Self-Feeding Living Materials Enabled by Cell Responsive Glycogen Nanoparticles as Metabolic Batteries.

Scaling engineered living materials to clinically relevant dimensions is limited by diffusion-dependent depletion of oxygen and nutrients, which rapidly induces metabolic failure. We introduce glycogen as a nutritional nanoparticle that provides cell-mediated, autonomous nutrient release to support long-term survival under extreme metabolic stress. We demonstrate that human mesenchymal stromal cells (hMSCs) survive for weeks in anoxia and serum deprivation when provided extracellular glycogen. Contrary to long-held assumptions, hMSCs secrete glycogen-degrading enzymes, enabling cell-density-co

CeO2-Loaded Shear-Thinning Hydrogel Combined with Dental Pulp Stem Cells Modulates the Oxidative-Inflammatory Microenvironment To Enhance Facial Nerve Regeneration.

Stem cell transplantation holds great promise for facial nerve repair, yet its efficacy is largely constrained by the excessive accumulation of reactive oxygen species (ROS) and the highly inflammatory microenvironment. Thus, developing bioactive scaffolds with robust antioxidant capacity for stem cell transplantation remains a challenge. In this study, an injectable and self-healing composite hydrogel composed of chitosan, graphene oxide, and cerium oxide nanoparticles (CeNPs) was developed to provide structural support for the neural conduit, create a favorable microenvironment for human den

T-cell exhaustion and senescence in cancer: Regulatory circuits and therapeutic state engineering.

In cancer, T-cell state strongly influences antitumor immunity and response to immunotherapy. High-parameter cytometry, single-cell transcriptomics, and multiomic profiling now resolve lineage identity, differentiation history, clonality, metabolic fitness, and tissue adaptation in parallel. These studies place naïve, activated, effector, memory, exhausted, and senescent T cells along continuous, context-dependent trajectories rather than within rigid subset boundaries. Their transitions are shaped by interacting transcriptional, epigenetic, signaling, and metabolic programs that integrate ant

Microglia-targeted Panax notoginseng polysaccharide nanoparticles alleviate Alzheimer's disease via AMPK-mTOR/HIF-1α-mediated immunometabolic reprogramming.

Alzheimer's disease (AD) is increasingly recognized as an immunometabolic disorder characterized by a self-reinforcing cycle of neuroinflammation and glycolytic reprogramming in microglia. Herein, we developed KPBIT@NPs, a microglia-targeted polysaccharide-based nanomedicine. Unlike many conventional nanocarriers that mainly serve as passive delivery vehicles, KPBIT@NPs employ the natural macromolecule Panax notoginseng polysaccharide (PNP) to construct a dual-functional, responsive nanoparticle platform. Through rational engineering via phenylboronic ester grafting, the polysaccharide backbon

Nanomedicine targeting neuroinflammatory pathways in Alzheimer's disease: a new frontier in inflammopharmacology.

Alzheimer's disease (AD) is a multifactorial neurodegenerative illness characterized by progressive cognitive impairment, synaptic compromise, and relentless neuroinflammation. Increasing evidence suggests that neuroinflammatory cascades orchestrated by microglial activation, astrocytic malfunction, cytokine hyperproduction, and inflammasome signalling are at the core of AD pathogenesis. Conventional anti-amyloid and cholinergic treatments are only symptomatic and neglect the inherent neuroimmune dysregulation. Nanomedicine is a revolutionary frontier in inflammopharmacology, which enables the

Mitochondrial dysfunction-driven inflammation and β-Cell apoptosis in type 2 diabetes mellitus: mechanistic insights and therapeutic implications.

Type 2 diabetes mellitus (T2DM) is a multifactorial metabolic syndrome characterized by chronic hyperglycemia, progressive pancreatic β-cell malfunction, and peripheral insulin resistance. There is growing evidence suggesting that mitochondrial dysfunction and apoptosis play a critical role in the development and advancement of T2DM. Mitochondria are essential for cellular energy metabolism and redox homeostasis. It is the metabolic stress and apoptotic cell death in insulin-producing β-cells and insulin-sensitive tissues induced by mitochondrial dysfunction caused by excessive reactive oxygen

Polystyrene microplastics induce auditory neurotoxicity in mammals: Integrated multi-omics profiling reveals oxidative damage and synaptic molecular dysregulation.

Microplastics (MPs) are ubiquitous environmental pollutants, yet their neurotoxic effects on the auditory system remain poorly understood. This study develops an integrated multi-level analytical framework combining auditory neurophysiology, behavioral assessment, tissue biochemistry, transcriptomics, and proteomics to investigate polystyrene (PS)-MPs-induced auditory neurotoxicity in rats. PS-MPs infiltrate the auditory system and significantly impair auditory processing, with central dysfunction emerging earlier and more prominently than peripheral alterations. Multi-omics analyses reveal co

Tissue-Resident Macrophages: Linking Physiology and Inflammation in Infection.

Macrophages are found in every tissue in the body, and their importance goes far beyond clearing pathogens. Developmental studies have shown that many macrophage populations originate during embryogenesis, persist through self-renewal, and adopt transcriptional and functional programs defined by their tissue environment. These tissue-resident macrophages (TRMs) are embedded within organ physiology, where they regulate, for instance, cardiac conduction, sculpt synaptic circuits, recycle iron, maintain epithelial barriers, and coordinate repair. During infection, TRMs connect to neural, metaboli

Bacterial Chondronecrosis with Osteomyelitis (BCO) in Broiler Chickens: Bacterial Causes, Pathophysiology and Control, with a Focus on the Potential of Electron Beam-Inactivated Vaccines.

Bacterial chondronecrosis with osteomyelitis (BCO) is a major cause of lameness in modern broiler chickens. It remains a persistent challenge in the broiler industry, resulting in substantial economic losses and serious animal health concerns. The pathogenesis of BCO involves rapid muscle growth, resulting in disproportionate body weight relative to skeletal maturity, thereby increasing mechanical stress and leading to microfractures and osteochondrotic clefts in the proximal growth plates of the femora and tibiae. Infection progresses via hematogenous dissemination and colonization of the leg

The Oral-Brain Axis: Mechanistic Insights Linking Periodontitis With Alzheimer's and Parkinson's Diseases.

Neurodegenerative diseases, including Alzheimer's disease (AD) and Parkinson's disease (PD), are major causes of disability and mortality worldwide. Emerging evidence suggests that chronic peripheral inflammation and microbial dysbiosis may contribute to neurodegenerative processes. The oral-brain axis has recently gained attention as a biological framework linking oral microbial communities, systemic inflammatory responses, immune regulation, and central nervous system function. Within this context, periodontitis, a prevalent chronic inflammatory disease driven by oral dysbiosis, has been pro

Malaria driven mechanisms shaping cancer risk and aggressiveness in African populations.

Malaria and cancer represent intersecting public health challenges in sub-Saharan Africa, where malaria remains endemic and cancer incidence is rapidly increasing. Emerging evidence indicates that chronic or recurrent malaria infection may influence carcinogenesis and tumour aggressiveness through complex biological mechanisms. This narrative review critically synthesizes data from PubMed, Scopus, and Web of Science to elucidate the mechanistic intersections between malaria and cancer risk, progression, and therapeutic response. The review highlights five principal axes linking malaria to onco

N-sulfonylated indole hydrazone hybrid targeting Staphylococcus aureus with compelling antibiofilm and membrane-disrupting properties.

Staphylococcus aureus is a highly adaptable opportunistic pathogen and a leading cause of infectious disease-related morbidity and mortality globally. This pathogen triggers a wide spectrum of infections, from mild skin infections to life-threatening conditions like sepsis and pneumonia. Thus, it is implicated in escalating the global antimicrobial resistance (AMR) crisis, and there is an urgent need for novel antistaphylococcal scaffolds. In response to this challenge, we previously reported a metal-free electrochemical strategy for regioselective synthesis of N-sulfonylated indole-based hydr

Butyrate improves dextran sulfate sodium-induced enterotoxicity in broilers.

BACKGROUND: Intestinal inflammation has emerged as a significant global concern in poultry farming, particularly due to its strong association with macrophage imbalance. Research has identified macrophages as critical target cells for butyrate, which collaboratively maintains intestinal immune homeostasis. This study delves into the regulatory effects of butyrate on macrophages to ameliorate enteritis in broiler chickens. RESULTS: Our findings indicate that broilers treated with dextran sulfate sodium (DSS) exhibit reduced body weight and compromised intestinal mucosal integrity, characterize

Advances in Probiotic and Prebiotic Applications for Sustainable Aquaculture: Harnessing Their Roles in China's Marine Pasture Restoration and Health.

Aquaculture is the fastest-growing food-producing sector globally, yet intensive farming in China has led to severe ecological degradation of coastal marine pastures, including habitat loss, disease outbreaks, and dysbiosis of cultured species. Probiotics (beneficial live microorganisms) and prebiotics (non-digestible substrates that promote beneficial microbiota) offer eco-friendly alternatives to antibiotics and chemicals, aligning with China's "Blue Granary" and marine ecological restoration initiatives. Recent advances demonstrate that multi-strain probiotics (including Lactobacillus, Baci

Engineering Chlorella-based consortia for sustainable food wastewater treatment: nutrient recovery, pollutant degradation and advanced harvesting.

Microalgae-based bioremediation of wastewater enables simultaneous nutrient removal and production of value-added biomass. This study systematically evaluated three geographically distinct Chlorella sp. strains for semi-continuous treatment of food wastewater, addressing critical challenges that hinder practical application. All strains demonstrated substantial nutrient removal (NO3--N > 73%, TP > 78%), with film-forming Chlorella sp. CQ uniquely sustaining complete nitrate elimination. This strain simultaneously produced biomass in which essential amino acids accounted for 40% of total amino

The oxidative stress-inflammation self-perpetuating cycle in multiple sclerosis: from mechanisms to emerging antioxidant strategies.

Multiple sclerosis (MS) is a complex multifactorial disease of the central nervous system (CNS) whose pathogenesis has not yet been fully elucidated. Current MS treatments primarily consist of disease-modifying therapies with anti-inflammatory and immunomodulatory properties, which effectively reduce relapse rates and disease activity. However, these therapies often exhibit limited long-term efficacy, may cause severe adverse effects, and remain largely insufficient in preventing the progressive accumulation of irreversible disability driven by axonal and neuronal damage. Although oxidative st

Inflammation and immune biomarkers: new frontiers in understanding and managing diabetes complications.

Chronic inflammation and immune dysregulation are central to the development and progression of diabetes complications, redefining existing paradigms of disease pathogenesis and management. Key pro-inflammatory cytokines, including tumour necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and interleukin-1 beta (IL-1β), contribute to tissue injury in the vasculature, kidneys, retina, and peripheral nerves by activating systemic and local inflammatory cascades. Emerging evidence highlights the utility of haematological inflammatory biomarkers such as the neutrophil-to-lymphocyte ratio (NLR),

Photobiomodulation repairs the blood-spinal cord barrier in a mouse model of spinal cord injury.

JOURNAL/nrgr/04.03/01300535-202606000-00062/figure1/v/2026-02-11T151048Z/r/image-tiff The blood-spinal cord barrier is crucial for preserving homeostasis of the central nervous system. After spinal cord injury, autophagic flux within endothelial cells is disrupted, compromising the integrity of the blood-spinal cord barrier. This disruption facilitates extensive infiltration of inflammatory cells, resulting in exacerbated neuroinflammatory responses, neuronal death, and impaired neuronal regeneration. Previous research has demonstrated that photobiomodulation promotes the regeneration of damag
