HSPA9 contributes to tumor progression and ferroptosis resistance by enhancing USP14-driven SLC7A11 deubiquitination in multiple myeloma.

Ferroptosis, a regulated cell death triggered by overload-dependent lipid peroxidation, is implicated in multiple human cancers. The mechanisms underlying ferroptosis in multiple myeloma (MM) remain enigmatic. Here, we confirmed that HSPA9 is overexpressed in MM samples and correlates with unfavorable outcomes. Functionally, HSPA9 enhances MM cell viability, ferroptosis resistance, and tumorigenicity, suggesting its oncogenic role. Proteomics screening identified SLC7A11, a key ferroptosis suppressor, as a HSPA9 interactor. Mechanistically, HSPA9 serves as a bridge to strengthen the interactio

Dual functioning Ru(II)/Ir(III) complexes for ferroptosis and apoptosis in triple-negative breast cancer: a proof of concept by glutathione depletion.

Although triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer, there is a lack of effective targeted therapies because of the negative expression of the targetable bioreceptors. Fighting TNBC requires innovative solutions beyond conventional treatments. Herein, we introduce novel imidazo-phenanthroline-based Ru(II)/Ir(III) complexes that exhibit a dual function on cancer cells by triggering both apoptosis and ferroptosis. By generating a surge of reactive oxygen species (ROS), depleting crucial antioxidants such as GSH (glutathione) and NADPH (nicotinamide adeni

Design, Synthesis, and Antibacterial Activity of Stilbene Derivatives against Phytopathogenic Bacteria.

The increasing resistance of phytopathogenic bacteria to chemical pesticides has underscored the urgent need for novel bactericidal agents. In this study, we designed, synthesized, and evaluated a series of stilbene derivatives for their antibacterial activity against six strains of plant-pathogenic bacteria. Among them, compound ST-A-8 showed the optimal activity against Xanthomonas oryzae pv oryzae (Xoo) and Erwinia aroideae, with both MIC values of 1.56 μg/mL, which was more potent than the commercialized antibacterial agent thiodiazole copper. Furthermore, the mechanism of ST-A-8 on Xoo wa

Senescent macrophages induce ferroptosis in skeletal muscle and accelerate osteoarthritis-related muscle atrophy.

Muscle atrophy around joints is a common issue for people with osteoarthritis (OA), but its causes are poorly understood. Here we demonstrate that chronic inflammation in quadriceps muscle coincides with OA in mice, characterized by an increase in macrophages, activation of inflammatory pathways and tissue vascularization. We show that, during OA progression, macrophages progressively exhibit increasing phenotypes of senescence and promote muscle atrophy through paracrine induction of ferroptosis. Mechanistically, iron overload-induced mitochondrial damage results in reduced asparagine metabol

Ferroptosis is a novel pathogenic mechanism of FDXR-related disease via disruption of the NRF2 pathway.

Loss-of-function variants in the ferredoxin reductase (FDXR) gene result in a primary mitochondrial disease in humans, involving abnormal mitochondrial iron accumulation. However, the molecular mechanism is not fully understood. To better understand the underlying pathology of FDXR-related disease, we generated a mouse model corresponding to the hotspot variant found in humans. We demonstrated increased lipid peroxidation in the inner mitochondrial and plasma membranes, resulting in susceptibility to ferroptosis. Closer examination revealed that disruption of the NRF2 pathway and its target ge

Ferroptosis regulation through Nrf2 and implications for neurodegenerative diseases.

This article provides an overview of the background knowledge of ferroptosis in the nervous system, as well as the key role of nuclear factor E2-related factor 2 (Nrf2) in regulating ferroptosis. The article takes Alzheimer's disease (AD), Parkinson's disease (PD), Huntington's disease (HD), and amyotrophic lateral sclerosis (ALS) as the starting point to explore the close association between Nrf2 and ferroptosis, which is of clear and significant importance for understanding the mechanism of neurodegenerative diseases (NDs) based on oxidative stress (OS). Accumulating evidence links ferroptos

Ferroptosis mechanisms and regulations in cardiovascular diseases in the past, present, and future.

Cardiovascular diseases (CVDs) are the main diseases that endanger human health, and their risk factors contribute to high morbidity and a high rate of hospitalization. Cell death is the most important pathophysiology in CVDs. As one of the cell death mechanisms, ferroptosis is a new form of regulated cell death (RCD) that broadly participates in CVDs (such as myocardial infarction, heart transplantation, atherosclerosis, heart failure, ischaemia/reperfusion (I/R) injury, atrial fibrillation, cardiomyopathy (radiation-induced cardiomyopathy, diabetes cardiomyopathy, sepsis-induced cardiac inju

Recent progress of ferroptosis in cancers and drug discovery.

Ferroptosis is a nonapoptotic form of cell death characterized by iron dependence and lipid peroxidation. Ferroptosis is involved in a range of pathological processes, such as cancer. Many studies have confirmed that ferroptosis plays an essential role in inhibiting cancer cell proliferation. In addition, a series of small-molecule compounds have been developed, including erastin, RSL3, and FIN56, which can be used as ferroptosis inducers. The combination of ferroptosis inducers with anticancer drugs can produce a significant synergistic effect in cancer treatment, and patients treated with th

Potential therapies for HCC involving targeting the ferroptosis pathway.

Liver cancer ranks as the third leading cause of cancer-related mortality worldwide, predominantly in the form of hepatocellular carcinoma (HCC). Conventional detection and treatment approaches have proven inadequate for addressing the elevated incidence and mortality rates associated with HCC. However, a significant body of research suggests that combating HCC through the induction of ferroptosis is possible. Ferroptosis is a regulated cell death process characterized by elevated levels of reactive oxygen species (ROS) and lipid peroxide accumulation, both of which are dependent on iron level

Fluorescent probes for formaldehyde based on formaldehyde-promoted C-N cleavage of azanyl carbamates.

Formaldehyde (FA) is an endogenous one-carbon metabolite and an environmental pollutant and carcinogen. Elevated FA levels are associated with many diseases. Methods for the convenient and in situ detection of FA levels are of great significance for understanding FA's biofunctions and signalling pathways. Herein, the NAP-FAP2 series of fluorescent probes for FA detection were developed based on FA-promoted C-N cleavage of 3-nitrophenylazanyl N-arylcarbamate via FA-induced intramolecularity, where the aryl group is the fluorophore 1,8-naphthalimide-4-yl. The 3-nitrophenylazanyl containing react

BACH1 inhibits senescence, obesity, and short lifespan by ferroptotic FGF21 secretion.

Ferroptosis is a type of regulated cell death characterized by iron-dependent lipid peroxidation. A model cell system is constructed to induce ferroptosis by re-expressing the transcription factor BACH1, a potent ferroptosis inducer, in immortalized mouse embryonic fibroblasts (iMEFs). The transfer of the culture supernatant from ferroptotic iMEFs activates the proliferation of hepatoma cells and other fibroblasts and suppresses cellular senescence-like features. The BACH1-dependent secretion of the longevity factor FGF21 is increased in ferroptotic iMEFs. The anti-senescent effects of the cul

Mannose metabolism inhibition sensitizes acute myeloid leukaemia cells to therapy by driving ferroptotic cell death.

Resistance to standard and novel therapies remains the main obstacle to cure in acute myeloid leukaemia (AML) and is often driven by metabolic adaptations which are therapeutically actionable. Here we identify inhibition of mannose-6-phosphate isomerase (MPI), the first enzyme in the mannose metabolism pathway, as a sensitizer to both cytarabine and FLT3 inhibitors across multiple AML models. Mechanistically, we identify a connection between mannose metabolism and fatty acid metabolism, that is mediated via preferential activation of the ATF6 arm of the unfolded protein response (UPR). This in

On the mechanisms of lysis triggered by perturbations of bacterial cell wall biosynthesis.

Inhibition of bacterial cell wall synthesis by antibiotics such as β-lactams is thought to cause explosive lysis through loss of cell wall integrity. However, recent studies on a wide range of bacteria have suggested that these antibiotics also perturb central carbon metabolism, contributing to death via oxidative damage. Here, we genetically dissect this connection in Bacillus subtilis perturbed for cell wall synthesis, and identify key enzymatic steps in upstream and downstream pathways that stimulate the generation of reactive oxygen species through cellular respiration. Our results also re
