macrophage polarisation. Conversely, Leishmania species have evolved multiple strategies to neutralize ROS, including the upregulation of host antioxidant enzymes like HO-1, inhibition of ROS-producing pathways, and expression of parasite-derived antioxidants such as SOD,
